Highly thermal conductivity materials with excellent electromagnetic interference shielding and Joule heating performances are ideal for thermal management in the next generation of communication industry,artificial i...Highly thermal conductivity materials with excellent electromagnetic interference shielding and Joule heating performances are ideal for thermal management in the next generation of communication industry,artificial intelligence and wearable electronics.In this work,silver nanowires(AgNWs)are prepared using silver nitrate as the silver source and ethylene glycol as the solvent and reducing agent,and boron nitride(BN)is performed to prepare BN nanosheets(BNNS)with the help of isopropyl alcohol and ultrasonication-assisted peeling method,which are compounded with aramid nanofibers(ANF)prepared by chemical dissociation,respectively,and the(BNNS/ANF)-(AgNWs/ANF)thermal conductivity and electromagnetic interference shielding composite films with Janus structures are prepared by the"vacuum-assisted filtration and hot-pressing"method.Janus(BNNS/ANF)-(AgNWs/ANF)composite films exhibit"one side insulating,one side conducting"performance,the surface resistivity of the BNNS/ANF surface is 4.7×10^(13) Ω,while the conductivity of the AgNWs/ANF surface is 5,275 S/cm.And Janus(BNNS/ANF)-(AgNWs/ANF)composite film with thickness of 95 pm has a high in-plane thermal conductivity coefficient of 8.12 W/(m·K)and superior electromagnetic interference shielding effectiveness of 70 dB.The obtained composite film also has excellent tensile strength of 122.9 MPa and tensile modulus and 2.7 GPa.It also has good temperature-voltage response characteristics(high Joule heating temperature at low supply voltage(5 V,215.0℃),fast response time(10 s)),excellent electrical stability and reliability(stable and constant real-time relative resistance under up to 300 cycles and 1,500 s of tensile-bending fatigue work tests).展开更多
Flexible multifunctional polymer-based electromagnetic interference(EMI)shielding composite films have important application values in the fields of 5G communication technology,wearable electronic devices and artifici...Flexible multifunctional polymer-based electromagnetic interference(EMI)shielding composite films have important application values in the fields of 5G communication technology,wearable electronic devices and artificial intelligence.In this work,Fe_(3)O_(4)/polyamic acid(PAA)nanofiber films are prepared by in-situ polymerization and electrospinning technology,and Ti_(3)C_(2)T_(x)nanosheets are deposited on the surface of the Fe_(3)O_(4)/PAA nanofiber films via vacuum-assisted filtration.Then,Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/polyimide(PI))composite films are obtained by thermal imidization.The two sides of the Janus films exhibit completely different properties.The Fe_(3)O_(4)/PI side has excellent hydrophobicity and insulation property,and the Ti_(3)C_(2)T_(x)side has hydrophilicity and terrific conductivity.When the mass fraction of Ti_(3)C_(2)T_(x)is 80 wt.%,the Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/PI)composite film has excellent EMI shielding performances and mechanical properties,with EMI shielding effectiveness,tensile strength and Young’s modulus reaching 66 dB,114.5 MPa and 5.8 GPa,respectively.At the same time,electromagnetic waves show different absorption shielding effectiveness(SEA)when incident from two sides of the Janus films.When the electromagnetic waves are incident from the Fe_(3)O_(4)/PI side,the SEA of the Janus film is 58 dB,much higher than that when the electromagnetic waves are incident from the Ti_(3)C_(2)T_(x)side(39 dB).In addition,the Ti_(3)C_(2)T_(x)side of the Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/PI)composite films also has excellent electrothermal and photothermal conversion performances.When the applied voltage is 4 V,the stable surface temperature reaches 108°C;when it is irradiated by simulated sunlight with power density of 200 mW/cm2,the stable surface temperature reaches 95℃.展开更多

Pancreatitis is represented as acute pancrea...Pancreatitis is inflammation of pancreas and caused by a number of factors including pancreatic duct obstruction, alcoholism, and mutation in the cationic trypsinogen gene. Pancreatitis is represented as acute pancreatitis with acute inflammatory responses and; chronic pan-creatitis characterized by marked stroma formation with a high number of infiltrating granulocytes(such as neutrophils, eosinophils), monocytes, macrophages and pancreatic stellate cells(PSCs). These inflammatory cells are known to play a central role in initiating and promoting inflammation including pancreatic fibrosis, i.e., a major risk factor for pancreatic cancer. A number of inflammatory cytokines are known to involve in pro-moting pancreatic pathogenesis that lead pancreatic fibrosis. Pancreatic fibrosis is a dynamic phenomenon that requires an intricate network of several autocrine and paracrine signaling pathways. In this review, we have provided the details of various cytokines and molecular mechanistic pathways(i.e., Transforming growth factor-β/SMAD, mitogen--activated protein kinases, Rho kinase, Janus kinase/signal transducers and activators, and phosphatidylinositol 3 kinase) that have a critical role in the activation of PSCs to promote chronic pancreatitis and trigger the phenomenon of pancreatic fibrogenesis. In this review of literature, we discuss the involvement of several pro-inflammatory and anti-inflammatory cytokines, such as in interleukin(IL)-1, IL-1β, IL-6, IL--8 IL-10, IL-18, IL--33 and tumor necrosis factor-α, in the pathogenesis of disease. Our review also highlights the significance of several experimental animal models that have an important role in dissecting the mechanistic pathways operating in the development of chronic pancreatitis, including pancreatic fibrosis. Additionally, we provided several intermediary molecules that are involved in major signaling pathways that might provide target molecules for future therapeutic treatment strategies for pancreatic pathogenesis.展开更多
OBJECTIVE: To observe effect of Liuweibuqi Capsule, a Traditional Chinese Medicine (TCM), on the janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway and matrix metalloproteinases ...OBJECTIVE: To observe effect of Liuweibuqi Capsule, a Traditional Chinese Medicine (TCM), on the janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway and matrix metalloproteinases (MMPs) in a chronic obstructive pulmonary disease (COPD) rat model with lung deficiency in terms of TCM's pattern differentiation. METHODS: Rats were randomly divided into a normal group, model group, Liuweibuqi group, Jinshuibao group, and spleen aminopeptidase group (n= 10). Aside from the normal group, all rats were ex-posed to smoke plus lipopolysaccharide tracheal instillation to establish the COPD model with lung deficiency. Models were established after 28 days and then the normal and model groups were given normal saline (0.09 g/kg), Liuweibuqi group was given Liuweibuqi capsule (0.35 g/kg), Jinshuibao group was given Jinshuibao capsules (0.495 g/kg), and the spleen group was given spleen aminopeptidase (0.33 mg/kg), once a day for 30 days. Changes in symptoms, signs, and lung histology were observed. Lung function was measured with a spirometer. Serum cytokines were detected using enzyme-linked immunosorbent assay, and changes in the JAK/STAT pathway, MMP-9, and MMPs inhibitor 1 (TIMP1) were detected by immunohistochemistry, RT-PCR, and western blotting, respectively.RESULTS: Compared with the normal group, lung tissue was damaged, and lung function was reduced in the model control group. Additionally, the levels of interleukin (IL)-1β, y interferon (IFN-γ), and IL-6 were higher, while IL-4 and IL-10 were lower in the model control group than those in the normal group. The expressions of JAK1, STAT3, ρ-STAT3, and MMP-9 mRNA and protein in lung tissue were higher, and TIMP1 mRNA and protein was lower in the model group compared with the normal group. After treatment, compared with the model group, the expression of inflammatory cytokines was lower in each treatment group, and expressions of JAK/ STAT pathway, MMPs were lower. Compared wi展开更多
In 2018,the pan-Janus kinase(JAK)inhibitor tofacitinib was launched for the treatment of ulcerative colitis(UC).Although tofacitinib has proven efficacious in patients with active UC,it failed in patients with Crohn’...In 2018,the pan-Janus kinase(JAK)inhibitor tofacitinib was launched for the treatment of ulcerative colitis(UC).Although tofacitinib has proven efficacious in patients with active UC,it failed in patients with Crohn’s disease(CD).This finding strongly hints at a different contribution of JAK signaling in both entities.Here,we review the current knowledge on the interplay between the JAK/signal transducer and activator of transcription(STAT)pathway and inflammatory bowel diseases(IBD).In particular,we provide a detailed overview of the differences and similarities of JAK/STAT-signaling in UC and CD,highlight the impact of the JAK/STAT pathway in experimental colitis models and summarize the published evidence on JAK/STAT-signaling in immune cells of IBD as well as the genetic association between the JAK/STAT pathway and IBD.Finally,we describe novel treatment strategies targeting JAK/STAT inhibition in UC and CD and comment on the limitations and challenges of the new drug class.展开更多

Methods: In this study, 205 cases of PMOP wer...Objectives: To investigate the mechanism of Liuwei Dihuang Pill (六味地黄丸, LDP) in treating postmenopausal osteoporosis (PMOP) with Shen (Kidney) yin deficiency. Methods: In this study, 205 cases of PMOP were divided into the PMOP Shen-yin deficiency group (Group A), PMOP Shen-yang deficiency group (Group B), PMOP without Shen deficiency group (Group C), and control group (Group N). Real-time polymerase chain reaction (RT-PCR) and Western blot techniques were used to observe the effects of LDP treatment on the cardiotrophin-like cytokine factor 1 (CLCF1), ankyrin repeat and SOCS box containing 1 (ASB1), and proldneticin 2 (PROK2) genes and the Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway. Results: The mRNA (P〈0.05) and protein (P〈0.01) expression levels of the CLCF1 gone in Group A were significantly lower than the corresponding levels in Group N. After LDP treatment for 3 months, the mRNA expression levels of the CLCF1 gone were obviously up-regulated (P〈0.01). After 6-month treatment, the expression levels of CLCF1 mRNA and protein were significantly up-regulated (both P〈0.01), and the average bone density of the top femur had significantly increased (P〈0.05). In vitro, CLCF1 overexpression resulted in a significant increase in the total protein and phosphorylated protein levels of JAK2 and STAT3. Conclusions: The CLCF1 gone is an important gone associated with PMOP Shen-yin deficiency and the therapeutic effects of LDP may be mediated by up-regulation of CLCF1 gone expression and activation of the JAK/STAT signaling pathway.展开更多

Some studies have demonstrated that the autoantibodies are involved in the immun...Background Autoantibodies against angiotensin AT1 receptor have been discovered in patients with preeclampsia or malignant hypertension. Some studies have demonstrated that the autoantibodies are involved in the immunopathogenesis of hypertension and have an agonist effect similar to angiotensin II. Methods Autoantibodies against AT1 receptor were purified from sera of patients with primary hypertension by affinity chromatography. Proliferation of cultured rat vascular smooth muscle cells was detected by bromodeoxyuridine incorporation and activation of signalling molecules detected by Western blotting and electrophoretic mobility shift assay. Results The AT1-RAb caused a significant proliferation similar to the Ang II during first 24 hours. The levels of nuclear factor-KB (NF-KB), phosphorylated JAK2., phosphorylated STAT1 (pSTAT1) and phosphorylated STAT3 (pSTAT3) molecules were increased in response to the autoantibodies. In contrast, the activations of NF-KB and JAK-STAT were blocked by Iosartan, pyrrolidinedithiocarbamate (a specific inhibitor of NF-KB) and AG490 (a specific inhibitor of the JAK2. tyrosine kinase). The expressions of NF-KB, pSTAT1 and pSTAT3 reached peak levels at different times. Moreover, the relative densities of electrophoretic bands showed that activation of pSTAT3 was more significant than STAT1 induced by AT1 -RAb. Conclusions These results suggest that the autoantibodies against AT1 receptor have an agonist effect similar to Ang II in proliferation of VSMCs and the NF-KB and JAK-STAT proteins play essential roles. The effect is different from Angll in that STAT3 is the main downstream activating molecule in JAK-STAT signalling pathway.展开更多

In this paper, we expl...Macrophages play an important role in inflammation, and excessive and chronic activation of macrophages leads to systemic inflammatory diseases, such as atherosclerosis and rheumatoid arthritis. In this paper, we explored the anti-inflammatory effect of broussonin E, a novel phenolic compound isolated from the barks of Broussonetia kanzinoki, and its underlying molecular mechanisms. We discovered that Broussonin E could suppress the LPS-induced pro-inflammatory production in RAW264.7 cells, involving TNF-α, IL-1β, IL-6, COX-2 and iNOS. And broussonin E enhanced the expressions of anti-inflammatory mediators such as IL-10, CD206 and arginase-1(Arg-1) in LPS-stimulated RAW264.7 cells. Further, we demonstrated that broussonin E inhibited the LPS-stimulated phosphorylation of ERK and p38 MAPK. Moreover, we found that broussonin E could activate janus kinase(JAK) 2, signal transducer and activator of transcription(STAT) 3. Downregulated pro-inflammatory cytokines and upregulated anti-inflammatory factors by broussonin E were abolished by using the inhibitor of JAK2-STAT3 pathway, WP1066. Taken together, our results showed that broussonin E could suppress inflammation by modulating macrophages activation state via inhibiting the ERK and p38 MAPK and enhancing JAK2-STAT3 signaling pathway, and can be further developed as a promising drug for the treatment of inflammation-related diseases such as atherosclerosis.展开更多
OBJECTIVE:Electroacupuncture effect on neurological behavior and the expression of tyrosine kinase Janus kinase 2(JAK 2) of ischemic cortex in rats with the focal cerebral ischemia were investigated in this study.METH...OBJECTIVE:Electroacupuncture effect on neurological behavior and the expression of tyrosine kinase Janus kinase 2(JAK 2) of ischemic cortex in rats with the focal cerebral ischemia were investigated in this study.METHODS:The model of focal cerebral ischemia was established by the heat-coagulation induced the occlusion of the middle cerebral artery.The electro-acupuncture was applied on Baihui(GV 20) and Dazhui(GV 14),and AG490 was applied by intracerebroventricular infusion.The expressions of JAK2 mRNA and phospharylatedJAK2(p-JAK2) in the ischemic cortex were observed by in situ hybridization and western blotting.RESULTS:The expressions of JAK2 mRNA and p-JAK2 were rarely found in sham surgery group.In model group,the expression of JAK2 mRNA and JAK2 phosphorylation had increased.After 1 day of cerebral ischemia,the expression had reached its peak.After cerebral ischemia,the expressions of JAK2 mRNA and p-JAK2 were consistent with the neurological deficit score.Electroacupuncture treatment and AG490 intervention were able to improve the neurological deficit score after cerebral ischemia,and down-regulate the expressions of JAK2 mRNAand JAK2 phosphorylation.CONCLUSION:After cerebral ischemia,the excessive expressions of JAK2 and the JAK2 phosphorylation would be one of mechanisms by which the brain injury got worse.The therapy of electro-acupuncture could reduce the expression of JAK2,and inhibit JAK2 phosphorylated activation,so as to block the abnormal activation of signal transduction pathway which was induced by JAK2.展开更多

METHODS: PSCs were isolated from rat pancreas tissue, and used in their culture-activated, myofibroblast-like phenotype. STAT-specific binding activity was assessed by electrophoretic mobility shift assay. Activation of Src, JAK2, STAT1, STAT3, and ERK was determined by Western blotting using anti-phosphospecific antibodies. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine. RESULTS: PDGF-BB induced STAT-specific binding activity, and activation of Src, JAK2, STAT1, STAB, and ERK. Ethanol and acetaldehyde at clinically relevant concentrations decreased basal activation of JAK2 and STAT3. PDGF-induced activation of STAT1 and STAT3 was inhibited by a Src inhibitor PP1 and a JAK2 inhibitor AG490, whereas PDGF-induced activation of ERK was inhibited by PP1, and not by AG490. PDGF-induced proliferation was inhibited by PP1 and AG490 as well as by STAT3 antisense oligonucleotide. CONCLUSION: PDGF-BB activated JAK2-STAT pathway via Src-dependent mechanism. Activation of 3AK2-STAT3 pathway, in addition to ERK, may play a role in PDGF-induced proliferation of PSCs.展开更多

Prolactin and interleukin-6 are described in detail as the best studied examples. Although improved nerve-sparing techniques have reduced the incidence of nerve injury, and the administration of phosphodiesterase type 5 inhibitors has revolutionized the treatment of erectile dysfunction, this problem remains a considerable challenge. In recent years, scientists have focused on brain-derived neurotrophic factor and vascular endothelial growth factor in the treatment of cavernous nerve injury in rat models. Results showed that both compounds were capable of enhancing the regeneration of the cavernous nerve and that activation of the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway played a major role in the process.展开更多

The aim of this study was to evaluate the effect of cerebrospinal fluid(CSF) containing QNYZD(CSF-QNYZD) on the differentiation of cultured NSCs and the involvement of the JAK2/STAT3 pathway.METHODS: The protein expression levels of glial fibrillary acidic protein(GFAP), tubulin, drosophila mothers against decapentaplegic protein(SMAD-1), STAT3, and phosphorylated-STAT3 were detected by western immunoblot analysis in the groups: control, CSF, JAK/STAT inhibitor(AG490),CSF-QNYZD, and CSF-XDZ(CSF-Xidezhen). The differentiation of NSCs was determined by immunofluorescence staining. The proliferation of NSCs was measured using the Cell Counting Kit-8 proliferation assay.RESULTS: Compared with the control group,CSF-QNYZD and AG490 significantly increased the number and expression of tubulin-positive cells, reduced the number and expression of GFAP-positive cells, and down-regulated the expression of p-STAT3. However, CSF-QNYZD also decreased the expression of SMAD-1 and STAT3.CONCLUSION: Enhanced neuronal differentiation may be associated with the down-regulation of glial differentiation instead of promoting proliferationin treated NSCs. Furthermore, QNYZD may play a direct role in suppressing the formation of GFAP-positive cells and enhancing neuronal differentiation by inhibiting JAK2/STAT3 activation. Overall, these results provide insights into the possible mechanism underlying QNYZD-mediated neurogenesis.展开更多

The Janus microcapsule motors were fabricated by template-assisted polyelectrolyte layer-by-layer assembly, followed by spraying of a gold layer on one side. The NIR-powered Janus motors achieved high propulsion with a maximum speed of 42μm.s-1 in water. The propulsion mechanism of the Janus motor was attributed to the self-thermophoresis effect: The asymmetric distribution of the gold layer generated a local thermal gradient, which in turn generated thermophoretic force to propel the Janus motor. Such NIR-propelled Janus capsule motors can move efficiently in cell culture medium and have no obvious effects on the cell at the power of the NIR laser, indicating considerable promise for future biomedical applications.展开更多

In order to reveal the roles of different cross-links with hierarchical bond en...Multi-bond network(MBN) which contains a single network with hierarchical cross-links is a suggested way to fabricate robust hydrogels. In order to reveal the roles of different cross-links with hierarchical bond energy in the MBN, here we fabricate poly(acrylic acid) physical hydrogels with dual bond network composed of ionic cross-links between carboxylFe3+ interactions and hydrogen bonds, and compare these dually cross-linked hydrogels with singly and ternarily cross-linked hydrogels. Simple models are employed to predict the tensile property, and the results confirm that the multi-bond network with hierarchical distribution in the bond energy of cross-links endows hydrogel with effective energy-dissipating mechanism. Moreover, the dually cross-linked MBN gels exhibit excellent mechanical properties(tensile strength up to 500 k Pa, elongation at break ~ 2400%) and complete self-healing after being kept at 50 °C for 48 h. The factors on promoting self-healing are deeply explored and the dynamic multi-bonds are regarded to trigger the self-healing along with the mutual diffusion of long polymer chains and ferric ions.展开更多

文摘Highly thermal conductivity materials with excellent electromagnetic interference shielding and Joule heating performances are ideal for thermal management in the next generation of communication industry,artificial intelligence and wearable electronics.In this work,silver nanowires(AgNWs)are prepared using silver nitrate as the silver source and ethylene glycol as the solvent and reducing agent,and boron nitride(BN)is performed to prepare BN nanosheets(BNNS)with the help of isopropyl alcohol and ultrasonication-assisted peeling method,which are compounded with aramid nanofibers(ANF)prepared by chemical dissociation,respectively,and the(BNNS/ANF)-(AgNWs/ANF)thermal conductivity and electromagnetic interference shielding composite films with Janus structures are prepared by the"vacuum-assisted filtration and hot-pressing"method.Janus(BNNS/ANF)-(AgNWs/ANF)composite films exhibit"one side insulating,one side conducting"performance,the surface resistivity of the BNNS/ANF surface is 4.7×10^(13) Ω,while the conductivity of the AgNWs/ANF surface is 5,275 S/cm.And Janus(BNNS/ANF)-(AgNWs/ANF)composite film with thickness of 95 pm has a high in-plane thermal conductivity coefficient of 8.12 W/(m·K)and superior electromagnetic interference shielding effectiveness of 70 dB.The obtained composite film also has excellent tensile strength of 122.9 MPa and tensile modulus and 2.7 GPa.It also has good temperature-voltage response characteristics(high Joule heating temperature at low supply voltage(5 V,215.0℃),fast response time(10 s)),excellent electrical stability and reliability(stable and constant real-time relative resistance under up to 300 cycles and 1,500 s of tensile-bending fatigue work tests).

文摘Flexible multifunctional polymer-based electromagnetic interference(EMI)shielding composite films have important application values in the fields of 5G communication technology,wearable electronic devices and artificial intelligence.In this work,Fe_(3)O_(4)/polyamic acid(PAA)nanofiber films are prepared by in-situ polymerization and electrospinning technology,and Ti_(3)C_(2)T_(x)nanosheets are deposited on the surface of the Fe_(3)O_(4)/PAA nanofiber films via vacuum-assisted filtration.Then,Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/polyimide(PI))composite films are obtained by thermal imidization.The two sides of the Janus films exhibit completely different properties.The Fe_(3)O_(4)/PI side has excellent hydrophobicity and insulation property,and the Ti_(3)C_(2)T_(x)side has hydrophilicity and terrific conductivity.When the mass fraction of Ti_(3)C_(2)T_(x)is 80 wt.%,the Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/PI)composite film has excellent EMI shielding performances and mechanical properties,with EMI shielding effectiveness,tensile strength and Young’s modulus reaching 66 dB,114.5 MPa and 5.8 GPa,respectively.At the same time,electromagnetic waves show different absorption shielding effectiveness(SEA)when incident from two sides of the Janus films.When the electromagnetic waves are incident from the Fe_(3)O_(4)/PI side,the SEA of the Janus film is 58 dB,much higher than that when the electromagnetic waves are incident from the Ti_(3)C_(2)T_(x)side(39 dB).In addition,the Ti_(3)C_(2)T_(x)side of the Janus Ti_(3)C_(2)T_(x)-(Fe_(3)O_(4)/PI)composite films also has excellent electrothermal and photothermal conversion performances.When the applied voltage is 4 V,the stable surface temperature reaches 108°C;when it is irradiated by simulated sunlight with power density of 200 mW/cm2,the stable surface temperature reaches 95℃.

文摘Pancreatitis is inflammation of pancreas and caused by a number of factors including pancreatic duct obstruction, alcoholism, and mutation in the cationic trypsinogen gene. Pancreatitis is represented as acute pancreatitis with acute inflammatory responses and; chronic pan-creatitis characterized by marked stroma formation with a high number of infiltrating granulocytes(such as neutrophils, eosinophils), monocytes, macrophages and pancreatic stellate cells(PSCs). These inflammatory cells are known to play a central role in initiating and promoting inflammation including pancreatic fibrosis, i.e., a major risk factor for pancreatic cancer. A number of inflammatory cytokines are known to involve in pro-moting pancreatic pathogenesis that lead pancreatic fibrosis. Pancreatic fibrosis is a dynamic phenomenon that requires an intricate network of several autocrine and paracrine signaling pathways. In this review, we have provided the details of various cytokines and molecular mechanistic pathways(i.e., Transforming growth factor-β/SMAD, mitogen--activated protein kinases, Rho kinase, Janus kinase/signal transducers and activators, and phosphatidylinositol 3 kinase) that have a critical role in the activation of PSCs to promote chronic pancreatitis and trigger the phenomenon of pancreatic fibrogenesis. In this review of literature, we discuss the involvement of several pro-inflammatory and anti-inflammatory cytokines, such as in interleukin(IL)-1, IL-1β, IL-6, IL--8 IL-10, IL-18, IL--33 and tumor necrosis factor-α, in the pathogenesis of disease. Our review also highlights the significance of several experimental animal models that have an important role in dissecting the mechanistic pathways operating in the development of chronic pancreatitis, including pancreatic fibrosis. Additionally, we provided several intermediary molecules that are involved in major signaling pathways that might provide target molecules for future therapeutic treatment strategies for pancreatic pathogenesis.

文摘OBJECTIVE: To observe effect of Liuweibuqi Capsule, a Traditional Chinese Medicine (TCM), on the janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway and matrix metalloproteinases (MMPs) in a chronic obstructive pulmonary disease (COPD) rat model with lung deficiency in terms of TCM's pattern differentiation. METHODS: Rats were randomly divided into a normal group, model group, Liuweibuqi group, Jinshuibao group, and spleen aminopeptidase group (n= 10). Aside from the normal group, all rats were ex-posed to smoke plus lipopolysaccharide tracheal instillation to establish the COPD model with lung deficiency. Models were established after 28 days and then the normal and model groups were given normal saline (0.09 g/kg), Liuweibuqi group was given Liuweibuqi capsule (0.35 g/kg), Jinshuibao group was given Jinshuibao capsules (0.495 g/kg), and the spleen group was given spleen aminopeptidase (0.33 mg/kg), once a day for 30 days. Changes in symptoms, signs, and lung histology were observed. Lung function was measured with a spirometer. Serum cytokines were detected using enzyme-linked immunosorbent assay, and changes in the JAK/STAT pathway, MMP-9, and MMPs inhibitor 1 (TIMP1) were detected by immunohistochemistry, RT-PCR, and western blotting, respectively.RESULTS: Compared with the normal group, lung tissue was damaged, and lung function was reduced in the model control group. Additionally, the levels of interleukin (IL)-1β, y interferon (IFN-γ), and IL-6 were higher, while IL-4 and IL-10 were lower in the model control group than those in the normal group. The expressions of JAK1, STAT3, ρ-STAT3, and MMP-9 mRNA and protein in lung tissue were higher, and TIMP1 mRNA and protein was lower in the model group compared with the normal group. After treatment, compared with the model group, the expression of inflammatory cytokines was lower in each treatment group, and expressions of JAK/ STAT pathway, MMPs were lower. Compared wi
文摘In 2018,the pan-Janus kinase(JAK)inhibitor tofacitinib was launched for the treatment of ulcerative colitis(UC).Although tofacitinib has proven efficacious in patients with active UC,it failed in patients with Crohn’s disease(CD).This finding strongly hints at a different contribution of JAK signaling in both entities.Here,we review the current knowledge on the interplay between the JAK/signal transducer and activator of transcription(STAT)pathway and inflammatory bowel diseases(IBD).In particular,we provide a detailed overview of the differences and similarities of JAK/STAT-signaling in UC and CD,highlight the impact of the JAK/STAT pathway in experimental colitis models and summarize the published evidence on JAK/STAT-signaling in immune cells of IBD as well as the genetic association between the JAK/STAT pathway and IBD.Finally,we describe novel treatment strategies targeting JAK/STAT inhibition in UC and CD and comment on the limitations and challenges of the new drug class.

文摘Objectives: To investigate the mechanism of Liuwei Dihuang Pill (六味地黄丸, LDP) in treating postmenopausal osteoporosis (PMOP) with Shen (Kidney) yin deficiency. Methods: In this study, 205 cases of PMOP were divided into the PMOP Shen-yin deficiency group (Group A), PMOP Shen-yang deficiency group (Group B), PMOP without Shen deficiency group (Group C), and control group (Group N). Real-time polymerase chain reaction (RT-PCR) and Western blot techniques were used to observe the effects of LDP treatment on the cardiotrophin-like cytokine factor 1 (CLCF1), ankyrin repeat and SOCS box containing 1 (ASB1), and proldneticin 2 (PROK2) genes and the Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway. Results: The mRNA (P〈0.05) and protein (P〈0.01) expression levels of the CLCF1 gone in Group A were significantly lower than the corresponding levels in Group N. After LDP treatment for 3 months, the mRNA expression levels of the CLCF1 gone were obviously up-regulated (P〈0.01). After 6-month treatment, the expression levels of CLCF1 mRNA and protein were significantly up-regulated (both P〈0.01), and the average bone density of the top femur had significantly increased (P〈0.05). In vitro, CLCF1 overexpression resulted in a significant increase in the total protein and phosphorylated protein levels of JAK2 and STAT3. Conclusions: The CLCF1 gone is an important gone associated with PMOP Shen-yin deficiency and the therapeutic effects of LDP may be mediated by up-regulation of CLCF1 gone expression and activation of the JAK/STAT signaling pathway.

文摘Background Autoantibodies against angiotensin AT1 receptor have been discovered in patients with preeclampsia or malignant hypertension. Some studies have demonstrated that the autoantibodies are involved in the immunopathogenesis of hypertension and have an agonist effect similar to angiotensin II. Methods Autoantibodies against AT1 receptor were purified from sera of patients with primary hypertension by affinity chromatography. Proliferation of cultured rat vascular smooth muscle cells was detected by bromodeoxyuridine incorporation and activation of signalling molecules detected by Western blotting and electrophoretic mobility shift assay. Results The AT1-RAb caused a significant proliferation similar to the Ang II during first 24 hours. The levels of nuclear factor-KB (NF-KB), phosphorylated JAK2., phosphorylated STAT1 (pSTAT1) and phosphorylated STAT3 (pSTAT3) molecules were increased in response to the autoantibodies. In contrast, the activations of NF-KB and JAK-STAT were blocked by Iosartan, pyrrolidinedithiocarbamate (a specific inhibitor of NF-KB) and AG490 (a specific inhibitor of the JAK2. tyrosine kinase). The expressions of NF-KB, pSTAT1 and pSTAT3 reached peak levels at different times. Moreover, the relative densities of electrophoretic bands showed that activation of pSTAT3 was more significant than STAT1 induced by AT1 -RAb. Conclusions These results suggest that the autoantibodies against AT1 receptor have an agonist effect similar to Ang II in proliferation of VSMCs and the NF-KB and JAK-STAT proteins play essential roles. The effect is different from Angll in that STAT3 is the main downstream activating molecule in JAK-STAT signalling pathway.

文摘Macrophages play an important role in inflammation, and excessive and chronic activation of macrophages leads to systemic inflammatory diseases, such as atherosclerosis and rheumatoid arthritis. In this paper, we explored the anti-inflammatory effect of broussonin E, a novel phenolic compound isolated from the barks of Broussonetia kanzinoki, and its underlying molecular mechanisms. We discovered that Broussonin E could suppress the LPS-induced pro-inflammatory production in RAW264.7 cells, involving TNF-α, IL-1β, IL-6, COX-2 and iNOS. And broussonin E enhanced the expressions of anti-inflammatory mediators such as IL-10, CD206 and arginase-1(Arg-1) in LPS-stimulated RAW264.7 cells. Further, we demonstrated that broussonin E inhibited the LPS-stimulated phosphorylation of ERK and p38 MAPK. Moreover, we found that broussonin E could activate janus kinase(JAK) 2, signal transducer and activator of transcription(STAT) 3. Downregulated pro-inflammatory cytokines and upregulated anti-inflammatory factors by broussonin E were abolished by using the inhibitor of JAK2-STAT3 pathway, WP1066. Taken together, our results showed that broussonin E could suppress inflammation by modulating macrophages activation state via inhibiting the ERK and p38 MAPK and enhancing JAK2-STAT3 signaling pathway, and can be further developed as a promising drug for the treatment of inflammation-related diseases such as atherosclerosis.

文摘OBJECTIVE:Electroacupuncture effect on neurological behavior and the expression of tyrosine kinase Janus kinase 2(JAK 2) of ischemic cortex in rats with the focal cerebral ischemia were investigated in this study.METHODS:The model of focal cerebral ischemia was established by the heat-coagulation induced the occlusion of the middle cerebral artery.The electro-acupuncture was applied on Baihui(GV 20) and Dazhui(GV 14),and AG490 was applied by intracerebroventricular infusion.The expressions of JAK2 mRNA and phospharylatedJAK2(p-JAK2) in the ischemic cortex were observed by in situ hybridization and western blotting.RESULTS:The expressions of JAK2 mRNA and p-JAK2 were rarely found in sham surgery group.In model group,the expression of JAK2 mRNA and JAK2 phosphorylation had increased.After 1 day of cerebral ischemia,the expression had reached its peak.After cerebral ischemia,the expressions of JAK2 mRNA and p-JAK2 were consistent with the neurological deficit score.Electroacupuncture treatment and AG490 intervention were able to improve the neurological deficit score after cerebral ischemia,and down-regulate the expressions of JAK2 mRNAand JAK2 phosphorylation.CONCLUSION:After cerebral ischemia,the excessive expressions of JAK2 and the JAK2 phosphorylation would be one of mechanisms by which the brain injury got worse.The therapy of electro-acupuncture could reduce the expression of JAK2,and inhibit JAK2 phosphorylated activation,so as to block the abnormal activation of signal transduction pathway which was induced by JAK2.

文摘AM: To clarify the role of Janus kinase-signal transducers and activators of transcription (JAK-STAT) pathway in platelet-derived growth factor (PDGF) induced proliferation in activated pancreatic stellate cells (PSCs). METHODS: PSCs were isolated from rat pancreas tissue, and used in their culture-activated, myofibroblast-like phenotype. STAT-specific binding activity was assessed by electrophoretic mobility shift assay. Activation of Src, JAK2, STAT1, STAT3, and ERK was determined by Western blotting using anti-phosphospecific antibodies. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine. RESULTS: PDGF-BB induced STAT-specific binding activity, and activation of Src, JAK2, STAT1, STAB, and ERK. Ethanol and acetaldehyde at clinically relevant concentrations decreased basal activation of JAK2 and STAT3. PDGF-induced activation of STAT1 and STAT3 was inhibited by a Src inhibitor PP1 and a JAK2 inhibitor AG490, whereas PDGF-induced activation of ERK was inhibited by PP1, and not by AG490. PDGF-induced proliferation was inhibited by PP1 and AG490 as well as by STAT3 antisense oligonucleotide. CONCLUSION: PDGF-BB activated JAK2-STAT pathway via Src-dependent mechanism. Activation of 3AK2-STAT3 pathway, in addition to ERK, may play a role in PDGF-induced proliferation of PSCs.
文摘The features of JAK-STAT signaling in liver cells are discussed in the current review. The role of this signaling cascade in carcinogenesis is accentuated. The possible involvement of this pathway and alteration of its elements are compared for normal cholangiocytes, cholangiocarcinoma predisposition and development. Prolactin and interleukin-6 are described in detail as the best studied examples. In addition, the non-classical nuclear translocation of cytokine receptors is discussed in terms of its possible implication to cholangiocarcinoma development.
文摘Retroperitoneal operations, such as radical prostatectomy, often damage the cavernous nerve, resulting in a high incidence of erectile dysfunction. Although improved nerve-sparing techniques have reduced the incidence of nerve injury, and the administration of phosphodiesterase type 5 inhibitors has revolutionized the treatment of erectile dysfunction, this problem remains a considerable challenge. In recent years, scientists have focused on brain-derived neurotrophic factor and vascular endothelial growth factor in the treatment of cavernous nerve injury in rat models. Results showed that both compounds were capable of enhancing the regeneration of the cavernous nerve and that activation of the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway played a major role in the process.

文摘OBJECTIVE: Inactivation of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) signaling axis plays a crucial role in determining the fate of neural stem cells(NSCs).Qingnaoyizhi decoction(QNYZD) has been used for the treatment of vascular dementia and has shown to improve synaptic remodeling. The aim of this study was to evaluate the effect of cerebrospinal fluid(CSF) containing QNYZD(CSF-QNYZD) on the differentiation of cultured NSCs and the involvement of the JAK2/STAT3 pathway.METHODS: The protein expression levels of glial fibrillary acidic protein(GFAP), tubulin, drosophila mothers against decapentaplegic protein(SMAD-1), STAT3, and phosphorylated-STAT3 were detected by western immunoblot analysis in the groups: control, CSF, JAK/STAT inhibitor(AG490),CSF-QNYZD, and CSF-XDZ(CSF-Xidezhen). The differentiation of NSCs was determined by immunofluorescence staining. The proliferation of NSCs was measured using the Cell Counting Kit-8 proliferation assay.RESULTS: Compared with the control group,CSF-QNYZD and AG490 significantly increased the number and expression of tubulin-positive cells, reduced the number and expression of GFAP-positive cells, and down-regulated the expression of p-STAT3. However, CSF-QNYZD also decreased the expression of SMAD-1 and STAT3.CONCLUSION: Enhanced neuronal differentiation may be associated with the down-regulation of glial differentiation instead of promoting proliferationin treated NSCs. Furthermore, QNYZD may play a direct role in suppressing the formation of GFAP-positive cells and enhancing neuronal differentiation by inhibiting JAK2/STAT3 activation. Overall, these results provide insights into the possible mechanism underlying QNYZD-mediated neurogenesis.

文摘Psoriatic arthritis(PsA)is a type of chronic inflammatory arthritis which is associated with psoriasis.The early recognition and treatment for PsA are of critical importance.Janus kinase(JAK)inhibitors,as a kind of orally small molecules,have emerged as an encouraging class of drug in PsA treatment.This review provides a discussion of the role and current status of JAK inhibitors in the control of PsA.There are three JAK inhibitors approved for use in autoimmune diseases,for example,tofacitinib,baricitinib,and upadacitinib,and only tofacitinib has been approved in PsA treatment.The clinical trials of upadacitinib and filgotinib in PsA patients are undergoing.The efficacy and safety of these agents were briefly discussed.Although there are still issues in terms of their efficacy and safety currently,JAK inhibitors are expected to benefit more PsA patients in future.

文摘We report a fuel-free, near-infrared (NIR)-driven Janus microcapsule motor. The Janus microcapsule motors were fabricated by template-assisted polyelectrolyte layer-by-layer assembly, followed by spraying of a gold layer on one side. The NIR-powered Janus motors achieved high propulsion with a maximum speed of 42μm.s-1 in water. The propulsion mechanism of the Janus motor was attributed to the self-thermophoresis effect: The asymmetric distribution of the gold layer generated a local thermal gradient, which in turn generated thermophoretic force to propel the Janus motor. Such NIR-propelled Janus capsule motors can move efficiently in cell culture medium and have no obvious effects on the cell at the power of the NIR laser, indicating considerable promise for future biomedical applications.

文摘Multi-bond network(MBN) which contains a single network with hierarchical cross-links is a suggested way to fabricate robust hydrogels. In order to reveal the roles of different cross-links with hierarchical bond energy in the MBN, here we fabricate poly(acrylic acid) physical hydrogels with dual bond network composed of ionic cross-links between carboxylFe3+ interactions and hydrogen bonds, and compare these dually cross-linked hydrogels with singly and ternarily cross-linked hydrogels. Simple models are employed to predict the tensile property, and the results confirm that the multi-bond network with hierarchical distribution in the bond energy of cross-links endows hydrogel with effective energy-dissipating mechanism. Moreover, the dually cross-linked MBN gels exhibit excellent mechanical properties(tensile strength up to 500 k Pa, elongation at break ~ 2400%) and complete self-healing after being kept at 50 °C for 48 h. The factors on promoting self-healing are deeply explored and the dynamic multi-bonds are regarded to trigger the self-healing along with the mutual diffusion of long polymer chains and ferric ions.
