Organ shortage is a major bottleneck in allotransplantation and causes many wait-listed patients to die or become too sick for transplantation.Genetically engineered pigs have been discussed as a potential alternative...Organ shortage is a major bottleneck in allotransplantation and causes many wait-listed patients to die or become too sick for transplantation.Genetically engineered pigs have been discussed as a potential alternative to allogeneic donor organs.Although xenotransplantation of pig-derived organs in nonhuman primates(NHPs)has shown sequential advances in recent years,there are still underlying problems that need to be completely addressed before clinical applications,including(i)acute humoral xenograft rejection;(ii)acute cellular rejection;(iii)dysregulation of coagulation and inflammation;(iv)physiological incompatibility;and(v)cross-species infection.Moreover,various genetic modifications to the pig donor need to be fully characterized,with the aim of identifying the ideal transgene combination for upcoming clinical trials.In addition,suitable pretransplant screening methods need to be confirmed for optimal donor-recipient matching,ensuring a good outcome from xenotransplantation.Herein,we summarize the understanding of organ xenotransplantation in pigs-to-NHPs and highlight the current status and recent progress in extending the survival time of pig xenografts and recipients.We also discuss practical strategies for overcoming the obstacles to xenotransplantation mentioned above to further advance transplantation of pig organs in the clinic.展开更多

Much is known about the regulation of gene expression in response to salt stress at the tran- scriptional level; however, little is known about this process at the posttranscriptional level. Recently, we demonstrated that SKIP is a component of spliceosome that interacts with clock gene pre-mRNAs and is essential for regulating their alternative splicing and mRNA maturation. In this study, we found that skip-1 plants are hypersensitive to both salt and osmotic stresses, and that SKIP is required for the alter- native splicing and mRNA maturation of several salt-tolerance genes, including NHXl, CBL1, P5CS1, RCl2A, and PATIO. A genome-wide analysis revealed that SKIP mediates the alternative splicing of many genes under salt-stress conditions, and that most of the alternative splicing events in skip-1 involve intron retention and can generate a premature termination codon in the transcribed mRNA. SKIP also controls alternative splicing by modulating the recognition or cleavage of 5' and 3' splice donor and acceptor sites under salt-stress conditions. Therefore, this study addresses the fundamental question of how the mRNA splicing machinery in plants contributes to salt-stress responses at the posttranscriptional level, and provides a link between alternative splicing and salt tolerance.展开更多
Objective:The transanal approach to specimen collection,combined with the prolapsing technique,is a wellestablished and minimally invasive surgery for treating rectal cancer.However,reports on outcomes for this approa...Objective:The transanal approach to specimen collection,combined with the prolapsing technique,is a wellestablished and minimally invasive surgery for treating rectal cancer.However,reports on outcomes for this approach are sparse.We compared short-and long-term outcomes of conventional laparoscopic surgery(CLS)vs.transanal natural orifice specimen extraction(NOSE)using the prolapsing technique for patients with middle-to low-rectal cancer.Methods:From January 2013 to December 2017,we enrolled consecutive patients with middle-to low-rectal cancer undergoing laparoscopic anterior resection.Totally,50 patients who underwent transanal NOSE using the prolapsing technique were matched with 50 patients who received CLS.Clinical parameters and survival outcomes between the two groups were compared.Results:Estimated blood loss(29.70±29.28 vs.52.80±45.09 mL,P=0.003),time to first flatus(2.50±0.79 vs.2.86±0.76,P=0.022),time to liquid diet(3.62±0.64 vs.4.20±0.76 d,P<0.001),and the need for analgesics(22%vs.48%,P=0.006)were significantly lower for the NOSE group compared to the CLS group.The incidences of overall complications and fecal incontinence were comparable in both groups.After a median follow-up of 44.52 months,the overall local recurrence rate(6%vs.5%,P=0.670),3-year disease-free survival(86.7%vs.88.0%,P=0.945)and 3-year overall survival(95.6%vs.96.0%,P=0.708),were not significantly different.Conclusions:For total laparoscopic rectal resection,transanal NOSE using the prolapsing technique is effective and safe,and associated with less trauma and pain,a faster recovery,and similar survival outcomes compared to CLS.展开更多

However, this does not address metastatic lymph nodes in the mesorectum. The aim of this study was to assess the incidence of lymph node metastases in ypT0 patients treated with NCRT and curative resection and to explore risk factors associated with survival.Methods: This was a retrospective study of patients with ypT0 rectal cancer after NCRT and curative resection at a tertiary care center in China from 2005 to 2014.Results: A total of 60(18.6%) patients who underwent surgery after NCRT and achieved ypT0 were enrolled in this study; one patient was excluded owing to lack of follow-up. Of these 59 patients, lymph node metastases were found in the mesorectum(ypT0N+) in eight(13.6%) patients. After a median follow-up of 52 months, 5-year recurrence-free survival(82.7% vs. 62.5%, P=0.014) and overall survival(OS)(90.9% vs. 70.0%, P=0.032) were much higher in ypN0 than yp N+ patients. Multivariate analyses showed that ypN+ status(P=0.009) and perioperative blood transfusion(BT)(P=0.001) were significantly independent risk factors associated with recurrence; however, no factor was correlated with 5-year OS.Conclusions: Patients with ypT0N0 rectal cancer can achieve excellent long-term outcomes; however, positive lymph nodes or tumor deposits can still be found in 13.6% of ypT0 patients. Nodal positivity in the mesorectum and perioperative BT are independent risk factors for recurrence.展开更多

The S (spike) protein is the largest structural protein, with 1,255 amino acids, in the viral genome. Its structure can be divided into three regions: a long N-terminal region in the exterior, a characteristic transmembrane (TM) region, and a short C-terminus in the interior of a virion. We detected fifteen substitutions of nucleotides by comparisons with the seventeen published SARS-CoV genome sequences, eight (53.3%) of which are non-synonymous mutations leading to amino acid alternations with predicted physiochemical changes. The possible antigenic determinants of the S protein are predicted, and the result is confirmed by ELISA (enzyme-linked immunosorbent assay) with synthesized peptides. Another profound finding is that three disulfide bonds are defined at the C-terminus with the N-terminus of the E (envelope) protein, based on the typical sequence and positions, thus establishing the structural connection with these two important structural proteins, if confirmed. Phyloge-netic analysis reveals several conserved regions that might be potent drug targets.展开更多
Background Elderly multiple organ dysfunction syndrome (MODS) patients receiving invasive mechanical ventilation have poor prognosis in intensive care units (ICUs).We studied the usefulness of four commonly used s...Background Elderly multiple organ dysfunction syndrome (MODS) patients receiving invasive mechanical ventilation have poor prognosis in intensive care units (ICUs).We studied the usefulness of four commonly used severity scores and extrapulmonary factors that affected weaning to predict outcome of such patients.Methods Clinical data of 197 patients on admission to ICUs (from January 2009 to June 2012) were used retrospectively.The Acute Physiology and Chronic Health Evaluation (APACHE) Ⅱ,APACHE Ⅲ,Sample Acute Physiological Score (SAPS) Ⅱ and MODS scores were calculated.All the patients were grouped into survivors and nonsurvivors according to the prognosis.Patients,who weaned from ventilator (n=154),were subdivided into a successful weaning group and a failed weaning group.The receiver operating characteristic (ROC) curves and Logistic regression was used for prognostic and weaning assessment.Results Based on the outcomes,the areas under the ROC of APACHE Ⅱ,APACHE Ⅲ,SAPS Ⅱ,and MODS were 0.837,0.833,0.824,and 0.837,respectively.The Logistic regression analysis revealed that the odds ratio (OR) of underlying lung diseases,serum albumin and creatinine,and the number of organ failures was 2.374,0.920,1.003,and 1.547.APACHE Ⅱ scores on admission performed excellent (ROC:0.921) on the weaning assessments.Conclusions APACHE Ⅱ and MODS systems were marginally better for evaluating the prognosis of elderly MODS patients who received invasive mechanical ventilation.Underlying lung diseases,serum albumin,serum creatinine and the number of organ failures were independent prognostic factors.Using the APACHE Ⅱ scores on admission before weaning may increase the likelihood of successful weaning.(ClinicalTrial.gov identifier NCT01802983).展开更多
In plants,the genome structure of hybrids changes compared with their parents,but the effects of these changes in hybrids remain elusive.Comparing reciprocal crosses between Col×C24 and C24×Col in Arabidopsi...In plants,the genome structure of hybrids changes compared with their parents,but the effects of these changes in hybrids remain elusive.Comparing reciprocal crosses between Col×C24 and C24×Col in Arabidopsis using high-throughput chromosome conformation capture assay(Hi-C)analysis,we found that hybrid three-dimensional(3D)chromatin organization had more long-distance interactions relative to parents,and this was mainly located in promoter regions and enriched in genes with heterosis-related pathways.The interactions between euchromatin and heterochromatin were increased,and the compartment strength decreased in hybrids.In compartment domain(CD)boundaries,the distal interactions were more in hybrids than their parents.In the hybrids of CURLY LEAF(clf)mutants clfCol×clfC24and clfC24×clfCol,the heterosis phenotype was damaged,and the long-distance interactions in hybrids were fewer than in their parents with lower H3K27me3.ChIP-seq data revealed higher levels of H3K27me3 in the region adjacent to the CD boundary and the same interactional homo-trans sites in the wild-type(WT)hybrids,which may have led to more long-distance interactions.In addition,the differentially expressed genes(DEGs)located in the boundaries of CDs and loop regions changed obviously in WT,and the functional enrichment for DEGs was different between WT and clf in the longdistance interactions and loop regions.Our findings may therefore propose a new epigenetic explanation of heterosis in the Arabidopsis hybrids and provide new insights into crop breeding and yield increase.展开更多
Background:The effect of intra-operative chemotherapy(IOC)on the long-term survival of patients with colorectal cancer(CRC)remains unclear.In this study,we evaluated the independent effect of intra-operative infusion ...Background:The effect of intra-operative chemotherapy(IOC)on the long-term survival of patients with colorectal cancer(CRC)remains unclear.In this study,we evaluated the independent effect of intra-operative infusion of 5-fluorouracil in combination with calcium folinate on the survival of CRC patients following radical resection.Methods:1820 patients were recruited,and 1263 received IOC and 557 did not.Clinical and demographic data were collected,including overall survival(OS),clinicopathological features,and treatment strategies.Risk factors for IOC-related deaths were identified using multivariate Cox proportional hazards models.A regression model was developed to analyze the independent effects of IOC.Results:Proportional hazard regression analysis showed that IOC(hazard ratio[HR]=0.53,95%confidence intervals[CI][0.43,0.65],P<0.001)was a protective factor for the survival of patients.The mean overall survival time in IOC group was 82.50(95%CI[80.52,84.49])months,and 71.21(95%CI[67.92,74.50])months in non-IOC group.The OS in IOC-treated patients were significantly higher than non-IOC-treated patients(P<0.001,log-rank test).Further analysis revealed that IOC decreased the risk of death in patients with CRC in a non-adjusted model(HR=0.53,95%CI[0.43,0.65],P<0.001),model 2(adjusted for age and gender,HR=0.52,95%CI[0.43,0.64],P<0.001),and model 3(adjusted for all factors,95%CI 0.71[0.55,0.90],P=0.006).The subgroup analysis showed that the HR for the effect of IOC on survival was lower in patients with stage II(HR=0.46,95%CI[0.31,0.67])or III disease(HR=0.59,95%CI[0.45,0.76]),regardless of pre-operative radiotherapy(HR=0.55,95%CI[0.45,0.68])or pre-operative chemotherapy(HR=0.54,95%CI[0.44,0.66]).Conclusions:IOC is an independent factor that influences the survival of CRC patients.It improved the OS of patients with stages II and III CRC after radical surgery.Trial registration:chictr.org.cn,ChiCTR 2100043775.展开更多
To adapt to the low-velocity water flow closely related to human life,the natural energy can be efficiently harvested and used to power monitoring devices.Herein,a triboelectric soft fishtail(TE-SFT)driven by flow-ind...To adapt to the low-velocity water flow closely related to human life,the natural energy can be efficiently harvested and used to power monitoring devices.Herein,a triboelectric soft fishtail(TE-SFT)driven by flow-induced vibration(FIV)effect is proposed based on the soft material synthesis technology.Specifically,inspired by the fishtail fin,a bluff body with the cross-section of fishtail-shaped is designed,and has a preferable vortex effect by fluid simulation.In power generation part,the triboelectric nanogenerator(TENG)is designed to act as an inertial pendulum structure by geometric method.Under the FIV effect,the TESFT driven by fishtail-shaped bluff body swings like a fish in the water and then brings the inertial pendulum to acquire the oscillation for harvesting energy from low-velocity water flow.The TE-SFT attains an open-circuit voltage(VOC)of 200 V to 313 V at the flow velocities of 0.24 to 0.89 m/s.Additionally,after 30 days of water immersion,the VOC of TE-SFT retains 96.81%.In demonstration,the TE-SFT is applied to power the temperature and humidity sensor through harvesting water flow energy.This work also provides a way for self-powered system based on the TENG and soft bionic fish in water environment.展开更多

The final R is 0.051. The MNDO MO calculation shows that the coordinate bonds of title compound possess certain extent of covalent character. The hydrokinetic energy of river current,as one of the essential and widespread renewable energies,is difficult to be harvested in low flow velocity and shallow water areas.In this work,a three-dimensional(3D)fully-en...The hydrokinetic energy of river current,as one of the essential and widespread renewable energies,is difficult to be harvested in low flow velocity and shallow water areas.In this work,a three-dimensional(3D)fully-enclosed triboelectric nanogenerator(FETENG)with bionic fish-like structure for harvesting hydrokinetic energy is reported,which is comprised of the triboelectric powergeneration unit,bionic fish-like structure and connection unit.Through the bionic structure,the FE-TENG realizes zero head power generation in shallow water with low flow velocity.What’s more,the effect of external excitations and bionic structures on the electrical performance are systematically studied in this work.The FE-TENG can generate peak power density of 7 and 0.36 W/m^(3)respectively under the simulated swing state with frequency of 1.25 Hz and simulated river current with flow velocity of 0.81 m/s.In practical applications,due to the 3D fully-enclosed design,the FE-TENG immersed in water for 35 days demonstrates excellent immersion durability with undiminished electrical performance.Therefore,the work proposes an efficient method realizing zero head power generation,and provides a good candidate for long-term service in the river current.展开更多
Safely and highly selective acetylene(C_(2)H_(2))capture is a great challenge,because of its highly explosive nature,as well as its nearly similar molecule size and boiling point toward the main impurity of carbon dio...Safely and highly selective acetylene(C_(2)H_(2))capture is a great challenge,because of its highly explosive nature,as well as its nearly similar molecule size and boiling point toward the main impurity of carbon dioxide(CO_(2)).Adsorption separation has shown a promising future.Herein,a new nanoporous coordination polymer(PCP)adsorbent with fixed and free Cu ions(termed NTU-66-Cu)was prepared through post-synthetic approach via cation exchanging from the pristine NTU-66,an anionic framework with new 3,4,6-c topology and two kinds of cages.The NTU-66-Cu shows significantly improved C_(2)H_(2)/CO_(2)selectivity from 6 to 32(v/v:1/1)or 4 to 4_(2)(v/v:1/4)at low pressure under 298 K,along with enhanced C_(2)H_(2)capacity(from 89.22to 111.53 cm^(3)·g^(-1)).More importantly,this observation was further validated by density functional theory(DFT)calculations and breakthrough experiments under continuous and dynamic conditions.Further,the excellent chemical stability enables this adsorbent to achieve recycle C_(2)H_(2)/CO_(2)separation without loss of C_(2)H_(2)capacity.展开更多

The effects of carbon and ionomer contents on the electrode micro-structure and fuel cell performance are investigated by physical characterization and single cell testing. The Pt nanowires are gradient distributed across the cathode thickness, and more Pt exists near the membrane. Both the carbon and ionomer contents can affect the Pt nanowires distribution and aggregation. In addition, the carbon loading dominates the transport distance of gas and proton, and the ionomer content affects the triple phase boundaries and porosity in the cathode. The optimal structure of Pt nanowire cathode is obtained at 0.10 mg·cm^-2 carbon loading and 10 wt% ionomer.展开更多

Photothermal therapy(PTT)has been proposed as an advanced patient-centered strategy for tumor treatment.Nevertheless,the uncertain safety of conventional photothermal conversion agents and the presence of intracellula...Photothermal therapy(PTT)has been proposed as an advanced patient-centered strategy for tumor treatment.Nevertheless,the uncertain safety of conventional photothermal conversion agents and the presence of intracellular self-protective autophagy mechanisms pose obstacles to the clinical application and efficacy of PTT.As we are deeply aware of the seriousness of these problems,we herein proposed an efficacy-enhancing strategy based on an implantable membrane platform(PPG@PB-HCQ)constructed from poly(lactic acid)(PLA),poly(ɛ-caprolactone)(PCL)and gelatin(Gel)electrospun nanofibers(PPG)and loaded with the biodegradable high-efficiency photothermal conversion agent Prussian blue(PB)and the autophagy inhibitor hydroxychloroquine sulfate(HCQ).Cellular experiments confirmed that the PPG@PB-HCQ nanofiber membrane exhibited a significantly stronger tumor cell-killing effect compared with the PTT alone.This enhancement features by of blocking the fusion of autophagosomes with lysosomes.The intracellular overexpression of the proteins microtubule-associated protein 1 light chain 3(LC3)-II and p62 and the low expression of the proteins LC3-I and Rab7(members of the RAS oncogene family)further demonstrated autophagic flux blockade.Importantly,the potent antitumor effect of the PPG@PB-HCQ therapeutic platform in B16 tumor-bearing model mice verified the efficacy-enhancing strategy of synergistic PTT and protective autophagy blockade.The present study provides a promising strategy for solving the difficulties of tumor treatment,as well as a new perspective for designing novel treatment platforms.展开更多
Objective To compare the short-term efficacy indicators of laparoscopic technique and open surgery in patients with rectal cancer surgery,and to further evaluate the safety and efficacy of laparoscopic rectal cancer s...Objective To compare the short-term efficacy indicators of laparoscopic technique and open surgery in patients with rectal cancer surgery,and to further evaluate the safety and efficacy of laparoscopic rectal cancer surgery.Methods This study adopted a prospective multicenter,open-label,non-randomized concurrent control method to analyze patients who received rectal cancer surgery from 10 colorectal tumor centers across the country from January 2017 to December 2018.The two groups of patients received laparoscopic-assisted surgery and conventional open surgery respectively.All surgeons were selected according to relevant standards and participated in the two groups of operations at the same time.Comprehensive evaluation and analysis of the operation and postoperative recovery,postoperative pathological results,survival information,postoperative related functional scores and other indicators.Results In the open surgery group,the proportion of tumors with a maximum diameter of more than 5 cm was higher(χ2=0.089,P=0.018),and the proportion of T4 was higher(χ2=0.478,P<0.001).In the laparoscopic group,the proportion of more than 12 harvested lymph nodes was higher(χ2=0.248,P<0.001).There were no significant differences in the length of postoperative hospital stay,intraoperative blood loss,time to first flatus after operation,time to first out of bed,time to first liquid diet,and preservation rate of anal sphincter between the two groups,but in the laparoscopic surgery group the operation time was shorter(t=6.750,P<0.001)and postoperative pain was less(t=0.896,0.63,0.964,and 0.989 on postoperative days 1,2,and 3,respectively,all P<0.001).The incidence of grade 2–4 adverse complication in the two groups was 12.5%and 14.6%,respectively,with no statistical difference(χ2=0.061,P=0.105),but the incidence of incision infection was lower in the laparoscopy group(χ2=0.19,P<0.001).There was no significant difference in 3-year disease-free survival between the two groups(HR=1.089,95%CI=0.962–1.232,P=0.170).Multivariate 展开更多
Objective:Through integrated bioinformatics analysis,the goal of this work was to find new,characterised N7-methylguanosine modification-related long non-coding RNAs(m7G-lncRNAs)that might be used to predict the progn...Objective:Through integrated bioinformatics analysis,the goal of this work was to find new,characterised N7-methylguanosine modification-related long non-coding RNAs(m7G-lncRNAs)that might be used to predict the prognosis of laryngeal squamous cell carcinoma(LSCC).Methods:The clinical data and LSCC gene expression data for the current investigation were initially retrieved from the TCGA database&sanitised.Then,using co-expression analysis of m7G-associated mRNAs&lncRNAs&differential expression analysis(DEA)among LSCC&normal sample categories,we discovered lncRNAs that were connected to m7G.The prognosis prediction model was built for the training category using univariate&multivariate COX regression&LASSO regression analyses,&the model’s efficacy was checked against the test category data.In addition,we conducted DEA of prognostic m7G-lncRNAs among LSCC&normal sample categories&compiled a list of co-expression networks&the structure of prognosis m7G-lncRNAs.To compare the prognoses for individuals with LSCC in the high-&low-risk categories in the prognosis prediction model,survival and risk assessments were also carried out.Finally,we created a nomogram to accurately forecast the outcomes of LSCC patients&created receiver operating characteristic(ROC)curves to assess the prognosis prediction model’s predictive capability.Results:Using co-expression network analysis&differential expression analysis,we discovered 774 m7G-lncRNAs and 551 DEm7G-lncRNAs,respectively.We then constructed a prognosis prediction model for six m7G-lncRNAs(FLG−AS1,RHOA−IT1,AC020913.3,AC027307.2,AC010973.2 and AC010789.1),identified 32 DEPm7G-lncRNAs,analyzed the correlation between 32 DEPm7G-lncRNAs and 13 DEPm7G-mRNAs,and performed survival analyses and risk analyses of the prognosis prediction model to assess the prognostic performance of LSCC patients.By displaying ROC curves and a nomogram,we finally checked the prognosis prediction model's accuracy.Conclusion:By creating novel predictive lncRNA signatures for clinical diagnosis展开更多

After being spiked ...Heterogeneous photocatalytic degradation of γ-HCH on soil surfaces was carried out to evaluate the photocatalytic effectiveness of α-Fe 2O 3 and TiO 2 toward degrading γ-HCH on soil surfaces. After being spiked with γ-HCH, soil samples were loaded with α-Fe 2O 3 or TiO 2 and exposed to UV-light irradiation. Different catalyst loads, 0%, 2%, 5%, 7%, and 10% (wt.)α-Fe 2O 3; 0%, 0.5%, 1%, 2:(wt.)TiO 2, were tested for up to 7 d irradiation. The effects of soil thickness, acidity, and humic substances were also investigated. The obtained results indicated that the γ-HCH photodegradation follows the pseudo-first-order kinetics. The addition of α-Fe 2O 3 or TiO 2 accelerates the photodegradation of γ-HCH, while the photodegradation rate decreases when the content of α-Fe 2O 3 exceeds 7%(wt.). The degradation rate increases with the soil pH value. Humic substances inhibit the photocatalytic degradation of γ-HCH. Pentachlorocyclohexene, tetrachlorocyclohexene, and trichlorobenzene are detected as photodegradation intermediates, which are gradually degraded with the photodegradation evolution.展开更多

文摘Organ shortage is a major bottleneck in allotransplantation and causes many wait-listed patients to die or become too sick for transplantation.Genetically engineered pigs have been discussed as a potential alternative to allogeneic donor organs.Although xenotransplantation of pig-derived organs in nonhuman primates(NHPs)has shown sequential advances in recent years,there are still underlying problems that need to be completely addressed before clinical applications,including(i)acute humoral xenograft rejection;(ii)acute cellular rejection;(iii)dysregulation of coagulation and inflammation;(iv)physiological incompatibility;and(v)cross-species infection.Moreover,various genetic modifications to the pig donor need to be fully characterized,with the aim of identifying the ideal transgene combination for upcoming clinical trials.In addition,suitable pretransplant screening methods need to be confirmed for optimal donor-recipient matching,ensuring a good outcome from xenotransplantation.Herein,we summarize the understanding of organ xenotransplantation in pigs-to-NHPs and highlight the current status and recent progress in extending the survival time of pig xenografts and recipients.We also discuss practical strategies for overcoming the obstacles to xenotransplantation mentioned above to further advance transplantation of pig organs in the clinic.
文摘Deciphering the mechanisms underlying plant responses to abiotic stress is key for improving plant stress resistance. Much is known about the regulation of gene expression in response to salt stress at the tran- scriptional level; however, little is known about this process at the posttranscriptional level. Recently, we demonstrated that SKIP is a component of spliceosome that interacts with clock gene pre-mRNAs and is essential for regulating their alternative splicing and mRNA maturation. In this study, we found that skip-1 plants are hypersensitive to both salt and osmotic stresses, and that SKIP is required for the alter- native splicing and mRNA maturation of several salt-tolerance genes, including NHXl, CBL1, P5CS1, RCl2A, and PATIO. A genome-wide analysis revealed that SKIP mediates the alternative splicing of many genes under salt-stress conditions, and that most of the alternative splicing events in skip-1 involve intron retention and can generate a premature termination codon in the transcribed mRNA. SKIP also controls alternative splicing by modulating the recognition or cleavage of 5' and 3' splice donor and acceptor sites under salt-stress conditions. Therefore, this study addresses the fundamental question of how the mRNA splicing machinery in plants contributes to salt-stress responses at the posttranscriptional level, and provides a link between alternative splicing and salt tolerance.

文摘Objective:The transanal approach to specimen collection,combined with the prolapsing technique,is a wellestablished and minimally invasive surgery for treating rectal cancer.However,reports on outcomes for this approach are sparse.We compared short-and long-term outcomes of conventional laparoscopic surgery(CLS)vs.transanal natural orifice specimen extraction(NOSE)using the prolapsing technique for patients with middle-to low-rectal cancer.Methods:From January 2013 to December 2017,we enrolled consecutive patients with middle-to low-rectal cancer undergoing laparoscopic anterior resection.Totally,50 patients who underwent transanal NOSE using the prolapsing technique were matched with 50 patients who received CLS.Clinical parameters and survival outcomes between the two groups were compared.Results:Estimated blood loss(29.70±29.28 vs.52.80±45.09 mL,P=0.003),time to first flatus(2.50±0.79 vs.2.86±0.76,P=0.022),time to liquid diet(3.62±0.64 vs.4.20±0.76 d,P<0.001),and the need for analgesics(22%vs.48%,P=0.006)were significantly lower for the NOSE group compared to the CLS group.The incidences of overall complications and fecal incontinence were comparable in both groups.After a median follow-up of 44.52 months,the overall local recurrence rate(6%vs.5%,P=0.670),3-year disease-free survival(86.7%vs.88.0%,P=0.945)and 3-year overall survival(95.6%vs.96.0%,P=0.708),were not significantly different.Conclusions:For total laparoscopic rectal resection,transanal NOSE using the prolapsing technique is effective and safe,and associated with less trauma and pain,a faster recovery,and similar survival outcomes compared to CLS.

文摘Objective: For patients with locally advanced rectal cancer treated with neoadjuvant chemoradiotherapy(NCRT), significant pathological response of the primary tumor has been proposed to identify candidates for organ preservation. However, this does not address metastatic lymph nodes in the mesorectum. The aim of this study was to assess the incidence of lymph node metastases in ypT0 patients treated with NCRT and curative resection and to explore risk factors associated with survival.Methods: This was a retrospective study of patients with ypT0 rectal cancer after NCRT and curative resection at a tertiary care center in China from 2005 to 2014.Results: A total of 60(18.6%) patients who underwent surgery after NCRT and achieved ypT0 were enrolled in this study; one patient was excluded owing to lack of follow-up. Of these 59 patients, lymph node metastases were found in the mesorectum(ypT0N+) in eight(13.6%) patients. After a median follow-up of 52 months, 5-year recurrence-free survival(82.7% vs. 62.5%, P=0.014) and overall survival(OS)(90.9% vs. 70.0%, P=0.032) were much higher in ypN0 than yp N+ patients. Multivariate analyses showed that ypN+ status(P=0.009) and perioperative blood transfusion(BT)(P=0.001) were significantly independent risk factors associated with recurrence; however, no factor was correlated with 5-year OS.Conclusions: Patients with ypT0N0 rectal cancer can achieve excellent long-term outcomes; however, positive lymph nodes or tumor deposits can still be found in 13.6% of ypT0 patients. Nodal positivity in the mesorectum and perioperative BT are independent risk factors for recurrence.
文摘The corona-like spikes or peplomers on the surface of the virion under electronic microscope are the most striking features of coronaviruses. The S (spike) protein is the largest structural protein, with 1,255 amino acids, in the viral genome. Its structure can be divided into three regions: a long N-terminal region in the exterior, a characteristic transmembrane (TM) region, and a short C-terminus in the interior of a virion. We detected fifteen substitutions of nucleotides by comparisons with the seventeen published SARS-CoV genome sequences, eight (53.3%) of which are non-synonymous mutations leading to amino acid alternations with predicted physiochemical changes. The possible antigenic determinants of the S protein are predicted, and the result is confirmed by ELISA (enzyme-linked immunosorbent assay) with synthesized peptides. Another profound finding is that three disulfide bonds are defined at the C-terminus with the N-terminus of the E (envelope) protein, based on the typical sequence and positions, thus establishing the structural connection with these two important structural proteins, if confirmed. Phyloge-netic analysis reveals several conserved regions that might be potent drug targets.
文摘Background Elderly multiple organ dysfunction syndrome (MODS) patients receiving invasive mechanical ventilation have poor prognosis in intensive care units (ICUs).We studied the usefulness of four commonly used severity scores and extrapulmonary factors that affected weaning to predict outcome of such patients.Methods Clinical data of 197 patients on admission to ICUs (from January 2009 to June 2012) were used retrospectively.The Acute Physiology and Chronic Health Evaluation (APACHE) Ⅱ,APACHE Ⅲ,Sample Acute Physiological Score (SAPS) Ⅱ and MODS scores were calculated.All the patients were grouped into survivors and nonsurvivors according to the prognosis.Patients,who weaned from ventilator (n=154),were subdivided into a successful weaning group and a failed weaning group.The receiver operating characteristic (ROC) curves and Logistic regression was used for prognostic and weaning assessment.Results Based on the outcomes,the areas under the ROC of APACHE Ⅱ,APACHE Ⅲ,SAPS Ⅱ,and MODS were 0.837,0.833,0.824,and 0.837,respectively.The Logistic regression analysis revealed that the odds ratio (OR) of underlying lung diseases,serum albumin and creatinine,and the number of organ failures was 2.374,0.920,1.003,and 1.547.APACHE Ⅱ scores on admission performed excellent (ROC:0.921) on the weaning assessments.Conclusions APACHE Ⅱ and MODS systems were marginally better for evaluating the prognosis of elderly MODS patients who received invasive mechanical ventilation.Underlying lung diseases,serum albumin,serum creatinine and the number of organ failures were independent prognostic factors.Using the APACHE Ⅱ scores on admission before weaning may increase the likelihood of successful weaning.(ClinicalTrial.gov identifier NCT01802983).

文摘In plants,the genome structure of hybrids changes compared with their parents,but the effects of these changes in hybrids remain elusive.Comparing reciprocal crosses between Col×C24 and C24×Col in Arabidopsis using high-throughput chromosome conformation capture assay(Hi-C)analysis,we found that hybrid three-dimensional(3D)chromatin organization had more long-distance interactions relative to parents,and this was mainly located in promoter regions and enriched in genes with heterosis-related pathways.The interactions between euchromatin and heterochromatin were increased,and the compartment strength decreased in hybrids.In compartment domain(CD)boundaries,the distal interactions were more in hybrids than their parents.In the hybrids of CURLY LEAF(clf)mutants clfCol×clfC24and clfC24×clfCol,the heterosis phenotype was damaged,and the long-distance interactions in hybrids were fewer than in their parents with lower H3K27me3.ChIP-seq data revealed higher levels of H3K27me3 in the region adjacent to the CD boundary and the same interactional homo-trans sites in the wild-type(WT)hybrids,which may have led to more long-distance interactions.In addition,the differentially expressed genes(DEGs)located in the boundaries of CDs and loop regions changed obviously in WT,and the functional enrichment for DEGs was different between WT and clf in the longdistance interactions and loop regions.Our findings may therefore propose a new epigenetic explanation of heterosis in the Arabidopsis hybrids and provide new insights into crop breeding and yield increase.

文摘Background:The effect of intra-operative chemotherapy(IOC)on the long-term survival of patients with colorectal cancer(CRC)remains unclear.In this study,we evaluated the independent effect of intra-operative infusion of 5-fluorouracil in combination with calcium folinate on the survival of CRC patients following radical resection.Methods:1820 patients were recruited,and 1263 received IOC and 557 did not.Clinical and demographic data were collected,including overall survival(OS),clinicopathological features,and treatment strategies.Risk factors for IOC-related deaths were identified using multivariate Cox proportional hazards models.A regression model was developed to analyze the independent effects of IOC.Results:Proportional hazard regression analysis showed that IOC(hazard ratio[HR]=0.53,95%confidence intervals[CI][0.43,0.65],P<0.001)was a protective factor for the survival of patients.The mean overall survival time in IOC group was 82.50(95%CI[80.52,84.49])months,and 71.21(95%CI[67.92,74.50])months in non-IOC group.The OS in IOC-treated patients were significantly higher than non-IOC-treated patients(P<0.001,log-rank test).Further analysis revealed that IOC decreased the risk of death in patients with CRC in a non-adjusted model(HR=0.53,95%CI[0.43,0.65],P<0.001),model 2(adjusted for age and gender,HR=0.52,95%CI[0.43,0.64],P<0.001),and model 3(adjusted for all factors,95%CI 0.71[0.55,0.90],P=0.006).The subgroup analysis showed that the HR for the effect of IOC on survival was lower in patients with stage II(HR=0.46,95%CI[0.31,0.67])or III disease(HR=0.59,95%CI[0.45,0.76]),regardless of pre-operative radiotherapy(HR=0.55,95%CI[0.45,0.68])or pre-operative chemotherapy(HR=0.54,95%CI[0.44,0.66]).Conclusions:IOC is an independent factor that influences the survival of CRC patients.It improved the OS of patients with stages II and III CRC after radical surgery.Trial registration:chictr.org.cn,ChiCTR 2100043775.

文摘To adapt to the low-velocity water flow closely related to human life,the natural energy can be efficiently harvested and used to power monitoring devices.Herein,a triboelectric soft fishtail(TE-SFT)driven by flow-induced vibration(FIV)effect is proposed based on the soft material synthesis technology.Specifically,inspired by the fishtail fin,a bluff body with the cross-section of fishtail-shaped is designed,and has a preferable vortex effect by fluid simulation.In power generation part,the triboelectric nanogenerator(TENG)is designed to act as an inertial pendulum structure by geometric method.Under the FIV effect,the TESFT driven by fishtail-shaped bluff body swings like a fish in the water and then brings the inertial pendulum to acquire the oscillation for harvesting energy from low-velocity water flow.The TE-SFT attains an open-circuit voltage(VOC)of 200 V to 313 V at the flow velocities of 0.24 to 0.89 m/s.Additionally,after 30 days of water immersion,the VOC of TE-SFT retains 96.81%.In demonstration,the TE-SFT is applied to power the temperature and humidity sensor through harvesting water flow energy.This work also provides a way for self-powered system based on the TENG and soft bionic fish in water environment.

The crystal is monoclinic, space group P2<sub>1</sub>/n with crystal parameters of α =0.7420(2) nm, b=0.344 9(1) nm, c=2.490 6(3) nm,β=94.89(1)°, Z=4, D<sub>c</sub>=1.799 g.cm<sup>-3</sup>, V=0.635 nm<sup>3</sup>,μ= 1.591 cm<sup>-1</sup>, F(000) = 392. The final R is 0.051. The MNDO MO calculation shows that the coordinate bonds of title compound possess certain extent of covalent character. O<sub>2</sub> atom of NTO anion is bonded to Li atom; the nitro group will be lost first when NTO is decomposed.

文摘The hydrokinetic energy of river current,as one of the essential and widespread renewable energies,is difficult to be harvested in low flow velocity and shallow water areas.In this work,a three-dimensional(3D)fully-enclosed triboelectric nanogenerator(FETENG)with bionic fish-like structure for harvesting hydrokinetic energy is reported,which is comprised of the triboelectric powergeneration unit,bionic fish-like structure and connection unit.Through the bionic structure,the FE-TENG realizes zero head power generation in shallow water with low flow velocity.What’s more,the effect of external excitations and bionic structures on the electrical performance are systematically studied in this work.The FE-TENG can generate peak power density of 7 and 0.36 W/m^(3)respectively under the simulated swing state with frequency of 1.25 Hz and simulated river current with flow velocity of 0.81 m/s.In practical applications,due to the 3D fully-enclosed design,the FE-TENG immersed in water for 35 days demonstrates excellent immersion durability with undiminished electrical performance.Therefore,the work proposes an efficient method realizing zero head power generation,and provides a good candidate for long-term service in the river current.

文摘Safely and highly selective acetylene(C_(2)H_(2))capture is a great challenge,because of its highly explosive nature,as well as its nearly similar molecule size and boiling point toward the main impurity of carbon dioxide(CO_(2)).Adsorption separation has shown a promising future.Herein,a new nanoporous coordination polymer(PCP)adsorbent with fixed and free Cu ions(termed NTU-66-Cu)was prepared through post-synthetic approach via cation exchanging from the pristine NTU-66,an anionic framework with new 3,4,6-c topology and two kinds of cages.The NTU-66-Cu shows significantly improved C_(2)H_(2)/CO_(2)selectivity from 6 to 32(v/v:1/1)or 4 to 4_(2)(v/v:1/4)at low pressure under 298 K,along with enhanced C_(2)H_(2)capacity(from 89.22to 111.53 cm^(3)·g^(-1)).More importantly,this observation was further validated by density functional theory(DFT)calculations and breakthrough experiments under continuous and dynamic conditions.Further,the excellent chemical stability enables this adsorbent to achieve recycle C_(2)H_(2)/CO_(2)separation without loss of C_(2)H_(2)capacity.
文摘Purpose Colorectal cancer is a common malignant tumor worldwide.In China,the ratio of rectal cancer to coloncancer in terms of incidence is close to 1:1.Low rectal cancer accounts for more than half of all cases of rectal cancer.In recent years,the proportion of rectal cancer has trended downward,however the incidence of rectal cancer inyounger adults is increasing.The CACA Guidelines for Holistic Integrative Management of Rectal Cancer were editedto help improve the diagnosis and comprehensive treatment in China.Methods This guideline has been prepared by consensuses reached by the CACA Committee of Colorectal CancerSociety,based on a careful review of the latest evidence including China’s studies,and referred to domestic and internationalrelative guidelines,also considered China’s specific national conditions and clinical practice.Results The CACA Guidelines for Holistic Integrative Management of Rectal Cancer include the epidemiology of rectalcancer,prevention and screening,diagnosis,treatment of nonmetastatic and metastatic rectal cancer,follow-up,and whole-course rehabilitation management.Conclusion Committee of Colorectal Cancer Society,Chinese Anti-Cancer Association,standardizes the diagnosisand treatment of rectal cancer in China through the formulation of the CACA Guidelines.
文摘High performance cathode for polymer electrolyte membrane fuel cell was prepared by depositing Pt nanowires in a carbon matrix coated on a substrate, and using decal transfer method to fabricate the membrane electrode assembly. The effects of carbon and ionomer contents on the electrode micro-structure and fuel cell performance are investigated by physical characterization and single cell testing. The Pt nanowires are gradient distributed across the cathode thickness, and more Pt exists near the membrane. Both the carbon and ionomer contents can affect the Pt nanowires distribution and aggregation. In addition, the carbon loading dominates the transport distance of gas and proton, and the ionomer content affects the triple phase boundaries and porosity in the cathode. The optimal structure of Pt nanowire cathode is obtained at 0.10 mg·cm^-2 carbon loading and 10 wt% ionomer.

文摘Small interfering RNA(siRNA)constitutes a promising therapeutic modality supporting the potential functional cure of hepatitis B.A novel ionizable lipidoid nanoparticle(RBP131)and a state-of-the-art lyophilization technology were developed in this study,enabling to deliver siRNA targeting apolipoprotein B(APOB)into the hepatocytes with an ED_(50)of 0.05 mg/kg after intravenous injection.In addition,according to the requirements of Investigational New Drug(IND)application,a potent siRNA targeting hepatitis B virus(HBV)was selected and encapsulated with RBP131 to fabricate a therapeutic formulation termed RB-HBV008.

文摘Photothermal therapy(PTT)has been proposed as an advanced patient-centered strategy for tumor treatment.Nevertheless,the uncertain safety of conventional photothermal conversion agents and the presence of intracellular self-protective autophagy mechanisms pose obstacles to the clinical application and efficacy of PTT.As we are deeply aware of the seriousness of these problems,we herein proposed an efficacy-enhancing strategy based on an implantable membrane platform(PPG@PB-HCQ)constructed from poly(lactic acid)(PLA),poly(ɛ-caprolactone)(PCL)and gelatin(Gel)electrospun nanofibers(PPG)and loaded with the biodegradable high-efficiency photothermal conversion agent Prussian blue(PB)and the autophagy inhibitor hydroxychloroquine sulfate(HCQ).Cellular experiments confirmed that the PPG@PB-HCQ nanofiber membrane exhibited a significantly stronger tumor cell-killing effect compared with the PTT alone.This enhancement features by of blocking the fusion of autophagosomes with lysosomes.The intracellular overexpression of the proteins microtubule-associated protein 1 light chain 3(LC3)-II and p62 and the low expression of the proteins LC3-I and Rab7(members of the RAS oncogene family)further demonstrated autophagic flux blockade.Importantly,the potent antitumor effect of the PPG@PB-HCQ therapeutic platform in B16 tumor-bearing model mice verified the efficacy-enhancing strategy of synergistic PTT and protective autophagy blockade.The present study provides a promising strategy for solving the difficulties of tumor treatment,as well as a new perspective for designing novel treatment platforms.

文摘Objective To compare the short-term efficacy indicators of laparoscopic technique and open surgery in patients with rectal cancer surgery,and to further evaluate the safety and efficacy of laparoscopic rectal cancer surgery.Methods This study adopted a prospective multicenter,open-label,non-randomized concurrent control method to analyze patients who received rectal cancer surgery from 10 colorectal tumor centers across the country from January 2017 to December 2018.The two groups of patients received laparoscopic-assisted surgery and conventional open surgery respectively.All surgeons were selected according to relevant standards and participated in the two groups of operations at the same time.Comprehensive evaluation and analysis of the operation and postoperative recovery,postoperative pathological results,survival information,postoperative related functional scores and other indicators.Results In the open surgery group,the proportion of tumors with a maximum diameter of more than 5 cm was higher(χ2=0.089,P=0.018),and the proportion of T4 was higher(χ2=0.478,P<0.001).In the laparoscopic group,the proportion of more than 12 harvested lymph nodes was higher(χ2=0.248,P<0.001).There were no significant differences in the length of postoperative hospital stay,intraoperative blood loss,time to first flatus after operation,time to first out of bed,time to first liquid diet,and preservation rate of anal sphincter between the two groups,but in the laparoscopic surgery group the operation time was shorter(t=6.750,P<0.001)and postoperative pain was less(t=0.896,0.63,0.964,and 0.989 on postoperative days 1,2,and 3,respectively,all P<0.001).The incidence of grade 2–4 adverse complication in the two groups was 12.5%and 14.6%,respectively,with no statistical difference(χ2=0.061,P=0.105),but the incidence of incision infection was lower in the laparoscopy group(χ2=0.19,P<0.001).There was no significant difference in 3-year disease-free survival between the two groups(HR=1.089,95%CI=0.962–1.232,P=0.170).Multivariate

文摘Objective:Through integrated bioinformatics analysis,the goal of this work was to find new,characterised N7-methylguanosine modification-related long non-coding RNAs(m7G-lncRNAs)that might be used to predict the prognosis of laryngeal squamous cell carcinoma(LSCC).Methods:The clinical data and LSCC gene expression data for the current investigation were initially retrieved from the TCGA database&sanitised.Then,using co-expression analysis of m7G-associated mRNAs&lncRNAs&differential expression analysis(DEA)among LSCC&normal sample categories,we discovered lncRNAs that were connected to m7G.The prognosis prediction model was built for the training category using univariate&multivariate COX regression&LASSO regression analyses,&the model’s efficacy was checked against the test category data.In addition,we conducted DEA of prognostic m7G-lncRNAs among LSCC&normal sample categories&compiled a list of co-expression networks&the structure of prognosis m7G-lncRNAs.To compare the prognoses for individuals with LSCC in the high-&low-risk categories in the prognosis prediction model,survival and risk assessments were also carried out.Finally,we created a nomogram to accurately forecast the outcomes of LSCC patients&created receiver operating characteristic(ROC)curves to assess the prognosis prediction model’s predictive capability.Results:Using co-expression network analysis&differential expression analysis,we discovered 774 m7G-lncRNAs and 551 DEm7G-lncRNAs,respectively.We then constructed a prognosis prediction model for six m7G-lncRNAs(FLG−AS1,RHOA−IT1,AC020913.3,AC027307.2,AC010973.2 and AC010789.1),identified 32 DEPm7G-lncRNAs,analyzed the correlation between 32 DEPm7G-lncRNAs and 13 DEPm7G-mRNAs,and performed survival analyses and risk analyses of the prognosis prediction model to assess the prognostic performance of LSCC patients.By displaying ROC curves and a nomogram,we finally checked the prognosis prediction model's accuracy.Conclusion:By creating novel predictive lncRNA signatures for clinical diagnosis
文摘Heterogeneous photocatalytic degradation of γ-HCH on soil surfaces was carried out to evaluate the photocatalytic effectiveness of α-Fe 2O 3 and TiO 2 toward degrading γ-HCH on soil surfaces. After being spiked with γ-HCH, soil samples were loaded with α-Fe 2O 3 or TiO 2 and exposed to UV-light irradiation. Different catalyst loads, 0%, 2%, 5%, 7%, and 10% (wt.)α-Fe 2O 3; 0%, 0.5%, 1%, 2:(wt.)TiO 2, were tested for up to 7 d irradiation. The effects of soil thickness, acidity, and humic substances were also investigated. The obtained results indicated that the γ-HCH photodegradation follows the pseudo-first-order kinetics. The addition of α-Fe 2O 3 or TiO 2 accelerates the photodegradation of γ-HCH, while the photodegradation rate decreases when the content of α-Fe 2O 3 exceeds 7%(wt.). The degradation rate increases with the soil pH value. Humic substances inhibit the photocatalytic degradation of γ-HCH. Pentachlorocyclohexene, tetrachlorocyclohexene, and trichlorobenzene are detected as photodegradation intermediates, which are gradually degraded with the photodegradation evolution.
