The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of trans...The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. Further efforts should focus on identifying the etiology of the SARS-associated virus and ruling out conclusively the existence of otherpossible SARS-related pathogen(s).展开更多

METHODS: Twenty patients with persistent HBV infection were included in this study, 1...AIM: To identify the property of dendritic cells (DCs) of peripheral blood monocytes (PBMC) in patients with chronic HBV infection. METHODS: Twenty patients with persistent HBV infection were included in this study, 10 healthy subjects being used as a control group. The peripheral blood mononuclear cells (PBMC) of T cell-depleted populations were incubated and induced into mature dendritic cells in the RPMI-1640 medium in the presence of cytokines GM-CSF, IL-4, FLt-3,TNF-alpha and 100mL.L(-1 )of fetal calf serum for a total of 10-12 days. The expressions of surface markers on DCs were evaluated using flow cytometric analysis. ELISA method was used to determine the cytokine levels of interleukin-12 (IL-12) and IL-10 in the supernatant produced by DCs. For detection of the stimulatory capacity of DCs to T cell proliferation, mytomycin C-treated DC were incubated with allogenic T cells. RESULTS: A typical morphology of mature DCs from healthy subjects and HBV-infected patients was induced in in vitro incubation, but the proliferation ability and cellular number of DCs from HBV-infected patients significantly decreased compared with healthy individuals. In particular, the expression levels of HLA-DR, CD80 (B7-1) and CD86 (B7-2) on DC surface from patients were also lower than that from healthy individuals (0.46 vs 0.92 for HLA-DR, 0.44 vs 0.88 for CD80 and 0.44 vs 0.84 for CD86,P【0.05). The stimulatory capacity and production of IL-12 of DCs from patients in allogenic mixed lymphocyte reaction (AMLR) significantly decreased, but the production level of nitric oxide (NO) by DCs simultaneously increased compared with healthy subjects (86 +/- 15 vs 170 +/- 22 micromol.L(-1), P 【0.05). CONCLUSION: The patients with chronic HBV infection have the defective function and immature phenotype of dendritic cells, which may be associated with the inability of efficient presentation of HBV antigens to host immune system for the clearance of HBV.展开更多
A field survey was conducted to investigate the metal and arsenic contamination in soils and vegetables on four villages (Shuichuan (SCH), Beiwan (BWA), Dongwan (DWA) and Wufe (WFE)) located along, Baiyin, C...A field survey was conducted to investigate the metal and arsenic contamination in soils and vegetables on four villages (Shuichuan (SCH), Beiwan (BWA), Dongwan (DWA) and Wufe (WFE)) located along, Baiyin, China, and to evaluate the possible health risks to local population through foodchain. Results show that the most significantly contaminated soils occurred upstream at SCH where Cd, Cu and As concentrations exceeded maximum allowable concentrations for Chinese agricultural soil. Further downstream the degree of contamination semi-systematically decreased in concentrations of metal. Generally, the leafy vegetables were more heavily contaminated than non-leafy vegetables. Chinese cabbage is the most severely contaminated, the concentrations of Cd exceeded the maximum permit levels (0.05 mg/kg) by 4.5 times. Bio-accumulate factor also shows that an entry of Cd to food chain plants is the greatest potential. Furthermore, the estimated daily intake amounts of the considered toxic elements (Cd, Pb and Cu) from the vegetables grown at SCH and BWA and DWA have exceeded the recommended dietary allowance levels. Thus, the vegetables grown in three villages above, which affected by Baiyin mining and smelting have a health hazard for human consumption.展开更多

Recently, extensive studies have focused on soil microbial diversity. However, understanding the diversity of this complex microbial community in the soil environment is a challenging task. Thus, it is important to master and comprehend appropriate methods for studying soil microbial diversity. Concepts of soil microbial diversity and major methods of study are briefly introduced in this paper. Then, the application of biochemical-based and molecular-based techniques in this area, and their advantages and disadvantages are evaluated. Based on recent related research, perspectives for studying microbial diversity in soils are presented.展开更多
目的了解河南省15岁及以上人群精神分裂症患病率、流行情况及影响因素。方法于2015年在河南省20个区县开展精神障碍流行病学调查,采用多阶段分层随机抽样方法,利用修订后的一般健康问卷(General Health Questionnaire,GHQ-12)作为初步...目的了解河南省15岁及以上人群精神分裂症患病率、流行情况及影响因素。方法于2015年在河南省20个区县开展精神障碍流行病学调查,采用多阶段分层随机抽样方法,利用修订后的一般健康问卷(General Health Questionnaire,GHQ-12)作为初步筛查工具,将调查对象按照评分分为高危、中危、低危3组,然后由精神科主治医师采用DSM-Ⅳ轴Ⅰ诊断定式临床访谈诊断量表(SCID)初步对被调查人员进行诊断,采用卡方检验对定性资料进行比较,采用Logistic回归分析对精神分裂症患病影响因素进行分析。结果完成本次调查共23846名,其中男11865名,女11981名,分别占总人数的49.76%和50.24%;其中精神分裂症患者124例,精神分裂症调整后终生患病率为5.56‰,时点患病率为5.04‰;农村83例,城市41例;男54例,女70例;受教育程度情况:文盲26例,小学39例,初中47例,高中或中专9例,大专或本科及以上3例;婚姻状态:未婚31例,初婚78例,再婚3例,离婚11例,丧偶1例。单因素分析结果显示,精神分裂症患者在不同受教育水平(χ^2=33.903,P<0.01)以及不同婚姻状况中患病率差异均有统计学意义(χ^2=137.143,P<0.01)。Logistic回归分析显示,城乡(Wald χ^2=37.296,P<0.01)、性别(Wald χ^2=13.131,P<0.01)、受教育程度(Wald χ^2=48.338,P<0.01)、婚姻状态(Wald χ^2=54.495,P<0.01)、家族史(Wald χ^2=107.340,P<0.01)是精神分裂症患者患病的影响因素。结论河南省精神分裂症患病率与国内外研究结果基本一致,精神分裂症防治应根据不同地区、不同年龄、不同性别等制定针对性措施。展开更多
