Mapping bruchid resistance(Br) will provide an important basis for cloning the responsible gene(s) and elucidating its functional mechanism, and will also facilitate marker-assisted selection in mung bean breeding. Here, we report the construction of the genetic linkage groups of mung bean and mapping of the Br1 locus using an RIL population derived from a cross between Berken, a bruchid-susceptible line, and ACC41, a bruchid-resistant line. A total of 560 markers were mapped onto 11 linkage groups,with 38.0% of the markers showing distorted segregation. The lengths of the linkage groups ranged from 45.2 to 117.0 c M with a total coverage of 732.9 c M and an average interval of1.3 c M between loci. Br1 was located on LG9 between BM202(0.7 c M) and Vr2-627(1.7 c M).Based on 270 shared SSR markers, most of the linkage groups were assigned to specific chromosomes. These results should further accelerate the genetic study of this crop.展开更多

Over a period spanning 2015 to 2...This study explores the use of genetic variability for advancing the genetic improvement of Cowpea (Vigna unguiculata (L.) Walp), particularly in response to insect infestation stress. Over a period spanning 2015 to 2017, forty accessions of cowpeas were evaluated to determine their variability under both insecticide spray and no insecticide spray conditions at the Teachings and Research Farms, Federal University of Agriculture, Abeokuta. The experimental design was a randomized complete block design in three replicates. The accessions were evaluated for plant height, leaf length, leaf width, number of days of 50% flowering, number of pods per plant, pod length, number of seeds per plant, 100-seed weight, and seed yield. Data collected were subjected to principal component and single linkage cluster analyses. Principal axis I (PCA1) accounted for 39% and 35% under insecticide spray and no insecticide spray respectively to the total variation in the accessions. Plant height with a factor score of 0.38, leaf length (0.41), number of leaves (0.37), and 100-seed, weight (0.30) was related to PCAI under insecticide spray while leaf width (0.32). Pod length (0.37) and number of seeds/plant (0.38) were significant to PCA1 under no insecticide spray. Notably, accessions such as SAMPEA6, SAMPEA10, IFE-Brown, and IFE-BPE exhibited consistent performance across both conditions, while others displayed condition-specific attributes. For instance, NGB1063, NGB1152, and NGB1093 demonstrated distinct traits under insecticide spray, while NGB1146 and NGB1124 exhibited notable characteristics under no insecticide spray conditions. Therefore, identifying these forty accessions with desirable traits hold promise for future genetic improvement efforts of cowpea cultivation in Nigeria and beyond.展开更多

Salt stress directly affects plant photosynthesis, which is an important physiological process to limit plant growth and crop yield. But the effects of salt stress on the mechanism of the photosynthesis is still not clear. In this experiment, the salt tolerant plant Alhagi pseudalhagi was selected as the experiment material, and the salt sensitive plant Vigna radiata as the control, to explore the effects of salt stress on photosynthetic parameters of A. pseudalhagi. Plants were grown in a greenhouse,cultured with 1/2 Hoagland nutrient solution, treated by 0(control), 50, 100 or 200 s of mmol/L of NaCl solution for 12 d. Then,the chlorophyll contents, gas exchange parameters and chlorophyll a fluorescence in each treatment were measured. The results showed that under the salt stress simulated by 50 mmol/L NaCl, the net photosynthetic rate(Pn) and stomatal conductance(gs)of A. pseudalhagi were significantly increased compared with the control, while intercellular CO_2 concentration(Ci) was significantly decreased, Fv'/Fm', Fv/Fm and Fv/Fo were increased over time, but had no significant differences with the control ФPSⅡ,ETR and qP were significantly increased, and NPQ was significantly decreased. Under the salt stress simulated by 100 and 200mmol/L NaCl, Fv'/Fm', Fv/Fm, Fv/Fo, ФPSⅡ and qP of A. pseudalhagi were gradually decreased over time. In contrary, under the salt stress simulated by 50 and 100 mmol/L NaCl, the Pn, gs, Ci, Fv'/Fm', Fv/Fm, Fv/Fo, ФPSII, qP and ETR were all significantly decreased, while NPQ was significantly increased.展开更多

This study aimed at evaluating the effect of applying bioinsecticides on the biological parameters of C. maculatus to reduce its damage to cowpea stocks. Four treatments including an untreated control and three doses of powdery extracts of C. nigricans, Z. zanthoxyloides and H. suaveolens, were compared in a Fischer block with four completely randomized repetitions. One hundred undamaged cowpea seeds were mixed with doses of each extract in Petri dishes and then fed as food substrates to ten pairs of C. maculatus for eight days. The extracts of H. suaveolens, C. nigricans and Z. zanthoxyloides all contain active ingredients including polyphenols, flavonoids, anthocyanins and alkaloids, etc. The extract of Z. zanthoxyloides was found to be more active on adults (100.00% mortality) followed by C. nigricans (98.75% mortality) and H. suaveolens (97.50% mortality) at 80 mg/g. Fecundity per female decreased significantly in the treated batches of 13.82 eggs for H. suaveolens at 10.12 eggs for C. nigricans at a dose of 80 mg/g. Germination rate of seeds was statistically raised to 100% at 60 and 80 mg/g by C. nigricans powder and lowered to 83.00% at 20 mg/g by Z. zanthoxyloides extracts. The application of Z. zanthoxyloides extract would be a less expensive alternative for the sustainable management of C. maculatus in cowpea stocks.展开更多

All the treatments significantly improved the growth of the plants as compared carbofuran and A. indica seed powder increased plant growth to untreated inoculated plants. Analysis of data showed that and yield significantly more in comparison to bavistin and P. fluorescens. Carbofuran was highly effective against nematode, bavistin against fungus, A. indica seed powder against both the pathogens and both the bioagents were moderately effective against both the pathogens.展开更多

Based on α-amylase gene structure and sequence, A pair of PCR primers, F (CCTACATTCTAACACACCCT) and R (GCATATTGTGCCAGTACAAT) were designed to amplify intron-3 fragments of α-amylase gene. 14 variant types were detected, including 13, 9, 10, 4 variant types in the wild, weed, locally cultivated and modern brought-up adzuki beans respectively, 9, 8, 7 variant types of the wild adzuki beans from Japan, China and Korea respectively, and some other variant types in the local adzuki beans from China and Bhutan. 60% of subjects of cultivated races were found to be EE type in the experiment. In addition, sequence analysis of intron-3 of α-amylase gene from 8 variant types reveals the evolution process of various variant types in adzuki beans.展开更多

The use of natural substances as pesticides is being encouraged to fight against cowpea field pests. The pesticidal potentials of Azadirachta indica and Boswellia dalzielii water extracts, Metarhizium anisopliae, alone and in combination with plant extracts, and a commercial synthetic pesticide (Decis<sup>®</sup>) were tested in field on two varieties of cowpeas, Vigna unguiculata in two agroecological zones (Guinean Savanah and Sahelian Savanah) of Cameroon. The field trials were carried out in a full randomized block design including nine treatments and a control. Four replications were made concerning the different treatments and control. At the flowering stage, the cowpea field was sprayed three times with different pesticidal formulations at the interval of five days. The number of ramifications per plant, and that of pods per block and seed yield were determined. The pesticide formulations considerably (p ®</sup>). Bafia variety treated with the combination of M. anisopliae and A. indica recorded the highest ramification rate (37.03 ± 1.59) in Maroua (Sahelian Savanah). The same variety also produced more important pods number (90.50 ± 16.66) in Ngaoundere (Guinean Savanah) with the binary combination of two plants used in this experiment. The highest seed yield (44.23 ± 2.31) was recorded in Ngaoundere with B125 variety treated with the combination of the three pesticidal formulations separately (A. indica, B. dalzielii, M. anisopliae). The plant products used in this work, M. anisopliae and their mixtures could supersede the synthetic pesticides considering environmental issue in cowpea crop protection. Then, such formulations would not only improve crop productivity but also preserve environment from the pollution due to the use of synthetic residual chemicals.展开更多

However, not much is known about the base index selection method in breeding cowpea for drought tolerance, which is important for yields. Consequently, the present study has been conducted to: 1) evaluate the yield performance of cowpea genotypes under artificial drought and well-watered condition, 2) ranke genotype performance using selection indices, and 3) assess relationship between agronomic traits and yield. The experiment was the 2 watering conditions laid out in a Randomized Complete Block Design (RCBD) with three replications. The experiment was carried out in pots under screen house at the Department of Horticulture at KNUST. The result showed that KPR1-96-73, Simbo, CZ06-4-16, Wilibaly and Agyenkwa were high yielding in well-water condition while Ghana Shoba, Sangaraka, Nketewade, Ghana Shoni and Korobalen were high yielding genotypes in water stress condition. The average yield reduction was 60.6% and 16% for grain and fodder yield respectively. The biplot displays revealed four groups among the genotypes tested which were based on their yielding capacity and drought tolerance. In cluster B high yielding and drought tolerant genotypes were identified, high yielding and drought susceptible have been identified in cluster A, low yielding and drought tolerant in cluster D, and lastly low yielding and drought susceptible in cluster C. Genotypes in cluster B were best due to the fact that it combines high yield and tolerance to drought. They were Ghana Shoni, Nketewade, Sangaraka and Ghana Shoba. These genotypes might be suitably employed in further drought tolerance breeding programs of cowpea. Significant relationships were observed between agronomic trait and yields under drought condition.展开更多

Data were collected on the Number of leaves, Plant height (cm), Growth habit (cm), Terminal leaflet length, Terminal leaflet width, Number of nodes/stem, Number of stems/plant, Number of pods/plant, Weight of 100 pods (g), Weight of 100 seeds (g), Length of seeds (mm), Width of seeds (mm), Weight of seeds/Plot (kg). There were significant differences for all characters, except Number of leaves, Terminal leaflet length, Terminal leaflet width, Length of seeds (mm) Weight of 100 pods (g), Weight of 100 seeds (g). The morphotypes UZ-VZ-04, UZ-VZ-03, UZ-VZ-06, UZ-VZ-02 and UZ-VZ-05 have demonstrated good performance for grain weight per plot and can be useful for a breeding program.展开更多

According to the cluster results, the 16 lines can be divided into four groups, including brucid resistant wild types, resistant cultivated lines, resistant progenies and a mixed group. BSA method was used to identify DNA markers that related with bruchid resistant gene by using resistant line and susceptible line and their F2 progeny. One codominant marker was identified, which generated a fragment of 1.79 kb in resistant lines and 1.03 kb in susceptible lines. Finally, this codominant marker was considered to be tightly linked with bruchid resistant gene and could be useful in resistant germplasm identification and marker-assisted selection.展开更多

Here we describe observations on the adaptability of mung bean germplasm obtained from the United States Department of Agriculture and their genetic diversity assessment using SSR markers. Several accessions were shown to be mixtures,based on their phenotypes for some characters. Most accessions were able to complete their lifecycles when grown in Beijing, China, making them ideal for crossbreeding without day length control. High diversity was revealed by the SSR markers, with an average of 4.2 alleles per locus and a PIC value of 0.650 per locus. STRUCTURE analysis divided the accessions into six groups.There was no obvious trend of accessions forming groups according to their geographical origin,owing mainly to germplasm exchange and an uneven distribution of accessions. The present results indicate that this germplasm would enrich the local gene pool, and provide information for the further use of germplasm in breeding programs.展开更多

Wilczek which grows flat along the ground can increase parasitism of the eggs of the Asian corn borer, Ostrinia furnacalis (Guenée), a crucial insect pest of maize in China, by Trichogramma ostriniae Pang Chen. This study investigated the behavioural responses of T. ostriniae to plant volatiles emitted from whole plants, leaves, and flowers of prostrate and erect varieties of mungbean in a 4-armed olfactometer. Female parasitiods were significantly attracted to the odours from some varieties of prostrate mungbean when the visit duration and the number of visits were considered, but were not attracted to the odours from any erect varieties. The attractive odours were shown to emanate from the leaves, rather than from the flowers of prostrate mungbean plants. These results have implications for the augmentation of Trichogramma parasitoids in bio-control of O. furnacalis and other crop pests.展开更多
Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family Fabaceae.It is a close relative of cultivated Vigna species such as adzuki bean(V.angularis),cowpea(V.unguiculata),mung bean(V.radiata),and blac...Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family Fabaceae.It is a close relative of cultivated Vigna species such as adzuki bean(V.angularis),cowpea(V.unguiculata),mung bean(V.radiata),and blackgram(V.mungo),and is distributed throughout the tropics.With its ability to tolerate salt stress,beach pea has great potential to contribute salt-tolerance genes for developing salt-tolerant cultivars in cultivated Vigna species.However,it is still underutilized in Vigna breeding programs.A draft genome sequence of beach pea was generated using a high-throughput next-generation sequencing platform,yielding 23.7 Gb of sequence from 79,929,868 filtered reads.A de novo genome assembly containing 68,731 scaffolds gave an N50 length of 10,272 bp and the assembled sequences totaled 365.6 Mb.A total of 35,448 SSRs,including 3574 compound SSRs,were identified and primer pairs for most of these SSRs were designed.Genome analysis identified 50,670 genes with mean coding sequence length 1042 bp.Phylogenetic analysis revealed highest sequence similarity with V.angularis,followed by V.radiata.Comparison with the V.angularis genome revealed 16,699 SNPs and 2253 InDels and comparison with the V.radiata genome revealed 17,538 SNPs and 2300 InDels.To our knowledge this is the first draft genome sequence of beach pea derived from an accession(ANBp-14-03)adapted locally in the Andaman and Nicobar Islands of India.The draft genome sequence may facilitate the genetic enhancement in cultivated Vigna species.展开更多
A set of representative 146 adzuki (Vigna angular is var. angularis, and var. nipponensis) germplasm from 6 Asian countries traditionally for adzuki bean production, together with an out group standard rice bean (Vign...A set of representative 146 adzuki (Vigna angular is var. angularis, and var. nipponensis) germplasm from 6 Asian countries traditionally for adzuki bean production, together with an out group standard rice bean (Vigna umbellata), were analyzed by AFLP methodology using 12 informative primer pairs. 313 unambiguous polymorphic bands were created. According to the dendrogram by cluster analysis based on AFLP banding, 143 of the accessions were distinct and revealed enough genetic diversity for identification and classification of accessions within Vigna angularis. A neighbor joining tree was generated using newly developed Innan's nucleotide diversity estimate from the AFLP data. From analysis, 7 distinct evolutionary groups, named as "Chinese cultivated", "Japanese cultivated", "Japanese complex-Korean cultivated", "Chinese wild", "China Taiwan wild", "Nepal-Bhutan cultivated" and "Hymalayan wild", were detected. Nucleotide diversity with geographical distribution of each group is discussed, regarding the evolutionary relationships between wild and cultivated adzuki beans. The preliminary results indicated that cultivated adzuki bean should be domesticated from at least 4 progenitors in at least 3 geographical origins.展开更多

Principal axis I (PCA1) accounted for 39% and 35% under insecticide spray and no insecticide spray respectively to the total variation in the accessions. Plant height with a factor score of 0.38, leaf length (0.41), number of leaves (0.37), and 100-seed, weight (0.30) was related to PCAI under insecticide spray while leaf width (0.32). Pod length (0.37) and number of seeds/plant (0.38) were significant to PCA1 under no insecticide spray. Notably, accessions such as SAMPEA6, SAMPEA10, IFE-Brown, and IFE-BPE exhibited consistent performance across both conditions, while others displayed condition-specific attributes. For instance, NGB1063, NGB1152, and NGB1093 demonstrated distinct traits under insecticide spray, while NGB1146 and NGB1124 exhibited notable characteristics under no insecticide spray conditions. Therefore, identifying these forty accessions with desirable traits hold promise for future genetic improvement efforts of cowpea cultivation in Nigeria and beyond.

文摘Soil salinity is one of the most important environmental factors limiting plant growth and productivity in many regions in the world. Salt stress directly affects plant photosynthesis, which is an important physiological process to limit plant growth and crop yield. But the effects of salt stress on the mechanism of the photosynthesis is still not clear. In this experiment, the salt tolerant plant Alhagi pseudalhagi was selected as the experiment material, and the salt sensitive plant Vigna radiata as the control, to explore the effects of salt stress on photosynthetic parameters of A. pseudalhagi. Plants were grown in a greenhouse,cultured with 1/2 Hoagland nutrient solution, treated by 0(control), 50, 100 or 200 s of mmol/L of NaCl solution for 12 d. Then,the chlorophyll contents, gas exchange parameters and chlorophyll a fluorescence in each treatment were measured. The results showed that under the salt stress simulated by 50 mmol/L NaCl, the net photosynthetic rate(Pn) and stomatal conductance(gs)of A. pseudalhagi were significantly increased compared with the control, while intercellular CO_2 concentration(Ci) was significantly decreased, Fv'/Fm', Fv/Fm and Fv/Fo were increased over time, but had no significant differences with the control ФPSⅡ,ETR and qP were significantly increased, and NPQ was significantly decreased. Under the salt stress simulated by 100 and 200mmol/L NaCl, Fv'/Fm', Fv/Fm, Fv/Fo, ФPSⅡ and qP of A. pseudalhagi were gradually decreased over time. In contrary, under the salt stress simulated by 50 and 100 mmol/L NaCl, the Pn, gs, Ci, Fv'/Fm', Fv/Fm, Fv/Fo, ФPSII, qP and ETR were all significantly decreased, while NPQ was significantly increased.
文摘About 80% of cowpea seeds are lost along with 30% of their weight after six months of unprotected storage due to attacks by Callosobruchus maculatus. This study aimed at evaluating the effect of applying bioinsecticides on the biological parameters of C. maculatus to reduce its damage to cowpea stocks. Four treatments including an untreated control and three doses of powdery extracts of C. nigricans, Z. zanthoxyloides and H. suaveolens, were compared in a Fischer block with four completely randomized repetitions. One hundred undamaged cowpea seeds were mixed with doses of each extract in Petri dishes and then fed as food substrates to ten pairs of C. maculatus for eight days. The extracts of H. suaveolens, C. nigricans and Z. zanthoxyloides all contain active ingredients including polyphenols, flavonoids, anthocyanins and alkaloids, etc. The extract of Z. zanthoxyloides was found to be more active on adults (100.00% mortality) followed by C. nigricans (98.75% mortality) and H. suaveolens (97.50% mortality) at 80 mg/g. Fecundity per female decreased significantly in the treated batches of 13.82 eggs for H. suaveolens at 10.12 eggs for C. nigricans at a dose of 80 mg/g. Germination rate of seeds was statistically raised to 100% at 60 and 80 mg/g by C. nigricans powder and lowered to 83.00% at 20 mg/g by Z. zanthoxyloides extracts. The application of Z. zanthoxyloides extract would be a less expensive alternative for the sustainable management of C. maculatus in cowpea stocks.
文摘Studies were conducted under pot conditions to determine the comparative efficacy ofcarbofuran at 1 mg a.i./kg soil, bavistin at 1 mg a.i./kg soil, neem (Azadirachta indica) seed powder at 50 mg/kg soil, green mould (Trichoderma harzianum) at 50.0 ml/kg soil, rhizobacteria (Pseudomonas fluorescens) at 50.0 ml/kg soil against root-knot nematode, Meloidogyne incognita-wilt fungus, Fusarium oxysporum disease complex on green gram, Vigna radiata cv ML-1108. All the treatments significantly improved the growth of the plants as compared carbofuran and A. indica seed powder increased plant growth to untreated inoculated plants. Analysis of data showed that and yield significantly more in comparison to bavistin and P. fluorescens. Carbofuran was highly effective against nematode, bavistin against fungus, A. indica seed powder against both the pathogens and both the bioagents were moderately effective against both the pathogens.
文摘This study describes variation of intron-3 of α-amylase gene from 156 breeds of adzuki beans using SSCP(single-strand conformation polymorphism)analysis. Based on α-amylase gene structure and sequence, A pair of PCR primers, F (CCTACATTCTAACACACCCT) and R (GCATATTGTGCCAGTACAAT) were designed to amplify intron-3 fragments of α-amylase gene. 14 variant types were detected, including 13, 9, 10, 4 variant types in the wild, weed, locally cultivated and modern brought-up adzuki beans respectively, 9, 8, 7 variant types of the wild adzuki beans from Japan, China and Korea respectively, and some other variant types in the local adzuki beans from China and Bhutan. 60% of subjects of cultivated races were found to be EE type in the experiment. In addition, sequence analysis of intron-3 of α-amylase gene from 8 variant types reveals the evolution process of various variant types in adzuki beans.
文摘Cowpea yield improvement is done by adding agricultural inputs. The use of natural substances as pesticides is being encouraged to fight against cowpea field pests. The pesticidal potentials of Azadirachta indica and Boswellia dalzielii water extracts, Metarhizium anisopliae, alone and in combination with plant extracts, and a commercial synthetic pesticide (Decis<sup>®</sup>) were tested in field on two varieties of cowpeas, Vigna unguiculata in two agroecological zones (Guinean Savanah and Sahelian Savanah) of Cameroon. The field trials were carried out in a full randomized block design including nine treatments and a control. Four replications were made concerning the different treatments and control. At the flowering stage, the cowpea field was sprayed three times with different pesticidal formulations at the interval of five days. The number of ramifications per plant, and that of pods per block and seed yield were determined. The pesticide formulations considerably (p ®</sup>). Bafia variety treated with the combination of M. anisopliae and A. indica recorded the highest ramification rate (37.03 ± 1.59) in Maroua (Sahelian Savanah). The same variety also produced more important pods number (90.50 ± 16.66) in Ngaoundere (Guinean Savanah) with the binary combination of two plants used in this experiment. The highest seed yield (44.23 ± 2.31) was recorded in Ngaoundere with B125 variety treated with the combination of the three pesticidal formulations separately (A. indica, B. dalzielii, M. anisopliae). The plant products used in this work, M. anisopliae and their mixtures could supersede the synthetic pesticides considering environmental issue in cowpea crop protection. Then, such formulations would not only improve crop productivity but also preserve environment from the pollution due to the use of synthetic residual chemicals.
文摘Cowpea [Vigna unguiculata (L.)] is one of the most important arid legumes cultivated for pulse and forage production. Drought is one of the most damageable constraints to crop production impacting negatively food security. The potential of cowpea to address food security is well established. However, not much is known about the base index selection method in breeding cowpea for drought tolerance, which is important for yields. Consequently, the present study has been conducted to: 1) evaluate the yield performance of cowpea genotypes under artificial drought and well-watered condition, 2) ranke genotype performance using selection indices, and 3) assess relationship between agronomic traits and yield. The experiment was the 2 watering conditions laid out in a Randomized Complete Block Design (RCBD) with three replications. The experiment was carried out in pots under screen house at the Department of Horticulture at KNUST. The result showed that KPR1-96-73, Simbo, CZ06-4-16, Wilibaly and Agyenkwa were high yielding in well-water condition while Ghana Shoba, Sangaraka, Nketewade, Ghana Shoni and Korobalen were high yielding genotypes in water stress condition. The average yield reduction was 60.6% and 16% for grain and fodder yield respectively. The biplot displays revealed four groups among the genotypes tested which were based on their yielding capacity and drought tolerance. In cluster B high yielding and drought tolerant genotypes were identified, high yielding and drought susceptible have been identified in cluster A, low yielding and drought tolerant in cluster D, and lastly low yielding and drought susceptible in cluster C. Genotypes in cluster B were best due to the fact that it combines high yield and tolerance to drought. They were Ghana Shoni, Nketewade, Sangaraka and Ghana Shoba. These genotypes might be suitably employed in further drought tolerance breeding programs of cowpea. Significant relationships were observed between agronomic trait and yields under drought condition.
文摘Bambara groundnut (Vigna subterranea (L.) Verdc.) has economic importance for the producers and the traders as cash crop since it generates substantial income for households in Niger. The objective of this study was to assess eight (8) Bambara groundnut morphotype from Université André Salifou de Zinder in Niger Republic by their morphological and agronomic characters. The experiment was conducted in a randomized complete block design with three replications during the rainy season of 2020-2021. Data were collected on the Number of leaves, Plant height (cm), Growth habit (cm), Terminal leaflet length, Terminal leaflet width, Number of nodes/stem, Number of stems/plant, Number of pods/plant, Weight of 100 pods (g), Weight of 100 seeds (g), Length of seeds (mm), Width of seeds (mm), Weight of seeds/Plot (kg). There were significant differences for all characters, except Number of leaves, Terminal leaflet length, Terminal leaflet width, Length of seeds (mm) Weight of 100 pods (g), Weight of 100 seeds (g). The morphotypes UZ-VZ-04, UZ-VZ-03, UZ-VZ-06, UZ-VZ-02 and UZ-VZ-05 have demonstrated good performance for grain weight per plot and can be useful for a breeding program.

文摘Sixteen mungbean lines were analyzed using 56 random primers. Different DNA bands were detected between Bruchid resistant lines and susceptible lines. According to the cluster results, the 16 lines can be divided into four groups, including brucid resistant wild types, resistant cultivated lines, resistant progenies and a mixed group. BSA method was used to identify DNA markers that related with bruchid resistant gene by using resistant line and susceptible line and their F2 progeny. One codominant marker was identified, which generated a fragment of 1.79 kb in resistant lines and 1.03 kb in susceptible lines. Finally, this codominant marker was considered to be tightly linked with bruchid resistant gene and could be useful in resistant germplasm identification and marker-assisted selection.

文摘Genetic resources from other countries or regions play an important role in broadening the genetic background of local breeding varieties. Here we describe observations on the adaptability of mung bean germplasm obtained from the United States Department of Agriculture and their genetic diversity assessment using SSR markers. Several accessions were shown to be mixtures,based on their phenotypes for some characters. Most accessions were able to complete their lifecycles when grown in Beijing, China, making them ideal for crossbreeding without day length control. High diversity was revealed by the SSR markers, with an average of 4.2 alleles per locus and a PIC value of 0.650 per locus. STRUCTURE analysis divided the accessions into six groups.There was no obvious trend of accessions forming groups according to their geographical origin,owing mainly to germplasm exchange and an uneven distribution of accessions. The present results indicate that this germplasm would enrich the local gene pool, and provide information for the further use of germplasm in breeding programs.

文摘Intercropping maize with the prostrate variety of mungbean, Vigna radiata (L.) Wilczek which grows flat along the ground can increase parasitism of the eggs of the Asian corn borer, Ostrinia furnacalis (Guenée), a crucial insect pest of maize in China, by Trichogramma ostriniae Pang Chen. This study investigated the behavioural responses of T. ostriniae to plant volatiles emitted from whole plants, leaves, and flowers of prostrate and erect varieties of mungbean in a 4-armed olfactometer. Female parasitiods were significantly attracted to the odours from some varieties of prostrate mungbean when the visit duration and the number of visits were considered, but were not attracted to the odours from any erect varieties. The attractive odours were shown to emanate from the leaves, rather than from the flowers of prostrate mungbean plants. These results have implications for the augmentation of Trichogramma parasitoids in bio-control of O. furnacalis and other crop pests.
文摘Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family Fabaceae.It is a close relative of cultivated Vigna species such as adzuki bean(V.angularis),cowpea(V.unguiculata),mung bean(V.radiata),and blackgram(V.mungo),and is distributed throughout the tropics.With its ability to tolerate salt stress,beach pea has great potential to contribute salt-tolerance genes for developing salt-tolerant cultivars in cultivated Vigna species.However,it is still underutilized in Vigna breeding programs.A draft genome sequence of beach pea was generated using a high-throughput next-generation sequencing platform,yielding 23.7 Gb of sequence from 79,929,868 filtered reads.A de novo genome assembly containing 68,731 scaffolds gave an N50 length of 10,272 bp and the assembled sequences totaled 365.6 Mb.A total of 35,448 SSRs,including 3574 compound SSRs,were identified and primer pairs for most of these SSRs were designed.Genome analysis identified 50,670 genes with mean coding sequence length 1042 bp.Phylogenetic analysis revealed highest sequence similarity with V.angularis,followed by V.radiata.Comparison with the V.angularis genome revealed 16,699 SNPs and 2253 InDels and comparison with the V.radiata genome revealed 17,538 SNPs and 2300 InDels.To our knowledge this is the first draft genome sequence of beach pea derived from an accession(ANBp-14-03)adapted locally in the Andaman and Nicobar Islands of India.The draft genome sequence may facilitate the genetic enhancement in cultivated Vigna species.
文摘A set of representative 146 adzuki (Vigna angular is var. angularis, and var. nipponensis) germplasm from 6 Asian countries traditionally for adzuki bean production, together with an out group standard rice bean (Vigna umbellata), were analyzed by AFLP methodology using 12 informative primer pairs. 313 unambiguous polymorphic bands were created. According to the dendrogram by cluster analysis based on AFLP banding, 143 of the accessions were distinct and revealed enough genetic diversity for identification and classification of accessions within Vigna angularis. A neighbor joining tree was generated using newly developed Innan's nucleotide diversity estimate from the AFLP data. From analysis, 7 distinct evolutionary groups, named as "Chinese cultivated", "Japanese cultivated", "Japanese complex-Korean cultivated", "Chinese wild", "China Taiwan wild", "Nepal-Bhutan cultivated" and "Hymalayan wild", were detected. Nucleotide diversity with geographical distribution of each group is discussed, regarding the evolutionary relationships between wild and cultivated adzuki beans. The preliminary results indicated that cultivated adzuki bean should be domesticated from at least 4 progenitors in at least 3 geographical origins.
