This study was designed to investigate whether ultrasound-mediated microbubble destruction could effectively deliver therapeutic plasmid into the retina of rat, and whether gene transfer of pigment epithelium-derived factor (PEDF) could inhibit CNV.Methods Human retinal pigment epithelial cells were isolated and treated either with ultrasound or plasmid alone, or with a combination of plasmid, ultrasound and microbubbles to approach feasibility of microbubble-enhanced ultrasound enhance PEDFgene expression; For in vivo animal studies, CNV was induced by argon lasgon laser in rats. These rats were randomly assigned to five groups and were treated by infusing microbubbles attached with the naked plasmid DNA of PEDF into the vitreous of rats followed by immediate ultrasound exposure (intravitreal injection); infusing liposomes with the naked plasmid DNA of PEDF into the vitreous (lipofectamine + PEDF); infusing microbubbles attached with PEDF into the orbit of rats with ultrasound irradiation immediately (retrobular injection); infusing microbubbles attached with PEDF into the femoral vein of rats with exposed to ultrasound immediately (vein injection). The CNV rats without any treatment served as control. Rats were sacrificed and eyes were enucleated at 7, 14, and 28 days after treatment. Gene and protein expression of PEDF was detected by quantitative real-time RT-PCR, Western blotting and immunofluorescence staining, respectively. The effect of PEDF gene transfer on CNV was examined by fluorescein fundus angiography.Results In vitro cell experiments showed that microbubbles with ultrasound irradiation could significantly enhance PEDF delivery as compared with microbubbles or ultrasound 展开更多
目的观察中医经筋手法结合运动疗法对小儿痉挛型脑瘫运动及步行能力的影响。方法选取2021年1月—2022年6月期间湖南省儿童医院收治的127例痉挛型脑瘫患儿为研究对象,以单纯随机抽样法分为对照组63例和观察组64例。对照组给予常规对症治...目的观察中医经筋手法结合运动疗法对小儿痉挛型脑瘫运动及步行能力的影响。方法选取2021年1月—2022年6月期间湖南省儿童医院收治的127例痉挛型脑瘫患儿为研究对象,以单纯随机抽样法分为对照组63例和观察组64例。对照组给予常规对症治疗和运动疗法,观察组在对照组基础上联合中医经筋手法治疗。治疗3个月后,观察比较两组患儿临床疗效,治疗前后改良Ashworth痉挛量表(Modified Ashworth Scale,MAS)评分、精细运动功能评定量表(Peabody developmental motor scale,PDMS-2)评分、粗大运动功能评定量表(Gross Motor Function Measure,GMFM)评分和步行能力。结果治疗后观察组临床总有效率93.75%(60/64)高于对照组80.95%(51/63),差异有统计学意义(P<0.05)。治疗1、2、3个月后两组患儿MAS评分均较治疗前下降,差异有统计学意义(P<0.05);且观察组低于对照组,差异有统计学意义(P<0.05)。治疗后两组患儿抓握能力、视觉运动能力评分均较治疗前升高,差异有统计学意义(P<0.05);两组患儿抓握能力、视觉运动能力评分比较,差异无统计学意义(P>0.05)。治疗后两组患儿GMFM行走和跑跳、站立位、坐位、爬与跪位、卧位与翻身评分均较治疗前升高,差异有统计学意义(P<0.05);且观察组站立位、坐位、爬与跪位评分高于对照组,差异有统计学意义(P<0.05)。治疗后两组患儿跨步长、1 min步行距离较治疗前增加,步宽、10 m步行时间较治疗前减少,差异有统计学意义(P<0.05);且观察组跨步长、1 min步行距离长于对照组,步宽、10 m步行时间短于对照组,差异有统计学意义(P<0.05)。结论中医经筋手法联合运动疗法治疗痉挛性脑瘫患儿疗效较好,有利于减轻肌张力、缓解肌肉痉挛状态,可提高粗大运动功能,具有较好的应用价值。展开更多
In order to understand the structural characteristics of lignin in triploid clones ofPopulus tomentosa and its changes in the processes of pulping and bleaching, milled wood lignin (MWL), lignin carbohydrate complex...In order to understand the structural characteristics of lignin in triploid clones ofPopulus tomentosa and its changes in the processes of pulping and bleaching, milled wood lignin (MWL), lignin carbohydrate complex (LCC) and the residual lignin from kraft pulp (KP) and sulfite pulp (SP) were isolated and analyzed by Fourier transform infrared (FTIR) spectrum and ^13C nuclear magnetic resonance (NMR). The most diagnostic peaks were assigned and the differences were discussed. The spectral patterns reveal that triploid P tomentosa shows the specific features of hardwood from temperate areas, but in the spectrum of FTIR, the strength ratio orAl270 cm^-1 to A1226 cm^-1 is 0.88, higher than the average of hardwood from temperate areas, which will make the lignin delignification more difficult during pulping and bleaching. The LCC from triploid P tomentosa is mainly composed of xyloglucan and glucuronic acid, and other glucides have much lower ratio. In LCC FTIR, there are three peaks at 1 427, 1 329 and 1 046 cm^-1, indicating that both semi-cellulose and cellulose could exist in LCC, and that there might be relationships between cellulose and lignin. Compared with the residual lignin from KP and SP, the condensed structure in KP is more than that in SP.展开更多

This makes cancer stem cells prime targets for therapeutic intervention. Eradicating cancer stem cells by efficient targeting agents may have the potential to cure cancer. In this review, we summarize recent breakthroughs that have improved our understanding of cancer stem cells, and we discuss the therapeutic strategy of targeting cancer stem cells, a promising future direction for cancer stem cell research.展开更多
Objective: To evaluate the immunoregulation of KangAi (KA) injection combined with chemo-treatment (chemo) for Non-small cell lung cancer (NSCLC). Methods: We systematically searched the literature of PubMed, EMBASE, ...Objective: To evaluate the immunoregulation of KangAi (KA) injection combined with chemo-treatment (chemo) for Non-small cell lung cancer (NSCLC). Methods: We systematically searched the literature of PubMed, EMBASE, CENTRAL, MEDLINE, CNKI, Wanfang, and VIP databases for all Randomized controlled trials (published from the earliest possible year to January 2019, no language restrictions) comparing KA injection combined with chemo and chemo alone in patients with NSCLC. Our main endpoints were immune function, clinical efficiency, KPS score and adverse events. The Cochrane Risk of Bias tool was applied for quality assessment. Results: 11 studies involving 1060 participants were included. The immune function (MD=3.18, 95% CI: 0.98-4.00, P<0.00001), clinical response rate (RR=1.28, 95%CI: 1.17-1.40, P<0.00001), KPS score (RR=1.86, 95%CI: 1.35-2.57, P=0.0002), and adverse events (RR=0.57, 95%CI: 0.50-0.65, P<0.00001) in the group of KA injection plus Chemo were vastly different from those in Chemo alone. Conclusion: KA injection combined with Chemo in the treatment of NSCLC improved the immune function, clinical efficiency and safety compared to Chemo alone. However, because many of the methodologies included in randomized controlled trials are of poor quality, more rigorous design and large randomized controlled trials are needed to test this benefit.展开更多
