In rice grains,the Waxy (Wx) gene is responsible for the synthesis of amylose,the most important determinant for eating and cooking quality.The effects of several Wx alleles on amylose content and the taste of cooked rice have been elucidated.However,the relationship between artificial selection and the evolution of various Wx alleles as well as their distribution remain unclear.Here we report the identification of an ancestral allele,Wx^lv,which dramatically affects the mouthfeel of rice grains by modulating the size of amylose molecules.We demonstrated that WF originated directly from wild rice,and the three major Wx alleles in cultivated rice (Wx^b,Wx^a,and Wx^in) differentiated after the substitution of one base pair at the functional sites.These data indicate that the Wx^lv allele played an important role in artificial selection and domestication.The findings also shed light on the evolution of various Wx alleles,which have greatly contributed to improving the eating and cooking quality of rice. 展开更多

In rice(Oryza sativa), amylose content(AC) is the major factor that determines eating and cooking quality(ECQ). The diversity in AC is largely attributed to natural allelic variation at the Waxy(Wx)locus. Here we identified a rare Wx allele, Wx^(mw) ,which combines a favorable AC, improved ECQ and grain transparency. Based on a phylogenetic analysis of Wx genomic sequences from 370 rice accessions, we speculated that Wx^(mw) may have derived from recombination between two important natural Wx alleles, Wx^(in) and Wx^(b). We validated the effects of Wx^(mw) on rice grain quality using both transgenic lines and near-isogenic lines(NILs). When introgressed into the japonica Nipponbare(NIP) background, Wx^(mw) resulted in a moderate AC that was intermediate between that of NILs carrying the Wx^(b)allele and NILs with the Wx^(mp) allele. Notably, mature grains of NILs fixed for Wx^(mw) had an improved transparent endosperm relative to soft rice. Further, we introduced Wx^(mw) into a high-yielding japonica cultivar via molecular marker-assisted selection: the introgressed lines exhibited clear improvements in ECQ and endosperm transparency. Our results suggest that Wx^(mw) is a promising allele to improve grain quality, especially ECQ and grain transparency of high-yielding japonica cultivars, in rice breeding programs. 展开更多

CDK4/6 inhibitors are the standard treatment in advanced HR+/HER2-breast cancer patients.Nevertheless,the resistance to CDK4/6 inhibitors is inevitable and the strategies to overcome resistance are of great interest.Here,we show that the palbociclibresistant breast cancer cells expressed significantly higher levels of Cyclin D1 and CDK4 proteins because of upregulated protein synthesis.Silencing Cyclin D1 or CDK4 led to cell cycle arrest while silencing Cyclin E1 or CDK2 restored the sensitivity to palbociclib.Furthermore,PI3K/mTOR pathway was hyper-activated in palbociclib-resistant cells,leading to more phosphorylated 4E-BP1 and higher levels of Cyclin D1 and CDK4 translation.Targeting PI3K/mTOR pathway with a specific PI3Kαinhibitor(BYL719)or an mTOR inhibitor(everolimus)reduced the protein levels of Cyclin D1 and CDK4,and restored the sensitivity to palbociclib.The tumor samples expressed significantly higher levels of Cyclin D1,CDK4,p-AKT and p-4E-BP1 after progression on palbociclib treatment.In conclusion,our findings suggest that overexpressed Cyclin D1 and CDK4 proteins lead to the resistance to CDK4/6 inhibitor and PI3K/mTOR inhibitors are able to restore the sensitivity to CDK4/6 inhibitors,which provides the biomarker and rationale for the combinational use of CDK4/6 inhibitors and PI3K/mTOR inhibitors after CDK4/6 inhibitor resistance in breast cancer. 展开更多

Starch is a vital energy source for living organisms and is a key raw material and additive in the food and non-food industries.Starch has received continuous attention in multiple research fields.The endosperm of cereals(e.g.,rice,corn,wheat,and barley)is the most important site for the synthesis of storage starch.Around 2010,several excellent reviews summarized key progress in various fields of starch research,serving as important references for subsequent research.In the past 10 years,many achievements have been made in the study of starch synthesis and regulation in cereals.The present review provides an update on research progress in starch synthesis of cereal endosperms over the past decade,focusing on new enzymes and non-enzymatic proteins involved in starch synthesis,regulatory networks of starch synthesis,and the use of elite alleles of starch synthesis-related genes in cereal breeding programs.We also provide perspectives on future research directions that will further our understanding of cereal starch biosynthesis and regulation to support the rational design of ideal quality grain. 展开更多

When plotted on the concordia diagram, the results give linear data array and the upper intercept age of (882±3)Ma, and the age was considered as the crystallization age of the protolith. Granitic gneiss has high ASI value (1.09), high LILE and LREE contents, significantly negative Eu depletion, distinctly negative Ba, Sr, P, Ti and Nb anomalies and indicate continental crust parentage, which is consistent with high initial 87Sr/86Sr ratios value (0.7170) and very negative εNd(t)=-14.1. The protolith magma is interpreted as a product of partial melting of the basement rocks of older basement crust. 展开更多

Silane coupling agent KH560 was used to modify the surface of nano-α-Al<sub>2</sub>O<sub>3</sub> in ethanol-aqueous solution with different proportions. The particle size of nano-α-Al<sub>2</sub>O<sub>3</sub> was determined by nano-particle size analyzer, and the effects of nano-α-Al<sub>2</sub>O<sub>3</sub> content, ethanol-aqueous solution ratio and KH560 dosage on the dispersion and particle size of nano-α-Al<sub>2</sub>O<sub>3</sub> were investigated. The material structure before and after modification was determined by Fourier transform infrared spectroscopy (FTIR). Aqueous polyurethane resin and inorganic components are combined with modified nano-α-Al<sub>2</sub>O<sub>3</sub> dispersion to form chromium-free passivation solution. The solution is coated on the galvanized sheet, the adhesion and surface hardness are tested, the bonding strength of the coating and the surface hardness of the substrate are discussed. The corrosion resistance and surface morphology of the matrix were investigated by electrochemical test, neutral salt spray test and scanning electron microscope test. The chromium-free passivation film formed after the modification of nano-α-Al<sub>2</sub>O<sub>3</sub> increases the surface hardness of galvanized sheet by about 85%. The corrosion resistance of the film is better than that of a single polyurethane film. The results show that the surface hardness and corrosion resistance of polyurethane resin composite passivation film are significantly improved by the introduction of nano-α-Al<sub>2</sub>O<sub>3</sub>. 展开更多

In this paper, an integrated desulfurization and denitrification technology is proposed for ultra-low emissions of SO2 and NOx in the steel, power and cement industries. A cost-effective and operationally efficient control strategy is realized through a forced oxidation-absorption-reduction process, which reduces equipment investment and operating costs. The technology was adapted to continuous and intermittent denitrification in different temperature zones, promoting the recycling of desulfurization and denitrification products. The study also explored the use of a highly active absorbent obtained by the hydration reaction of coal ash and lime from a power company for the desulfurization and denitrification of sintered flue gases in iron and steel mills, which produces by-products that can be used as retarding agents in the cement industry, resulting in a circular economy. The article emphasizes the importance of improving the lime digestion process and developing new denitrification agents for environmentally safe and cost-effective flue gas treatment. 展开更多

The standing time during the pretreatment of primary sample has a great influence on the concentration of nitrite tested by spectrophotometric method. In this context, three kinds of food samples are prepared, including canned mustard, canned fish and home-made pickled water. A series of standing times are placed during the sample pretreatments and the corresponding nitrite contents in these samples are detected by spectrophotometric method based on N-ethylenediamine dihydrochloride. This study aims to find out a reasonable standing time during the pretreatment of food sample, providing influence factor for precise detection of nitrite. 展开更多

In this study, the adsorption effect of β-cyclodextrin modified biochar (BC) on phenanthrene (PHE) in contaminated soil was investigated, aiming to provide an efficient and environmentally friendly remediation strategy for Polycyclic Aromatic Hydrocarbons (PAHs) contaminated soil. Through kinetic and isotherm analysis, β-CDBC-CA showed excellent phenanthrene adsorption performance, and the adsorption effect increased with the increase of time and was affected by temperature. The results show that β-CDBC-CA can not only effectively adsorb phenanthrene in soil, but also serve as a surfactant to help desorption phenanthrene adsorbed by soil organic matter and improve the efficiency of microbial degradation. The experimental data showed that the Elovich model could describe the adsorption behavior of β-CDBC-CA on phenanthrene well, while Langmuir and Freundlich models performed better in fitting parameters, revealing the adsorption mechanism of phenanthrene in contaminated soil by β-cyclodextrin-modified biochar. In addition, temperature has a significant effect on the adsorption capacity of β-CDBC-CA, and its application in soil remediation can be optimized by adjusting temperature. This study not only provides new materials and technical means for soil remediation but also provides important data support for an in-depth understanding of the environmental behavior of PAHs. By citing relevant research results, this study further improves the control and understanding of environmental risks of PAHs, which is of great significance for the protection of ecological environment and human health. 展开更多

At present, the commonly used denitrification methods include selective catalytic reduction (SCR), active coke, etc. As a newly developed denitrification technology, oxidation denitrification is not widely used, and the technical level is mixed, and there might be problems such as yellow smoke, secondary pollution and ozone escape in the practical application. In this paper, problems existing in the denitrification process of sintering flue gas oxidation are analyzed, and a 320 m2 sintering machine is taken as an example. Comparing the denitrification technology of sintering industry, it could be seen that the denitrification technology route of oxidation method has low pollution, low cost and high comprehensive environmental benefits, and has greatly potential development. 展开更多

In this paper, biochar (BC) was used as raw material, activated by deionizing aqueous solution, NaCl solution, CA solution and HCl solution respectively. Epichlorohydrin (EPI) was used as crosslinking agent, and β-cyclodextrin (β-CD) was used to modify biochar (BC). The prepared modified biochar materials were labeled with β-CDBC, β-CDBC-Na, β-CDBC-CA and β-CDBC-H, respectively. The infrared spectrum, X-ray diffractometer, scanning electron microscope and specific surface area of the four modified materials were tested. The results showed that the C-O stretching vibration peak at 1020 cm<sup>−</sup><sup>1</sup> of the modified materials was slightly offset compared with that of biochar. The characteristic absorption peaks of XRD pattern decrease obviously at 2θ = 26.7˚ and 29.5˚. It can be obviously observed on the electron microscope image that the surface is loaded or formed clathrates, and BET data and graphs also show that the specific surface area of the modified biochar is larger. Therefore, β-cyclodextrin successfully modified biochar and formed clathrates on the surface of biochar or was loaded in the pore structure of biochar, especially β-CDBC-CA achieved better modification effect. Because biochar and β-cyclodextrin raw materials are cheap, easy to prepare and green, and less prone to secondary pollution, it has a good advantage in environmental governance. 展开更多

The area of the macro-film is larger than 30 cm×15 cm, and this large film can be bended, or folded without any damage, and even can be tailored freely. After a simple modification of perfluoroalkysilane, the surface of the macro-film shows excellent superhydrophobicity with a water contact angle of 165.74-2 deg. and sliding angle lower than 3 deg., the prepared superhydrophobic films showing excellent antifouling, self-cleaning and water-repellent functions. The topographic roughness and perfluoroalkysilane modification are found to contribute to the observed superhydrophobicity. Considering the outstanding electronic, chemical and mechanical properties of DWCNTs, it is expected that this multifunctional DWCNT macro-film has potential applications in many fields. 展开更多

The construction of a pest detection algorithm is an important step to couple"ground-space"characteristics,which is also the basis for rapid and accurate monitoring and detection of pest damage.In four experimental areas in Sanming City,Jiangle County,Sha County and Yanping District in Fujian Province,sample data on pest damage in 182 sets of Dendrolimus punctatus were collected.The data were randomly divided into a training set and testing set,and five duplicate tests and one eliminating-indicator test were done.Based on the characterization analysis of the host for D.punctatus damage,seven characteristic indicators of ground and remote sensing including leaf area index,standard error of leaf area index(SEL)of pine forest,normalized difference vegetation index(NDVI),wetness from tasseled cap transformation(WET),green band(B2),red band(B3),near-infrared band(B4)of remote sensing image are obtained to construct BP neural networks and random forest models of pest levels.The detection results of these two algorithms were comprehensively compared from the aspects of detection precision,kappa coefficient,receiver operating characteristic curve,and a paired t test.The results showed that the seven indicators all were responsive to pest damage,and NDVI was relatively weak;the average pest damage detection precision of six tests by BP neural networks was 77.29%,the kappa coefficient was 0.6869 and after the RF algorithm,the respective values were 79.30%and 0.7151,showing that the latter is more optimized,but there was no significant difference(p>0.05);the detection precision,kappa coefficient and AUC of the RF algorithm was higher than the BP neural networks for three pest levels(no damage,moderate damage and severe damage).The detection precision and AUC of BP neural networks were a little higher for mild damage,but the difference was not significant(p>0.05)except for the kappa coefficient for the no damage level(p<0.05).An"over-fitting"phenomenon tends to occur in BP neural networks,while RF method is more robust,provi 展开更多

Seed germination is the beginning of a new lifecycle,and involves many complex physiological and biochemical reactions including seed reserve mobilization in the endosperm and nutrient transport and reuse in the embryo.Although glutelin is a dominant storage protein in rice,its contribution to seed germination and its regulatory mechanisms are mostly unknown.Gibberellin (GA) and brassinosteroid (BR),two major growth-promoting phytohormones,also play positive roles in controlling seed germination.However,how GA and BR interact and coordinate seed germination and facilitate glutelin mobilization remains unclear.In the present study,biochemical and physiological analyses of seed germination indicated that both GA and BR promote seed germination and post-germination growth.Exogenous application of GA restored germination defects caused by BR deficiency or insensitivity.Proteomic and q RT-PCR results showed that the expression of several glutelin proteins and their encoding genes was induced by BR and GA in the embryo.Expression assays suggested that the increased accumulation of glutelin protein in the embryo was due to the accelerated degradation of glutelin by a cysteine proteinase (REP-1) in the endosperm.The breakdown of glutelin in the endosperm showed a strict positive correspondence with the length of the shoot.The GluA2 mutation led to reduced degradation rate of glutelin and defects in seed germination,and the promotion effect of GA on seed germination was weakened in the glua2mutant.In vitro culture assay of rice embryos showed that glutelin mobilization functioned downstream of the GA and BR pathways to promote shoot elongation.These findings suggest a mechanism that mediates crosstalk between BR and GA in co-regulating rice seed germination and embryo growth. 展开更多

The influence of Axial-Slot Casing Treatment(ASCT)on the performance and stability enhancement mechanisms of ASCT were experimentally and numerically investigated in a highspeed mixed-flow compressor under three different tip clearances.Unsteady simulations showed the compressor stalled through end-wall stall route,i.e.the spike stall inception originating from rotor tip region,which was validated by dynamical measurements.When the ASCT was applied,greater than 20%of Stall Margin Improvement(SMI)could be achieved for the compressor under each tip clearance size.The streamwise velocity contours and flow structures in the tip region and axial slots were deeply analyzed to explore how the so called‘‘suction and injection effects"generated by the ASCT manipulate tip clearance flow and enhance the stability of compressor under different tip clearances.It was found that the dominant stability enhancement mechanisms of ASCT varies with tip clearance size for the mixed-flow compressor.(A)For the small tip clearance,the dominant mechanism of stability enhancement is the blockage reduction generated in the blade passage by the suction effect of ASCT.(B)For the large tip clearance,the injection effect of the ASCT is the dominant mechanism of stability enhancement with ASCT,which plays the leading role in delaying the spillage of incoming/tip leakage flow interface at the rotor Leading Edge(LE)plane. 展开更多

Despite the great potential of anti-PD-L1 antibodies for immunotherapy,their low response rate due to an immunosuppressive tumor microenvironment has hampered their application.To address this issue,we constructed a cell membrane-coated nanosystem(mB4S)to reverse an immunosuppressive microenvironment to an immuno-supportive one for strengthening the anti-tumor effect.In this system,Epirubicin(EPI)as an immunogenic cell death(ICD)inducer was coupled to a branched glycopolymer via hydrazone bonds and diABZI as a stimulator of interferon genes(STING)agonist was encapsulated into mB4S.After internalization of mB4S,EPI was acidic-responsively released to induce ICD,which was characterized by an increased level of calreticulin(CRT)exposure and enhanced ATP secretion.Meanwhile,diABZI effectively activated the STING pathway.Treatment with mB4S in combination with an anti-PD-L1 antibody elicited potent immune responses by increasing the ratio of matured dendritic cells(DCs)and CD8+T cells,promoting cytokines secretion,up-regulating M1-like tumor-associated macrophages(TAMs)and down-regulating immunosuppressive myeloid-derived suppressor cells(MDSCs).Therefore,this nanosystem for co-delivery of an ICD inducer and a STING agonist achieved promotion of DCs maturation and CD8+T cells infiltration,creating an immuno-supportive microenvironment,thus potentiating the therapy effect of the anti-PD-L1 antibody in both 4T1 breast and CT26 colon tumor mice. 展开更多
