穿支动脉粥样硬化病(branch athero-matous disease,BAD)是急性缺血性卒中的常见类型,最早由Caplan[1]于1989年提出,Caplan认为BAD是一个“被忽视的、疏于研究且未被应用的概念”。常见的穿支动脉包括豆纹动脉(lenticulostriate arterie... 穿支动脉粥样硬化病(branch athero-matous disease,BAD)是急性缺血性卒中的常见类型,最早由Caplan[1]于1989年提出,Caplan认为BAD是一个“被忽视的、疏于研究且未被应用的概念”。常见的穿支动脉包括豆纹动脉(lenticulostriate arteries,LSA)、脑桥旁正中动脉(paramedian pontine arteries,P PA)、丘脑膝状体动脉、脉络膜前动脉、Heubner’s动脉和丘脑穿通动脉等。BAD占所有缺血性卒中病因的10%~15%,临床上多表现为以进行性运动功能缺失(progressive motor deficiency,PDM)为主的早期神经功能恶化(early neurological deterioration,END),多发生在发病48~72 h,临床预后差[2-3]。传统影像学很难发现BAD的血管病变,导致临床上早期诊断困难。基于临床及头颅MRI表现,目前临床研究较多的是发生在LSA和PPA部位的BAD。 展开更多

地球表层是由山水林田湖草等要素构成的互动和互馈的复杂系统,这些要素中化学元素的互动和互馈过程均可驱动化学元素分布模式的变化。本文以系统的1∶25万土地质量地球化学调查数据为基础,讨论了中国西南地区土壤中常量元素、亲生物元... 地球表层是由山水林田湖草等要素构成的互动和互馈的复杂系统,这些要素中化学元素的互动和互馈过程均可驱动化学元素分布模式的变化。本文以系统的1∶25万土地质量地球化学调查数据为基础,讨论了中国西南地区土壤中常量元素、亲生物元素、卤族元素、重金属元素分布模式的驱动机制和互动过程。结果发现成土母质决定了土地资源中化学元素的自然状况,表生地球化学动力学过程重塑了表层土壤中元素分布模式,生物地球化学过程驱动了土地质量的演化趋势,强烈的人类活动(如矿业活动)破坏了元素分布的自然演化规律。表生地球化学动力学理论的提出将引领山水林田湖草生命共同体知识体系构建和土地资源地球化学综合调查技术的革新。 展开更多

There is considerable potential for integrating transarterial chemoembolization(TACE),programmed death-(ligand)1(PD-[L]1)inhibitors,and molecular targeted treatments(MTT)in hepatocellular carcinoma(HCC).It is necessary to investigate the therapeutic efficacy and safety of TACE combined with PD-(L)1 inhibitors and MTT in real-world situations.In this nationwide,retrospective,cohort study,826 HCC patients receiving either TACE plus PD-(L)1 blockades and MTT(combination group,n=376)or TACE monotherapy(monotherapy group,n=450)were included from January 2018 to May 2021.The primary endpoint was progression-free survival(PFS)according to modified RECIST.The secondary outcomes included overall survival(OS),objective response rate(ORR),and safety.We performed propensity score matching approaches to reduce bias between two groups.After matching,228 pairs were included with a predominantly advanced disease population.Median PFS in combination group was 9.5 months(95%confidence interval[CI],8.4-11.0)versus 8.0 months(95%CI,6.6-9.5)(adjusted hazard ratio[HR],0.70,P=0.002).OS and ORR were also significantly higher in combination group(median OS,19.2[16.1-27.3]vs.15.7 months[13.0-20.2];adjusted HR,0.63,P=0.001;ORR,60.1%vs.32.0%;P<0.001).Grade 3/4 adverse events were observed at a rate of 15.8%and 7.5%in combination and monotherapy groups,respectively.Our results suggest that TACE plus PD-(L)1 blockades and MTT could significantly improve PFS,OS,and ORR versus TACE monotherapy for Chinese patients with predominantly advanced HCC in real-world practice,with an acceptable safety profile. 展开更多

为探究重金属、微塑料和二者的复合效应对农作物种子生长特性的影响,选取水稻种子为实验对象,探究微塑料聚苯乙烯(mPS)和聚对苯二甲酸类塑料(mPET,100、300、500、1000 mg·L^-1和1500 mg·L^-1)与重金属镉(Cd,2、5、10 mg·... 为探究重金属、微塑料和二者的复合效应对农作物种子生长特性的影响,选取水稻种子为实验对象,探究微塑料聚苯乙烯(mPS)和聚对苯二甲酸类塑料(mPET,100、300、500、1000 mg·L^-1和1500 mg·L^-1)与重金属镉(Cd,2、5、10 mg·L^-1和50 mg·L^-1)单一及复合对种子萌发及生长的影响。结果表明:单一实验中,镉胁迫对水稻种子萌发特性的影响基本表现为“低促高抑”的规律,随着镉浓度的升高,发芽率、发芽势、发芽指数、活力指数以及根长和芽长等各项指标均呈现下降的趋势;单一微塑料暴露下,总体表现为低浓度(100 mg·L^-1)促进发芽,中浓度(500 mg·L^-1)抑制发芽,高浓度(>1000 mg·L^-1)无影响,即发芽率与对照组持平;两种微塑料对水稻种子的活力指数、根长和芽长均有所促进。微塑料(100 mg·L^-1和500 mg·L^-1)和镉(2 mg·L^-1和10 mg·L^-1)复合污染实验中,与对照组相比,低浓度MPs(100 mg·L^-1)-Cd(2 mg·L^-1)复合产生的协同作用促进了水稻种子发芽,其余复合污染对种子的发芽均表现为拮抗作用;对于低浓度的微塑料(100 mg·L^-1),其与镉复合会对水稻种子芽和根的生长起协同作用,只有mPS(100 mg·L^-1)与高浓度的镉(10 mg·L^-1)复合才会对根的生长起到拮抗作用;高浓度的微塑料与高浓度的镉复合后没有对水稻种子根和芽产生影响。研究表明,相比于单一污染,微塑料-镉复合效应对水稻种子生长特性、根长和芽长的影响总体表现为拮抗作用,一定程度上降低了单一污染物的毒害作用。 展开更多

The Large sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST) general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio(SNR) ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters: late A,FGK-type stars with high quality spectra(1 061 918 entries), A-type stars(100 073 entries), and M-type stars(121 522 entries). This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided. 展开更多

Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. This evidencebased consensus would be valuable to assist front-line surgeons and medical specialists in the optimal management of GLM. 展开更多

沙库巴曲缬沙坦对射血分数降低性心力衰竭患者心力衰竭易损期的疗效 被引量:25

目的观察沙库巴曲缬沙坦对射血分数降低性心力衰竭(HF-REF)患者心力衰竭易损期的治疗效果。方法患者分为对照组32例,试验组31例。对照组在常规治疗方法基础上给予盐酸贝那普利片,每次10 mg,每日1次,口服。试验组在常规治疗方法基础上给... 目的观察沙库巴曲缬沙坦对射血分数降低性心力衰竭(HF-REF)患者心力衰竭易损期的治疗效果。方法患者分为对照组32例,试验组31例。对照组在常规治疗方法基础上给予盐酸贝那普利片,每次10 mg,每日1次,口服。试验组在常规治疗方法基础上给予沙库巴曲颉沙坦钠片,停用其他血管紧张素转化酶抑制剂、血管紧张素Ⅱ受体阻滞剂后48 h开始给药,起始剂量50 mg,每日2次,口服;服药2周后,提升剂量至100 mg,每日2次,口服;之后逐步提升剂量至200 mg,每日2次,口服。2组的治疗效果检测时间均为出院后3个月(心力衰竭易损期)。比较2组的心功能指标、血清学指标、运动能力指标、预后指标、治疗效果和药物不良反应发生情况。结果治疗后,试验组和对照组的左心室射血分数(LVEF)分别为(55.13±6.35)%和(49.77±6.18)%,心脏指数(CI)分别为(3.63±0.15)和(3.48±0.27)L·min^(-1)·m^(-2),血清氨基末端脑钠肽前体(NT-proBNP)分别为(380.61±98.37)和(462.35±107.42)pg·mL^(-1),6 min步行试验(6MWT)分别为(360.22±61.15)和(315.65±54.51)m,差异有统计学意义(均P<0.05)。试验组和对照组的心力衰竭易损期再入院率、心力衰竭易损期心血管不良事件发生率比较,差异有统计学意义(均P<0.05),试验组和对照组的心力衰竭易损期心血管不良事件死亡率比较,差异无统计学意义(P>0.05)。试验组出现症状性低血压2例,高钾血症1例,血管性水肿1例,药物不良反应发生率为12.90%(4例/31例);对照组出现症状性低血压1例,肝肾功能损伤1例,高钾血症2例,血管性水肿1例,药物不良反应发生率为15.63%(5例/32例)。2组药物不良反应发生率比较,差异无统计学意义(P>0.05)。结论对于HF-REF患者,在常规治疗方法的基础上使用沙库巴曲缬沙坦,有助于促进住院期间的治疗效果提升,改善心功能指标,预防患者在心力衰竭易损期之内的各项不良事件,药物的使用安全性� 展开更多

目的观察通络活血虫类药(全蝎、蜈蚣、守宫)对乏氧环境下肺癌血管生成相关细胞因子的影响。方法采用低氧工作站体外模拟肿瘤乏氧微环境培养A549肺癌细胞,加入不同浓度全蝎、蜈蚣、守宫药物血清,MTT法检测细胞增殖,筛选最佳药物浓度及作... 目的观察通络活血虫类药(全蝎、蜈蚣、守宫)对乏氧环境下肺癌血管生成相关细胞因子的影响。方法采用低氧工作站体外模拟肿瘤乏氧微环境培养A549肺癌细胞,加入不同浓度全蝎、蜈蚣、守宫药物血清,MTT法检测细胞增殖,筛选最佳药物浓度及作用时间。A549肺癌细胞经3种药物血清处理后,收集细胞培养上清液,ELISA检测上清液中血管内皮生长因子(VEGF)、转化生长因子-β1(TGF-β1)、碱性成纤维细胞生长因子(b FGF)的含量。结果 3种药物血清对常氧及乏氧培养的A549肺癌细胞增殖均有一定的抑制作用。最终筛选7.5%浓度药物血清、作用24 h进行后续实验。3种药物血清较对照组A549细胞VEGF、TGF-β1、b FGF含量均降低(P<0.05,P<0.01)。结论 3种虫类药在常氧和乏氧状态下均有抑制癌细胞和对血管生成相关细胞因子调控的作用。 展开更多

Finding the electromagnetic (EM) counterpart of binary compact star merger, especially the binary neutron star (BNS) merger, is critically important for gravitational wave (GW) astronomy, cosmology and fundament... Finding the electromagnetic (EM) counterpart of binary compact star merger, especially the binary neutron star (BNS) merger, is critically important for gravitational wave (GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017, Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart, GRB 170817A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope (HE) onboard Insight-HXMT (Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart (SSS17a/AT2017gfo) with very large collection area (M000 cm2) and microsecond time resolution in 0.2-5 MeV. In addition, Insight-HXMT quickly implemented a Target of Opportunity (TOO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy (0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817A. Meanwhile, Insight-HXMT/HE provides one of the most stringent constraints (-10-7 to 104 erg/cm2/s) for both GRB170817A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. Therefore the observation of Insight-HXMT constitutes an important chapter in the full context of multi-wavelength and multi-messenger observation of this historical GW event. 展开更多

AIM: To assess the effect of notoginsenoside R1 on hepatic microcirculatory disturbance induced by gut ischemia/reperfusion (I/R) in mice. METHODS: The superior mesenteric artery (SMA) of C57/BL mice was ligated for 15 min to induce gut ischemia followed by 30-rain reperfusion. In another set of experiments, R1 was continuously infused (10 mg/kg per hour) from 10 min before I/R until the end of the investigation to study the influence of R1 on hepatic microcirculatory disturbance induced by gut I/R. Hepatic microcirculation was observed by inverted microscopy, and the vascular diameter, red blood cell (RBC) velocity and sinusoid perfusion were estimated. Leukocyte rolling and adhesion were observed under a laser confocal microscope. Thirty and 60 min after reperfusion, lactate dehydrogenase (LDH), alanine aminotransferase (ALl') and aspartate transaminase (AST) in peripheral blood were determined. The expression of adhesion molecules CD11b/CD18 in neutrophils and tumor necrosis factor- alpha (TNF-α), interleukin-6 (IL-6) and monocyte chemotactic protein-1 (MCP-1) in plasma were evaluated by flow Oltometry. E-selectin and intercellular adhesion molecule-1 (ICAM-1) in hepatic tissue were examined by immunofluorescence.RESULTS: After gut I/R, the diameters of terminal portal venules and central veins, RBC velocity and the number of perfused sinusoids were decreased, while the leukocyte rolling and adhesion, the expression of E-selectin in hepatic vessels and CD18 in neutrophils, IL-6, MCP-1, LDH, ALT and AST were increased. R1 treatment attenuated these alterations except for IL-6 and MCP-1. CONCLUSION: R1 prevents I/R-induced hepatic microcirculation disturbance and hepatocyte injury, The effect of R1 is related to its inhibition of leukocyte rolling and adhesion by inhibiting the expression of E-selectin in endothelium and CD18 in neutrophils. 展开更多

In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. In particular, its wide field monitoring capabilities will be highly instrumental to detect the electro-magnetic counterparts of gravitational wave sources.The paper provides a detailed description of:(1) the technological and technical aspects, and the expected performance of the instruments of the scientific payload;(2) the elements and functions of the mission, from the spacecraft to the ground segment. 展开更多

We describe the current plans for a spectroscopic survey of millions of stars in the Milky Way galaxy using the Guo Shou Jing Telescope (GSJT, formerly calledthe Large sky Area Multi-Object fiber Spectroscopic Telescope -- LAMOST). The survey will obtain spectra for 2.5 million stars brighter than r 〈 19 during dark/grey time, and 5 million stars brighter than r 〈 17 or J 〈 16 on nights that are moonlit or have low transparency. The survey will begin in the fall of 2012, and will run for at least four years. The telescope's design constrains the optimal declination range for observations to 10~ 〈 di 〈 50~, and site conditions lead to an emphasis on stars in the direction of the Galactic anticenter. The survey is divided into three parts with different target selection strategies: disk, anticenter, and spheroid. The resulting dataset will be used to study the merger history of the Milky Way, the substructure and evolution of the disks, the nature of the first generation of stars through identification of the lowest metallicity stars, and star formation through study of open clusters and OB associations. Detailed design of the LAMOST Experiment for Galactic Understanding and Exploration (LEGUE) survey will be completed in summer 2012, after a review of the results of the pilot survey. 展开更多

目的研究麻黄-甘草药对对过敏性哮喘的影响,并初步探讨其作用机制。方法建立屋尘螨(HDM)诱导的过敏性哮喘模型,造模同时给予麻黄-甘草药对提取物(MG)。使用无创肺功能仪检测小鼠气道反应性,HE染色观察小鼠肺组织病理学变化,Masson染色... 目的研究麻黄-甘草药对对过敏性哮喘的影响,并初步探讨其作用机制。方法建立屋尘螨(HDM)诱导的过敏性哮喘模型,造模同时给予麻黄-甘草药对提取物(MG)。使用无创肺功能仪检测小鼠气道反应性,HE染色观察小鼠肺组织病理学变化,Masson染色观察小鼠肺组织的胶原沉积情况,进行血液嗜酸性粒细胞(EOS)计数,ELISA法检测支气管肺泡灌洗液(BALF)及肺组织IL-4、IL-5、IL-13、TGF-β1及血清IgE的表达,免疫组化检测肺组织E-cadherin、N-cadherin、Vimentin、α-SMA和TGF-β1蛋白表达情况。结果 MG可显著降低气道高反应性,抑制BALF及肺组织IL-4、IL-5和IL-13的表达,降低血液EOS及血清IgE的表达,并且减轻肺组织的炎症浸润及胶原沉积。进一步结果表明MG可抑制肺组织TGF-β1的表达,上调E-cadherin以及下调N-cadherin、Vimentin、α-SMA的表达。结论 MG能够通过抑制支气管上皮间质转化,从而缓解气道的炎症状态以及改善哮喘的气道重塑,进而发挥对过敏性哮喘的治疗作用。 展开更多

AIM:To investigate the effect of total salvianolic acid(TSA) on ischemia-reperfusion(I/R)-induced rat mesenteric microcirculatory dysfunctions.METHODS:Male Wistar rats were randomly distributed into 5 groups(n = 6 each):Sham group and I/R group(infused with saline),TSA group,TSA + I/R group and I/R + TSA group(infused with TSA,5 mg/kg per hour).Mesenteric I/R were conducted by a ligation of the mesenteric artery and vein(10 min) and subsequent release of the occlusion.TSA was continuously infused either starting from 10 min before the ischemia or 10 min after reperfusion.Changes in mesenteric microcirculatory variables,including diameter of venule,velocity of red blood cells in venule,leukocyte adhesion,free radicals released from venule,albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope.Meanwhile,the expression of adhesion molecules CD11b/CD18 on neutrophils was evaluated by flow cytometry.Ultrastructural evidence of mesenteric venules damage was assessed after microcirculation observation.RESULTS:I/R led to multiple responses in mesenteric post-capillary venules,including a significant increase in the adhesion of leukocytes,production of oxygen radicals in the venular wall,albumin efflux and enhanced mast cell degranulation in vivo.All the I/R-induced manifestations were significantly reduced by pre-or post-treatment with TSA,with the exception that the I/R-induced increase in mast cell degranulation was inhibited only by pre-treatment with TSA.Moreover,preor post-treatment with TSA significantly attenuated the expression of CD11b/CD18 on neutrophils,reducing the increase in the number of caveolae in the endothelial cells of mesentery post-capillary venules induced by I/R.CONCLUSION:The results demonstrated that TSA protects from and ameliorates the microcirculation disturbance induced by I/R,which was associated with TSA inhibiting the production of oxygen-free radicals in the venular wall and the expression of CD11b/CD18 on neutrophils. 展开更多

【目的】本研究旨在利用Oxford Nanopore测序技术组装和注释东方蜜蜂微孢子虫Nosema ceranae的高质量全长转录组。【方法】采用Nanopore PromethION系统对东方蜜蜂微孢子虫的纯净孢子进行转录组测序。通过识别每条clean read两端引物鉴... 【目的】本研究旨在利用Oxford Nanopore测序技术组装和注释东方蜜蜂微孢子虫Nosema ceranae的高质量全长转录组。【方法】采用Nanopore PromethION系统对东方蜜蜂微孢子虫的纯净孢子进行转录组测序。通过识别每条clean read两端引物鉴定全长转录本序列。利用Blast工具将全长转录本比对Nr,Swiss-Prot,KOG,eggNOG,Pfam,GO和KEGG数据库,获得相应注释信息。分别利用蛋白结构域分析方法CPC,CNCI,CPAT和Pfam对长链非编码RNA(long noncoding RNA,lncRNA)进行预测,获得高可信度lncRNA。利用CPM(counts per million)法计算每一条全长转录本的表达量。【结果】利用Nanopore PromethION系统对东方蜜蜂微孢子虫转录组测序共测得6988795条raw reads,经质控获得6953469条clean reads,其中包含5143999条全长转录本。共鉴定到10243条非冗余全长转录本,N50和平均读长分别为1042 bp和894 bp,最大读长为4855 bp。有9342,4038,4283,2569,4859和3450条全长转录本分别注释到Nr,KOG,eggNOG,Pfam,GO和KEGG数据库。注释到东方蜜蜂微孢子虫、蜜蜂微孢子虫Nosema apis和家蚕微孢子虫Nosema bombycis的全长转录本数量最多。共鉴定到87条高可信度lncRNA,包含49条正义链lncRNA(sense lncRNA)、25条反义链lncRNA(anti-sense lncRNA)和13条基因间区lncRNA。本研究的测序量足以检测到全部表达的全长转录本,全长转录本的表达量(CPM)范围在0.1到10000以上。【结论】本研究构建和注释了东方蜜蜂微孢子虫的高质量全长转录组数据,可为病原的比较转录组分析、转录本的可变剪接和可变腺苷酸化分析、简单重复序列(simple sequence repeat,SSR)位点挖掘、基因结构优化以及基因全长序列克隆及功能研究提供关键基础。 展开更多

目的:观察双参宁心胶囊通过调控线粒体三磷酸腺苷(ATP)敏感性钾通道来减轻大鼠心肌缺血/再灌注损伤。方法:成年雄性SD大鼠56只,随机分为假手术组(sham),模型组(model),双参宁心组(SSNX)给予SSNX 90 mg·kg^-1,线粒体ATP敏感性钾通道... 目的:观察双参宁心胶囊通过调控线粒体三磷酸腺苷(ATP)敏感性钾通道来减轻大鼠心肌缺血/再灌注损伤。方法:成年雄性SD大鼠56只,随机分为假手术组(sham),模型组(model),双参宁心组(SSNX)给予SSNX 90 mg·kg^-1,线粒体ATP敏感性钾通道(MitoKATP)通道抑制剂组(SSNX+5-HD),给予SSNX 90 mg·kg^-1和5-HD 5 mg·kg^-1;每组各14只。除假手术组外其余3组均阻断冠状动脉左前降支(LAD)45 min,分别于再灌注后3 h后处死,氯化2,3,5-三苯基四氮唑(TTC)染色观察心肌缺血和梗死面积,苏木素-伊红(HE)染色观察心肌组织损伤程度。试剂盒检测血清乳酸盐脱氢酶(LDH),肌酸激酶(CK)及肌酸激酶同工酶(CK-MB)的活性。透射电镜下观察心肌细胞线粒体及线粒体自噬超微结构变化,荧光探针法检测心肌细胞中线粒体膜电位水平的变化。结果:与sham组比较,model组心肌梗死面积与心肌缺血面积百分率明显增大,心肌组织排列紊乱,疏松,个别心肌纤维断裂,心肌细胞坏死,血清中CK,CK-MB,LDH活性显著升高(P<0.01),线粒体膜电位显著降低(P<0.01),透射电镜观察线粒体结构破坏明显。与model组比较,SSNX组心肌组织排列有序,少数区域细胞水肿轻度变性,心肌梗死面积与心肌缺血面积百分率显著降低,血清中CK,CK-MB,LDH活性显著降低(P<0.01),线粒体膜电位升高(P<0.01),SSNX+5-HD组心肌组织排列轻度无序,部分区域细胞水肿轻度变性,心肌梗死面积与心肌缺血面积百分率显著降低,血清中CK,CK-MB,LDH活性显著降低(P<0.01),线粒体膜电位升高(P<0.01)。而SSNX+5-HD组较SSNX组,血清中CK,CK-MB,LDH活性显著升高(P<0.01),心肌梗死面积与心肌缺血面积百分率显著增大,线粒体膜电位有所降低(P<0.05)。结论:双参宁心胶囊通过开放线粒体ATP敏感性钾通道来保护大鼠心肌缺血再灌注损伤。 展开更多
