Since its initial release in 2001,the human reference genome has undergone continuous improvement in quality,and the recently released telomere-to-telomere(T2T)version-T2T-CHM13—reaches its highest level of continuit...Since its initial release in 2001,the human reference genome has undergone continuous improvement in quality,and the recently released telomere-to-telomere(T2T)version-T2T-CHM13—reaches its highest level of continuity and accuracy after 20 years of effort by working on a simplified,nearly homozygous genome of a hydatidiform mole cell line.Here,to provide an authentic complete diploid human genome reference for the Han Chinese,the largest population in the world,we assembled the genome of a male Han Chinese individual,T2T-YAO,which includes T2T assemblies of all the 22+X+M and 22+Y chromosomes in both haploids.The quality of T2T-YAO is much better than those of all currently available diploid assemblies,and its haploid version,T2T-YAO-hp,generated by selecting the better assembly for each autosome,reaches the top quality of fewer than one error per 29.5 Mb,even higher than that of T2T-CHM13.Derived from an individual living in the aboriginal region of the Han population,T2T-YAO shows clear ancestry and potential genetic continuity from the ancient ancestors.Each haplotype of T2TYAO possesses330-Mb exclusive sequences,3100 unique genes,and tens of thousands of nucleotide and structural variations as compared with CHM13,highlighting the necessity of a population-stratified reference genome.The construction of T2T-YAO,an accurate and authentic representative of the Chinese population,would enable precise delineation of genomic variations and advance our understandings in the hereditability of diseases and phenotypes,especially within the context of the unique variations of the Chinese population.展开更多
In the aerospace field,for aerospace engines and other high-end manufacturing equipment working in extreme environments,like ultrahigh temperatures,high pressure,and high-speed airflow,in situ temperature measurement ...In the aerospace field,for aerospace engines and other high-end manufacturing equipment working in extreme environments,like ultrahigh temperatures,high pressure,and high-speed airflow,in situ temperature measurement is of great importance for improving the structure design and achieving the health monitoring and the fault diagnosis of critical parts.Optical fiber sensors have the advantages of small size,easy design,corrosion resistance,anti-electromagnetic interference,and the ability to achieve distributed or quasi-distributed sensing and have broad application prospects for temperature sensing in extreme environments.In this review,first,we introduce the current research status of fiber Bragg grating-type and Fabry–Perot interferometer-type high-temperature sensors.Then we review the optical fiber hightemperature sensor encapsulation techniques,including tubular encapsulation,substrate encapsulation,and metalembedded encapsulation,and discuss the extreme environmental adaptability of different encapsulation structures.Finally,the critical technological issues that need to be solved for the application of optical fiber sensors in extreme environments are discussed.展开更多
Purpose:To report a rare case of biliary candidiasis in a host with normal immunity.Methods:Laboratory tests indicated an elevated G level of 1000 pg/ml.Microscopic examination of pathological sections demonstrated mu...Purpose:To report a rare case of biliary candidiasis in a host with normal immunity.Methods:Laboratory tests indicated an elevated G level of 1000 pg/ml.Microscopic examination of pathological sections demonstrated multiple,narrow base,budding yeast cells following Grocott's methenamine silver staining and periodic acid-Schiff staining.Subsequent sequence analysis supported the diagnosis of biliary candidiasis.Results:A 31-year-old previously healthy man developed obstructive jaundice under no obvious inducement.He experienced cholecystectomy and biliary drainage.Surgical specimens revealed granulomatous chronic inflammation in the gallbladder,liver,bile ducts and mesenteric lymph nodes.He was misdiagnosed with tuberculosis and received antituberculosis treatment.However,his condition did not improve.He was finally diagnosed with biliary candidiasis and administered the antifungal treatment with fluconazole.His symptoms improved after such treatment.Conclusions:We reported a case of biliary candidiasis that mimicked IgG4-RD in a host with normal immunity.Our findings highlighted the need for pathological diagnosis in patients with symptoms resembling IgG4-RD.展开更多

[Method] Totally 288 individuals of 43dayold black muscovy duck were randomly divided intonine treatments, four repeats each treatment and eight ducks each repeat. UsingL9 (34) orthogonal experiment design (four factors and three levels),three metabolic energy levels (12.32, 12.72, 13.12 MJ/kg), three crude protein levels (14%, 16%, 18%), three calcium levels (0.6%, 0.8%, 1.0%)and three available phosphorus levels (0.30%, 0.45%, 0.6%) were designed, and a total of nine feeding groups (C1C9) were designed according todifferent nutritional levels. Through combination of feeding test (49 d) and metabolic test (pretest 4 d and formal test 3 d), the growth performance,meat performance and apparent nutrient digestibility of ducks were determined. [Result] The ducks in experimental group C2 had the highest average daily gain, the lowest feed gain ratio, the largest body oblique length, chest width, chest depth and tibia length, which had significant or extremely significant differences with other groups (P〈0.05 or P〈0.01). The slaughter rate, semieviscerated rate, eviscerated rate and meat quality ofducks in C2 group were higher (P〈0.05). The apparent digestibility of crude protein and dietary metabolic energy in C2 group were 88.68% and63.07%, respectively, both were significantly or extremely significantly higher than the other groups(P〈0.05 or P〈0.01), while the interaction effectof calcium and phosphorus level on apparent digestibility was not obvious.
