METHODS: The profiles of serum miRNA expression were first generated with serum samples collected from 10 patients with CHB and 10 healthy donors (Ctrls) by microarray analysis. The levels of several miRNAs were further quantitated by real-time reverse transcription polymerase chain reaction with serum samples from another 24 CHB patients and 24 Ctrls. Serum samples of 20 patients with nonalcohlic steatohepatitis (NASH) were also included for comparison. The comparison in the levels of miRNAs between groups (CHB, NASH and Ctrl) was analyzed with Mann-Whitney U-test. The cor- relation between miRNAs and clinical pathoparameters was analyzed using Spearman correlation analysis or canonical correlation analysis. The receiver-operator characteristic (ROC) curves were also generated to de- termine the specificity and sensitivity of each individual miRNA in distinguishing patients with CriB from Ctrls. RESULTS: miRNA profile analysis showed that 34 miR- NAs were differentially expressed between CriB and Ctrl subjects, in which 12 were up-regulated and 22 down-regulated in CriB subject (fold change 〉 2.0 and P 〈 0.01). The median levels of miR-122, -572, -575 and -638 were significantly higher (P 〈 1.00 × 10-5) while miR-744 significantly lower (P 〈 1.0× 10-6) in Crib compared with the Ctrl. The levels of miR-122, -572 and -638 were also higher (P 〈 1.00×10-3) while the level of miR-744 lower in CriB (P 〈 0.05) than in NASH, although the difference between them was not as significant as that between CHB and Ctrl. ROC curve analysis revealed that the levels of miR-122, -572, -575, -638 and -744 in serum were sensitive and specific enough to distinguish CriB, NASH and Ctrl. This study aimed to elucidate the molecular mechanisms by which berberine protects against cerebral ischemia/reperfusion(I/R)injury.The oxygen-glucose deprivation/reperfusion(OGD/R)PC12 model was established.Cell coun...This study aimed to elucidate the molecular mechanisms by which berberine protects against cerebral ischemia/reperfusion(I/R)injury.The oxygen-glucose deprivation/reperfusion(OGD/R)PC12 model was established.Cell counting kit-8(CCK-8)was used to detect the toxicity of berberine and the viability of PC12 cells.Hoechst 33258 staining and flow cytometry were used to observe the nuclear morphology,and changes of apoptosis and reactive oxygen species(ROS),respectively.Western blotting and immunofluorescence assay were employed to detect autophagy-related proteins[microtubule-associated protein 1A/1B-light chain 3(LC3),P62/SQSTM-1,Beclin-1]and endoplasmic reticulum(ER)stress-related markers[glucose-regulated protein 78(GRP78),C/EBP homologous protein(CHOP),Bcl-2-associated X(Bax)and cleaved caspase-3].The GFP-RFP-LC3 adenovirus was used to assay the change of autophagic flux.Our results showed that berberine could increase the viability of PC12 cells,decrease the concentrations of ROS after OGD/R treatment,and suppress OGD/R-induced ER stress and autophagy.Moreover,the results revealed the involvement of the mammalian target of rapamycin(mTOR)pathway in the induction of autophagy,and berberine could activate the phosphorylation of mTOR and thus mitigate autophagy.In conclusion,our study suggested that berberine may protect against OGD/R-induced apoptosis by regulating ER stress and autophagy,and it holds promises in the treatment of cerebral I/R injury.展开更多
The microstructures and textures of four rectangular extruded Mg alloys, Mg-3Gd, Mg-3Nd, Mg-1Gd-1Nd and Mg-1.5Gd-1.5Nd(all in wt%) were investigated by using optical microscopy(OM), X-ray diffraction(XRD) and el...The microstructures and textures of four rectangular extruded Mg alloys, Mg-3Gd, Mg-3Nd, Mg-1Gd-1Nd and Mg-1.5Gd-1.5Nd(all in wt%) were investigated by using optical microscopy(OM), X-ray diffraction(XRD) and electron backscatter diffraction(EBSD). The results demonstrate that the extruded alloys all reveal fully recrystallized microstructure and weak non-basal texture.For textures of the four extruded alloys, it is shown that the basal planes of majority grains tilt away from the sheet plane normal toward the extrusion direction. Compared with two binary alloys, the ternary alloys possess weaker texture strengths. Through orientation distribution function analysis, two types of rare earth textures, {0113} 〈0223〉and {1215} 〈1213〉, were found in the extruded alloys.Texture component {1215} 〈1213〉 is relatively weak, but it plays a critical role in the low texture intensities of two ternary alloys. A pairing effect of Gd and Nd elements has an influence on the texture modification. Additionally,there is a difference in the sensitivities of experimental parameters between the microstructure and texture.展开更多
