Dysregulated expression of microRNAs (miRNAs) in various tissues has been associated with a variety of diseases, including cancers. Here we demonstrate that miRNAs are present in the serum and plasma of humans and other animals such as mice, rats, bovine fetuses, calves, and horses. The levels of miRNAs in serum are stable, reproducible, and consistent among individuals of the same species. Employing Solexa, we sequenced all serum miRNAs of healthy Chinese subjects and found over 100 and 91 serum miRNAs in male and female subjects, respectively. We also identified specific expression patterns of serum miRNAs for lung cancer, colorectal cancer, and diabetes, providing evidence that serum miRNAs contain fingerprints for various diseases. Two non-small cell lung cancer-specific serum miRNAs obtained by Solexa were further validated in an independent trial of 75 healthy donors and 152 cancer patients, using quantitative reverse transcription polymerase chain reaction assays. Through these analyses, we conclude that serum miRNAs can serve as potential biomarkers for the detection of various cancers and other diseases. 展开更多

Intelligent manufacturing is a general concept that is under continuous development. It can be categorized into three basic paradigms: digital manufacturing, digital-networked manufacturing, and newgeneration intelligent manufacturing. New-generation intelligent manufacturing represents an indepth integration of new-generation artificial intelligence (AI) technology and advanced manufacturing technology. It runs through every link in the full life-cycle of design, production, product, and service. The concept also relates to the optimization and integration of corresponding systems; the continuous improvement of enterprises' product quality, performance, and service levels; and reduction in resources consumption. New-generation intelligent manufacturing acts as the core driving force of the new indus- trial revolution and will continue to he the main pathway for the transformation and upgrading of the manufacturing industry in the decades to come. Human-cyher-physical systems (HCPSs) reveal the tech- nological mechanisms of new-generation intelligent manufacturing and can effectively guide related the- oretical research and engineering practice. Given the sequential development, cross interaction, and iterative upgrading characteristics of the three basic paradigms of intelligent manufacturing, a technol- ogy roadmap for "parallel promotion and integrated development" should he developed in order to drive forward the intelligent transformation of the manufacturing industry in China. 展开更多

The primary pattern of the late Cenozoic to the present tectonic deformation of China is characterized by relative movements and interactions of tectonic blocks. Active tectonic blocks are geological units that have been separated from each other by active tectonic zones. Boundaries between blocks are the highest gradient of differential movement. Most of tectonic activity occurs on boundaries of the blocks. Earthquakes are results of abrupt releases of accumulated strain energy that reaches the threshold of strength of the earth's crust. Boundaries of tectonic blocks are the locations of most discontinuous deformation and highest gradient of stress accumulation, thus are the most likely places for strain energy accumulation and releases, and in turn, devastating earthquakes. Almost all earthquakes of magnitude greater than 8 and 80%-90% of earthquakes of magnitude over 7 occur along boundaries of active tectonic blocks. This fact indicates that differential movements and interactions of active tectonic blocks are the primary mechanism for the occurrences of devastating earthquakes. 展开更多

Grain weight is a major determinant of crop grain yield and is controlled by naturally occurring quantitative trait loci (QTLs). We earlier identified a major QTL that controls rice grain width and weight, GW5, which was mapped to a recombination hotspot on rice chromosome 5. To gain a better understanding of how GW5 controls rice grain width, we conducted fine mapping of this locus and uncovered a 1 212-bp deletion associated with the increased grain width in the rice cultivar Asominori, in comparison with the slender grain rice IR24. In addition, genotyping analyses of 46 rice cultivars revealed that this deletion is highly correlated with the grain-width phenotype, suggesting that the GW5 deletion might have been selected during rice domestication. GW5 encodes a novel nuclear protein of 144 amino acids that is localized to the nucleus. Furthermore, we show that GW5 physically interacts with polyubiquitin in a yeast two-hybrid assay. Together, our results suggest that GW5 represents a major QTL underlying rice width and weight, and that it likely acts in the ubiquitin-proteasome pathway to regulate cell division during seed development. This study provides novel insights into the molecular mechanisms controlling rice grain development and suggests that GW5 could serve as a potential tool for high-yield breeding of crops. 展开更多

This study aimed to obtain the first national estimate of the prevalence of autism spectrum disorder(ASD) in Chinese children.We targeted the population of 6 to 12-year-old children for this prevalence study by multistage convenient cluster sampling.The Modified Chinese Autism Spectrum Rating Scale was used for the screening process.Of the target population of 142,086 children,88.5%(n=125,806) participated in the study.A total of 363 children were confirmed as having ASD.The observed ASD prevalence rate was 0.29%(95% CI:0.26%-0.32%) for the overall population.After adjustment for response rates,the estimated number of ASD cases was867 in the target population sample,thereby achieving an estimated prevalence of 0.70%(95% CI:0.64%-0.74%).The prevalence was significantly higher in boys than in girls(0.95%;95% CI:0.87%-1.02% versus 0.30%;95%CI:0.26%-0.34%;P <0.001).Of the 363 confirmed ASD cases,43.3% were newly diagnosed,and most of those(90.4%) were attending regular schools,and 68.8% of the children with ASD had at least one neuropsychiatric comorbidity.Our findings provide reliable data on the estimated ASD prevalence and comorbidities in Chinese children. 展开更多

Background:National Cancer Center(NCC)of China annually reports the nationwide statistics for cancer inci-dence and mortality using population-based cancer registry data from all available cancer registries in China.Methods:There were a total of 487 registries which reported high quality data of cancer incidence and mortality across China in 2016.The nationwide numbers of new cancer cases and deaths were estimated using the pooled cancer registry data,which were stratified by area(urban/rural),sex,age group(0,1-4,5-9,10-14…85+)and cancer site for incidence and mortality,and then multiplied by corresponding national population.The world Segi’s population was applied for the calculation of age-standardized rates.Results:About 4,064,000 new cancer cases and 2,413,500 new cancer deaths occurred in China in 2016.Cancers of the lung,colon-rectum,stomach,liver and female breast were the top five common cancers,accounting for 57.4%of total cancer new cases.Cancers of the lung,liver,stomach,colon-rectum and esophagus were the five leading causes of cancer deaths,accounting for 69.3%of total cancer deaths.The crude and age-standardized incidence rates(ASIR)were 293.91 and 186.46 per 100,000 population,respectively.The crude mortality rate was 174.55/100,000 and the age-standardized mortality rate(ASMR)was 105.19/100,000.The ASIR was higher but the ASMR was lower in urban areas than that in rural areas.In past decades,the ASIR was relatively stable in males,but significantly increased by about 2.3%per year in females for overall cancers combined.In contrast,the ASMR significantly decreased by about 1.2%per year for both sexes during 2000-2016.Notably,the cancer-specific ASIR and ASMR of esophageal,stomach,and liver cancers decreased significantly,whereas both rates for cancers of the colon-rectum,prostate,female breast,cervix,and thyroid increased significantly.Conclusions:Cancer remains a major public health problem in China,which demands long-term collaborative efforts of a broad community.With the national guideline 展开更多

Liquid chromatography-mass spectrometry (LC-MS)-based metabolomics has been facilitated by the con- struction of MSz spectral tag (MS2T) library from the total scan ESI MS/MS data, and the development of widely targeted metabolomics method using MS/MS data gathered from authentic standards. In this report, a novel strategy called step- wise multiple ion monitoring-enhanced product ions (stepwise MIM-EPI) was developed to construct the MS2T library, in which stepwise MIM was used as survey scans to trigger the acquisition of EPI. A total number of 698 (almost) non- redundant metabolites with MS2 spectra were obtained, of which 135 metabolites were identified/annotated. Integrating the data gathered from our MS2T library and other available multiple reaction monitoring (MRM) information, a widely targeted metabolomics method was developed to quantify 277 metabolites, including some phytohormones. Evaluation of the dehydration responses and natural variations of these metabolites in rice leaf not only suggested the coordinated regulation of abscisic acid (ABA) with metabolites such as serotonin derivative(s), polyamine conjugates under drought stress, but also revealed some C-glycosylated flavones as the potential markers for the discrimination of indica and japonica rice subspecies. The new MS2T library construction and widely targeted metabolomics strategy could be used as a tool for rice functional genomics. 展开更多

QTL Ici Mapping is freely available public software capable of building high-density linkage maps and mapping quantitative trait loci(QTL) in biparental populations. Eight functionalities are integrated in this software package:(1) BIN: binning of redundant markers;(2) MAP: construction of linkage maps in biparental populations;(3) CMP: consensus map construction from multiple linkage maps sharing common markers;(4) SDL: mapping of segregation distortion loci;(5) BIP: mapping of additive, dominant, and digenic epistasis genes;(6) MET: QTL-by-environment interaction analysis;(7) CSL: mapping of additive and digenic epistasis genes with chromosome segment substitution lines; and(8) NAM: QTL mapping in NAM populations. Input files can be arranged in plain text, MS Excel 2003, or MS Excel 2007 formats. Output files have the same prefix name as the input but with different extensions. As examples, there are two output files in BIN, one for summarizing the identified bin groups and deleted markers in each bin, and the other for using the MAP functionality. Eight output files are generated by MAP, including summary of the completed linkage maps, Mendelian ratio test of individual markers, estimates of recombination frequencies, LOD scores, and genetic distances, and the input files for using the BIP, SDL,and MET functionalities. More than 30 output files are generated by BIP, including results at all scanning positions, identified QTL, permutation tests, and detection powers for up to six mapping methods. Three supplementary tools have also been developed to display completed genetic linkage maps, to estimate recombination frequency between two loci,and to perform analysis of variance for multi-environmental trials. 展开更多

我国正面临着全球最大的卒中挑战。全球疾病负担(global burden of disease,GBD)研究结果显示,2019年我国新发卒中394万例,卒中患者达到2876万例,卒中死亡人数为219万例。此外,卒中也是我国伤残调整生命年(disability-adjusted life yea... 我国正面临着全球最大的卒中挑战。全球疾病负担(global burden of disease,GBD)研究结果显示,2019年我国新发卒中394万例,卒中患者达到2876万例,卒中死亡人数为219万例。此外,卒中也是我国伤残调整生命年(disability-adjusted life years,DALYs)的首位原因,2019年卒中所致DALYs达到4590万。近期的几项大型流行病学调查对与卒中有关的既往疾病数据进行了更新。2018年我国18~69岁成人的超重和肥胖率分别为34.4%和16.8%;18岁及以上无高血压病史的成人中50.9%处于高血压前期,成人高血压的加权患病率为27.5%。根据美国糖尿病学会诊断标准,2017年我国18岁及以上成人中,总糖尿病和糖尿病前期的加权患病率分别为12.8%和35.2%;45岁以上成人的心房颤动加权患病率为1.8%,患者人数相当于790万人。医院质量监测系统(hospital quality monitoring system,HQMS)中1672家三级公立医院的数据显示,2019年收治卒中病例3411168例,其中缺血性卒中(ischemic stroke,IS)2818875例(82.6%),脑出血(intracebral hemorrhage,ICH)485474例(14.2%),蛛网膜下腔出血(subarachnoid hemorrhage,SAH)106819例(3.1%)。中位年龄为66岁,59.6%为男性。在IS、ICH和SAH中儿童卒中(年龄<18岁)分别有1379例(<0.1%)、2604例(0.5%)和1250例(1.2%)。超过三分之一[1231519例(36.1%)]的卒中病例参加了城镇居民基本医疗保险,其次是城镇职工基本医疗保险[891103例(26.1%)]和新型农村合作医疗保险[543108例(15.9%)]。主要危险因素是高血压(IS 57.3%、ICH 69.9%、SAH 44.1%),主要并发症是肺炎或肺部感染(IS 10.4%、ICH 34.6%、SAH29.7%)。总体的院内死亡/非医嘱离院率为8.5%,从IS的6.0%到SAH的20.6%不等。住院时间为9.0(6.0~13.0)d,范围从IS的10.0(7.0~13.0)d到ICH的14.0(8.0~22.0)d。HQMS中2847家二级公立或民营医院的数据也有类似的结果。HQMS的数据显示,内蒙古自治区、安徽省、西藏自治区和北京市去外省就医比例较高。北京市、� 展开更多

There exist differences in the epidemiological characteristics,clinicopathological features,tumor biological characteristics,treatment patterns,and drug selections between gastric cancer patients from the Eastern and Western countries.The Chinese Society of Clinical Oncology(CSCO)has organized a panel of senior experts specializing in all sub-specialties of gastric cancer to compile a clinical guideline for the diagnosis and treatment of gastric cancer since 2016 and renews it annually.Taking into account regional differences,giving full consideration to the accessibility of diagnosis and treatment resources,these experts have conducted expert consensus judgment on relevant evidence and made various grades of recommendations for the clinical diagnosis and treatment of gastric cancer to reflect the value of cancer treatment and meeting health economic indexes in China.The 2021 CSCO Clinical Practice Guidelines for Gastric Cancer covers the diagnosis,treatment,follow-up,and screening of gastric cancer.Based on the 2020 version of the CSCO Chinese Gastric Cancer guidelines,this updated guideline integrates the results ofmajor clinical studies from China and overseas for the past year,focused on the inclusion of research data from the Chinese population for more personalized and clinically relevant recommendations.For the comprehensive treatment of non-metastatic gastric cancer,attentions were paid to neoadjuvant treatment.The value of perioperative chemotherapy is gradually becoming clearer and its recommendation level has been updated.For the comprehensive treatment of metastatic gastric cancer,recommendations for immunotherapy were included,and immune checkpoint inhibitors fromthird-line to the first-line of treatment for different patient groups with detailed notes are provided. 展开更多

In January 2013, a long-lasting episode of severe haze occurred in central and eastern China, and it attracted attention from all sectors of society. The process and evolution of haze pollution episodes were observed by the "Forming Mechanism and Con- trol Strategies of Haze in China" group using an intensive aerosol and trace gases campaign that simultaneously obtained data at 11 ground-based observing sites in the CARE-China network. The characteristics and formation mechanism of haze pollu- tion episodes were discussed. Five haze pollution episodes were identified in the Beijing-Tianjin-Hebei (Jing-Jin-Ji) area; the two most severe episodes occurred during 9-15 January and 25-31 January. During these two haze pollution episodes, the maximum hourly PMz5 mass concentrations in Beijing were 680 and 530 ~tg m-3, respectively. The process and evolution of haze pollution episodes in other major cities in the Jing-Jin-Ji area, such as Shijiazhuang and Tianjin were almost the same as those observed in Beijing. The external cause of the severe haze episodes was the unusual atmospheric circulation, the depres- sion of strong cold air activities and the very unfavorable dispersion due to geographical and meteorological conditions. How- ever, the internal cause was the quick secondary transformation of primary gaseous pollutants to secondary aerosols, which contributed to the "explosive growth" and "sustained growth" of PM2.5. Particularly, the abnormally high amount of nitric ox- ide (NOx) in the haze episodes, produced by fossil fuel combustion and vehicle emissions, played a direct or indirect role in the quick secondary transformation of coal-burning sulphur dioxide (SO2) to sulphate aerosols. Furthermore, gaseous pollutants were transformed into secondary aerosols through heterogeneous reactions on the surface of fine particles, which can change the particle's size and chemical composition. The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection. 展开更多

Granite is one of the most important components of the continental crust on our Earth; it thus has been an enduring studied subject in geology. According to present knowledge, granite shows a great deal of heterogeneity in terms of its texture,structure, mineral species and geochemical compositions at different scales from small dike to large batholith. However, the reasons for these variations are not well understood although numerous interpretations have been proposed. The key point of this debate is whether granitic magma can be effectively differentiated through fractional crystallization, and, if so, what kind of crystallization occurred during the magmatic evolution. Although granitic magma has high viscosity because of its elevated SiO2 content, we agree that fractional crystallization is effectively processed during its evolution based on the evidence from field investigation,mineral species and its chemical variations, and geochemical compositions. These data indicate that crystal settling by gravitation is not the only mechanism dominating granitic differentiation. On the contrary, flow segregation or dynamic sorting may be more important. Accordingly, granite can be divided into unfractionated, fractionated(including weakly fractionated and highly fractionated) and cumulated types, according to the differentiation degree. Highly fractionated granitic magmas are generally high in primary temperature or high with various volatiles during the later stage, which make the fractional crystallization much easier than the common granitic melts. In addition, effective magmatic differentiation can be also expected when the magma emplaced along a large scale of extensional structure. Highly fractionated granitic magma is easily contaminated by country rocks due to its relatively prolonged crystallization time. Thus, granites do not always reflect the characteristics of the source areas and the physical and chemical conditions of the primary magma. Background:The National Cancer Center(NCC)of China regularly reports the nationwide statistics on cancer incidence and mortality in China.The International Agency for Research on Cancer(IARC)calculates and publishes the cancer burden of countries around the world every two years.To ensure consistency between the actual surveillance data in China and the data published by IARC,NCC has received approval from the National Health Commission and IARC to simultaneously release the cancer burden data for China in GLOBOCAN 2022.Methods:There were a total of 700 registries reporting high-quality data on cancer incidence and mortality across China in 2018,of which 106 registries with continuous monitoring from 2010 to 2018 were used to establish an age-period-cohort model to simulate the trend of cancer incidence and mortality and to estimate the incidence and mortality in China in 2022.In addition,we analyzed the temporal trends of age-standardized cancer incidence and mortality from 2000 to 2018 using data from 22 continuous cancer registries.Results:It was estimated about 4,824,700 new cancer cases and 2,574,200 new cancer deaths occurred in China in 2022.Cancers of the lung,colon-rectum,thyroid,liver and stomach were the top five cancer types,accounting for 57.42%of new cancer cases.Cancers of the lung,liver,stomach,colon-rectum and esophagus were the five leading causes of cancer deaths,accounting for 67.50%of total cancer deaths.The crude rate and age-standardized incidence rate(ASIR)were 341.75 per 100,000 and 201.61 per 100,000,respectively.The crude mortality rate was 182.34 per 100,000 and the age-standardized mortality rate(ASMR)was 96.47 per 100,000.The ASIR of all cancers combined increased by approximately 1.4%per year during 2000–2018,while the ASMR decreased by approximately 1.3%per year.We observed decreasing trends in ASIR and ASMR for cancers of the esophagus,stomach,and liver,whereas the ASIR increased significantly for cancers of the thyroid,prostate,and cervix.Conclusions:Cancer remains a major publ 展开更多
