However, most CRISPR/Cas9 systems reported for plants can only modify one or a few target sites. Here, we report a robust CRISPR/Cas9 vector system, utilizing a plant codon optimized Cas9 gene, for convenient and high- efficiency multiplex genome editing in monocot and dicot plants. We designed PCR-based procedures to rapidly generate multiple sgRNA expression cassettes, which can be assembled into the binary CRISPR/ Cas9 vectors in one round of cloning by Golden Gate ligation or Gibson Assembly. With this system, we edi- ted 46 target sites in rice with an average 85.4% rate of mutation, mostly in biallelic and homozygous status. We reasoned that about 16% of the homozygous mutations in rice were generated through the non-homol- ogous end-joining mechanism followed by homologous recombination-based repair. We also obtained uni- form biallelic, heterozygous, homozygous, and chimeric mutations in Arabidopsis T1 plants. The targeted mutations in both rice and Arabidopsis were heritable. We provide examples of loss-of-function gene mu- tations in To rice and T1Arabidopsis plants by simultaneous targeting of multiple (up to eight) members of a gene family, multiple genes in a biosynthetic pathway, or multiple sites in a single gene. This system has provided a versatile toolbox for studying functions of multiple genes and gene families in plants for basic research and genetic improvement. 展开更多

The strongly peraluminous granites (SPGs) of Eastern Nanling Range (ENR) are a characteristic of all bearing highly aluminous minerals, such as muscovite±AI-rich biotite± tourmaline±garnet, and lack of ... The strongly peraluminous granites (SPGs) of Eastern Nanling Range (ENR) are a characteristic of all bearing highly aluminous minerals, such as muscovite±AI-rich biotite± tourmaline±garnet, and lack of cordierite. In respect of petrography, geochemistry, Nd isotope, and single grain zircon U-Pb dating, the representative granite bodies of them are studied. The research shows that these granites were emplaced in two stages, namely 228-225 Ma BP and J2-3 159-156 Ma BP, belonging to Indosinian and early Yanshanian periods, respectively, and they have low εNd(t) values (-10.6--11.1), high A/CNK, Rb/Sr ratios and tDM values (1887-1817 Ma), and REE's tetrad effect (TE1,3=1.13-1.34). In comparison with related geology, petrology and chronology of granites in adjacent regions, it is suggested that Indosinian SPGs of ENR formed in the circumstance of post-collisional extension 20 Ma after the major collision of Indosinian Movement (258-243 Ma BP) in Indo-China Peninsula, and early Yanshanian SPGs formed in the background of back-arc extension setting controlled by paleo-Pacific tectonic domain, and J1, the interval of two stages, is the interim from Tethyan to Pacific tectonic domains in South China. These SPGs have similar geological and geochemical characteristics, because they all crystallized from the magma of partial melting of early Proterozoic metasedimentary rocks when the thickened crust (≤50 km) became thinning, decompression, and transmitting of water. 展开更多

Structural health monitoring (SHM) is a multi-discipline field that involves the automatic sensing of structural loads and response by means of a large number of sensors and instruments, followed by a diagnosis of the structural health based on the collected data. Because an SHM system implemented into a structure automatically senses, evaluates, and warns about structural conditions in real time, massive data are a significant feature of SHM. The techniques related to massive data are referred to as data science and engineering, and include acquisition techniques, transition techniques, management techniques, and processing and mining algorithms for massive data. This paper provides a brief review of the state of the art of data science and engineering in SHM as investigated by these authors, and covers the compressive sampling-based data-acquisition algorithm, the anomaly data diagnosis approach using a deep learning algorithm, crack identification approaches using computer vision techniques, and condition assessment approaches for bridges using machine learning algorithms. The Hujierte-Chaganhadamiao volcanic-plutonic sequence is located in the Mandula area of northen Damaoqi, Inner Mongo- lia. It contains mainly mafic volcanics, with several ultramafic blocks in its central part and mafic- ntermediate intrusions in the east. The zircon U-Pb ages of the gabbro and basalt samples are 278.5±3.0 Ma (MSWD=).66) and 273.7±1.0 Ma (MSWD=0.36), respectively. These ages constrain the magmatism occurred in the Early Permian. The Mandula Permian mafic rocks are characterized by low abundances of REE and slightly LREE-enriched chondrite-normal zed N-MORB-type REE patterns. And these mafic samples have high (143Nd/144Nd)i (0.51262-0.51270), relatively high pos tive εNd(t) (3.4-8.0), and high Mg# (49-54), suggesting derivation from the depleted asthenosphere mantle source. But they al, show the enrichment in large ion lithophile elements (LILE, e.g. Rb, Ba and Sr), depletion in high field strength elements (I-FSE); distinctly negative Nb, Ta anomalies and slightly negative P, Ti anomalies; high (875r/86Sr)i (0.70490-0.70537), low (206pb/204pb)i (17.39-17.93). All of these resemble the characteristics of the enriched mantle source or involvement of arc materials in the magma genesis. Furthermore, as shown in the correlation plots of eNd(t) VS (87Sr/86Sr)i, (206pb/204Pb)ivs (207pb/204Pb)i and (208pb/204pb)i, La/Ba La/Nb vs La/Nb, and La/Nb ratio, the magma source could have been experienced the contamination and metasomation from the previous subduction process. And the positive correlation between the selected major oxides and trace elements could be explained by the contamination from continental crust and arc materials during the magmatic emplacement. Combined with the distinct geochemistry features of Mandula mafic samples and many previous researches in the study area, the Early Permian magmatism in Mandula occurred under an extensional rift setting, and a juvenile ocean basin probably had already been formed after 展开更多

Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,"Huang Qin," are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. The reference genome of S. baicalensis is also useful for improving the genome assemblies for other members of the mint family and offers an important foundation for decoding the synthetic pathways of bioactive compounds in medicinal plants. 展开更多

Granitoid gneisses are widespread in Precambrian metamorphic blocks of eastern segment of the Central Tianshan Tectonic Zone, and they have intrusive contact relationships with their metamorphic sedimentary country rocks of Proterozoic Xingxingxia and Kawabulag groups. Zircon U-Pb ages from a granodioritic gneiss (IW11-1) and a parametamorphic schist (W05-9) are determined at the Weiya area. Euhedral prismatic zircons from the granodioritic gneiss (IW11-1) provide U-Pb isotopic discordia intercept ages of 1218±17 Ma and 426±26 Ma, respectively, and euhedral prismatic zircons from the parametamorphic schist (W05-9) display U-Pb isotopic discordia intercept ages of 1216±74 Ma and 290±15 Ma, respectively. A whole-rock Sm-Nd isotopic isochron is determined in augen granitoid gneiss samples at the Gang- gou-Kumishi area and we obtain the isochron age of 1142±120 Ma, and its ε Nd (t) = -4.3. These geochronological data suggest that these Precambrian metamorphic basement blocks within eastern segment of the Central Tianshan Tectonic Zone can be produced during 1140—1220 Ma, and occur a nearly homochronous metamorphism. Integrated to these geochronological data, Nd depleted mantle model ages (T DM ) and epsilon Nd(t) values of these granitoid gneiss samples indicate that they can derive from mixing in various scales both magmas from mantle and crust sources at a late Mesoproterozoic active continental margin tectonic environment. Similarity in geochronology, Sm-Nd isotopic geochemistry between Weiya-Xingxingxia, Pargangtag and Ganggou-Kumishi areas suggests that they could be a bigger uniform metamorphic basement block, which could be formed by the assembly of the supercontinent Rodinia and be separated by late geological processes. 展开更多

Zipingpu Dam is located 17.17 km from the epicenter and the influence intensity at the damsite reached IX―X degrees. It is the first time in the world for such a high concrete-faced rockfill dam (CFRD) in China to experience such a strong earthquake occurring in such a short distance. Due to its location of special importance to the down- stream areas, the safety state and damaging condition of the Zipingpu Dam has attracted great atten- tions from various circles. Based on site investigation of the damages to the Zipingpu Dam and com- parison analysis of the observation data obtained before and after the earthquake, the paper makes a comprehensive assessment of the key technical issues relevant to this high CFRD, including its safety state and the high slope stability after the earthquake. 展开更多

Inspired by the nature,lotus leaf-derived gradient hierarchical porous C/MoS2 morphology genetic composites(GHPCM)were successfully fabricated through an in situ strategy.The biological microstructure of lotus leaf was well preserved after treatment.Different pores with gradient pore sizes ranging from 300 to 5μm were hierarchically distributed in the composites.In addition,the surface states of lotus leaf resulted in the Janus-like morphologies of MoS2.The GHPCM exhibit excellent electromagnetic wave absorption performance,with the minimum reflection loss of−50.1 dB at a thickness of 2.4 mm and the maximum effective bandwidth of 6.0 GHz at a thickness of 2.2 mm.The outstanding performance could be attributed to the synergy of conductive loss,polarization loss,and impedance matching.In particularly,we provided a brand-new dielectric sum-quotient model to analyze the electromagnetic performance of the non-magnetic material system.It suggests that the specific sum and quotient of permittivity are the key to keep reflection loss below−10 dB within a certain frequency range.Furthermore,based on the concept of material genetic engineering,the dielectric constant could be taken into account to seek for suitable materials with designable electromagnetic absorption performance. 展开更多

Application of Artificial Bee Colony Algorithm to Maximum Likelihood DOA Estimation 被引量:18

Maximum Likelihood (ML) method has an excellent performance for Direction-Of-Arrival (DOA) estimation, but a mul- tidimensional nonlinear solution search is required which complicates the computation and prevents the method from practical use. To reduce the high computational burden of ML method and make it more suitable to engineering applications, we apply the Artificial Bee Colony (ABC) algorithm to maximize the likelihood function for DOA estimation. As a recently proposed bio-inspired computing algorithm, ABC algorithm is originally used to optimize multivariable functions by imitating the be- havior of bee colony finding excellent nectar sources in the nature environment. It offers an excellent alternative to the con- ventional methods in ML-DOA estimation. The performance of ABC-based ML and other popular meta-heuristic-based ML methods for DOA estimation are compared for various scenarios of convergence, Signal-to-Noise Ratio (SNR), and number of iterations. The computation loads of ABC-based ML and the conventional ML methods for DOA estimation are also investi- gated. Simulation results demonstrate that the proposed ABC based method is more efficient in computation and statistical performance than other ML-based DOA estimation methods. 展开更多

The Qilian Shan lies along the northeastern edge of the Tibetan Plateau. To constrain its deformation history, we conducted integrated research on Mesozoic-Cenozoic stratigraphic sections in the Jiuxi Basin immediately north of the mountain range. Paleocurrent measurements, sandstone compositional data, and facies analysis of Cenozoic stratigraphic sections suggest that the Jiuxi Basin received sediments from the Altyn Tagh Range in the northwest, initially in the Oligocene (-33 Ma), depositing the Huoshaogou Formation in the northern part of the basin. Later, the source area of the Jiuxi Basin changed to the Qilian Shan in the south during Late Oligocene (-27 Ma), which led to the deposition of the Baiyanghe Formation. We suggest that uplift of the northern Qilian Shan induced by thrusting began no later than the Late Oligocene. Fission-track analysis of apatite from the Qilian Shan yields further information about the deformation history of the northern Qilain Shan and the Jiuxi Basin. It shows that a period of rapid cooling, interpreted as exhumation, initiated in the Oligocene. We suggest that this exhumation marked the initial uplift of the Qilian Shan resulting from the India-Asia collision. 展开更多

Electromagnetic pollution has been causing a series of problems in people’s life,and electromagnetic absorbers with lightweight and broad absorbing bandwidth properties are widely desired.In this work,novel sandwich-like 2D laminated Fe&TiO2 nanoparticles@C nanocomposites were rationally designed and successfully developed from the MXene–MOFs hybrids.The formation of Fe and rutile-TiO2 nanoparticles sandwiched by the two-dimensional carbon nanosheets provided strong electromagnetic energy attenuation and good impedance matching for electromagnetic wave(EMW)absorption.As expected,the nanocomposites achieved a broad effective absorption bandwidth of 6.5 GHz at a thickness of only 1.6 mm and the minimum reflection loss(RL)value of−51.8 dB at 6.6 GHz with a thickness of 3 mm.This work not only provides a good design and fabricating concept for the laminated metal and functional nanoparticles@C nanocomposites with good EMW absorption,but also offers an important guideline to fabricate various two-dimensional nanocomposites derived from the MXene precursors. 展开更多

Complete resection could be achieved in virtually all myasthenic patients with Masaoka stage I and II thymoma us- ing the trans-sternal technique. Whether this is appropriate for minimally invasive approach is not yet clear. We evalu- ated the feasibility of complete video-assisted thoracoscopic surgery (VATS) thymectomy for the treatment of Ma- saoka stage I and Ⅱ thymoma-associated myasthenia gravis, compared to conventional trans-sternal thymectomy. We summarized 33 patients with Masaoka stage I and II thymoma-associated myasthenia gravis between April 2006 and September 2011. Of these, 15 patients underwent right-sided complete VATS (the VATS group) by us- ing adjuvant pneuomomediastinum, comparing with 18 patients using the trans-sternal approach (the T3b group). No intraoperative death was found and no VATS case required conversion to median sternotomy. Significant differences between the two groups regarding duration of surgery and volume of intraoperative blood loss (P = 0.001 and P 〈 0.001, respectively) were observed. Postoperative morbidities were 26.7% and 33.3% for the VATS and T3b groups, respectively. All 33 patients were followed up for 12 to 61 months in the study. The cumulative probabilities of reaching complete stable remission and effective rate were 26.7% (4/15) and 93.3% (14/15) in the VATS group, which had a significantly higher complete stable remission and effective rate than those in the T3b group (P = 0.026 and P = 0.000, respectively). We conclude that VATS thymectomy utilizing adjuvant pneuomo- mediastinum for the treatment of stage I and II thymoma-associated myasthenia gravis is technically feasible but deserves further investigation in a large series with long-term follow-up. 展开更多

In the search of alternative resources to make commodity chemicals and transportation fuels for a low carbon future,lignocellulosic biomass with over 180-billion-ton annual production rate has been identified as a promising feedstock.This review focuses on the state-of-the-art catalytic transformation of lignocellulosic biomass into value-added chemicals and fuels.Following a brief introduction on the structure,major resources and pretreatment methods of lignocellulosic biomass,the catalytic conversion of three main components,i.e.,cellulose,hemicellulose and lignin,into various compounds are comprehensively discussed.Either in separate steps or in one-pot,cellulose and hemicellulose are hydrolyzed into sugars and upgraded into oxygen-containing chemicals such as 5-HMF,furfural,polyols,and organic acids,or even nitrogen-containing chemicals such as amino acids.On the other hand,lignin is first depolymerized into phenols,catechols,guaiacols,aldehydes and ketones,and then further transformed into hydrocarbon fuels,bioplastic precursors and bioactive compounds.The review then introduces the transformations of whole biomass via catalytic gasification,catalytic pyrolysis,as well as emerging strategies.Finally,opportunities,challenges and prospective of woody biomass valorization are highlighted. 展开更多

Network virtualization(NV) is considered as an enabling tool to remove the gradual ossification of current Internet. In the network virtualization environment, a set of heterogeneous virtual networks(VNs), isolated from each other, share the underlying resources of one or multiple substrate networks(SNs) according to the resource allocation strategy. This kind of resource allocation strategy is commonly known as so called Virtual Network Embedding(VNE) algorithm in network virtualization. Owing to the common sense that VNE problem is NP-hard in nature, most of VNE algorithms proposed in the literature are heuristic. This paper surveys and analyzes a number of representative heuristic solutions in the literature. Apart from the analysis of representative heuristic solutions, a taxonomy of the heuristic solutions is also presented in the form of table. Future research directions of VNE, especially for the heuristics, are emphasized and highlighted at the end of this survey. 展开更多

Geochronological researches were made on gabbro and basalt. Sm-Nd isochron age of gabbro is (829 ± 60) Ma, while the age of gabbro mixed basalt is (949 ± 62) Ma. The dating of Sm-Nd isochron proves that ophiolite formed in (829 ± 60) Ma, which implies that the northern half of Tarim (or north Tarim Block) had been separated by an ocean from the southern half of Tarim and Qaidam (or south Tarim Block) until (829 ± 60) Ma. The south Tarim Block could accrete to the north Tarim Block at the beginning of Sinian,thereby forming the north Altun Tagh suture. The Sinian system would be the first cover on the amalgamated Tarim craton. 展开更多

Three-dimensional (3D) hierarchical Pt-Cu tetragonal, highly branched, and dendritic superstructures have been synthesized by a facile template-free hydrothermal approach, showing growth patterns along (111, 110), (111), and (100) planes, respectively. These structures have been characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), energy dispersive X-ray spectroscopy (EDX), inductively coupled plasma optical emission spectrometry (ICP-OES) and a detailed formation mechanism has been developed, which shows that the in situ formed 12 and the galvanic replacement reaction between Cu and Pt4, may guide the formation of these superstructures. The comparative electrocatalytic properties have been investigated for methanol and ethanol oxidation. Due to their interconnected arms, sufficient absorption sites, and exposed surfaces, these superstructures exhibit enhanced electrocatalytic performance for electro-oxidation of methanol and ethanol when compared with commercial Pt/C and Pt black. 展开更多

The ^(25)Mg(p,γ)^(26)Al reaction plays an important role in the study of cosmic 1.809 MeV γ-ray as a signature of ongoing nucleosynthesis in the Galaxy.At astrophysical temperature around 0.1 GK,the ^(25)Mg(p,γ)^(26)Al reaction rates are dominated by the 92 keV resonance capture process.We report a precise measurement of the 92 keV ^(25)Mg(p,γ)^(26)Al resonance in the day-one experiment at Jinping Underground Nuclear Astrophysics experiment(JUNA)facility in the China Jinping Underground Laboratory(CJPL).The resonance strength and ground state feeding factor are determined to be 3.8±0.3×10^(-10) eV and 0:660:04,respectively.The results are in agreement with those reported in the previous direct underground measurement within uncertainty,but with significantly reduced uncertainties.Consequently,we recommend new ^(25)Mg(p,γ)^(26)Al reaction rates which are by a factor of 2.4 larger than those adopted in REACLIB database at the temperature around 0.1 GK.The new results indicate higher production rates of ^(26g)Al and the cosmic 1.809 MeV γ-ray.The implication of the new rates for the understanding of other astrophysical situations is also discussed. 展开更多

In this paper, an archetypal aseismic system is proposed with 2-degree of freedom based on a smooth and discontinuous(SD)oscillator to avoid the failure of electric power system under the complex excitation of seismic waves. This model comprises two vibration isolation units for the orthogonal horizontal directions, and each of them admits the stable quasi-zero stiffness(SQZS)with a pair of inclined linear elastic springs. The equation of motion is formulated by using Lagrange equation, and the SQZS condition is obtained by optimizing the parameters of the system. The analysis shows that the system behaves a remarkable vibration isolation performance with low resonant frequency and a large stroke of SQZS interval. The experimental investigations are carried out to show a high sonsistency with the theoretical results, which demonstrates the improvement of aseismic behavior of the proposed model under the seismic wave. 展开更多
