We earlier identified a major QTL that controls rice grain width and weight, GW5, which was mapped to a recombination hotspot on rice chromosome 5. To gain a better understanding of how GW5 controls rice grain width, we conducted fine mapping of this locus and uncovered a 1 212-bp deletion associated with the increased grain width in the rice cultivar Asominori, in comparison with the slender grain rice IR24. In addition, genotyping analyses of 46 rice cultivars revealed that this deletion is highly correlated with the grain-width phenotype, suggesting that the GW5 deletion might have been selected during rice domestication. GW5 encodes a novel nuclear protein of 144 amino acids that is localized to the nucleus. Furthermore, we show that GW5 physically interacts with polyubiquitin in a yeast two-hybrid assay. Together, our results suggest that GW5 represents a major QTL underlying rice width and weight, and that it likely acts in the ubiquitin-proteasome pathway to regulate cell division during seed development. This study provides novel insights into the molecular mechanisms controlling rice grain development and suggests that GW5 could serve as a potential tool for high-yield breeding of crops.展开更多
China faces the greatest challenge from stroke in the world.The death rate for cerebrovascular diseases in China was 149.49 per 100000,accounting for 1.57 million deaths in 2018.It ranked third among the leading cause...China faces the greatest challenge from stroke in the world.The death rate for cerebrovascular diseases in China was 149.49 per 100000,accounting for 1.57 million deaths in 2018.It ranked third among the leading causes of death behind malignant tumours and heart disease.The age-standardised prevalence and incidence of stroke in 2013 were 1114.8 per 100000 population and 246.8 per 100000 person-years,respectively.According to the Global Burden of Disease Study 2017,the years of life lost(YLLs)per 100000 population for stroke increased by 14.6%;YLLs due to stroke rose from third highest among all causes in 1990 to the highest in 2017.The absolute numbers and rates per 100000 population for all-age disability-adjusted life years(DALYs)for stroke increased substantially between 1990 and 2017,and stroke was the leading cause of all-age DALYs in 2017.The main contributors to cerebrovascular diseases include behavioural risk factors(smoking and alcohol use)and pre-existing conditions(hypertension,diabetes mellitus,dyslipidaemia and atrial fibrillation(AF)).The most prevalent risk factors among stroke survivors were hypertension(63.0%-84.2%)and smoking(31.7%-47.6%).The least prevalent was AF(2.7%-7.4%).The prevalences for major risk factors for stroke are high and most have increased over time.Based on the latest national epidemiological data,26.6%of adults aged≥15 years(307.6 million adults)smoked tobacco products.For those aged≥18 years,age-adjusted prevalence of hypertension was 25.2%;adjusted prevalence of hypercholesterolaemia was 5.8%;and the standardised prevalence of diabetes was 10.9%.For those aged≥40 years,the standardised prevalence of AF was 2.31%.Data from the Hospital Quality Monitoring System showed that 3010204 inpatients with stroke were admitted to 1853 tertiary care hospitals during 2018.Of those,2466785(81.9%)were ischaemic strokes(ISs);447609(14.9%)were intracerebral haemorrhages(ICHs);and 95810(3.2%)were subarachnoid haemorrhages(SAHs).The average age of patients admitted was 66 years old,and展开更多
It has been known that,the novel coronavirus,2019-nCoV,which is considered similar to SARS-CoV,invades human cells via the receptor angiotensin converting enzyme II(ACE2).Moreover,lung cells that have ACE2 expression ...It has been known that,the novel coronavirus,2019-nCoV,which is considered similar to SARS-CoV,invades human cells via the receptor angiotensin converting enzyme II(ACE2).Moreover,lung cells that have ACE2 expression may be the main target cells during 2019-nCoV infection.However,some patients also exhibit non-respiratory symptoms,such as kidney failure,implying that 2019-nCoV could also invade other organs.To construct a risk map of different human organs,we analyzed the single-cell RNA sequencing(scRNA-seq)datasets derived from major human physiological systems,including the respiratory,cardiovascular,digestive,and urinary systems.Through scRNA-seq data analyses,we identified the organs at risk,such as lung,heart,esophagus,kidney,bladder,and ileum,and located specific cell types(i.e.,type II alveolar cells(AT2),myocardial cells,proximal tubule cells of the kidney,ileum and esophagus epithelial cells,and bladder urothelial cells),which are vulnerable to 2019-nCoV infection.Based on the findings,we constructed a risk map indicating the vulnerability of different organs to 2019-nCoV infection.This study may provide potential clues for further investigation of the pathogenesis and route of 2019-nCoV infection.展开更多

The process and evolution of haze pollution episodes were observed ...In January 2013, a long-lasting episode of severe haze occurred in central and eastern China, and it attracted attention from all sectors of society. The process and evolution of haze pollution episodes were observed by the "Forming Mechanism and Con- trol Strategies of Haze in China" group using an intensive aerosol and trace gases campaign that simultaneously obtained data at 11 ground-based observing sites in the CARE-China network. The characteristics and formation mechanism of haze pollu- tion episodes were discussed. Five haze pollution episodes were identified in the Beijing-Tianjin-Hebei (Jing-Jin-Ji) area; the two most severe episodes occurred during 9-15 January and 25-31 January. During these two haze pollution episodes, the maximum hourly PMz5 mass concentrations in Beijing were 680 and 530 ~tg m-3, respectively. The process and evolution of haze pollution episodes in other major cities in the Jing-Jin-Ji area, such as Shijiazhuang and Tianjin were almost the same as those observed in Beijing. The external cause of the severe haze episodes was the unusual atmospheric circulation, the depres- sion of strong cold air activities and the very unfavorable dispersion due to geographical and meteorological conditions. How- ever, the internal cause was the quick secondary transformation of primary gaseous pollutants to secondary aerosols, which contributed to the "explosive growth" and "sustained growth" of PM2.5. Particularly, the abnormally high amount of nitric ox- ide (NOx) in the haze episodes, produced by fossil fuel combustion and vehicle emissions, played a direct or indirect role in the quick secondary transformation of coal-burning sulphur dioxide (SO2) to sulphate aerosols. Furthermore, gaseous pollutants were transformed into secondary aerosols through heterogeneous reactions on the surface of fine particles, which can change the particle's size and chemical composition. PTEN expression wascompared with tumor stage, lymph node metastasis, Lauren'sand WHO's histological classification of gastric carcinoma.Expression of VEGF was also detected in 60 cases of gastriccarcinoma and its correlation with PTEN was concerned.RESULTS: The positive rates of PTEN protein were 100 %(102/102), 98.5 %(65/66), 66.7 % (4/6) and 47.8 %(88/184)in normal mucosa, IM, dysplasia and carcinoma of the stomach,respectively. The positive rates in dysplasia and carcinomawere lower than in normal mucosa and IM (P<0.01).Advanced gastric cancers expressed less frequent PTEN thanearly gastric cancer (42.9 % v567.6 %, P<0.01). The positiverate of PTEN protein was lower in gastric cancer with thanwithout lymph node metastasis (40.3 % v563.3 %, P<0.01).PTEN was less expressed in diffuse-type than in intestinal-type gastric cancer (41.5 % v557.8 %,P<0.05). Signet ringcell carcinoma showed the expression of PTEN at the lowestlevel (25.0 %, 7/28); less than well and moderatelydifferentiated ones (P<0.01). Expression of PTEN was notcorrelated with expression of VEGF (P>0.05).CONCLUSION: Loss or reduced expression of PTEN proteinoccures commonly in tumorigenesis and progression of gastriccarcinoma. It is suggested that PTEN can be an objective markerfor pathologically biological behaviors of gastric carcinoma.展开更多

By synthetically considering the size of energy density, the frequency of energy level and the stability of energy density, the resources of wind energy and wave energy in the East China Sea and South China Sea were analyzed and regionalized. Vegetation cover changes on the Loess Plateau have experienced four stages as follows: (1) vegetation cover showed a continued increasing phase during 1981―1989; (2) vegetation cover changes came into a relative steady phase with small fluctuations during 1990―1998; (3) vegetation cover declined rapidly during 1999―2001; and (4) vegetation cover increased rapidly during 2002―2006. The vegetation cover changes of the Loess Plateau show a notable spatial difference. The vegetation cover has obviously increased in the Inner Mongolia and Ningxia plain along the Yellow River and the ecological rehabilitated region of Ordos Plateau, however the vegetation cover evidently decreased in the hilly and gully areas of Loess Plateau, Liupan Mountains region and the northern hillside of Qinling Mountains. The response of NDVI to climate changes varied with different vegetation types. NDVI of sandy land vegetation, grassland and cultivated land show a significant increasing trend, but forest shows a decreasing trend. The results obtained in this study show that the spatiotemporal variations of vegetation cover are the outcome of climate changes and human activities. Temperature is a control factor of the seasonal change of vegetation growth. The increased temperature makes soil drier and unfavors vegetation growth in summer, but it favors vegetation growth in spring and autumn because of a longer growing period. There is a significant correlation between vegetation cover and precipitation and thus, the change in precipitation is an important factor for vegetation variation. The improved agricultural production has resulted in an increase of NDVI in the farmland, and the implementation of large-scale vegetation construction has led to some beneficial effect in ecology展开更多

The subject areas and main results were overviewed including biodiversity and eco-geological distribution of soil fauna communities. Studies of ecological distributions of soil fauna and dynamic ranges in space from tropical, subtropical to temperate regions, and in categories from forest, grassland, desert, wetland, farmland to urban ecosystems, the responses and indications of soil fauna to soil environments. Effects of intensive disturbance such as fire, grazing, farming, fertilization on soil fauna include sensitive (e.g. nematode) and rare groups and community indexes. The functions of soil fauna were discussed including environmental construction, environmental purification, litter decomposition and elements cycling. Interactions between soil fauna and other biota in soil ecosystems and linking between aboveground and belowground diversity and the effects of global change on soil fauna community in China were also included. Finally, the authors pointed out common interests in soil fauna eco-geographical studies, which include application of molecule biology into soil fauna taxa; function and mechanism of soil fauna community diversity; interaction between aboveground and belowground ecosystems; effects of disturbance, pollution, biological invasion, and global change on soil fauna community and function. Examining China's five-year plans during 1991–2010, we find that state-owned firms in government supported industries enjoy faster growth in initial public offerings and higher offer prices. Further, they enjoy faster growth in loans granted by major national banks. However, this preferential access to capital by state-owned firms appears to be achieved at the expense of non-state-owned firms which are crowded out. Government support induces more investment but also brings more overinvestment, which mainly comes from the non-state sector.Finally, supported industries have higher stock market returns and cash flow growth that dampen when state ownership increases.展开更多

Here we report a highly efficient approach to induce human embryonic stem (ES) cells and induced pluripo- tent stem (iPS) cells to differentiate into mature insulin-producing cells in a chemical-defined culture system. The differentiated human ES cells obtained by this approach comprised nearly 25% insulin-positive cells as assayed by flow cytometry analysis, which released insulin/C-peptide in response to glucose stimuli in a manner comparable to that of adult human islets. Most of these insulin-producing cells co-expressed mature β cell-specific markers such as NKX6-1 and PDX1, indicating a similar gene expression pattern to adult islet β cells in vivo. In this study, we also demonstrated that EGF facilitates the expansion of PDXl-positive pancreatic progenitors. Moreover, our protocol also succeeded in efficiently inducing human iPS cells to differentiate into insuIin-producing ceils. Therefore, this work not only provides a new model to study the mechanism of human pancreatic specialization and maturation in vitro, but also enhances the possibility of utilizing patient-specific iPS cells for the treatment of diabetes.展开更多

METHODS Male C57BL/6J mice we...AIM To investigate whether gut microbiota metabolite sodium butyrate (NaB) is an effective substance for attenuating non-alcoholic fatty liver disease (NAFLD) and the internal mechanisms. METHODS Male C57BL/6J mice were divided into three groups, normal control were fed standard chow and model group were fed a high-fat diet (HFD) for 16 wk, the intervention group were fed HFD for 16 wk and treated with NaB for 8 wk. Gut microbiota from each group were detected at baseline and at 16 wk, liver histology were evaluated and gastrointestinal barrier indicator such as zonula occluden-1 (ZO-1) were detected by immunohistochemistry and realtime-PCR, further serum or liver endotoxin were determined by ELISA and inflammation-or metabolism-associated genes were quantified by real-time PCR. RESULTS NaB corrected the HFD-induced gut microbiota imbalance in mice, while it considerably elevated the abundances of the beneficial bacteria Christensenellaceae, Blautia and Lactobacillus. These bacteria can produce butyric acid in what seems like a virtuous circle. And butyrate restored HFD induced intestinal mucosa damage, increased the expression of ZO-1 in small intestine, further decreased the levels of gut endotoxin in serum and liver compared with HF group. Endotoxin-associated genes such as TLR4 and Myd88, pro-inflammation genes such as MCP-1, TNF-alpha, IL-1, IL-2, IL-6 and IFN-gamma in liver or epididymal fat were obviously downregulated after NaB intervention. Liver inflammation and fat accumulation were ameliorated, the levels of TG and cholesterol in liver were decreased after NaB intervention, NAS score was significantly decreased, metabolic indices such as FBG and HOMA-IR and liver function indicators ALT and AST were improved compared with HF group. CONCLUSION NaB may restore the dysbiosis of gut microbiota to attenuate steatohepatitis, which is suggested to be a potential gut microbiota modulator and therapeutic substance for NAFLD.展开更多

Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infectio...Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections.This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China,from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records,laboratory assessments,and chest CT scans were reviewed.As a result,none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever,cough,fatigue,etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs.The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization.These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died.The median communicable period,defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests,was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members,which even caused severe COVID-19 pneumonia.Overall,the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization.However,the communicable period could be up to three weeks and the communicated patients could develop severe illness.These results highlighted the importance of close cont展开更多
A field survey was conducted to investigate the metal and arsenic contamination in soils and vegetables on four villages (Shuichuan (SCH), Beiwan (BWA), Dongwan (DWA) and Wufe (WFE)) located along, Baiyin, C...A field survey was conducted to investigate the metal and arsenic contamination in soils and vegetables on four villages (Shuichuan (SCH), Beiwan (BWA), Dongwan (DWA) and Wufe (WFE)) located along, Baiyin, China, and to evaluate the possible health risks to local population through foodchain. Results show that the most significantly contaminated soils occurred upstream at SCH where Cd, Cu and As concentrations exceeded maximum allowable concentrations for Chinese agricultural soil. Further downstream the degree of contamination semi-systematically decreased in concentrations of metal. Generally, the leafy vegetables were more heavily contaminated than non-leafy vegetables. Chinese cabbage is the most severely contaminated, the concentrations of Cd exceeded the maximum permit levels (0.05 mg/kg) by 4.5 times. Bio-accumulate factor also shows that an entry of Cd to food chain plants is the greatest potential. Furthermore, the estimated daily intake amounts of the considered toxic elements (Cd, Pb and Cu) from the vegetables grown at SCH and BWA and DWA have exceeded the recommended dietary allowance levels. Thus, the vegetables grown in three villages above, which affected by Baiyin mining and smelting have a health hazard for human consumption.展开更多
文摘Grain weight is a major determinant of crop grain yield and is controlled by naturally occurring quantitative trait loci (QTLs). We earlier identified a major QTL that controls rice grain width and weight, GW5, which was mapped to a recombination hotspot on rice chromosome 5. To gain a better understanding of how GW5 controls rice grain width, we conducted fine mapping of this locus and uncovered a 1 212-bp deletion associated with the increased grain width in the rice cultivar Asominori, in comparison with the slender grain rice IR24. In addition, genotyping analyses of 46 rice cultivars revealed that this deletion is highly correlated with the grain-width phenotype, suggesting that the GW5 deletion might have been selected during rice domestication. GW5 encodes a novel nuclear protein of 144 amino acids that is localized to the nucleus. Furthermore, we show that GW5 physically interacts with polyubiquitin in a yeast two-hybrid assay. Together, our results suggest that GW5 represents a major QTL underlying rice width and weight, and that it likely acts in the ubiquitin-proteasome pathway to regulate cell division during seed development. This study provides novel insights into the molecular mechanisms controlling rice grain development and suggests that GW5 could serve as a potential tool for high-yield breeding of crops.

文摘China faces the greatest challenge from stroke in the world.The death rate for cerebrovascular diseases in China was 149.49 per 100000,accounting for 1.57 million deaths in 2018.It ranked third among the leading causes of death behind malignant tumours and heart disease.The age-standardised prevalence and incidence of stroke in 2013 were 1114.8 per 100000 population and 246.8 per 100000 person-years,respectively.According to the Global Burden of Disease Study 2017,the years of life lost(YLLs)per 100000 population for stroke increased by 14.6%;YLLs due to stroke rose from third highest among all causes in 1990 to the highest in 2017.The absolute numbers and rates per 100000 population for all-age disability-adjusted life years(DALYs)for stroke increased substantially between 1990 and 2017,and stroke was the leading cause of all-age DALYs in 2017.The main contributors to cerebrovascular diseases include behavioural risk factors(smoking and alcohol use)and pre-existing conditions(hypertension,diabetes mellitus,dyslipidaemia and atrial fibrillation(AF)).The most prevalent risk factors among stroke survivors were hypertension(63.0%-84.2%)and smoking(31.7%-47.6%).The least prevalent was AF(2.7%-7.4%).The prevalences for major risk factors for stroke are high and most have increased over time.Based on the latest national epidemiological data,26.6%of adults aged≥15 years(307.6 million adults)smoked tobacco products.For those aged≥18 years,age-adjusted prevalence of hypertension was 25.2%;adjusted prevalence of hypercholesterolaemia was 5.8%;and the standardised prevalence of diabetes was 10.9%.For those aged≥40 years,the standardised prevalence of AF was 2.31%.Data from the Hospital Quality Monitoring System showed that 3010204 inpatients with stroke were admitted to 1853 tertiary care hospitals during 2018.Of those,2466785(81.9%)were ischaemic strokes(ISs);447609(14.9%)were intracerebral haemorrhages(ICHs);and 95810(3.2%)were subarachnoid haemorrhages(SAHs).The average age of patients admitted was 66 years old,and

文摘It has been known that,the novel coronavirus,2019-nCoV,which is considered similar to SARS-CoV,invades human cells via the receptor angiotensin converting enzyme II(ACE2).Moreover,lung cells that have ACE2 expression may be the main target cells during 2019-nCoV infection.However,some patients also exhibit non-respiratory symptoms,such as kidney failure,implying that 2019-nCoV could also invade other organs.To construct a risk map of different human organs,we analyzed the single-cell RNA sequencing(scRNA-seq)datasets derived from major human physiological systems,including the respiratory,cardiovascular,digestive,and urinary systems.Through scRNA-seq data analyses,we identified the organs at risk,such as lung,heart,esophagus,kidney,bladder,and ileum,and located specific cell types(i.e.,type II alveolar cells(AT2),myocardial cells,proximal tubule cells of the kidney,ileum and esophagus epithelial cells,and bladder urothelial cells),which are vulnerable to 2019-nCoV infection.Based on the findings,we constructed a risk map indicating the vulnerability of different organs to 2019-nCoV infection.This study may provide potential clues for further investigation of the pathogenesis and route of 2019-nCoV infection.

文摘In January 2013, a long-lasting episode of severe haze occurred in central and eastern China, and it attracted attention from all sectors of society. The process and evolution of haze pollution episodes were observed by the "Forming Mechanism and Con- trol Strategies of Haze in China" group using an intensive aerosol and trace gases campaign that simultaneously obtained data at 11 ground-based observing sites in the CARE-China network. The characteristics and formation mechanism of haze pollu- tion episodes were discussed. Five haze pollution episodes were identified in the Beijing-Tianjin-Hebei (Jing-Jin-Ji) area; the two most severe episodes occurred during 9-15 January and 25-31 January. During these two haze pollution episodes, the maximum hourly PMz5 mass concentrations in Beijing were 680 and 530 ~tg m-3, respectively. The process and evolution of haze pollution episodes in other major cities in the Jing-Jin-Ji area, such as Shijiazhuang and Tianjin were almost the same as those observed in Beijing. The external cause of the severe haze episodes was the unusual atmospheric circulation, the depres- sion of strong cold air activities and the very unfavorable dispersion due to geographical and meteorological conditions. How- ever, the internal cause was the quick secondary transformation of primary gaseous pollutants to secondary aerosols, which contributed to the "explosive growth" and "sustained growth" of PM2.5. Particularly, the abnormally high amount of nitric ox- ide (NOx) in the haze episodes, produced by fossil fuel combustion and vehicle emissions, played a direct or indirect role in the quick secondary transformation of coal-burning sulphur dioxide (SO2) to sulphate aerosols. Furthermore, gaseous pollutants were transformed into secondary aerosols through heterogeneous reactions on the surface of fine particles, which can change the particle's size and chemical composition. 文摘There exist differences in the epidemiological characteristics,clinicopathological features,tumor biological characteristics,treatment patterns,and drug selections between gastric cancer patients from the Eastern and Western countries.The Chinese Society of Clinical Oncology(CSCO)has organized a panel of senior experts specializing in all sub-specialties of gastric cancer to compile a clinical guideline for the diagnosis and treatment of gastric cancer since 2016 and renews it annually.Taking into account regional differences,giving full consideration to the accessibility of diagnosis and treatment resources,these experts have conducted expert consensus judgment on relevant evidence and made various grades of recommendations for the clinical diagnosis and treatment of gastric cancer to reflect the value of cancer treatment and meeting health economic indexes in China.The 2021 CSCO Clinical Practice Guidelines for Gastric Cancer covers the diagnosis,treatment,follow-up,and screening of gastric cancer.Based on the 2020 version of the CSCO Chinese Gastric Cancer guidelines,this updated guideline integrates the results ofmajor clinical studies from China and overseas for the past year,focused on the inclusion of research data from the Chinese population for more personalized and clinically relevant recommendations.For the comprehensive treatment of non-metastatic gastric cancer,attentions were paid to neoadjuvant treatment.The value of perioperative chemotherapy is gradually becoming clearer and its recommendation level has been updated.For the comprehensive treatment of metastatic gastric cancer,recommendations for immunotherapy were included,and immune checkpoint inhibitors fromthird-line to the first-line of treatment for different patient groups with detailed notes are provided.

文摘AIM: To detect the expression of PTEN encoding productin normal mucosa, intestinal metaplasia (IM), dysplasia andcarcinoma of the stomach, and to investigate its clinicalimplication in tumorigenesis and progression of gastriccarcinoma.METHODS: Formalin-fixed paraffin embedded specimens from184 cases of gastric carcinoma, their adjacent normal mucosa,IM and dysplasia were evaluated for PTEN protein expressionby SABC immunohistochemistry. PTEN expression wascompared with tumor stage, lymph node metastasis, Lauren'sand WHO's histological classification of gastric carcinoma.Expression of VEGF was also detected in 60 cases of gastriccarcinoma and its correlation with PTEN was concerned.RESULTS: The positive rates of PTEN protein were 100 %(102/102), 98.5 %(65/66), 66.7 % (4/6) and 47.8 %(88/184)in normal mucosa, IM, dysplasia and carcinoma of the stomach,respectively. The positive rates in dysplasia and carcinomawere lower than in normal mucosa and IM (P<0.01).Advanced gastric cancers expressed less frequent PTEN thanearly gastric cancer (42.9 % v567.6 %, P<0.01). The positiverate of PTEN protein was lower in gastric cancer with thanwithout lymph node metastasis (40.3 % v563.3 %, P<0.01).PTEN was less expressed in diffuse-type than in intestinal-type gastric cancer (41.5 % v557.8 %,P<0.05). Signet ringcell carcinoma showed the expression of PTEN at the lowestlevel (25.0 %, 7/28); less than well and moderatelydifferentiated ones (P<0.01). Expression of PTEN was notcorrelated with expression of VEGF (P>0.05).CONCLUSION: Loss or reduced expression of PTEN proteinoccures commonly in tumorigenesis and progression of gastriccarcinoma. It is suggested that PTEN can be an objective markerfor pathologically biological behaviors of gastric carcinoma.

文摘In this paper, the third-generation wave model WAVEWATCH-Ⅲ (WW3) was used to simulate the wave field of the East China Sea and South China Sea from January 1988 to December 2009, with wind input of CCMP wind field. Then, the wind energy density and wave energy density were calculated by using the simulated 22-years' wave-field data and CCMP data. By synthetically considering the size of energy density, the frequency of energy level and the stability of energy density, the resources of wind energy and wave energy in the East China Sea and South China Sea were analyzed and regionalized. The result can be a guide to searching location of wind & wave power plant.

文摘Spatiotemporal variations of Chinese Loess Plateau vegetation cover during 1981-2006 have been investigated using GIMMS and SPOT VGT NDVI data and the cause of vegetation cover changes has been analyzed, considering the climate changes and human activities. Vegetation cover changes on the Loess Plateau have experienced four stages as follows: (1) vegetation cover showed a continued increasing phase during 1981―1989; (2) vegetation cover changes came into a relative steady phase with small fluctuations during 1990―1998; (3) vegetation cover declined rapidly during 1999―2001; and (4) vegetation cover increased rapidly during 2002―2006. The vegetation cover changes of the Loess Plateau show a notable spatial difference. The vegetation cover has obviously increased in the Inner Mongolia and Ningxia plain along the Yellow River and the ecological rehabilitated region of Ordos Plateau, however the vegetation cover evidently decreased in the hilly and gully areas of Loess Plateau, Liupan Mountains region and the northern hillside of Qinling Mountains. The response of NDVI to climate changes varied with different vegetation types. NDVI of sandy land vegetation, grassland and cultivated land show a significant increasing trend, but forest shows a decreasing trend. The results obtained in this study show that the spatiotemporal variations of vegetation cover are the outcome of climate changes and human activities. Temperature is a control factor of the seasonal change of vegetation growth. The increased temperature makes soil drier and unfavors vegetation growth in summer, but it favors vegetation growth in spring and autumn because of a longer growing period. There is a significant correlation between vegetation cover and precipitation and thus, the change in precipitation is an important factor for vegetation variation. The improved agricultural production has resulted in an increase of NDVI in the farmland, and the implementation of large-scale vegetation construction has led to some beneficial effect in ecology
文摘In order to further promote the standardization of diagnosis and treatment of gastrointestinal stromal tumor (GIST) in China, the members of Chinese Society of Clinical Oncology (CSCO) Expert Committee on GIST thoroughly discussed the key contents of the consensus guidelines, and voted on the controversial issue. In final, the Chinese consensus guidelines for the diagnosis and management of GIST (2017 edition) was formed on the basis of 2013 edition consensus guidelines, which is hereby announced. The consensus included the pathological diagnosis, recurrence risk classification evaluation, targeted agent therapy, surgery and principles of surveillance of GIST.

文摘This review summarizes main research findings in soil fauna eco-geography in China in the past 30 years. The subject areas and main results were overviewed including biodiversity and eco-geological distribution of soil fauna communities. Studies of ecological distributions of soil fauna and dynamic ranges in space from tropical, subtropical to temperate regions, and in categories from forest, grassland, desert, wetland, farmland to urban ecosystems, the responses and indications of soil fauna to soil environments. Effects of intensive disturbance such as fire, grazing, farming, fertilization on soil fauna include sensitive (e.g. nematode) and rare groups and community indexes. The functions of soil fauna were discussed including environmental construction, environmental purification, litter decomposition and elements cycling. Interactions between soil fauna and other biota in soil ecosystems and linking between aboveground and belowground diversity and the effects of global change on soil fauna community in China were also included. Finally, the authors pointed out common interests in soil fauna eco-geographical studies, which include application of molecule biology into soil fauna taxa; function and mechanism of soil fauna community diversity; interaction between aboveground and belowground ecosystems; effects of disturbance, pollution, biological invasion, and global change on soil fauna community and function. The review is to provide a scientific basis for promoting soil fauna eco-geographical studies in China.

文摘A careful study on the integral properties of the primitive hydrostatic balance equations for baroclinic atmosphere is carried out, and a new scheme todesign the global adiabatic model of atmospheric dynamics ispresented. This scheme includes a method of weighted equal-areamesh and a fully discrete finite difference method with quadraticand linear conservations for solving the primitive equationsystem. Using this scheme, we established a new dynamical corewith adjustable high resolution acceptable to the availablecomputer capability, which can be very stable without anyfiltering and smoothing. Especially, some important integralproperties are kept unchanged, such as the anti-symmetries of thehorizontal advection operators and the vertical convectionoperator, the mass conservation, the effective energy conservationunder the standard stratification approximation, and so on. Somenumerical tests on the new dynamical core, respectively regardingits global conservations and its integrated performances inclimatic modeling, incorporated with the physical packagesfrom the Community Atmospheric Model Version 2 (CAM2) of NationalCenter for Atmospheric Research (NCAR), are included.

文摘An important factor influencing corporate finance and economic growth in China lies in its government sponsored industrial policies. Examining China's five-year plans during 1991–2010, we find that state-owned firms in government supported industries enjoy faster growth in initial public offerings and higher offer prices. Further, they enjoy faster growth in loans granted by major national banks. However, this preferential access to capital by state-owned firms appears to be achieved at the expense of non-state-owned firms which are crowded out. Government support induces more investment but also brings more overinvestment, which mainly comes from the non-state sector.Finally, supported industries have higher stock market returns and cash flow growth that dampen when state ownership increases.
文摘Human pluripotent stem cells represent a potentially unlimited source of functional pancreatic endocrine lineage cells. Here we report a highly efficient approach to induce human embryonic stem (ES) cells and induced pluripo- tent stem (iPS) cells to differentiate into mature insulin-producing cells in a chemical-defined culture system. The differentiated human ES cells obtained by this approach comprised nearly 25% insulin-positive cells as assayed by flow cytometry analysis, which released insulin/C-peptide in response to glucose stimuli in a manner comparable to that of adult human islets. Most of these insulin-producing cells co-expressed mature β cell-specific markers such as NKX6-1 and PDX1, indicating a similar gene expression pattern to adult islet β cells in vivo. In this study, we also demonstrated that EGF facilitates the expansion of PDXl-positive pancreatic progenitors. Moreover, our protocol also succeeded in efficiently inducing human iPS cells to differentiate into insuIin-producing ceils. Therefore, this work not only provides a new model to study the mechanism of human pancreatic specialization and maturation in vitro, but also enhances the possibility of utilizing patient-specific iPS cells for the treatment of diabetes.

文摘AIM To investigate whether gut microbiota metabolite sodium butyrate (NaB) is an effective substance for attenuating non-alcoholic fatty liver disease (NAFLD) and the internal mechanisms. METHODS Male C57BL/6J mice were divided into three groups, normal control were fed standard chow and model group were fed a high-fat diet (HFD) for 16 wk, the intervention group were fed HFD for 16 wk and treated with NaB for 8 wk. Gut microbiota from each group were detected at baseline and at 16 wk, liver histology were evaluated and gastrointestinal barrier indicator such as zonula occluden-1 (ZO-1) were detected by immunohistochemistry and realtime-PCR, further serum or liver endotoxin were determined by ELISA and inflammation-or metabolism-associated genes were quantified by real-time PCR. RESULTS NaB corrected the HFD-induced gut microbiota imbalance in mice, while it considerably elevated the abundances of the beneficial bacteria Christensenellaceae, Blautia and Lactobacillus. These bacteria can produce butyric acid in what seems like a virtuous circle. And butyrate restored HFD induced intestinal mucosa damage, increased the expression of ZO-1 in small intestine, further decreased the levels of gut endotoxin in serum and liver compared with HF group. Endotoxin-associated genes such as TLR4 and Myd88, pro-inflammation genes such as MCP-1, TNF-alpha, IL-1, IL-2, IL-6 and IFN-gamma in liver or epididymal fat were obviously downregulated after NaB intervention. Liver inflammation and fat accumulation were ameliorated, the levels of TG and cholesterol in liver were decreased after NaB intervention, NAS score was significantly decreased, metabolic indices such as FBG and HOMA-IR and liver function indicators ALT and AST were improved compared with HF group. CONCLUSION NaB may restore the dysbiosis of gut microbiota to attenuate steatohepatitis, which is suggested to be a potential gut microbiota modulator and therapeutic substance for NAFLD.

文摘Traditional Chinese medicine(TCM)is a precious treasure of the Chinese nation and has unique advantages in the prevention and treatment of diseases.The holistic view of TCM coincides with the new generation of medical research paradigm characterized by network and system.TCM gave birth to a new method featuring holistic and systematic"network target",a core theory and method of network pharmacology.TCM is also an important research object of network pharmacology.TCM network pharmacology,which aims to understand the network-based biological basis of complex diseases,TCM syndromes and herb treatments,plays a critical role in the origin and development process of network pharmacology.This review introduces new progresses of TCM network pharmacology in recent years,including predicting herb targets,understanding biological foundation of diseases and syndromes,network regulation mechanisms of herbal formulae,and identifying disease and syndrome biomarkers based on biological network.These studies show a trend of combining computational,experimental and clinical approaches,which is a promising direction of TCM network pharmacology research in the future.Considering that TCM network pharmacology is still a young research field,it is necessary to further standardize the research process and evaluation indicators to promote its healthy development.

文摘Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections.This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China,from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records,laboratory assessments,and chest CT scans were reviewed.As a result,none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever,cough,fatigue,etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs.The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization.These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died.The median communicable period,defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests,was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members,which even caused severe COVID-19 pneumonia.Overall,the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization.However,the communicable period could be up to three weeks and the communicated patients could develop severe illness.These results highlighted the importance of close cont

文摘A field survey was conducted to investigate the metal and arsenic contamination in soils and vegetables on four villages (Shuichuan (SCH), Beiwan (BWA), Dongwan (DWA) and Wufe (WFE)) located along, Baiyin, China, and to evaluate the possible health risks to local population through foodchain. Results show that the most significantly contaminated soils occurred upstream at SCH where Cd, Cu and As concentrations exceeded maximum allowable concentrations for Chinese agricultural soil. Further downstream the degree of contamination semi-systematically decreased in concentrations of metal. Generally, the leafy vegetables were more heavily contaminated than non-leafy vegetables. Chinese cabbage is the most severely contaminated, the concentrations of Cd exceeded the maximum permit levels (0.05 mg/kg) by 4.5 times. Bio-accumulate factor also shows that an entry of Cd to food chain plants is the greatest potential. Furthermore, the estimated daily intake amounts of the considered toxic elements (Cd, Pb and Cu) from the vegetables grown at SCH and BWA and DWA have exceeded the recommended dietary allowance levels. Thus, the vegetables grown in three villages above, which affected by Baiyin mining and smelting have a health hazard for human consumption.
