Grapevine is one of the most economically important crops worldwide.However,the previous versions of the grapevine reference genome tipically consist of thousands of fragments with missing centromeres and telomeres,li...Grapevine is one of the most economically important crops worldwide.However,the previous versions of the grapevine reference genome tipically consist of thousands of fragments with missing centromeres and telomeres,limiting the accessibility of the repetitive sequences,the centromeric and telomeric regions,and the study of inheritance of important agronomic traits in these regions.Here,we assembled a telomere-to-telomere(T2T)gap-free reference genome for the cultivar PN40024 using PacBio HiFi long reads.The T2T reference genome(PN_T2T)is 69 Mb longer with 9018 more genes identified than the 12X.v0 version.We annotated 67%repetitive sequences,19 centromeres and 36 telomeres,and incorporated gene annotations of previous versions into the PN_T2T assembly.We detected a total of 377 gene clusters,which showed associations with complex traits,such as aroma and disease resistance.Even though PN40024 derives from nine generations of selfing,we still found nine genomic hotspots of heterozygous sites associated with biological processes,such as the oxidation–reduction process and protein phosphorylation.The fully annotated complete reference genome therefore constitutes an important resource for grapevine genetic studies and breeding programs.展开更多

These complexation behaviors have been indicated by proton nuclear magnetic resonance(~1H NMR) spectroscopy and isothermal titration calorimetry(ITC),and have been confirmed by X-ray single crystal structural analysis of Q[8]_(3)/G2_(2). While free guests G1–G3 have no ability to sensitize singlet oxygen, they all become effective singlet oxygen sensitizers when complexed with Q[8]. By irradiation of white light, Q[8]_(3)/G_(2) complexes induced selective oxidation of aryl sulfide to aryl sulfoxide in good yields and selective oxidation of dimethyl sulfide(DMS) to dimethyl sulfoxide(DMSO) in excellent yields without over-oxidation to dimethyl sulfone(DMSO_(2))even for elongated irradiation. High-yield photo-oxidation of DMS to DMSO_(2) was only achieved by irradiation of blue light in the presence of Q[8]_(3)/G1_(2), which was also effective in photobleaching of Rhodamine B in water. OpenStreetMap has a large number of volunteers.There is a hypothesis that volunteers with different cultural backgrounds may have different editing behaviors when contributing to OSM.It may be strongly related to data...OpenStreetMap has a large number of volunteers.There is a hypothesis that volunteers with different cultural backgrounds may have different editing behaviors when contributing to OSM.It may be strongly related to data quality and data reliability on OSM.As for the heterogeneity and the reliability of OSM data,previous research usually focuses on the geometric accuracy,spatial location accuracy and semantic integrity of OSM data,while few researchers have analyzed these problems from the perspective of editing behavior.On the grounds of relationship between mapping motivation and editing behavior,the dispersion of editing trajectory and clockwise direction index are proposed in the paper to explore whether the volunteers are sufficiently motivated and knowledgeable.In the experiments,the historical OSM data of four countries suggested that developed countries have lower trajectory dispersion.The lower degree of trajectory dispersion reflects the higher concentration and professionalism of volunteers.A high degree of drawing direction consistency shows volunteers who mapped French data were natives with local knowledge.From this point of view,this paper verifies that volunteer editing behavior is an effective method to analyze data quality heterogeneity and data reliability.展开更多
Ectopic accumulation of fat can cause a variety of metabolic diseases,and the emerging non-alcoholic fatty pancreas disease(NAFPD)is increasingly being recognized by clinicians as a cause for concern.NAFPD is a diseas...Ectopic accumulation of fat can cause a variety of metabolic diseases,and the emerging non-alcoholic fatty pancreas disease(NAFPD)is increasingly being recognized by clinicians as a cause for concern.NAFPD is a disease caused by abnormal accumulation of adipose tissue in the pancreas,which is related to obesity.The main feature of NAFPD is death of acinar cells,which are then replaced by adipose cells.However,the underlying molecular mechanisms have not been fully explored.Obesity,aging,and metabolic syndrome are independent risk factors for the occurrence and development of NAFPD.Studies have shown that NAFPD leads to insulin resistance and pancreatic dysfunction,increases the risk of diabetes mellitus,worsens the severity of pancreatitis,and is significantly correlated with pancreatic cancer and postoperative pancreatic fistula.There is no standard treatment for NAFPD;exercise,a balanced diet,and lifestyle can help reduce pancreatic fat;however,other treatment modalities such as drugs and bariatric surgery are still being explored.The specific pathological mechanism of NAFPD remains unclear,and its potential association with various clinical diseases requires further study.This review summarizes the etiology,diagnosis,clinical consequences,and potential therapeutic strategies of NAFPD.展开更多

Neutrophil extracellular traps(NETs)can capture and kill viruses,such as influenza viruses,human immunodeficiency virus(HIV),and respiratory syncytial virus(RSV),thus contributing to host defense.Contrary to our expec...Neutrophil extracellular traps(NETs)can capture and kill viruses,such as influenza viruses,human immunodeficiency virus(HIV),and respiratory syncytial virus(RSV),thus contributing to host defense.Contrary to our expectation,we show here that the histones released by NETosis enhance the infectivity of SARS-CoV-2,as found by using live SARS-CoV-2 and two pseudovirus systems as well as a mouse model.The histone H3 or H4 selectively binds to subunit 2 of the spike(S)protein,as shown by a biochemical binding assay,surface plasmon resonance and binding energy calculation as well as the construction of a mutant S protein by replacing four acidic amino acids.Sialic acid on the host cell surface is the key molecule to which histones bridge subunit 2 of the S protein.Moreover,histones enhance cell-cell fusion.Finally,treatment with an inhibitor of NETosis,histone H3 or H4,or sialic acid notably affected the levels of sgRNA copies and the number of apoptotic cells in a mouse model.These findings suggest that SARS-CoV-2 could hijack histones from neutrophil NETosis to promote its host cell attachment and entry process and may be important in exploring pathogenesis and possible strategies to develop new effective therapies for COVID-19.展开更多
SMARCA4-deficient tumors are gaining attention,and this type of tumor is highly malignant with a poor prognosis.The frequency of SMARCA4 deletion varies in different tumors.However,it has never been identified in the ...SMARCA4-deficient tumors are gaining attention,and this type of tumor is highly malignant with a poor prognosis.The frequency of SMARCA4 deletion varies in different tumors.However,it has never been identified in the pancreas.A 67-year-old woman pre-sented with upper abdominal pain lasting for half a month.Computed tomography indicated a space-occupying lesion in the head of the pancreas,considered a malignant tumor.Further Endoscopic ultrasound fine-needle aspiration was performed and report-ed as SMARCA4-deficient undifferentiated pancreatic tumor.The patient received no treatment and survived only two and a half months from diagnosis to death.We are the first to report SMARCA4-deficient pancreatic undifferentiated tumor in which this new tumor entity presents with poorer overall survival.We also summarized the advances of SMARCA4 in oncology and treatment.展开更多
Analysis of urban spatial structures is an effective way to explain and solve increasingly serious urban problems.However,many of the existing methods are limited because of data quality and availability,and usually y...Analysis of urban spatial structures is an effective way to explain and solve increasingly serious urban problems.However,many of the existing methods are limited because of data quality and availability,and usually yield inaccurate results due to the unclear description of urban social functions.In this paper,we present an investigation on urban social function based spatial structure analysis using building footprint data.An improved turning function(TF)algorithm and a selforganizing clustering method are presented to generate the variable area units(VAUs)of high-homogeneity from building footprints as the basic research units.Based on the generated VAUs,five spatial metrics are then developed for measuring the morphological characteristics and the spatial distribution patterns of buildings in an urban block.Within these spatial metrics,three models are formulated for calculating the social function likelihoods of each urban block to describe mixed social functions in an urban block,quantitatively.Consequently,the urban structures can be clearly observed by an analysis of the spatial distribution patterns,the development trends,and the hierarchy of different social functions.The results of a case study conducted for Munich validate the effectiveness of the proposed method.展开更多
Autoimmune pancreatitis(AIP)is a rare and emerging immune-mediated pancreatitis.The involvement of the pancreas with lymph-oplasmacytic infiltration and fibrosis on histology is the most common features and has been w...Autoimmune pancreatitis(AIP)is a rare and emerging immune-mediated pancreatitis.The involvement of the pancreas with lymph-oplasmacytic infiltration and fibrosis on histology is the most common features and has been well studied in the literature.It is characterized by a nonspecific presentation that mimics a malignant process.The purpose of this report is to explore the different clinical and paraclinical aspects of this disease,as well as the challenges posed by its management.The series are made up of three different cases of patients with elevated serum IgG4 levels and pancreatic changes.The first case initially revealed cholestatic jaundice with extrahepatic and intrahepatic cholangitis,and ultrasound reflected multiple occupying changes in the pancreas.The elevated serum IgG4 level was brilliant.Patient improved under the influence of corticosteroids.The second case was clinically asymptomatic,just found weight loss.Magnetic resonance revealed a round mass in the head of the pancreas and was considered as a neuroendocrine tumor.The patient showed slightly elevated serum IgG4 level,but was sensitive to hormone therapy.The third case concerned epigastric pain and enlarged lymph nodes.It was initially diagnosed as pancreatic cancer with distant metastasis,but also finally resolved by corticosteroid treatment.The clinical presentation of AIP is heterogeneous,as evidenced by our three clinical cases.Further observational and interventional studies are needed to better identify and manage this disease.展开更多
Background and objective:The incidence of pancreatic neuroendocrine neoplasms(pNENs)has been increasing year by year,and pancreatic steatosis has been paid more and more attention to by clinicians.Our study aims to re...Background and objective:The incidence of pancreatic neuroendocrine neoplasms(pNENs)has been increasing year by year,and pancreatic steatosis has been paid more and more attention to by clinicians.Our study aims to reveal the correlation between pancreatic steatosis,clinicopathological parameters,and the prognosis of patients with pNENs.Methods:The clinicopathological data of patients with pNENs who underwent surgical resections in our institution from January 2013 to May 2022 were retrospectively analyzed.The mean computed tomography(CT)values of the pancreas and spleen were used to assess the severity of the pancreatic steatosis.Pearson Chi-square test,T test,Mann Whitney U test,Univariate Cox re-gression,and Multivariate Cox regression were used to explore the relationship between pancreatic steatosis and age,sex,body mass index,tumor type,tumor location,tumor stage,blood lipid,prognosis,and other factors.Results:A total of 122 patients were included in our study,and 19.67%have pancreatic steatosis.The incidence of pancreatic steatosis was significantly higher in patients with fatty liver than in patients without fatty liver(36.8%vs 16.5%,P=.04).In insu-linoma,the incidence of pancreatic steatosis in elderly patients was significantly higher than in young and middle-aged patients(41.7%vs 9.3%,P=.025).There was no significant difference in lipid levels between the pancreatic steatosis group and the non-pancreatic steatosis group(P>.05).Kaplan-Meier curves show that the prognosis of stage III/IV patients was significantly worse than that of stage I/II patients(P<.001).However,there was no significant difference in prognosis between patients with and without pancreatic steatosis(P=.404).Conclusion:The incidence of pancreatic steatosis was significantly higher in those with fatty liver than those without fatty liver.There is a high incidence of pancreatic steatosis in elderly patients with insulinoma.American Joint Committee on Cancer(AJCC)stage,but not pancreatic steatosis,significantly affected the prognosis of 展开更多

文摘SMARCA4-deficient tumors are gaining attention,and this type of tumor is highly malignant with a poor prognosis.The frequency of SMARCA4 deletion varies in different tumors.However,it has never been identified in the pancreas.A 67-year-old woman pre-sented with upper abdominal pain lasting for half a month.Computed tomography indicated a space-occupying lesion in the head of the pancreas,considered a malignant tumor.Further Endoscopic ultrasound fine-needle aspiration was performed and report-ed as SMARCA4-deficient undifferentiated pancreatic tumor.The patient received no treatment and survived only two and a half months from diagnosis to death.We are the first to report SMARCA4-deficient pancreatic undifferentiated tumor in which this new tumor entity presents with poorer overall survival.We also summarized the advances of SMARCA4 in oncology and treatment.

文摘Analysis of urban spatial structures is an effective way to explain and solve increasingly serious urban problems.However,many of the existing methods are limited because of data quality and availability,and usually yield inaccurate results due to the unclear description of urban social functions.In this paper,we present an investigation on urban social function based spatial structure analysis using building footprint data.An improved turning function(TF)algorithm and a selforganizing clustering method are presented to generate the variable area units(VAUs)of high-homogeneity from building footprints as the basic research units.Based on the generated VAUs,five spatial metrics are then developed for measuring the morphological characteristics and the spatial distribution patterns of buildings in an urban block.Within these spatial metrics,three models are formulated for calculating the social function likelihoods of each urban block to describe mixed social functions in an urban block,quantitatively.Consequently,the urban structures can be clearly observed by an analysis of the spatial distribution patterns,the development trends,and the hierarchy of different social functions.The results of a case study conducted for Munich validate the effectiveness of the proposed method.

文摘Autoimmune pancreatitis(AIP)is a rare and emerging immune-mediated pancreatitis.The involvement of the pancreas with lymph-oplasmacytic infiltration and fibrosis on histology is the most common features and has been well studied in the literature.It is characterized by a nonspecific presentation that mimics a malignant process.The purpose of this report is to explore the different clinical and paraclinical aspects of this disease,as well as the challenges posed by its management.The series are made up of three different cases of patients with elevated serum IgG4 levels and pancreatic changes.The first case initially revealed cholestatic jaundice with extrahepatic and intrahepatic cholangitis,and ultrasound reflected multiple occupying changes in the pancreas.The elevated serum IgG4 level was brilliant.Patient improved under the influence of corticosteroids.The second case was clinically asymptomatic,just found weight loss.Magnetic resonance revealed a round mass in the head of the pancreas and was considered as a neuroendocrine tumor.The patient showed slightly elevated serum IgG4 level,but was sensitive to hormone therapy.The third case concerned epigastric pain and enlarged lymph nodes.It was initially diagnosed as pancreatic cancer with distant metastasis,but also finally resolved by corticosteroid treatment.The clinical presentation of AIP is heterogeneous,as evidenced by our three clinical cases.Further observational and interventional studies are needed to better identify and manage this disease.

文摘Background and objective:The incidence of pancreatic neuroendocrine neoplasms(pNENs)has been increasing year by year,and pancreatic steatosis has been paid more and more attention to by clinicians.Our study aims to reveal the correlation between pancreatic steatosis,clinicopathological parameters,and the prognosis of patients with pNENs.Methods:The clinicopathological data of patients with pNENs who underwent surgical resections in our institution from January 2013 to May 2022 were retrospectively analyzed.The mean computed tomography(CT)values of the pancreas and spleen were used to assess the severity of the pancreatic steatosis.Pearson Chi-square test,T test,Mann Whitney U test,Univariate Cox re-gression,and Multivariate Cox regression were used to explore the relationship between pancreatic steatosis and age,sex,body mass index,tumor type,tumor location,tumor stage,blood lipid,prognosis,and other factors.Results:A total of 122 patients were included in our study,and 19.67%have pancreatic steatosis.The incidence of pancreatic steatosis was significantly higher in patients with fatty liver than in patients without fatty liver(36.8%vs 16.5%,P=.04).In insu-linoma,the incidence of pancreatic steatosis in elderly patients was significantly higher than in young and middle-aged patients(41.7%vs 9.3%,P=.025).There was no significant difference in lipid levels between the pancreatic steatosis group and the non-pancreatic steatosis group(P>.05).Kaplan-Meier curves show that the prognosis of stage III/IV patients was significantly worse than that of stage I/II patients(P<.001).However,there was no significant difference in prognosis between patients with and without pancreatic steatosis(P=.404).Conclusion:The incidence of pancreatic steatosis was significantly higher in those with fatty liver than those without fatty liver.There is a high incidence of pancreatic steatosis in elderly patients with insulinoma.American Joint Committee on Cancer(AJCC)stage,but not pancreatic steatosis,significantly affected the prognosis of
