Background:Cervical cancer has the fourth highest incidence and mortality rate of all cancers in women worldwide;让seriously harms their physical and mental health.The aim of this study was to observe the roles and preliminary mechanism of Taurine(Tau)-induced apoptosis in cervical cancer cells.Methods:Cells from the human cervical cancer cell line SiHa were transfected with the recombinant plasmid pEGFP-N1-MST1(mammalian sterile 20-like kinase 1);then,the cell proliferation activity was analyzed by the MTT assay,cell apoptosis by flow cytometry,and the related protein levels by Western blotting.Results:Tau inhibited the proliferation of SiHa cells and induced apoptosis in these cells(the apoptotic rate was 21.95%in the Tau 160 mmol/L group and 30%in the Tau 320 mmol/L group),upregulated the expression of the MST1(control,0.53;Tau 40-320 mmol/L groups,0.84-1.45)and Bax(control,0.45;Tau 40-320 mmol/L groups,0.64-1.51)proteins(P<0.01),and downregulated the expression of Bcl-2(control,1.28,Tau 40-320 mmol/L groups,0.93-0.47)(P<0.01).The overexpression of MST1 promoted the apoptosis of SiHa cells,enhanced the apoptosis-inductive effects of Tau(P<0.01),upregulated the expression of the proapoptotic proteins p73,p53,PUMA(p53 upregulated modulator of apoptosis),and caspase-3,and promoted the phosphorylation of YAP(Yes-associated protein).Conclusions:Tau inhibited the proliferation and induced the apoptosis of cervical cancer SiHa cells.The MST1 protein plays an important role in the Tau-induced apoptosis of cervical cancer cells. 展开更多

目的探讨地塞米松对哮喘小鼠中性粒细胞性气道炎症和高迁移率族蛋白B1(HMGB1)表达的影响。方法40只SPF级雌性Balb/c小鼠随机分为对照组、哮喘组、1 mg/kg地塞米松干预组、5 mg/kg地塞米松干预组,每组10只,采用无鸡蛋卵清蛋白诱导建立哮... 目的探讨地塞米松对哮喘小鼠中性粒细胞性气道炎症和高迁移率族蛋白B1(HMGB1)表达的影响。方法40只SPF级雌性Balb/c小鼠随机分为对照组、哮喘组、1 mg/kg地塞米松干预组、5 mg/kg地塞米松干预组,每组10只,采用无鸡蛋卵清蛋白诱导建立哮喘模型。采用小鼠有创肺功能仪测定小鼠气道高反应性;检测小鼠支气管肺泡灌洗液(BALF)中炎性细胞的数量及分类;HE染色观察肺组织病理学变化;免疫组化和Western blotting检测小鼠气道上皮及肺组织HMGB1的表达。结果与对照组比较,哮喘组小鼠气道反应性明显增强(P<0.01),BALF中炎性细胞总数明显增加,且以中性粒细胞为主(P<0.001),气道周围大量炎性细胞浸润,上皮细胞不同程度坏死脱落,气道上皮及肺组织HMGB1表达明显增加(P<0.05)。与哮喘组比较,地塞米松干预组小鼠气道反应性降低(P<0.05),BALF中炎性细胞总数和中性粒细胞计数均减少(P<0.01),气道周围炎性细胞浸润明显减少,气道炎症明显减轻,气道上皮及肺组织HMGB1表达差异无统计学意义(P>0.05)。结论以中性粒细胞性气道炎症为主的哮喘小鼠气道上皮及肺组织中HMGB1表达明显增加,地塞米松干预后,哮喘小鼠仍表现出较强的中性粒细胞性气道炎症及肺组织HMGB1高表达,提示HMGB1可能与哮喘中性粒细胞性气道炎症和糖皮质激素抵抗有关。 展开更多

MRI定量肝脏及心脏铁过载对地中海贫血全相合造血干细胞移植的影响 被引量:5

背景:大部分的重型β-地中海贫血患儿都存在铁过载的情况,而铁过载可对造血干细胞移植产生不利影响。目的:利用MRI T2*检测肝脏及心脏铁过载情况,从而评估肝脏、心脏铁过载对重型β-地中海贫血患儿全相合异基因造血干细胞移植的影响。方... 背景:大部分的重型β-地中海贫血患儿都存在铁过载的情况,而铁过载可对造血干细胞移植产生不利影响。目的:利用MRI T2*检测肝脏及心脏铁过载情况,从而评估肝脏、心脏铁过载对重型β-地中海贫血患儿全相合异基因造血干细胞移植的影响。方法:对81例全相合异基因造血干细胞移植、年龄>3岁,能配合MRI检测的重型β-地中海贫血患儿,进行肝脏及心脏MRI T2*检测。根据检测结果计算出肝脏及心脏铁浓度,作为评价肝心铁过载的指标,并将肝脏、心脏铁浓度与患儿血清铁蛋白、造血重建时间、死亡率、植入率、移植后常见并发症的发病率,如移植物抗宿主病、感染、免疫性溶血、全血细胞减少、肝静脉阻塞病、败血症进行相关因素分析。结果与结论:肝脏铁浓度与血红蛋白植入时间呈正相关(r=0.229,P=0.043),心脏铁浓度与死亡率呈正相关(r=0.266,P=0.017),血清铁蛋白与植入率呈负相关(r=-0.289,P=0.009),血清铁蛋白与败血症发病率呈正相关(r=0.251,P=0.024),血清铁蛋白与全血细胞减少发病率呈正相关(r=0.276,P=0.013)。由此可见,铁过载可对重型β-地中海贫血患儿全相合异基因造血干细胞移植产生不利影响。因此,对行造血干细胞移植的β-地中海贫血患儿综合进行血清铁蛋白、肝脏及心脏铁过载的评估非常必要。 展开更多

Nkx2-1 (Nkx homeobox-1 gene), also known as TTF-1 (thyroid transcription factor-1), is a tissue-specific transcription factor of the thyroid, lung, and ventral forebrain. While it has been shown to play a critical role in lung development and lung cancer differentiation and morphogenesis, molecular mechanisms mediating Nkx2-1 cell- and tissue-specific expression in normal and cancerous lungs have yet to be fully elucidated. The recent identification of prognostic biomarkers in lung cancer, particularly in lung adenocarcinoma (ADC), and the different reactivity of patients to chemotherapeutic drugs have opened new avenues for evaluating patient survival and the development of novel effective therapeutic strategies. The function of Nkx2-1 as a proto-oncogene was recently characterized and the gene is implicated as a contributory factor in lung cancer development. In this review, we summarize the role of this transcription factor in the development, diagnosis, and prognosis of lung cancer in the hope of providing insights into the utility of Nkx2-1 as a novel biomarker of lung cancer. 展开更多

目的:探讨血清铁蛋白水平(SF)在异基因造血干细胞移植(allo-HSCT)后血小板减少中的临床意义。方法:回顾性分析本院35例移植后血小板减少患者临床资料及移植情况,并从血小板计数正常的424例患者中按照年龄及性别匹配方式选取35名同时期... 目的:探讨血清铁蛋白水平(SF)在异基因造血干细胞移植(allo-HSCT)后血小板减少中的临床意义。方法:回顾性分析本院35例移植后血小板减少患者临床资料及移植情况,并从血小板计数正常的424例患者中按照年龄及性别匹配方式选取35名同时期移植患者作为对照,参照移植前SF水平,通过卡方检验和Fisher精确检验,以及单因素、多因素Logistic回归分析分析危险因素。采用Kaplan-Meier模型估算生存曲线以探讨其临床意义,并应用ROC曲线验证SF的预测效能。结果:移植后血小板减少组与对照组相比,移植前SF水平显著增高(P=0.001)。多因素分析结果显示,移植前SF水平为移植后血小板减少的危险因素。ROC曲线分析表明,SF水平可作为移植后血小板减少的预测指标。SF≥1 000 ng/ml的移植后血小板减少患者有较高的死亡风险(P=0.014)。结论:移植前的SF水平与移植后患者血小板减少的发生与预后相关,检测SF水平可以为移植后血小板减少的干预提供指导。 展开更多

中速磁悬浮列车的滑模周期自适应学习控制方法 被引量:1

为了提高中速磁悬浮列车的运行控制性能,考虑到中速磁悬浮列车沿固定线路往返运行时的周期特性,提出了一种基于滑模周期自适应学习的中速磁悬浮列车运行控制算法,对应的滑模周期自适应控制器由等效比例积分微分(PID)和速度前馈控制部分... 为了提高中速磁悬浮列车的运行控制性能,考虑到中速磁悬浮列车沿固定线路往返运行时的周期特性,提出了一种基于滑模周期自适应学习的中速磁悬浮列车运行控制算法,对应的滑模周期自适应控制器由等效比例积分微分(PID)和速度前馈控制部分、磁阻力与空气阻力补偿部分以及坡道阻力滑模周期自适应补偿部分组成;采用粒子群优化算法辨识了运行控制器的参数,采用滑模周期自适应学习控制器学习上一周期内的列车运行信息,实时估计并补偿列车运行过程中的坡道阻力,消除坡道阻力对列车运行性能的影响;利用全长5 076 m的中速磁悬浮列车半实物仿真试验线进行了数值仿真,并将设计的滑模周期自适应控制器与PID控制器进行仿真对比。仿真结果显示:滑模周期自适应控制器和PID控制器作用下的列车最大位置跟踪误差分别为0.004和0.007 m,最大速度跟踪误差分别为0.007和0.036 m·s^(-1);经过4个迭代周期后,滑模周期自适应控制器已准确地估计了给定的坡道阻力;在控制系统受到扰动的情况下,滑模周期自适应控制器与PID控制器作用下的位置与速度跟踪曲线均有波动,相比于PID控制器,滑模周期自适应控制器控制下的跟踪曲线波动更小。可见,相比于传统的PID控制算法,提出的滑模周期自适应学习控制方法可以提高中速磁悬浮列车的运行控制性能。 展开更多

Purpose:The purpose of this study was to assess and compare elbow range of motion,triceps extension strength and functional results of type C(AO/OTA)distal humerus fractures treated with bilateral triceps tendon(BTT)approach and olecranon osteotomy(00).At the same time,we are also trying to know whether BTT approach can provide sufficient vision for comminuted intra-articular fractures of the distal humerus,and whether it is convenient to convert to the treatment to total elbow arthroplasty(TEA)or 00.Methods:Patients treated with 00 and BTT approaches for type C distal humerus fractures between July 2014 and December 2017 were retrospectively reviewed.Inclusion criteria include:(1)patients'age were more than 18 years old,(2)follow-up was no less than 6 months,and(3)patients were diagnosed with type C fractures(based on the AO/OTA classification).Exclusion criteria include:(1)open fractures(Gustillo type 2 or type 3),(2)treated by other approaches,and(3)presented with combined injuries of ipsilateral upper extremities,such as ulnar nerve.Elbow range of motion and triceps extension strength testing were completely valuated,when the fractures had healed.Assessment of functional results using the Mayo elbow performance score and complications were conducted in final follow-up.The data were compared using the two tailed Student's t-test.All data were presented as mean±standard deviation.Results:Eighty-six patients of type C distal humerus fractures,treated by 00 and BTT approach were retrospectively reviewed between July 2014 and December 2017.Fifty-five distal humerus fractures(23 males and 32 females,mean age 52.7 years)treated by BTT approach or 00 were included in this study.There were 10 fractures of type Cl,16 type C2 and 29 type C3 according to the AO/OTA classification.Patients were divided into two surgical approach groups chosen by the operators:BTT group(28 patients)and 00 group(27 patients).And the mean follow-up time of all patients was 15.6 months(range,6-36 months).Three cases in BTT group were convert 展开更多

Alpinia officinarum Hance extract alleviates particulate matter-induced lung injury in mice

Objective: To evaluate the effect of Alpinia officinarum Hance(A. officinarum) extract on lung injury caused by particulate matter(PM). Methods: The Kunming mice were intranasally instilled with PM and treated with A. officinarum extract for 3 weeks. Bronchoalveolar lavage fluid, blood and lung samples were collected for biochemical, serological and histopathological studies. Results: Serological analysis showed that albumin levels, lactate dehydrogenase and alkaline phosphatase activities in bronchoalveolar lavage fluid were significantly reduced after administrations of 50, 100 and 200 mg/kg of A. officinarum extracts to the PM injured mice. Markers of oxidative stress, nitric oxide, malondialdehyde levels and nitric oxide synthase activities, were significantly decreased. Correspondingly, total superoxide dismutase activity was improved dramatically. The expressions of interleukin-6 and tumor necrosis factor alpha were also down-regulated obviously. In addition, pathological sections of lung tissue showed that A. officinarum could reduce the infiltration of inflammatory cells, pulmonary edema and pulmonary fibrosis. These results showed that A. officinarum extract could alleviate PMinduced lung injury via reducing the permeability of cell membranes in lung tissue, eliminating oxidative stress and relieving inflammatory response.Conclusions: A. officinarum extract was an efficient treatment for PM-induced lung injury in mice, and it may be a promising therapeutic agent in future. 展开更多
