In the present study, we investigated for the first time two single nucleotide polymorphisms(SNPs) in the regulatory region of genes encoding NADPH oxidase 4(NOX4) and p22 phox(CYBA) in NAFLD.Methods: A total of 207 biopsy-proven NAFLD patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(NASH)(n = 180)] were evaluated. Genomic DNA was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of PCR.Results: Associations of CYBA-675 T/A with high-density lipoprotein(HDL)(TT vs TA vs AA; P < 0.01) and triglycerides(TGL)(TT vs XA; P < 0.01) were observed only in NASH patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(ALT)(CA + AA vs CC; P = 0.02). However, there was no association of SNPs in the CYBA and NOX4 genes encoding the NADPH oxidase system proteins and the presence of NASH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, P < 0.01) and those who were more obese(32.2 vs 29.0 kg/m2,P < 0.01). In addition, serum glucose and insulin levels increased significantly in the presence of NASH.Conclusions: There were associations between the presence of the allele A in the NOX4 SNP and a higher concentration of ALT in the NAFLD population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGL and lower HDL in NASH patients. Fire blight disease,caused by the bacterium Erwinia amylovora(E.amylovora),is responsible for substantial losses in cultivated apples worldwide.An important mechanism of plant immunity is based on the recognition of c...Fire blight disease,caused by the bacterium Erwinia amylovora(E.amylovora),is responsible for substantial losses in cultivated apples worldwide.An important mechanism of plant immunity is based on the recognition of conserved microbial molecules,named pathogen-associated or microbe-associated molecular patterns(PAMPs or MAMPs),through pattern recognition receptors(PRRs),leading to pattern-triggered immunity(PTI).The interspecies transfer of PRRs represents a promising strategy to engineer broad-spectrum and durable disease resistance in crops.EFR,the Arabidopsis thaliana PRR for the PAMP elf18 derived from the elongation factor thermal unstable(EF-Tu)proved to be effective in improving bacterial resistance when expressed into Solanaceae and other plant species.In this study,we tested whether EFR can affect the interaction of apple with E.amylovora by its ectopic expression in the susceptible apple rootstock M.26.Stable EFR expression led to the activation of PAMP-triggered immune response in apple leaves upon treatment with supernatant of E.amylovora,as measured by the production of reactive oxygen species and the induction of known defense genes.The amount of tissue necrosis associated with E.amylovora infection was significantly reduced in the EFR transgenic rootstock compared to the wild-type.Our results show that the expression of EFR in apple rootstock may be a valuable biotechnology strategy to improve the resistance of apple to fire blight.展开更多

The polymorphism of lactase non-persistence/lactase persistence(LCT-13910C>T) was examined by PCR-restriction fragment length polymorphism technique in 102 liver biopsy-proven NAFLD patients(steatosis in 9 and NASH in 93) and compared to those of 501 unrelated healthy volunteers. Anthropometric, clinical, biochemical and liver histology data were analyzed. Continuous variables were compared using the t or Mann-Whitney tests, and categorical data were compared with the Fisher's exact test. Univariate logistic regression and multivariate logistic regression adjusted for gender and age were performed.RESULTS No differences in the LCT-13910 genotype frequencies were noted between the NAFLD patients(66.67% of the patients with steatosis were CC, 33.33% were CT, and none were TT; 55.91% of the patients with NASH were CC, 39.78% were CT, and 4.3% were TT; P = 0.941) and the healthy controls(59.12% were CC, 35.67% were CT, and 5.21% were TT) or between the steatosis and NASH patients. That is, the distribution of the lactase non-persistence/lactase persistence polymorphism(LCT-13910C>T) in the patients with NAFLD was equal to that in the general population. In the NASH patients, the univariate analysis revealed that the lactase nonpersistence(low lactase activity or hypolactasia) phenotype was associated with higher insulin levels(23.47 ± 15.94 μU/m L vs 15.8 ± 8.33 μU/m L, P = 0.027) and a higher frequency of insulin resistance(91.84% vs 72.22%, P = 0.02) compared with the lactase persistence phenotype. G4 are stabilized by the presence of cations and are characterized by a high degree of structural polymorphism with different patterns of groove, loop arrangement, strand orientations and stoichiometry. G-rich sequences are over-represented in the promoter regions of many oncogenes as well as at human telomeres, d(TTAGGG) repeats, ranging in size from 3 to 15 kb, involved in protecting chromosomal ends. A specialized enzyme, called telomerase, provides a telomere maintenance mechanism by elongating the end of the G-strand and it is activated in the majority of cancer cells. Therefore there are two general strategies of telomerase targeting in cancer treatment. One is a direct targeting of telomerase to cause its inhibition;the other one is the use of G4 stabilizers which block telomerase access to telomere, thus causing an indirect enzyme inhibition. Here, we evaluated the molecular recognition of some phenanthroline-based ligands against four different experimental models of the human telomeric sequence d[AG3(T2AG3)3] by means of docking simulations. Our theoretical analysis was able to reproduce the experimental affinity measurements, with a linear squared correlation factor r2 equal to 0.719 among all the studied models. These findings highlighted the importance to consider the polymorphism of the DNA G4. Interestingly, this correlation resulted always improved with respect to that of the single folds, with the exception of the parallel structure, thus suggesting a key role of this G4 conformation in the interaction network of the tested binders. Moreover, we identified the moieties of the phenanthroline scaffold directly involved in the complex formation. This allowed to rationalize the improved binding affinity always associated with a bis-phenanthroline system and to explain why a phenanthroline substituted with a pyridine ring is favored with respect to the pyrimidine one.展开更多

Smart Micro Electro-Mechanical Systems (MEMS) are also characterized by different physical phenomena affecting their properties at different scales. Data-driven formulations can then be helpful to deal with the complexity of the multi-physics governing their response to the external stimuli, and optimize their performances. As an example, Lorentz force micro-magnetometers working principle rests on the interaction of a magnetic field with a current flowing inside a semiconducting, micro-structured medium. If an alternating current with a properly set frequency is let to flow longitudinally in a slender beam, the system is driven into resonance and the sensitivity to the magnetic field may result largely enhanced. In our former activity, a reduced-order physical model of the movable structure of a single-axis Lorentz force MEMS magnetometer was developed, to feed a multi-objective topology optimization procedure. That model-based approach did not account for stochastic effects, which lead to the scattering in the experimental data at the micrometric length-scale. The formulation is here improved to allow for stochastic effects through a two-scale deep learning model designed as follows: at the material scale, a neural network is adopted to learn the scattering in the mechanical properties of polysilicon induced by its polycrystalline morphology;at the device scale, a further neural network is adopted to learn the most important geometric features of the movable parts that affect the overall performance of the magnetometer. The media used in tissue culture can be modified through the addition of inexpensive organic materi...In-vitro propagation is a technique that provides a vital solution for the conservation of endangered orchid species. The media used in tissue culture can be modified through the addition of inexpensive organic materials as an alternative to expensive synthetic additives. Some organic sources, such as coconut water and fruit juice, contain significant amounts of vitamins, amino acids, and organic compounds which can act as growth regulators, making these organic sources excellent additives for in-vitro cultivation. The aim of this study was to develop a protocol for in-vitro micropropagation and acclimatization of Epidendrum nocturnum using organic supplements in the growth media and various substrates at the acclimatization stage. Banana powder, coconut water, and potato dextrose were added to a basal seed sowing media and evaluated for seed germination percentage and plantlet growth. In addition, various substrates such as coconut coir, horticultural charcoal, sphagnum moss, and wood bark were evaluated for height, number of leaves, and number of shoots in the acclimatization portion of this study. The culture medium with coconut water showed a greater germination percentage (71.00% and 76.75%) compared with the control (37.50% and 45.50%) at 60 and 90 days after seed sowing, respectively. Media with organic supplements showed greater values of plant length and number of roots compared with the control. The combination of coconut coir and horticultural charcoal was shown to be more efficient than the combination of sphagnum moss, horticultural charcoal, and wood bark, as results showed greater values of plant height and number of leaves at 30, 90, and 120 days after transplantation in acclimatization of E. nocturnum.展开更多

Media with organic supplements showed greater values of plant length and number of roots compared with the control. The combination of coconut coir and horticultural charcoal was shown to be more efficient than the combination of sphagnum moss, horticultural charcoal, and wood bark, as results showed greater values of plant height and number of leaves at 30, 90, and 120 days after transplantation in acclimatization of E. nocturnum.
