沙带鱼是带鱼科中一种重要的海洋经济鱼类,广泛分布于印度-西太平洋海域。近年来,过度捕捞和渔业生态环境变化使沙带鱼的自然资源和种群结构受到了严重影响。因此,为了制定出科学合理的资源保护和管理措施,急需开发出大量多态的沙带鱼... 沙带鱼是带鱼科中一种重要的海洋经济鱼类,广泛分布于印度-西太平洋海域。近年来,过度捕捞和渔业生态环境变化使沙带鱼的自然资源和种群结构受到了严重影响。因此,为了制定出科学合理的资源保护和管理措施,急需开发出大量多态的沙带鱼微卫星位点以促进其种群遗传学研究。本研究采用SLAF-seq技术,首次在沙带鱼基因组DNA中筛选出28个多态性微卫星位点。每个位点的等位基因数为2~18,平均值9.11;观测杂合度和期望杂合度分别为0.077~0.871(平均值0.634)和0.151~0.942(平均值0.711);各位点多态信息含量为0.137~0.919,平均值0.670。经过Bonferroni校正后,有3个位点显著偏离哈迪-温伯格平衡(HWE),并显示出较高的近交系数和无效等位基因频率,其余25个位点均符合HWE。这些多态性微卫星DNA标记可为沙带鱼的种群遗传资源研究提供有效的分子标记和技术支撑。此外,跨物种扩增结果表明,有19个微卫星标记在4种带鱼属鱼类中具有较好的通用性,这些标记可为阐明带鱼科属间或种间的系统进化关系提供新的研究手段和视角。 展开更多

【目的】开发黑老虎SSR标记,为其群体遗传学研究以及种质资源收集、保存、评价以及良种选育提供依据。【方法】基于高通量测序获取候选SSR位点,设计引物进行PCR扩增及其产物多态性检测;以湖南会同、城步和桂东3个天然群体66个黑老虎植... 【目的】开发黑老虎SSR标记,为其群体遗传学研究以及种质资源收集、保存、评价以及良种选育提供依据。【方法】基于高通量测序获取候选SSR位点,设计引物进行PCR扩增及其产物多态性检测;以湖南会同、城步和桂东3个天然群体66个黑老虎植株为供试材料,选取多态位点开展遗传多样性分析,并应用5个近缘种25个植株探讨SSR标记的通用性。【结果】通过高通量测序检索到152207个候选SSR位点,以二核苷酸重复比例(57.69%)最大,其次为单核苷酸(17.30%)和四核苷酸重复(10.68%);单、二、三、四、五和六核苷酸重复单元分别以A/T(98.06%)、AC/GT(57.30%)、AAG/CTT(41.91%)、AAAT/ATTT(29.55%)、AAAAT/ATTTT(29.18%)和ACACAT/ATGTGT(40.31%)最为普遍。从160对SSR引物中筛选出28对,能成功进行PCR扩增且获得多态位点;3个天然群体的遗传多样性分析结果表明,28个位点共有168个等位基因,每位点等位基因数为2~14,实际杂合度和期望杂合度分别为0.000~0.900和0.000~0.860;会同、城步和桂东群体分别有9、13和7个位点显著偏离Hardy-Weinberg平衡(P<0.05),未发现位点间连锁不平衡现象(P≥0.05)。28个位点中,有24和14个可应用于南五味子和异形南五味子,而对于翼梗五味子、华中五味子和狭叶五味子,则通用性偏低,仅6~9个位点得以成功扩增。【结论】开发出的28个SSR标记,可应用于黑老虎及其近缘种的遗传多样性分析、种质资源评价、种质鉴定以及分子标记辅助育种等。 展开更多

Korean freshwater snails of the genus Semisulcospira are widely distributed across East Asia.It has been a very popular nutritional food in Korea,and is an ecologically important water quality indicator because it lives only in clean water.However,no microsatellite markers have been generated to study the population genetic diversity of this genus.In the present study,we developed and characterized 18 novel microsatellite loci from Semisulcospira coreana genomic DNA.The microsatellites were isolated using 454 GS-FLX titanium sequencing and 18 markers were used for genotyping in S.coreana.In addition,we also tested the cross-species transferability of the microsatellite markers in four additional Semisulcospira spp.We identified 18 polymorphic loci and the number of alleles per loci,and their polymorphism information content values ranged from 2 to 17 and 0.203 to 0.902,respectively.The observed and expected heterozygosities of the loci ranged from 0.063 to 0.924 and 0.226 to 0.924,respectively.According to the analysis of the cross-species transferability of these markers,four species,S.forticosta,S.gottschei,S.tegulata,and S.libertina,showed a very high transferability(80%–85%).These results show that this set of nuclear markers could be useful for population genetics studies of this species and closely related species. 展开更多

We characterized 14 anonymous nuclear loci from Pinus thunbergii Parl., an important pine species native to Japan. One hundred and twenty-six single nucleotide polymorphisms (SNPs) were identified from these loci, giving a frequency of 1 SNP per 51 bp. Nucleotide di- versity (0) ranged from 1.06 × 10^-3 to 11.87 × 10^-3, with all average of 4.99 × 10^-3. Only one locus (mK45) deviated significantly from the Hardy-Weinberg equilibrium. Thirteen of 14 loci were applicable in other pine species. These loci will be useful for nucleotide variation studies and will provide material for SNP-based marker development in P. thun- bergii and related species. 展开更多

As a powerful molecular tool, microsatellites offer the great potentials for various purposes in plants. This review provides a scenario of microsatellites in medicinal plants including development from genomic or expressed sequence tag libraries, cross-species transferability, genotyping, and potential applications.
