In order to investigate the differential expression of BLM protein in hematopoietic tumor cell strains and study the effects of BLM gene on ultraviolet (UV)- or hydroxyurea (HU)-induced apoptosis, Western blot was used to detect the expression of BLM protein in normal human bone marrow mononuclear cells and 4 kinds of hematopoietic tumor cell strains. The 4 kinds of hematopoietic tumor cells were exposed to UV light with a germicidal UV lamp or treated with 2 mmol/L hydroxyurea and the apoptotic rate was detected by using AnnexinV-FITC. The results showed that these tumor cells expressed BLM protein higher than the normal human bone marrow mononuclear cells (P〈0.01). In the 4 hematopoietic tumor cells, BLM protein was all specially cleaved in response to UV- or HU-induced apoptosis. The increase of BLM protein expression may play an important role in the development of these tumors, and BLM proteolysis is likely to be a general feature of the apoptotic response.展开更多
Protons(H^+)as well as different major and trace elements may inhibit cadmium(Cd)uptake in aquatic organisms and thus alleviate Cd toxicity.However,little is known about such interactions in soil organisms.In thi...Protons(H^+)as well as different major and trace elements may inhibit cadmium(Cd)uptake in aquatic organisms and thus alleviate Cd toxicity.However,little is known about such interactions in soil organisms.In this study,the independent effects of the cations calcium(Ca^2+),magnesium(Mg^2+),potassium(K^+),H^+ and zinc(Zn^2+)on Cd toxicity were investigated with 5-day long barley root elongation tests in nutrient solutions.The tested concentrations of selected cations and trace metal ions were based on the ranges that occur naturally in soil pore water.The toxicity of Cd decreased with increasing activity of Ca^2+,Mg^2+,H^+and Zn^2+,but not K^+.Accordingly,conditional binding constants were obtained for the binding of Cd^2+,Ca^2+,Mg^2+,H^+,and Zn^2+ with the binding ligand:log K(CdBL)5.19,logK(CaBL)2.87,logK(MgBL)2.98,logK(HBL)5.13 and logK(ZnBL)5.42,respectively.Furthermore,it was calculated that on average 29% of the biotic ligand sites needed to be occupied by Cd to induce a 50% decrease in root elongation.Using the estimated constants,a biotic ligand model was successfully developed to predict the Cd toxicity to barley root elongation as a function of solution characteristics.The feasibility and accuracy of its application for predicting Cd toxicity in soils were discussed.展开更多

The direct and indirect dissolved methods result in irreversible and revers...In the TCNQ-modified BLM, the voltammetric response is different due to the dif-ferent methods used to prepare the membrane forming solution. The direct and indirect dissolved methods result in irreversible and reversible responses respectively. These results can be explained by the different styles of the orientation of TCNQ in the membrane. The reversible response is controlled by the diffusion of electroactive species in the interior of the membrane. When MB is used to modify the BLM, very complex voltammograms are obtained. The intersection of the voltammetric curves can be regarded to be owing to the appearance of new phase in the membrane caused by MB. But it disappears at lower scan rate. Peak current increases with decreasing scan rate. This indicates that the resistance of the membrane at lower scan rate is lower than that at higher scan rate. Asymmetric curve of MB incorporated BLM is ascribed to the different rates of redox reaction at the two membrane/solution interfaces.展开更多
