Firstly, based on the traffic loading investigation on the expressway bridge within 24 hours a day, all the critical parameters of traffic flow, such as the vehicle type, weight, separation space and speed are all recorded and analyzed to extract its statistical characteristics, which are used to work out random traffic flow simulation program RTF. This RTF program can be embedded with the other general FEM software. Secondly, a dynamic analysis module RTFWVB of the wind-vehicle-bridge coupling vibration under random traffic flow is presented, which can consider arbitrary number of vehicles, multi-lanes and traffic flow direction. Finally, Hangzhou Bay Bridge in China is selected as a numerical example to demonstrate dynamic interaction of the RTFWVB system. The results indicate that the traffic flow direction has just a little influence on bridge dynamic response, that the mean responses are mainly determined by the moving vehicle loads, and that the fluctuating components will increase with the increase of wind speed.展开更多

In the context of the increasing scale of bridges and the increasing service life of bridges,it is very important to carry out efficient,accurate and intelligent bridge operation and maintenance.In recent years,advanc...In the context of the increasing scale of bridges and the increasing service life of bridges,it is very important to carry out efficient,accurate and intelligent bridge operation and maintenance.In recent years,advanced equipment,technology and intelligent algorithms have developed rapidly.It is necessary to apply advanced equipment and algorithms to bridge operation and maintenance business to facilitate the digitalization and intelligence of bridge operation and maintenance.To grasp the research progress on the bridge intelligent operation and maintenance,this paper summarizes the research progress in recent years from the aspects of intelligent detection equipment and technology,intelligent monitoring equipment and technology,intelligent data analysis,intelligent evaluation and early warning,and intelligent repair and maintenance.According to the review,more and more smart devices have been used to replace human beings to detect dangerous and hidden bridge components.At the same time,image processing,radar and other technologies have been used to analyze component damage more objectively and quantitatively.To solve the shortcomings of traditional sensors such as short life and low robustness,more non-contact measurement methods have been proposed.Scholars have proposed various intelligent algorithms to process the massive amount of bridge health monitoring data to improve the quality of the data.To achieve the rapid perception of bridge status and timely early warning of structural abnormalities,different from traditional theoretical calculations,scholars have tried to use data-driven methods to intelligently evaluate and early warning of bridge structural status.In terms of intelligent repair and maintenance,more intelligent algorithms have been used to optimize structural maintenance strategies and determine the best maintenance time by integrating multisource heterogeneous data.All these provide strong support for the automation,digitization and intelligence of bridge operation and maintenance.展开更多
Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional land...Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.展开更多
Background Acute lymphoblastic leukemia (ALL) and chemotherapy can cause immune imbalance,and gaseous molecule hydrogen sulfide (H2S) can participate in the process of immune response.This study aimed to investiga...Background Acute lymphoblastic leukemia (ALL) and chemotherapy can cause immune imbalance,and gaseous molecule hydrogen sulfide (H2S) can participate in the process of immune response.This study aimed to investigate the immune regulation of H2S in pediatric ALL.Methods Children (n=78) with ALL admitted during 2010-2013 were included in this study.Two blood samples were collected in period of before chemotherapy,bone marrow remission and two days after chemotherapy,respectively.Serum contents of H2S and cytokines,including interleukin-1β (IL-13),interleukin-2 (IL-2),interferon-γ (IFN-γ),tumor necrosis factor (TNF-α),interleukin-4 (IL-4),interleukin-6 (IL-6),interleukin-10 (IL-10) and macrophage inflammatory protein-1α (MIP-1a),were detected using ELISA method.Stepwise regression was used to analyze the correlation between H2S and cytokines.Furthermore,human Jurkat cells were cultured in vitro,and nucleoprotein of Jurkat cells and peripheral blood mononuclear cells (PBMCs) were collected,contents of cystathionine γ-lyase (CSE) and certain cytokines were measured by Western blotting.Results Serum concentrations of H2S,IL-13,IL-6,IL-10 and MIP-1α in children with ALL were increased significantly (P <0.01),while concentrations of IL-2,TNF-α,IFN-γ and IL-4 decreased obviously (P <0.01).In patients after chemotherapy,concentrations of H2S and IL-10 were decreased significantly (P <0.05),but IL-4 and IFN-γ concentrations increased markedly (P <0.05).At remission stage,H2S,IL-13,IL-4,IL-6,IL-10 and MIP-1α concentrations were further decreased markedly (P <0.05),but concentrations of IL-2,TNF-α and IFN-γ increased again (P <0.05).Protein contents of CSE,IL-10,IL-4 and IL-2 of PBMCs also increased markedly in children with ALL.Moreover,changes of CSE protein contents of PBMCs were consistent with serum H2S contents,and there were significant correlation between H2S and certain cytokines based on stepwise regression analysis.展开更多
Background:The aim of this study was to review clinical features of adolescent malignant germ cell tumors(MGCTs)in Beijing and analyze the peculiar characteristics of this age group.Methods:Clinical characteristics,pa...Background:The aim of this study was to review clinical features of adolescent malignant germ cell tumors(MGCTs)in Beijing and analyze the peculiar characteristics of this age group.Methods:Clinical characteristics,pathological presentations,and survival outcomes of 34 patients were analyzed retrospectively.Results:Of 34 patients,12 girls and 22 boys,18(52.9%)had an extra-cranial tumor,including one testicular tumor,five ovarian tumors,one sacrococcygeal tumor,and 11 mediastinal tumors.Histologically,we found immature teratomas(n=6),yolk sac tumors(n=5),mixed malignant tumors(n=5),an embryonic carcinoma(n=1),and seminoma(n=1).Three-year event-free survival(EFS)and overall survival(OS)were 48.8%and 62.9%,respectively.Another 16(47.1%)patients had an intracranial tumor,including nine in the pineal region,five in the suprasellar region,one in basal ganglia,and one in cerebellopontine.All patients had localized disease and an excellent outcome with 3-year EFS and OS of 93.7%and 100%,respectively.Conclusions:Adolescent MGCTs are rare with a strong dependence on gender,and the mediastina and pineal region are the most common tumor locations.The prognosis is promising compared with that of other adolescent tumors and MGCTs in other age groups.MGCTs in mediastina have a tendency to companion with other hematological malignancies,and the prognosis is extremely poor in these patients.展开更多

Background Bacterial inflammation is a common complication in patients with leukemia,and sulfur dioxide (SO2) is a bioactive molecule in modulating Gram-negative bacilli infection.This study aimed to examine the cha...Background Bacterial inflammation is a common complication in patients with leukemia,and sulfur dioxide (SO2) is a bioactive molecule in modulating Gram-negative bacilli infection.This study aimed to examine the changes in SO2,nuclear factor-κB (NF-κB),and interleukin-8 (IL-8) levels in pediatric acute lymphoblastic leukemia (ALL) with Gram-negative bacterial inflammation.Methods Fifty-five ALL children were enrolled in this study,including 30 males and 25 females,aged 3-13 years,and the median age was 7.8 years.All these children who accepted chemotherapy for ALL were divided into the control group (before chemotherapy),the infection group (after chemotherapy with infection),and the recovery group (the infection was controlled after 1 week).The serum level of SO2 was detected using high performance liquid chromatography with fluorescence assay,and NF-κB and IL-8 levels were measured by enzyme-linked immunosorbent assay (ELISA).Human THP-1 cells were cultured,induced,and differentiated into macrophages,which were divided into five groups and each group was cultured with different stimulators:lipopolysaccharide (LPS) group,LPS+L-aspartate-β-hydroxamate (HDX) group,LPS+SO2 group,SO2,and control groups.NF-κB level and IL-8 protein contents by ELISA were examined in each group.Results In comparison with those of the control group,levels of serum SO2,NF-κB,and IL-8 of the infection group were significantly increased (P <0.05),while those of the recovery group were significantly decreased (P <0.05).A positive correlation was found between levels of serum SO2 and intracellular NF-κB/IL-8,and the correlation coefficients were 0.671 and 0.798 (P <0.05),respectively.According to the results found in human THP-1 cells,levels of NF-κB and IL-8 in LPS group were significantly increased compared with those of the control group (P <0.05); when compared with those in LPS group,levels of NF-κB in LPS+HDX group further increased significantly (P <展开更多
Importance:Cancer is the main cause of death by disease in children.Children experience the highest incidence of cancer in the first year of life.However,there is no comprehensive registration system for children with...Importance:Cancer is the main cause of death by disease in children.Children experience the highest incidence of cancer in the first year of life.However,there is no comprehensive registration system for children with tumors in China.Objective:To summarize the diagnosis and treatment of infant cancer and analyze the status of standardized diagnosis and management among several treatment centers in Beijing,China,thereby providing evidence to guide further clinical research.Methods:From January 1,2010 to December 31,2019,patients with newly diagnosed infantile malignant solid tumors were admitted to six large tertiary pediatric solid tumor diagnosis and treatment centers in Beijing.The epidemiology,clinical features,and therapeutic effects of tumors in these patients were analyzed retrospectively.All patients were followed up until March 31,2020.Results:In total,938 patients were enrolled in this study.There were 530 boys(56.5%)and 408 girls(43.5%);the median age was 6.0 months(range,0-12.0 months).The three most common tumors were retinoblastoma in 366 patients(39.0%),neuroblastoma in 266 patients(28.4%),hepatoblastoma in 133 patients(14.2%),and central nervous system tumors in 52 patients(5.5%).The estimated 5-year overall survival rate was 81.3%±1.8%,and the 5-year event-free survival rate was 71.8%±2.9%.The 5-year overall survival rates of non-rhabdomyosarcoma soft tissue sarcoma,neuroblastoma,and retinoblastoma were 100%,88%±2.2%,and 86.9%±2.1%,respectively.The 5-year event-free survival rates were 81.1%±2.7%for neuroblastoma,81.6%±9.8%for non-rhabdomyosarcoma soft tissue sarcoma,and 72.7%±14.1%for extracranial malignant germ cell tumors.Interpretation:The three most common infantile malignant solid tumors were retinoblastoma,neuroblastoma,and hepatoblastoma.Multidisciplinary combined diagnosis and treatment is needed for infantile tumors.展开更多

Due to the particular mode of inheritance, X-chromosomal X-STR has the advantage in female traces identification against male contamination and in complex kinship cases, such as deficiency cases (mother-son or father-daughter), grandparent-grandchild comparisons, half-sisters testing, etc. In this study, 4 blood samples from two paternity cases without father were examined with TYPER19 Amplification kit first, from the result, it is hard to judge the relationship between the mother-sons, then TYPERX19 kit was conducted to obtain the full profiles of the two pairs of the mother-son, from the genetypes of the samples, it can be inferred that neither of the two pairs of mother-son were in line with the kinship, which provides important evidence for the two paternity cases. All together, the X-chromosome marks included in the TYPERX19 kit can offer the possibility to solve complex kinship cases where autosomal STR markers cannot provide the information needed.展开更多

Interleukin-26(IL-26),originally called AK155,is one of the cytokines of the IL-10 cytokine family,and its gene is located in human chromosome region 12q15.IL-26 signals through the receptors IL-20R1 and IL-10R2,which...Interleukin-26(IL-26),originally called AK155,is one of the cytokines of the IL-10 cytokine family,and its gene is located in human chromosome region 12q15.IL-26 signals through the receptors IL-20R1 and IL-10R2,which form a heterodimer,targets specific cells and induces activation of the JAK-STAT pathway,allowing the rapid phosphorylation of STAT1 and STAT3 and the initiation of their effects.1 This signaling plays an important role in the regulation of host defense and inflammatory diseases.However,whether the role of IL-26 in the progression of hepatocellular carcinoma(HCC)occurs via autophagy remains unclear.Here,we analyzed the association between IL-26 levels and the clinicopathological characteristics of 115 patients with HCC and found that IL-26 levels were particularly associated with tumor stage and survival.We found that IL-26 levels were associated with the prognosis of HCC patients.Furthermore,we found that increased autophagy by IL-26 was dependent on the JAK-STAT3 signaling pathway,which could result in the inhibition of the migration and invasion abilities of these HCC cells.These findings suggest that the levels of IL-26 may be a useful prognostic marker for HCC patients and may offer insights into the clinical application of IL-26 in the future.展开更多
For comprehensive understanding of practical application and evaluation on the power of30 commonly used InDeis(Qiagen Investigator DIPplex®kit),we captured population data from 25 Chinese populations and employed...For comprehensive understanding of practical application and evaluation on the power of30 commonly used InDeis(Qiagen Investigator DIPplex®kit),we captured population data from 25 Chinese populations and employed F-statistics for population genetics analysis.The results indicated that the distributions of allelic frequencies among populations were in different levels.Furthermore,the phylogeny confoiming pairwise FST distances showed that the difierentiation of majority populations were consistent with their geographic locations and historic dispersals.We conduct the comprehensive correlation analysis between FST and heterozygosity of30 InDel loci and provided strong evidence for ongoing InDei loci selection.The Fst values of 30 InDels were calculated within 25 Chinese populations,and then,these loci were characterized definitely based on their roles in population genetics or individual identification.Data indicated that 17 InDels with FST<0.01 could be utilized regarding Chinese individual identification(total discrimination power=0.999985 and cumulative matching probability=0.00000009).We comprehensively reconstructed the population structure and filled the gap of evaluating the ability of InDels in personal as well as population identification.The application of InDel loci in the forensic area would convincingly promote the development matter of forensic population identification and personal discrimination.展开更多

文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.
文摘Background Acute lymphoblastic leukemia (ALL) and chemotherapy can cause immune imbalance,and gaseous molecule hydrogen sulfide (H2S) can participate in the process of immune response.This study aimed to investigate the immune regulation of H2S in pediatric ALL.Methods Children (n=78) with ALL admitted during 2010-2013 were included in this study.Two blood samples were collected in period of before chemotherapy,bone marrow remission and two days after chemotherapy,respectively.Serum contents of H2S and cytokines,including interleukin-1β (IL-13),interleukin-2 (IL-2),interferon-γ (IFN-γ),tumor necrosis factor (TNF-α),interleukin-4 (IL-4),interleukin-6 (IL-6),interleukin-10 (IL-10) and macrophage inflammatory protein-1α (MIP-1a),were detected using ELISA method.Stepwise regression was used to analyze the correlation between H2S and cytokines.Furthermore,human Jurkat cells were cultured in vitro,and nucleoprotein of Jurkat cells and peripheral blood mononuclear cells (PBMCs) were collected,contents of cystathionine γ-lyase (CSE) and certain cytokines were measured by Western blotting.Results Serum concentrations of H2S,IL-13,IL-6,IL-10 and MIP-1α in children with ALL were increased significantly (P <0.01),while concentrations of IL-2,TNF-α,IFN-γ and IL-4 decreased obviously (P <0.01).In patients after chemotherapy,concentrations of H2S and IL-10 were decreased significantly (P <0.05),but IL-4 and IFN-γ concentrations increased markedly (P <0.05).At remission stage,H2S,IL-13,IL-4,IL-6,IL-10 and MIP-1α concentrations were further decreased markedly (P <0.05),but concentrations of IL-2,TNF-α and IFN-γ increased again (P <0.05).Protein contents of CSE,IL-10,IL-4 and IL-2 of PBMCs also increased markedly in children with ALL.Moreover,changes of CSE protein contents of PBMCs were consistent with serum H2S contents,and there were significant correlation between H2S and certain cytokines based on stepwise regression analysis.
文摘Background:The aim of this study was to review clinical features of adolescent malignant germ cell tumors(MGCTs)in Beijing and analyze the peculiar characteristics of this age group.Methods:Clinical characteristics,pathological presentations,and survival outcomes of 34 patients were analyzed retrospectively.Results:Of 34 patients,12 girls and 22 boys,18(52.9%)had an extra-cranial tumor,including one testicular tumor,five ovarian tumors,one sacrococcygeal tumor,and 11 mediastinal tumors.Histologically,we found immature teratomas(n=6),yolk sac tumors(n=5),mixed malignant tumors(n=5),an embryonic carcinoma(n=1),and seminoma(n=1).Three-year event-free survival(EFS)and overall survival(OS)were 48.8%and 62.9%,respectively.Another 16(47.1%)patients had an intracranial tumor,including nine in the pineal region,five in the suprasellar region,one in basal ganglia,and one in cerebellopontine.All patients had localized disease and an excellent outcome with 3-year EFS and OS of 93.7%and 100%,respectively.Conclusions:Adolescent MGCTs are rare with a strong dependence on gender,and the mediastina and pineal region are the most common tumor locations.The prognosis is promising compared with that of other adolescent tumors and MGCTs in other age groups.MGCTs in mediastina have a tendency to companion with other hematological malignancies,and the prognosis is extremely poor in these patients.

文摘To promote and develop the theoretical basis and application of the wind-vehicle-bridge coupling vibration system,the corresponding research status and prospects are reviewed and discussed from five aspects,i.e.,the analytical framework,the aerodynamic interference,the evaluation criteria,the design loads of long-span bridge and the double-deck railcum-road bridge.The refining process of analysis system is reviewed from the aspects of simulation wind load,vehicle load and bridge structure,and the corresponding coupling relationship.For aerodynamic interference,the development process is summarized from the simulative precision of the elements(wind,vehicle and bridge),the load cases and the object of interference.For evaluation criteria,the corresponding development course is summarized from the certain evaluation method to uncertain one.For long-span bridge design load,the wind and vehicle loads are reviewed and summarized from current multinational codes and theoretical research.For double-deck rail-cum-road bridge,the mechanism of multi-element coupling relationship and corresponding aerodynamic interference are both reviewed.By comprehensive review and summary,the analytical framework is in the process from simplification to refinement.The simulation and consideration of the objects of structural interference gradually become complex.The corresponding simulation theory,wind tunnel scale,test equipment and technology are the key factors to limit its development.For systematic evaluation of vehicle and bridge,the structural and systemic security are the basis of the evaluation,and the auxiliary components and functional evaluation need to be paid more attention.The evaluation criterion will be developed from certain method to reliability assessment.For design load of long-span bridge,the vehicle load is gradually transferred from the simple application of the design load of small-medium span bridge into a complex model considering the load characteristics.For double-deck rail-cum-road bridge,the basic theory and
文摘Background Bacterial inflammation is a common complication in patients with leukemia,and sulfur dioxide (SO2) is a bioactive molecule in modulating Gram-negative bacilli infection.This study aimed to examine the changes in SO2,nuclear factor-κB (NF-κB),and interleukin-8 (IL-8) levels in pediatric acute lymphoblastic leukemia (ALL) with Gram-negative bacterial inflammation.Methods Fifty-five ALL children were enrolled in this study,including 30 males and 25 females,aged 3-13 years,and the median age was 7.8 years.All these children who accepted chemotherapy for ALL were divided into the control group (before chemotherapy),the infection group (after chemotherapy with infection),and the recovery group (the infection was controlled after 1 week).The serum level of SO2 was detected using high performance liquid chromatography with fluorescence assay,and NF-κB and IL-8 levels were measured by enzyme-linked immunosorbent assay (ELISA).Human THP-1 cells were cultured,induced,and differentiated into macrophages,which were divided into five groups and each group was cultured with different stimulators:lipopolysaccharide (LPS) group,LPS+L-aspartate-β-hydroxamate (HDX) group,LPS+SO2 group,SO2,and control groups.NF-κB level and IL-8 protein contents by ELISA were examined in each group.Results In comparison with those of the control group,levels of serum SO2,NF-κB,and IL-8 of the infection group were significantly increased (P <0.05),while those of the recovery group were significantly decreased (P <0.05).A positive correlation was found between levels of serum SO2 and intracellular NF-κB/IL-8,and the correlation coefficients were 0.671 and 0.798 (P <0.05),respectively.According to the results found in human THP-1 cells,levels of NF-κB and IL-8 in LPS group were significantly increased compared with those of the control group (P <0.05); when compared with those in LPS group,levels of NF-κB in LPS+HDX group further increased significantly (P <

文摘Importance:Cancer is the main cause of death by disease in children.Children experience the highest incidence of cancer in the first year of life.However,there is no comprehensive registration system for children with tumors in China.Objective:To summarize the diagnosis and treatment of infant cancer and analyze the status of standardized diagnosis and management among several treatment centers in Beijing,China,thereby providing evidence to guide further clinical research.Methods:From January 1,2010 to December 31,2019,patients with newly diagnosed infantile malignant solid tumors were admitted to six large tertiary pediatric solid tumor diagnosis and treatment centers in Beijing.The epidemiology,clinical features,and therapeutic effects of tumors in these patients were analyzed retrospectively.All patients were followed up until March 31,2020.Results:In total,938 patients were enrolled in this study.There were 530 boys(56.5%)and 408 girls(43.5%);the median age was 6.0 months(range,0-12.0 months).The three most common tumors were retinoblastoma in 366 patients(39.0%),neuroblastoma in 266 patients(28.4%),hepatoblastoma in 133 patients(14.2%),and central nervous system tumors in 52 patients(5.5%).The estimated 5-year overall survival rate was 81.3%±1.8%,and the 5-year event-free survival rate was 71.8%±2.9%.The 5-year overall survival rates of non-rhabdomyosarcoma soft tissue sarcoma,neuroblastoma,and retinoblastoma were 100%,88%±2.2%,and 86.9%±2.1%,respectively.The 5-year event-free survival rates were 81.1%±2.7%for neuroblastoma,81.6%±9.8%for non-rhabdomyosarcoma soft tissue sarcoma,and 72.7%±14.1%for extracranial malignant germ cell tumors.Interpretation:The three most common infantile malignant solid tumors were retinoblastoma,neuroblastoma,and hepatoblastoma.Multidisciplinary combined diagnosis and treatment is needed for infantile tumors.
文摘A completely new finite element software, Finite ElementProgram Generator (FEPG), is introduced and its designing thought and organizing structure is presented.FEPG uses the method of components and the technique of artificial intelligence to generate finite element program automatically by a computer according to the general principles of mathematic and internal rules of finite element method,as is similar to the deduction of mathematics.FEPG breaks through the limitation of present finite element software,which only applies to special discipline,while FEPG is suitable for all kinds of differential equations solved by finite element method.Now FEPG has been applied to superconductor research,electromagnetic field study,petroleum exploration,transportation,structure engineering,water conservancy,ship mechanics, solid-liquid coupling problems and liquid dynamics,etc.in China.
文摘The usefulness of the X-chromosomal STRs (short tandem repeats) for forensic purposes seems to be restricted, but may be valuable in some paternity cases. Due to the particular mode of inheritance, X-chromosomal X-STR has the advantage in female traces identification against male contamination and in complex kinship cases, such as deficiency cases (mother-son or father-daughter), grandparent-grandchild comparisons, half-sisters testing, etc. In this study, 4 blood samples from two paternity cases without father were examined with TYPER19 Amplification kit first, from the result, it is hard to judge the relationship between the mother-sons, then TYPERX19 kit was conducted to obtain the full profiles of the two pairs of the mother-son, from the genetypes of the samples, it can be inferred that neither of the two pairs of mother-son were in line with the kinship, which provides important evidence for the two paternity cases. All together, the X-chromosome marks included in the TYPERX19 kit can offer the possibility to solve complex kinship cases where autosomal STR markers cannot provide the information needed.
文摘Body fluid identification through messenger RNA(mRNA)has been proposed as a useful supplement to presumptive and confirmatory tests by previous laboratory studies;however,its application in routine clinical forensic examination was rare.We report a case of sexual assault in which body fluid identification by mRNA profiling was used.Vaginal secretions mRNA markers(MUC4,HBD1,and CYP2B7P1)were used to test the sample,being obtained positive results.This case demonstrates that mRNA profiling of body fluids could be applied to routine case examinations as an aid,acting as a scientific collaborative evidence to strengthen the medicolegal opinion.
文摘Hepatocellular carcinoma(HCC)is the sixth most common malignancy and the fourth leading cause of cancer related death worldwide.China covers over half of cases,leading HCC to be a vital threaten to public health.Despite advances in diagnosis and treatments,high recurrence rate remains a major obstacle in HCC management.Multi-omics currently facilitates surveillance,precise diagnosis,and personalized treatment decision making in clinical setting.Non-invasive radiomics utilizes preoperative radiological imaging to reflect subtle pixel-level pattern changes that correlate to specific clinical outcomes.Radiomics has been widely used in histopathological diagnosis prediction,treatment response evaluation,and prognosis prediction.High-throughput sequencing and gene expression profiling enabled genomics and proteomics to identify distinct transcriptomic subclasses and recurrent genetic alterations in HCC,which would reveal the complex multistep process of the pathophysiology.The accumulation of big medical data and the development of artificial intelligence techniques are providing new insights for our better understanding of the mechanism of HCC via multi-omics,and show potential to convert surgical/intervention treatment into an antitumorigenic one,which would greatly advance precision medicine in HCC management.

文摘Interleukin-26(IL-26),originally called AK155,is one of the cytokines of the IL-10 cytokine family,and its gene is located in human chromosome region 12q15.IL-26 signals through the receptors IL-20R1 and IL-10R2,which form a heterodimer,targets specific cells and induces activation of the JAK-STAT pathway,allowing the rapid phosphorylation of STAT1 and STAT3 and the initiation of their effects.1 This signaling plays an important role in the regulation of host defense and inflammatory diseases.However,whether the role of IL-26 in the progression of hepatocellular carcinoma(HCC)occurs via autophagy remains unclear.Here,we analyzed the association between IL-26 levels and the clinicopathological characteristics of 115 patients with HCC and found that IL-26 levels were particularly associated with tumor stage and survival.We found that IL-26 levels were associated with the prognosis of HCC patients.Furthermore,we found that increased autophagy by IL-26 was dependent on the JAK-STAT3 signaling pathway,which could result in the inhibition of the migration and invasion abilities of these HCC cells.These findings suggest that the levels of IL-26 may be a useful prognostic marker for HCC patients and may offer insights into the clinical application of IL-26 in the future.

文摘For comprehensive understanding of practical application and evaluation on the power of30 commonly used InDeis(Qiagen Investigator DIPplex®kit),we captured population data from 25 Chinese populations and employed F-statistics for population genetics analysis.The results indicated that the distributions of allelic frequencies among populations were in different levels.Furthermore,the phylogeny confoiming pairwise FST distances showed that the difierentiation of majority populations were consistent with their geographic locations and historic dispersals.We conduct the comprehensive correlation analysis between FST and heterozygosity of30 InDel loci and provided strong evidence for ongoing InDei loci selection.The Fst values of 30 InDels were calculated within 25 Chinese populations,and then,these loci were characterized definitely based on their roles in population genetics or individual identification.Data indicated that 17 InDels with FST<0.01 could be utilized regarding Chinese individual identification(total discrimination power=0.999985 and cumulative matching probability=0.00000009).We comprehensively reconstructed the population structure and filled the gap of evaluating the ability of InDels in personal as well as population identification.The application of InDel loci in the forensic area would convincingly promote the development matter of forensic population identification and personal discrimination.
