The transwell chamber migration assay and CCD digital camera imaging techniques were used to investigate the relationship between regulatory volume decrease (RVD) and cell migration in nasopharyngeal carcinoma cells (...The transwell chamber migration assay and CCD digital camera imaging techniques were used to investigate the relationship between regulatory volume decrease (RVD) and cell migration in nasopharyngeal carcinoma cells (CNE-2Z cells). Both migrated and non-migrated CNE-2Z cells, when swollen by 47% hypotonic solution, exhibited RVD which was inhibited by extracellular application of chloride channel blockers adenosine 5’-triphosphate (ATP), 5-nitro-2-(3- phenylpropylamino) benzoic acid (NPPB) and tamoxifen. However, RVD rate in migrated CNE-2Z cells was bigger than that of non-migrated cells and the sensitivity of migrated cells to NPPB and tamoxifen was higher than that of non- migrated cells. ATP, NPPB and tamoxifen also inhibited migration of CNE-2Z cells. The inhibition of migration was positively correlated to the blockage of RVD, with a correlation coefficient (r) = 0.99, suggesting a functional relation- ship between RVD and cell migration. We conclude that RVD is involved in cell migration and RVD may play an important role in migratory process in CNE-2Z cells.展开更多

On the basis of the effective feature subset selected, a method is described to extract the objects which are higher than their surroundings, such as trees or forest, in the color aerial images. The efficacy of nanoparticle(NP)-based drug delivery technology is hampered by aberrant tumor stromal microenvironments(TSMs)that hinder NP transportation.Therefore,the promotion of NP permeation into deep tumor sites...The efficacy of nanoparticle(NP)-based drug delivery technology is hampered by aberrant tumor stromal microenvironments(TSMs)that hinder NP transportation.Therefore,the promotion of NP permeation into deep tumor sites via the regulation of tumor microenvironments is of critical importance.Herein,we propose a potential solution using a dihydralazine(HDZ)-loaded nanoparticle drug delivery system containing a pH-responsive,cyclic RGD peptide-modified prodrug based on doxorubicin(cRGD-Dex-DOX).With a combined experimental and theoretical approach,we find that the designed NP system can recognize the acid tumor environments and precisely release the encapsulated HDZ into tumor tissues.HDZ can notably downregulate the expression levels of hypoxia-inducible factor 1α(HIF1α),α-smooth muscle actin,and fibronectin through the dilation of tumor blood vessels.These changes in the TSMs enhance the enrichment and penetration of NPs and also unexpectedly promote the infiltration of activated T cells into tumors,suggesting that such a system may offer an effective“multifunctional therapy”through both improving the chemotherapeutic effect and enhancing the immune response to tumors.In vivo experiments on 4T1 breast cancer bearing mice indeed validate that this therapy has the most outstanding antitumor effects over all the other tested control regimens,with the lowest side effects as well.展开更多

Accurate segmentation and detection of rice seedlings is essential for precision agriculture and high-yield cultivation.However,current methods suffer from high computational complexity and poor robustness to differen...Accurate segmentation and detection of rice seedlings is essential for precision agriculture and high-yield cultivation.However,current methods suffer from high computational complexity and poor robustness to different rice varieties and densities.This article proposes 2 Iightweight neural network architectures,LW-Segnet and LW-Unet,for high-precision rice seedling segmentation.The networks adopt an encoder-decoderstructure with hybrid lightweight convolutions and spatial pyramid dilated convolutions,achieving accurate segmentation while reducing model parameters.Multispectral imagery acquired by unmanned aerial vehicle(UAV)was used to train and test the models covering 3 rice varieties and different planting densities.Experimental results demonstrate that the proposed LW-Segnet and LW-Unet models achieve higher F1-scores and intersection over union values for seedling detection and row segmentation across varieties,indicating improved segmentation accuracy.Furthermore,the models exhibit stable performance when handling different varieties and densities,showing strong robustness.In terms of efficiency,the networks have lower graphics processing unit memory usage,complexity,and parameters but faster inference speeds,reflecting higher computational efficiency.In particular,the fast speed of LW-Unet indicates potential for real-time applications.The study presents lightweight yet effective neural network architectures for agricultural tasks.By handling multiple rice varieties and densities with high accuracy,efficiency,and robustness,the models show promise for use in edge devices and UAVs to assist precision farming and crop management.The findings provide valuable insights into designing lightweight deep learning models to tackle complex agricultural problems.展开更多
Compared to conventional cancer treatment, combination therapy based on well-designed nanoscale platforms may offer an opportunity to eliminate tumors and reduce recurrence and metastasis.In this study, we prepared mu...Compared to conventional cancer treatment, combination therapy based on well-designed nanoscale platforms may offer an opportunity to eliminate tumors and reduce recurrence and metastasis.In this study, we prepared multifunctional microspheres loading ^(131)I-labeled hollow copper sulfide nanoparticles and paclitaxel( ^(131)I-HCu SNPs-MS-PTX) for imaging and therapeutics of W256/B breast tumors in rats.18 F-fluordeoxyglucose(18 F-FDG) positron emission tomography/computed tomography(PET/CT) imaging detected that the expansion of the tumor volume was delayed(Po0.05) following intra-tumoral(i.t.) injection with ^(131)I-HCu SNPs-MS-PTX plus near-infrared(NIR) irradiation. The immunohistochemical analysis further confirmed the anti-tumor effect. The single photon emission computed tomography(SPECT)/photoacoustic imaging mediated by ^(131)I-HCu SNPs-MS-PTX demonstrated that microspheres were mainly distributed in the tumors with a relatively low distribution in other organs. Our results revealed that ^(131)I-HCu SNPs-MS-PTX offered combined photothermal, chemo-and radio-therapies, eliminating tumors at a relatively low dose, as well as allowing SPECT/CT and photoacoustic imaging monitoring of distribution of the injected agents non-invasively. The copper sulfide-loaded microspheres, ^(131)I-HCu SNPs-MS-PTX, can serve as a versatile theranostic agent in an orthotopic breast cancer model.展开更多
Recent change detection(CD)methods focus on the extraction of deep change semantic features.However,existing methods overlook the fine-grained features and have the poor ability to capture long-range space–time infor...Recent change detection(CD)methods focus on the extraction of deep change semantic features.However,existing methods overlook the fine-grained features and have the poor ability to capture long-range space–time information,which leads to the micro changes missing and the edges of change types smoothing.In this paper,a potential transformer-based semantic change detection(SCD)model,Pyramid-SCDFormer is proposed,which precisely recognizes the small changes and fine edges details of the changes.The SCD model selectively merges different semantic tokens in multi-head self-attention block to obtain multiscale features,which is crucial for extraction information of remote sensing images(RSIs)with multiple changes from different scales.Moreover,we create a well-annotated SCD dataset,Landsat-SCD with unprecedented time series and change types in complex scenarios.Comparing with three Convolutional Neural Network-based,one attention-based,and two transformer-based networks,experimental results demonstrate that the Pyramid-SCDFormer stably outperforms the existing state-of-the-art CD models and obtains an improvement in MIoU/F1 of 1.11/0.76%,0.57/0.50%,and 8.75/8.59%on the LEVIR-CD,WHU_CD,and Landsat-SCD dataset respectively.For change classes proportion less than 1%,the proposed model improves the MIoU by 7.17–19.53%on Landsat-SCD dataset.The recognition performance for small-scale and fine edges of change types has greatly improved.展开更多

SARS-CoV-2 has caused more than 3.8 million deaths worldwide,and several types of COVID-19 vaccines are urgently approved for use,including adenovirus vectored vaccines.However,the thermal instability and pre-existing...SARS-CoV-2 has caused more than 3.8 million deaths worldwide,and several types of COVID-19 vaccines are urgently approved for use,including adenovirus vectored vaccines.However,the thermal instability and pre-existing immunity have limited its wide applications.To circumvent these obstacles,we constructed a self-biomineralized adenovirus vectored COVID-19 vaccine(Sad23L-n Co V-S-Ca P)by generating a calcium phosphate mineral exterior(Ca P)based on Sad23L vector carrying the full-length gene of SARS-Co V-2 spike protein(S)under physiological condition.This Sad23L-n Co V-S-Ca P vaccine was examined for its characteristics of structure,thermostability,immunogenicity and avoiding the problem of preexisting immunity.In thermostability test,Sad23L-n Co V-S-Ca P could be stored at 4°C for over 45 days,26°C for more than 8 days and 37°C for approximately 2 days.Furthermore,Sad23L-n Co V-S-Ca P induced higher level of S-specific antibody and T cell responses,and was not affected by the pre-existing anti-Sad23L immunity,suggesting it could be used as boosting immunization on Sad23L-n Co V-S priming vaccination.The boosting with Sad23L-n Co V-S-Ca P vaccine induced high titers of 10^(5.01)anti-S1,10^(4.77)anti-S2 binding antibody,10^(3.04) pseudovirus neutralizing antibody(IC_(50)),and robust T-cell response of IFN-c(1466.16 SFCs/10^(6)cells)to S peptides,respectively.In summary,the selfbiomineralization of the COVID-19 vaccine Sad23L-n Co V-S-Ca P improved vaccine efficacy,which could be used in prime-boost regimen for prevention of SARS-Co V-2 infection in humans.展开更多

Trigeminal nerve schwannomas, which are rare, slowly growing, benign tumors, account for 0.2% to 1.0% of all intracranial tumors and 0.8% to 8.0% of intracranial schwannomas.1-5 These tumors are treated surgically.1-4...Trigeminal nerve schwannomas, which are rare, slowly growing, benign tumors, account for 0.2% to 1.0% of all intracranial tumors and 0.8% to 8.0% of intracranial schwannomas.1-5 These tumors are treated surgically.1-4 The development of microsurgery and skull base surgery has made complete resection possible in most patients. Nevertheless, cranial nerve sequelae appear after complete resection of these tumors because they are located close to the cavernous sinus and usually adhere to the vital vascular and neural structures. As an alternative to microsurgical resection, Leksell gamma knife (LGK) radiosurgery has been performed for patients with intracranial schwannomas to minimize the treatment-related morbidity and achieve a long-term control of tumor growth.6,7 In this report, we describe our 6-year experience in the treatment of 38 patients with trigeminal schwannomas by LGK.展开更多

The single photon emission computed tomography(SPECT)/photoacoustic imaging mediated by ^(131)I-HCu SNPs-MS-PTX demonstrated that microspheres were mainly distributed in the tumors with a relatively low distribution in other organs. Our results revealed that ^(131)I-HCu SNPs-MS-PTX offered combined photothermal, chemo-and radio-therapies, eliminating tumors at a relatively low dose, as well as allowing SPECT/CT and photoacoustic imaging monitoring of distribution of the injected agents non-invasively. The copper sulfide-loaded microspheres, ^(131)I-HCu SNPs-MS-PTX, can serve as a versatile theranostic agent in an orthotopic breast cancer model.

文摘Recent change detection(CD)methods focus on the extraction of deep change semantic features.However,existing methods overlook the fine-grained features and have the poor ability to capture long-range space–time information,which leads to the micro changes missing and the edges of change types smoothing.In this paper,a potential transformer-based semantic change detection(SCD)model,Pyramid-SCDFormer is proposed,which precisely recognizes the small changes and fine edges details of the changes.The SCD model selectively merges different semantic tokens in multi-head self-attention block to obtain multiscale features,which is crucial for extraction information of remote sensing images(RSIs)with multiple changes from different scales.Moreover,we create a well-annotated SCD dataset,Landsat-SCD with unprecedented time series and change types in complex scenarios.Comparing with three Convolutional Neural Network-based,one attention-based,and two transformer-based networks,experimental results demonstrate that the Pyramid-SCDFormer stably outperforms the existing state-of-the-art CD models and obtains an improvement in MIoU/F1 of 1.11/0.76%,0.57/0.50%,and 8.75/8.59%on the LEVIR-CD,WHU_CD,and Landsat-SCD dataset respectively.For change classes proportion less than 1%,the proposed model improves the MIoU by 7.17–19.53%on Landsat-SCD dataset.The recognition performance for small-scale and fine edges of change types has greatly improved.
文摘This paper introduces a novel technique for object detection using genetic algorithms and morphological processing. The method employs a kind of object oriented structure element, which is derived by genetic algorithms. The population of morphological filters is iteratively evaluated according to a statistical performance index corresponding to object extraction ability, and evolves into an optimal structuring element using the evolution principles of genetic search. Experimental results of road extraction from high resolution satellite images are presented to illustrate the merit and feasibility of the proposed method.
文摘目的了解四川凉山彝族成人脂肪和肥胖相关基因(fat mass and obesity associated gene,FTO)多态性与肥胖的关联。方法采用病例对照研究方法,病例组为彝族移民流行病学调查中收集的肥胖者[体重指数(body mass index,BMI)≥28 kg/m^2],共322例;对照组为非肥胖非超重者(BMI<24 kg/m^2),按年龄和性别与病例组进行1∶2频数匹配,共644例。采用TaqMan实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)法对FTO基因(rs1121980和rs17817449)进行基因分型。采用非条件Logistic回归分析模型分析两位点多态性与肥胖的关联。结果彝族农民中,加性模型和显性模型下,rs1121980位点与肥胖均存在统计学关联(加性模型:OR=1.45,95%CI:1.01~2.07,P=0.042;显性模型:OR=1.52,95%CI:1.01~2.31,P=0.048)。彝族农民中未见rs17817449与肥胖存在统计学关联(均有P>0.05)。彝族移民中,两位点(rs1121980和rs17817449)与肥胖均未见统计学关联(均有P>0.05)。结论彝族农民FTO rs1121980多态性与成人肥胖存在统计学关联,A等位基因是肥胖的危险因素。彝族移民FTO两位点与成人肥胖均未见关联。

文摘SARS-CoV-2 has caused more than 3.8 million deaths worldwide,and several types of COVID-19 vaccines are urgently approved for use,including adenovirus vectored vaccines.However,the thermal instability and pre-existing immunity have limited its wide applications.To circumvent these obstacles,we constructed a self-biomineralized adenovirus vectored COVID-19 vaccine(Sad23L-n Co V-S-Ca P)by generating a calcium phosphate mineral exterior(Ca P)based on Sad23L vector carrying the full-length gene of SARS-Co V-2 spike protein(S)under physiological condition.This Sad23L-n Co V-S-Ca P vaccine was examined for its characteristics of structure,thermostability,immunogenicity and avoiding the problem of preexisting immunity.In thermostability test,Sad23L-n Co V-S-Ca P could be stored at 4°C for over 45 days,26°C for more than 8 days and 37°C for approximately 2 days.Furthermore,Sad23L-n Co V-S-Ca P induced higher level of S-specific antibody and T cell responses,and was not affected by the pre-existing anti-Sad23L immunity,suggesting it could be used as boosting immunization on Sad23L-n Co V-S priming vaccination.The boosting with Sad23L-n Co V-S-Ca P vaccine induced high titers of 10^(5.01)anti-S1,10^(4.77)anti-S2 binding antibody,10^(3.04) pseudovirus neutralizing antibody(IC_(50)),and robust T-cell response of IFN-c(1466.16 SFCs/10^(6)cells)to S peptides,respectively.In summary,the selfbiomineralization of the COVID-19 vaccine Sad23L-n Co V-S-Ca P improved vaccine efficacy,which could be used in prime-boost regimen for prevention of SARS-Co V-2 infection in humans.
文摘Intramolecular cyclization of N-alkoxyl amines are studied for the stereoselective preparation of 2, 4-disubstituted pyrrolidine derivatives. Reduction of oximes under acidic conditions by NaBH3CN afforded the corresponding nucleophilic hydroxylamine derivatives, which subsequently cyclized via SN2' mechanism to give the desired N-alkoxyl pyrrolidines.
文摘Trigeminal nerve schwannomas, which are rare, slowly growing, benign tumors, account for 0.2% to 1.0% of all intracranial tumors and 0.8% to 8.0% of intracranial schwannomas.1-5 These tumors are treated surgically.1-4 The development of microsurgery and skull base surgery has made complete resection possible in most patients. Nevertheless, cranial nerve sequelae appear after complete resection of these tumors because they are located close to the cavernous sinus and usually adhere to the vital vascular and neural structures. As an alternative to microsurgical resection, Leksell gamma knife (LGK) radiosurgery has been performed for patients with intracranial schwannomas to minimize the treatment-related morbidity and achieve a long-term control of tumor growth.6,7 In this report, we describe our 6-year experience in the treatment of 38 patients with trigeminal schwannomas by LGK.
