The aim of this study was to investigate the role of the 5-HT7 receptor in the pathogenesis of IBS. Methods A rat model of irritable bowel syndrome with diarrhea (IBS-D) was established by colonic instillation of acetic acid and restraint stress. A rat model with irritable bowel syndrome with constipation (IBS-C) was established by stomach irrigated with 0-4℃ cool water daily for 14 days. The content and distribution of 5-HT in the brain and gut were examined by immunohistochemistry and the mRNA expression of the 5-HT7 receptor was determined by fluorescent quantitative reverse transcription polymerase chain reaction. The accumulation of cyclic adenosine monophosphate (cAMP) in all the same tissues was measured by radioimmunity. Results The models of IBS were reliable by identification. The immunohistochemistry results showed that there were significantly more 5-HT positive cells in the IBS-D group than in the control group in the hippocampus, hypothalamus, jejunum, ileum, proximate colon and distal colon (P〈0.05), as well as more than were found in the IBS-C group in jejunum and ileum (P〈0.05). There were more 5-HT positive cells in the IBS-C group than in the control hippocampus, hypothalamus, ileum, proximate colon, and distal colon (P〈0.05). Real time-PCR results showed that the expression level of the 5-HT7 receptor in both the IBS-C and IBS-D groups were enhanced compared with the control group in the hippocampus and hypothalamus (P〈0.05). The expression level of 5-HT7 receptors in the IBS-C group was notably greater when compared with the controls in the ileum and colon (P〈0.05). METHODS: A total of 31 patients newly diagnosed with ECA and 26 healthy subjects were included in the current study. The frequencies of MDSCs and Th17 cells in peripheral blood were determined by flow cytometry. The mRNA expression of cytokines, arginase 1 (Arg1) and inducible NO synthase (iNOS) in peripheral blood mononuclear cells (PBMCs) and plasma Arg1 were assessed by real-time polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. RESULTS: There was an increased prevalence of MDSCs in the peripheral blood from ECA patients (15.21% ± 2.25%) when compared with healthy control (HC) (1.10% ± 0.12%, P < 0.0001). The plasma levels of Arg1 in ECA patients were significantly higher than those in HC (28.28 ± 4.10 ng/mL vs 9.57 ± 1.51 ng/ mL, P=0.0003). iNOS mRNA levels in the peripheral blood of ECA patients also showed a threefold increase compared with HC (P=0.0162). The frequencies of Th17 cells (CD4 + IL-17A + ) were significantly elevated in ECA patients versus HC (3.50% ± 0.33% vs 1.82% ± 0.19%, P=0.0001). Increased mRNA expression of IL-17 and ROR-γt was also observed in ECA patients compared with HC (P=0.0041 and P=0.0004, respectively), while the mRNA expression of IL-6 and tumor necrosis factor-α (TNF-α) showed significant decreases (P=0.0049 and P < 0.0001, respectively). No obvious correlations were found between the frequencies of MDSCs and Th17 cells in the peripheral blood from ECA patients(r=-0.1725, P=0.3534). Arg1 mRNA levels were positively correlated with levels of IL-6 (r=0.6404, P=0.0031) and TNF-α (r=0.7646, P=0.0001). Similarly, iNOS mRNA levels were also positively correlated with levels of IL-6 (r=0.6782, P=0.0007) and TNF-α (r=0.7633, P < 0.0001). CONCLUSION: This study reveals the relationship between circulating MDSCs and Th17 cells, which may lead to new immunotherapy approac展开更多

Three sets of filters are installed in a dual layer filterwheel that use Johnson-Cousins UBVRI, Sloan Digital Sky Survey u′g′r′i′z′ and Stomgren uvby. The photometric system and the CCD camera are introduced, followed by detailed analysis of their performances, and determination of the relevant parameters, including gain, readout noise, dark current and linearity of the CCD cam- era. In addition, the parameters describing the site's astro-climate, including typical seeing, statistics on the number of clear nights and average sky brightness, based on data gathered from Sep. 2007 to Aug. 2013, are systematically studied and reported in this work. Photometric calibrations were done using Landolt standard star obser- vations spanning eight nights, which yielded transformation coefficients, photometric precision and system throughput. The limiting magnitudes are simulated using the derived calibration parameters and classic observation conditions at WHO.展开更多

In total, 33 compounds of volaUles emitted from three developmental stage plants were separated and identified, and quantitatively analyzed by the internal standard addition method. All of the samples of volatile were found to be rich in monoterpenes and sesquiterpenes.β-phellandrene and caryophyllene predominated in the volatiles of the leaves of plants at the two- and ten-leaf stages. Furthermore, (E)-2-hexenal were the dominant components in the volatiles emitted from anthesis plants. The results of volatiles analyzed show that the compositions varied depending on the developmental stages. The volatiles emitted from crushed tomato leaves of plants at the anthesis stage had the most strongly inhibitory activity against the spore germination and hyphal growth of Botrytis cinerea and Fusarium oxysporum, followed by ten- and two-leaf plants. However, the activity of volatiles, emitted from the leaves of plants at the two-leaf stage, in inhibiting F. oxysporum was greater than B. cinerea.展开更多

Methods: Three siRNA sequences against NgR were designed and transfected into cerebellar granule ceils (CGCs) to screen for the most efficient sequence of NgR siRNA by using reverse transcription polymerase chain reaction (RT-PCR) and immunofluorescence staining. NgR siRNA sequence I was found the most efficient which was then transfected into the CGCs grown on CNS myelin substrate to observe its disinhihition for neurite outgrowth. Results: Compared with the scrambled control sequence of siRNA, the NgR siRNA sequence 1 signifi- cantly decreased NgR mRNA level at 24 b and 48 h (p 〈 0.05), which was recovered by 96 h after transfection. NgR immunoreactivity was also markedly reduced at 24 and 48 h after tile transfection of siRNA sequence 1 compared with that before transfection (p 〈 0.05). The NgR immunoreactiviW was recovered after 72 h post-transfection. Moreover, the neurite outgrowth on tire myelin substrate was greatly improved within 72 h after the transfection with siRNA sequence 1 compared with the scrambled sequence-transfected group or non-transfected group (p 〈 0.05). Conclusion: : siRNA-mediated knockdown of NgR expression contributes to neurite outgrowth in vitro.展开更多

CONCLUSION: This study reveals the relationship between circulating MDSCs and Th17 cells, which may lead to new immunotherapy approac

文摘The one-meter telescope at Weihai Observatory (WHO) of Shandong University is an f/8 Cassegrain telescope. The limiting magnitudes are simulated using the derived calibration parameters and classic observation conditions at WHO.

文摘High alumina slag will cause severe corrosion at the interface of alumina refractory, and the wetting behavior of slag is a key factor influencing the corrosion resistance of refractory ceramics. The static magnetic field is a promising solution for improvement in the slag resistance of refractory. The wetting of alumina refractory ceramics with different basicities of high alumina slags under a weak static magnetic field was analyzed, given that a weak static magnetic field can affect the corrosion behavior of refractory ceramics.Taking slag S_(3) as an example, when there was an external static magnetic field of 1.0 mT at 1600 ℃,the thickness of calcium aluminate reaction layer at the interface decreased by 36.7%,the denting depth of interface decreased by 35.6%,and the apparent wetting angle increased by 20%.The living radicals and their formation path in oxide melts were verified by first-principles calculation combined with electron paramagnetic resonance spectroscopy analysis. The influence of the flux density of a weak static magnetic field on the wetting behavior of slags was also explored. The contact angle of the slags increased owing to the inhibitory effect of magnetic field on the radicalinvolved reaction at the interface of the slag and the alumina refractory ceramic. The relationships between the magnetic flux density, diffusion coefficient, slag microstructure(hyperfine coupling constant),and contact angle were established. This provides a theoretical basis for the field control of radical involved reactive wetting between inorganic oxide slags and solid oxide ceramics.

文摘Fe-doped BiVO4with hierarchical flower-like structure was prepared via a hydrothermal method using sodium dodecylbenzene sulfonate(SDBS)as structure directing agent.X-ray diffraction(XRD),scanning electron microscope(SEM),transmissionelectron microscope(TEM),high resolution transmission electron microscope(HRTEM),X-ray photoelectron spectroscopy(XPS)and UV-Vis were applied for characterization of the as-prepared samples. The formation mechanism of flower-like structure wasproposed based on the evolution of morphology as a function of hydrothermal time.Fe-doped into substitutional sites of BiVO4effectively improved the migration and separation of photogenerated carrier and enhanced the utilization of visible light.Flower-likeFe-doped BiVO4showed much higher visible-light-driven photocatalytic efficiency for degradation of methyl blue compared withthe pristine BiVO4.And the sample with a Fe/Bi mole ratio of2.5%showed the highest photocatalytic efficiency.

文摘Hydrogen-bonded organic frameworks(HOFs)are a recent class of porous materials that have garnered considerable research interest owing to their distinctive characteristics.HOFs can be constructed through judicious selection of H-bonding motifs, which are further enforced by other weak intermolecular interactions such asπ–πstacking, van der Waals forces, and framework interpenetration.Taking advantage of these interactions,we can expand the functional field of HOFs by introducing active molecules.Recently, researchers have made substantial advancements in using HOFs for chemical sensing, catalysis, proton conduction, biological applications, and others. The low bonding energy of H-bonds allows for precise control over the concentration of ligands in solvents, forming diverse HOF structures. These varied structures offer significant advantages for producing HOFs with photo-responsive and electro-responsive properties. However, the presence of H-bonds in HOFs results in their inherent lower stability compared to metal-organic frameworks(MOFs)and covalent-organic frameworks(COFs)formed via coordination and covalent bonds, respectively. As a result, the pursuit of stable and innovative HOF materials with novel functional sites remains an ongoing challenge. This review provides an overview of recent research progress in the development of new strategies for stable HOF synthesis and applications of HOFs with stimuli-responsive properties. We first classified all synthetic methods reported to date and discussed the stable HOFs synthesized,as well as their unique properties and applications. In addition,we summarized the applications of HOFs utilizing their synergistic responses to external stimuli, including photo, electrical, pressure, and chemical stimuli. We systematically reviewed stable HOF synthesis and applications, which may lead to a deeper understanding of the structure–activity relationship for these materials and guide future HOF design.

文摘Volatiles emitted from the leaves of Lycopersicon esculentum at the two-, ten-leaf and anthesis periods were collected by a gas absorbing method and analyzed by gas chromatography (GC)-mass spectrometry. However, the activity of volatiles, emitted from the leaves of plants at the two-leaf stage, in inhibiting F. oxysporum was greater than B. cinerea.

文摘Monoclinic BiPO4with rod-like shape was prepared via a CTAB-assisted hydrothermal route.MnOx nanoparticles were loaded on the surfaces of BiPO4rods by a photo-deposition process to form MnOx/BiPO4heterojunctions.The as-prepared samples were characterized by XRD,SEM,TEM,XPS,FL,and UV-Vis diffuse reflectance measurements. The results showed that MnOx nanoparticles were strongly anchored to the surfaces of BiPO4rods when the mole ratio of Mn to Bi was controlled at a low level, forming MnOx/BiPO4heterojunctions with effective and sound interfaces. The MnOx/BiPO4heterojunctions exhibited higher photoactivity than pristine BiPO4for photodegradation of methyl blue under UV irradiation, which could be attributed to the efficient charge transfer at the heterojunction interfaces. The higher light absorption ability of MnOx/BiPO4in the range of300?420nm compared with pristine BiPO4was also responsible for the enhanced photocatalytic activities of MnOx/BiPO4heterojunctions.
文摘Positive nucleic acid(NA)results have been found in recovered and discharged COVID-19 patients, but the proportion is unclear. This study was designed to analyze the recurrent positive rate of NA results after consecutively negative results, and the relationship between the specific antibody production and positive NA rate.According to Strengthening the Reporting of Observational Studies in Epidemiology guidelines, data of inpatients in Sino-French New City Branch of Tongji Hospital between Jan.28 and Mar.6,2020 were collected.A total of 564 COVID-19 patients over 14 years old who received the examinations of NA and antibodies against SARS-CoV-2 were included.Days of viral shedding and specific antibodies were recorded and assessed.Among NA tests in respiratory samples(throat swabs, nasopharyngeal swabs, sputum and flushing fluid in alveoli),the patients with all-negative NA results accounted for 17.20%,those with single-positive results for 46.63%,and those with multiple-positive results for 36.17%respectively.Besides,the recurrent positive NA results after consecutively negative results appeared in 66 patients(11.70%).For multiple-positive patients, median viral shedding duration was 20 days(range:1 to 57 days).Of the 205 patients who received 2 or more antibody tests,141(68.78%)had decreased IgG and IgM concentrations.IgM decreased to normal range in 24 patients, with a median of 44 days from symptom onset.Viral shedding duration was not significantly correlated with gender, age, disease severity, changes in pulmonary imaging, and antibody concentration. It is concluded that antibody level and antibody change had no significant correlation with the positive rate of NA tests and the conversion rate after continuous negative NA tests. In order to reduce the recurrent positive proportion after discharge,3 or more consecutive negative NA test results with test interval more than 24 h every time are suggested for the discharge or release from quarantine.

文摘This paper proposes a reasonable radiation-resistant composite channel structure for In P HEMTs.The simulation results show that the composite channel structure has excellent electrical properties due to increased modulation doping efficiency and carrier confinement. Moreover, the direct current(DC)and radio frequency(RF)characteristics and their reliability between the single channel structure and the composite channel structure after 75-ke V proton irradiation are compared in detail. The results show that the composite channel structure has excellent radiation tolerance.Mechanism analysis demonstrates that the composite channel structure weakens the carrier removal effect. This phenomenon can account for the increase of native carrier and the decrease of defect capture rate.

文摘The electronic properties of xPA/nPPP/yPA sandwiched copolymers with a well-barrier-well structure have been studied by using a tight-binding calculation. It was found that the electronic properties of the neutral states of these sandwiched copolymers are sensitive to the constitutions of PPP and PA monomers and the interface coupling between PA and PPP. It is verified that the quantum tunnelling effect will occur at the lowest conductive state of xPA/nPPP/xPA copolymers.

文摘We investigate quantum phase transitions for q-state quantum Potts models(q=2,3,4)on a square lattice and for the Ising model on a honeycomb lattice by using the infinite projected entangled-pair state algorithm with a simplified updating scheme. We extend the universal order parameter to a two-dimensional lattice system, which allows us to explore quantum phase transitions with symmetry-broken order for any translation-invariant quantum lattice system of the symmetry group G.The universal order parameter is zero in the symmetric phase, and it ranges from zero to unity in the symmetry-broken phase. The ground-state fidelity per lattice site is computed, and a pinch point is identified on the fidelity surface near the critical point. The results offer another example highlighting the connection between(i)critical points for a quantum many-body system undergoing a quantum phase-transition and(ii)pinch points on a fidelity surface. In addition,we discuss three quantum coherence measures:the quantum Jensen–Shannon divergence, the relative entropy of coherence, and the l1norm of coherence, which are singular at the critical point, thereby identifying quantum phase transitions.
文摘Purpose: To investigate the in vitro effect of short interfering RNAs (siRNAs) against Nogo receptor (NgR) on neurite outgrowth under an inhibitory substrate of central nervous system (CNS) myelin. Conclusion: : siRNA-mediated knockdown of NgR expression contributes to neurite outgrowth in vitro.
文摘BACKGROUND Squamous cell carcinoma(SCC)of bone is usually caused by metastasis from the lungs, bladder,or other sites.Primary SCC of bone most frequently involves the skull bones, and primary involvement of other sites in the skeletal system is extremely rare.To date, only three such cases have been reported, which makes the diagnosis, treatment, and prognosis of this disease a challenge.CASE SUMMARY A 76-year-old Chinese man presented to our hospital with nonspecific pain and limited mobility in the right shoulder for 4 mo.He underwent three-dimensional computed tomography reconstruction and magnetic resonance imaging of the right shoulder, which revealed an osteolytic destructive lesion in the right scapula with invasion into the surrounding muscles and soft tissues.Ultrasound-guided core needle biopsy detected a malignant tumor, and immunohistochemical analysis revealed a poorly differentiated SCC.Wide excision of the right scapular bone was performed, and pathological examination of the surgical specimen confirmed the diagnosis. At the last follow-up examination within 2 years, the patient was doing well with the pain significantly relieved in the right shoulder.CONCLUSION Primary SCC of bone is extremely rare at sites other than the skull.Clinicians must exhaust all available means for the diagnosis of primary SCC of the bone,so greater attention can be paid to its timely and effective management.Regular and adequate follow-up is essential to help rule out metastasis and judge the prognosis.

文摘A novel memristor-based multi-scroll hyperchaotic system is proposed.Based on a voltage-controlled memristor and a modulating sine nonlinear function,a novel method is proposed to generate the multi-scroll hyperchaotic attractors.Firstly,a multi-scroll chaotic system is constructed from a three-dimensional chaotic system by designing a modulating sine nonlinear function. Then,a voltage-controlled memristor is introduced into the above-designed multi-scroll chaotic system. Thus,a memristor-based multi-scroll hyperchaotic system is generated, and this hyperchaotic system can produce various coexisting hyperchaotic attractors with different topological structures. Moreover, different number of scrolls and different topological attractors can be obtained by varying the initial conditions of this system without changing the system parameters. The Lyapunov exponents, bifurcation diagrams and basins of attraction are given to analyze the dynamical characteristics of the multi-scroll hyperchaotic system.Besides, the field programmable gate array(FPGA)based digital implementation of the memristor-based multi-scroll hyperchaotic system is carried out.The experimental results of the FPGA-based digital circuit are displayed on the oscilloscope.

文摘This study examined the inhibitory effect of triptolid(TP)on tumor necrosis factor-α(TNF-α)secreted from peripheral blood monocular cells(PBMCs)and the association of the inhibitory effect with TNF-α-308 gene polymorphisms in rheumatoid arthritis(RA)patients.Gene polymorphism at A-G site 308 in the promoter region of TNF-αgene was detected in 42 RA patients by using allele specific polymerase chain reaction(AS-PCR)assay.PBMCs were harvested from these patients and treatedfirst with lipopolysaccharides(LPS)and then with different doses of TP(1,5.4 and 15 ng/mL).The TNF-αlevel in the supernatants was measured by enzyme-linked immuno-sorbent assay(ELISA).The results showed that TNF-αlevel in the supernatants of TP(1 ng/mL)-treated PBMCs was decreased by 3.80%and 4.91%,respectively,in the patients with AA and AG genotypes, when compared with those treated with LPS alone(P>0.05).Moreover,the TNF-αlevel in the patients with GG genotype was reduced by 20.74%(P<0.05).When PBMCs were treated with TP at 5.4 ng/mL,TNF-αlevels in the patients with AA,AG,and GG genotypes were decreased by 20.42%,34.73%,and 41.69%,respectively(P<0.05).The TNF-αlevel was slightly higher in the PBMCs treated with 15 ng/mL of TP than those in the two TP groups in the patients carrying AA,AG,and GG genotypes(P>0.05).It was concluded that gene polymorphism at TNF-α-308 sites may relate to the secretion of TNF-αin RA patients.TP has different inhibitory effects on the secretion of TNF-αin the patients harboring different genotypes, which may be one of the reasons for individual variation in response to TP.
