The current availability of genotypes for a very large number of single n...Background: Runs of homozygosity(ROHs) are homozygous segments of the genome where the two haplotypes inherited from the parents are identical. The current availability of genotypes for a very large number of single nucleotide polymorphisms(SNPs) is leading to more accurate characterization of ROHs in the whole genome. Here,we investigated the occurrence and distribution of ROHs in 3,692 Large White pigs and compared estimates of inbreeding coefficients calculated based on ROHs(FROH), homozygosity(FHOM), genomic relationship matrix(FGRM)and pedigree(FPED). Furthermore, we identified genomic regions with high ROH frequencies and annotated their candidate genes.Results: In total, 176,182 ROHs were identified from 3,569 animals, and all individuals displayed at least one ROH longer than 1 Mb. The ROHs identified were unevenly distributed on the autosomes. The highest and lowest coverages of Sus scrofa chromosomes(SSC) by ROH were on SSC14 and SSC13, respectively. The highest pairwise correlation among the different inbreeding coefficient estimates was 0.95 between FROH_totaland FHOM, while the lowest was-0.083 between FGRMand FPED. The correlations between FPEDand FROHusing four classes of ROH lengths ranged from 0.18 to 0.37 and increased with increasing ROH length, except for ROH > 10 Mb. Twelve ROH islands were located on four chromosomes(SSC1, 4, 6 and 14). These ROH islands harboured genes associated with reproduction, muscular development, fat deposition and adaptation, such as SIRT1, MYPN, SETDB1 and PSMD4.Conclusion: FROHcan be used to accurately assess individual inbreeding levels compared to other inbreeding coefficient estimators. In the absence of pedigree records, FROHcan provide an alternative to inbreeding estimates.Our findings can be used not only to effectively increase the response to selection by appropriately managing the rate of inbreeding and minimizing the negative effects of inbreeding depression but also to help detect genomic regions with an effect on traits under selection.展开更多

The model was verified by the data of the reach 25km upstream the Jialingjia ng estuary and the 44km long main stream of the Chongqing reach of the Yangtze r iver. To investigate the impact of overexpression of heterologous SUS on the growth and developm...In plants, sucrose synthase (SUS) enzymes catalyze conversion of sucrose into fructose and UDP-glucose in the presence of UDP. To investigate the impact of overexpression of heterologous SUS on the growth and development of Arabidopsis, we transformed Arabidopsis plants with an overexpression vector containing an aspen SUS gene (PtrSUS1). The genomic PCR confirmed the successful integration of PtrSUS1 transgene in the Arabidopsis genome. PtrSUS1 expression in transgenic Arabidopsis plants was confirmed by RT-PCR. The SUS activity was dramatically increased in all transgenic lines examined. The three selected transgenic PtrSUS1 lines exhibited faster growth and flowered about 10 days earlier compared to untransformed controls, and also possessed 133%, 139%, and 143% SUS activity compared to controls. Both fresh weights and dry biomass yields of the whole plants from these three selected transgenic lines were significantly increased to 125% of the controls. Transgenic PtrSUS1 lines also had a higher tolerance to higher concentration of sucrose which was reflective of the increased SUS activity in transgenic versus wild-type plants. The growth differences between wild-type and transgenic plants, either in root and hypocotyl length or in fresh and dry weight of whole plant, became more pronounced on the media containing higher sucrose concentrations. Taken together, these results showed that the early flowering, faster growth and increased tolerance to higher sucrose in transgenic lines were caused by the genome integration and constitutive expression of the aspen PtrSUS1 gene in transgenic Arabidopsis.展开更多

Based on the WShler curve of smooth materials, a modified method for fatigue life prediction is approached. The characteristic fatigue length of material under cyclic loading is associated with the polycrystalline material. Rather than the stress at a point, the average stress within the characteristic fatigue length is implemented for the fatigue life prediction. The method can be applied to both the smooth and the defected material. The fatigue life prediction is also verified experimentally by specimens with various small circular holes. Through the comparison, it is found that the method can be adopted to predict the fatigue lives with different size effects.展开更多

Fur- thermore, after soils softening, seedling slinging or direct seeding was adopted, which is dominated by natural rainfall and supplemented by artificial irrigation. The test proved that the technology help increasing yield and profits over 20%, with chemical fertilizer reduced by more than 10%, and labor cost reduced by 10%. What's more, if the technology applied once, no-tillage or slight tillage is recom- mended in the field, which would maintain original eco-conditions of soils and reach high yield, with energy, water, labor costs reduced in an environment-friendly way.展开更多
Distance traveled and home range size describe how animals move in space.The seasonal variations of these parameters are important to comprehensively understand animal ecology and its connection with reproductive beha...Distance traveled and home range size describe how animals move in space.The seasonal variations of these parameters are important to comprehensively understand animal ecology and its connection with reproductive behavior and energy costs.Researchers usually estimate the distance traveled as the sum of the straight-line displacements between sampled positions,but this approach is sensitive to the sampling frequency and does not account for the tortuosity of the animal’s movements.By means of the continuous-time movement modeling which takes into account autocorrelation and tortuosity of movement data,we estimated the distance traveled and monthly home range size of 28 wild boar Sus scrofa and modeled their inter-sexual seasonal variability.Males traveled longer distances and used larger home ranges than females,particularly during the rut in autumn-winter,consistently with the different biological cycles of males and females.Males enlarged their home ranges during the rut but traveled constant average distances along the year,whereas females traveled shorter distances in correspondence with the peak of food resources and birth periods but exhibited constant home range size across seasons.The differences between the seasonal variation patterns of distance traveled and home range size,observed in both sexes,revealed the complex relationship between these two aspects of spatial behavior and the great opportunity of including both distance traveled and home range size in behavioral ecology investigations.We provided a detailed analysis of wild boar spatial behavior and its relationships with the reproductive cycles of males and females,promoting a deeper comprehension of their behavioral ecology.展开更多
