A reliable serum marker for cholangiocarcinoma would be a useful diagnostic test. The aims of our study were to evaluate the usefulness of a serum CA19-9 determination in the diagnosis of cholangiocarcinoma.METHODS: We prospectively measured serum CA19-9 and CEA concentrations in patients with cholangiocarcinoma (n=35), benign biliary diseases (n=92), and healthy individuals (n=15). Serum CA19-9 and CEA concentrations were measured by an immunoradiometric assay without knowledge of the clinical diagnosis.RESULTS:The sensitivity of a CA19-9 value>37KU·L^-1 and a CEA value >22μg·L^-1 in diagnosing cholangiocarcinoma were 77.14% and 68.57%, respectively. When compared with the benign biliary diseases group,the true negative rates of serum CA19-9 and CEA were 84.78% and 81.52%,respectively. The false positive rates of serum CA19-9 and CEA were 15.22% and 18.48%, whereas the accuracy of serum CA19-9 and CEA were 82.68% and 77.95%,respectively. Serum CA19-9 and CEA concentrations were significantly elevated (P<0.001 and P<0.05) in patients with cholangiocarcinoma (290.31±5.34KU·L^-1 and 36.46±18.03μg·L^-1) compared with patients with benign biliary diseases (13.38±2.59KU·L^-1 and 13.84±3.85μg·L^-1) and healthy individuals (12.78±3.69KU·L^-1 and 11.48±3.37μg·L^-1). In 15 patients undergoing curative resection of cholangiocarcinoma,the mean serum CA19-9 concentration was decreased from a preoperative level of 286.41±4.36KU·L^-1 to a postoperative level of 62.01±17.43KU·L^-1 (P<0.001), and the mean serum CEA concentration from 39.41±24.35μg·L^-1 to 28.69±11.03μg·L6-1(P<0.05). In patients with cholangiocarcinoma,however, no correlation was found between serum CEA and CA19-9 concentrations (r=-0.036).CONCLUSION:These data suggest that the serum CA19-9 determination is a useful addition to the available tests for the differential diagnosis of cholangiocarcinoma. In addition to their many well-known advantages(e.g.,ultra-high porosity,good pore size distribution,easy functionalization,and structural tolerability),metal-organic frameworks(MOFs)are a new class of advanced functi...In addition to their many well-known advantages(e.g.,ultra-high porosity,good pore size distribution,easy functionalization,and structural tolerability),metal-organic frameworks(MOFs)are a new class of advanced functional materials.However,their backbones are highly susceptible to deformation after exposure to acidic or alkaline conditions.As a result of lithium-ion batteries embedding or detaching directly from MOFs,they irreversibly collapse.As a result,they fail to maintain their electrochemical performance.These factors have hindered the development of MOFs as direct electrode materials,making the design of MOF materials with controlled morphology and stable dimensions a new challenge.In this study,we adopted a versatile and effective method to synthesize a novel MOF material(NiCo-BP(BP=BTC/phen and BTC=1,3,5-benzenetricarboxylic acid))using the rigid ligands 1,10-phenanthroline and homobenzotrizoic acid,and the emergence of the Ni-O/N and Co-O/N coordination layers was observed by extended X-ray absorption fine structure(EXAFS)tests,indicating that Ni and Co were coordinated with heterocyclic N-given atoms to form a stable p-πconjugated structure.Meanwhile,the metal-ion is attached to the carboxylic acid ligand on the other side,making the metal-organic skeleton complete and robust.The nanosphere structure of NiCo-BP(~400 nm)allows for full exposure and utilisation of the active sites,especially the Ni,Co,and phenanthroline units,and exhibit impressively high specific capacity and cycling stability.At a high current density of 1.0 A·g^(−1),a high discharge specific capacity of 631.6 mAh·g^(−1)was obtained after 1000 cycles.The co-participation of two organic ligands in the coordination is in accordance with the theory of soft and hard acids and bases,which contributes to the ability of the material to maintain a high capacity in cycling as well as its cyclic stability.展开更多
This study aims to investigate the influence of CO_(2)-mixing dose(mass fractions of 0.3%,0.6%,and 0.9%)and prolonged mixing time on the fresh and hardened properties of cement pastes.The CO_(2)-mixing can act as coag...This study aims to investigate the influence of CO_(2)-mixing dose(mass fractions of 0.3%,0.6%,and 0.9%)and prolonged mixing time on the fresh and hardened properties of cement pastes.The CO_(2)-mixing can act as coagulant in fresh cement mixtures,resulting in a significant reduction in workability associated with the formation of a rich calcium carbonate network on the surface of cement particles.The CO_(2)-mixing cement pastes were found to be much stiffer and more difficult to handle,place,and compact than the control mixture,which had a negative effect on the mechanical strength performance of the hardened pastes.However,prolonging the mixing time for 1 min(immediately after CO_(2)-mixing)can effectively improve the workability(by~53%–85%)by breaking up the flocculation network of deposited calcium carbonates.As a result,the presence of detached calcium carbonate accelerated early cement hydration and densified the microstructure;this improved early-age compressive strength by~6%–32%,depending on the CO_(2)-mixing dose used.Therefore,it seems that the CO_(2)-mixing dose should be controlled at≤0.6%with the mixing time prolonged in order to attain satisfactory workability and mechanical strength.展开更多

The initiation of the floral elements is a centripetal spiral and the direction of the spiral is clockwise or anti-clockwise. However, the development of the androecium is highly unusual: in a longitudinal series of four stamens, the second stamen develops first from the inner to outer, then the third one, the fourth one and the first one in turn. The microsporogenesis and anther maturation follows the same developmental sequence. The tepals are different from the bracts and the stamens in both shape and size in the early developmental stage, but there is no difference between the stamens and carpels in the early developmental stage. Therefore, we established a spatio-temporal process of the floral morphogenesis of A. rivularis var. flore-minore and offer another meaning of the floral diversity patterns attributed to the level of the genus.展开更多
Pseudomonas aeruginosa is one of the leading nosocomial pathogens that causes both severe acute and chronic infections.The strong capacity of P.aeruginosa to form biofilms can dramatically increase its antibiotic resi...Pseudomonas aeruginosa is one of the leading nosocomial pathogens that causes both severe acute and chronic infections.The strong capacity of P.aeruginosa to form biofilms can dramatically increase its antibiotic resistance and lead to treatment failure.The biofilm resident bacterial cells display distinct gene expression profiles and phenotypes compared to their free-living counterparts.Elucidating the genetic determinants of biofilm formation is crucial for the development of antibiofilm drugs.In this study,a highthroughput transposon-insertion site sequencing(Tn-seq)approach was employed to identify novel P.aeruginosa biofilm genetic determinants.When analyzing the novel biofilm regulatory genes,we found that the cell division factor ZapE(PA4438)controls the P.aeruginosa pqs quorum sensing system.The ΔzapE mutant lost fitness against the wild-type PAO1 strain in biofilms and its production of 2-heptyl-3-hydroxy-4(1H)-quinolone(PQS)had been reduced.Further biochemical analysis showed that ZapE interacts with PqsH,which encodes the synthase that converts 2-heptyl-4-quinolone(HHQ)to PQS.In addition,site-directed mutagenesis of the ATPase active site of ZapE(K72A)abolished the positive regulation of ZapE on PQS signaling.As ZapE is highly conserved among the Pseudomonas group,our study suggests that it is a potential drug target for the control of Pseudomonas infections.展开更多
Background: A comprehensive metabolism network of engineered E. coli is very important in systems biology and metabolomics studies. Many tools focus on two-dimensional space to display pathways in metabolic network. However, the usage of three-dimensional visualization may help to understand better the intricate topology of metabolic and regulatory networks. Methods: We manually curated large amount of experimental data (including pathways, reactions and metabolites) from literature related with different types of engineered E. coli and then utilized a novel technology of three dimensional visualization to develop a comprehensive metabolic network named SynBioEcoll. Results: SynBioEoli contains 740 biosynthetic pathways, 3,889 metabolic reactions, 2,255 chemical compounds manually curated from about 11,000 metabolism publications related with different types of engineered E. coil Furthermore, SynBioEcoli integrates with various informatics techniques. Conclusions: SynBioEcoli could be regarded as a comprehensive knowledgebase of engineered E. coli and represents the next generation cellular metabolism network visualization technology. To investigate the feasibility and safety of alginate fibers loaded with chlorhexidine gluconate(CHG)for biomedical applications,vitro cytotoxic test and drug release pattern were studied.Discovery Studio(DS),a comput...To investigate the feasibility and safety of alginate fibers loaded with chlorhexidine gluconate(CHG)for biomedical applications,vitro cytotoxic test and drug release pattern were studied.Discovery Studio(DS),a computer aided software,was utilized to briefly investigate the energy state and solvation at the molecular level.Alginate fiber loaded with CHG presented different drug release patterns in PBS and A solution.Cytotoxicity results indicated that CHG-loaded alginate fibers can promote the growth of L-929 mouse fibroblast cells with better biocompatibility.Through the simulation,the lowest energy conformation of three representative molecular bond regions was optimized with stronger stability.As the torsional energy increased,the bond energy of the target regions decreased.Besides,the transition energy of the target regions was greatly reduced by solvation.In a word,alginate fibers loaded with CHG expressed better biocompatibility and drug sustained-release ability,which indicates that CHG-loaded alginate fibers has a broad application prospect in medical dressing.展开更多

In Study A comparing SR-test with IR-reference in a crossover design T-max and t(1/2) of propafenone for SR-test were significantly higher than those for IR-reference while C-max and AUC were lower demonstrating the sustained release properties of the new formulation. In Study B comparing SR-test with SR-reference the observed C and A TIC of propafenone for SR test (124.5 +/- 140.0 ng/mL and 612.0 +/- 699.2 ng.h/mL, respectively) were higher than for SR-reference (78.52 +/- 72.92 ng/mL and 423.6 +/- 431.6 ng.h/mL, respectively) although the differences were not significant. Overall, the new formulation has as good if not better sustained release characteristics to the market leader formulation. 背景与目的:恶性肿瘤严重威胁着居民健康,已成为重大的公共卫生问题。本研究旨在描述和分析2015年上海市恶性肿瘤流行特征。方法:根据上海市恶性肿瘤病例报告登记系统收集的恶性肿瘤发病和死亡资料,按地区、性别分层,分别计算恶性肿瘤...背景与目的:恶性肿瘤严重威胁着居民健康,已成为重大的公共卫生问题。本研究旨在描述和分析2015年上海市恶性肿瘤流行特征。方法:根据上海市恶性肿瘤病例报告登记系统收集的恶性肿瘤发病和死亡资料,按地区、性别分层,分别计算恶性肿瘤发病与死亡粗率、标化率、前10位恶性肿瘤发病与死亡顺位和构成等,并应用Joinpoint统计软件分析2002—2015年上海市肺癌发病和死亡趋势,估算总体和分阶段的年度变化百分比(annual percent change,APC)。采用Segi’s世界标准人口年龄构成计算标化率。结果:2015年上海市共报告恶性肿瘤新发病例71 610例,死亡病例38 445例。病理学诊断比例(percentage of morphologically verified cases,MV%)为78.42%,只有死亡医学证明书比例(percentage of death certifications only,DCO%)为0.21%,死亡发病比(mortality to incidence ratio,M/I)为0.55。上海市恶性肿瘤粗发病率为497.33/10万,标化发病率为228.82/10万,男性标化发病率低于女性,市区低于郊区。恶性肿瘤发病在40岁以后快速上升,在80~84岁年龄组达到高峰。全市发病前10位恶性肿瘤依次为肺癌、结直肠癌、甲状腺癌、胃癌、乳腺癌、肝癌、前列腺癌、胰腺癌、脑和中枢神经系统肿瘤以及膀胱癌,前10位恶性肿瘤占全部恶性肿瘤发病的76.59%。全市恶性肿瘤粗死亡率为267.00/10万,标化死亡率为95.99/10万,男性标化死亡率高于女性,市区和郊区基本持平。死亡率在45岁以后快速上升,在≥85岁年龄组达到高峰。死亡前10位恶性肿瘤依次为肺癌、结直肠癌、胃癌、肝癌、胰腺癌、乳腺癌、食管癌、胆囊癌、前列腺癌以及脑和中枢神经系统肿瘤,前10位恶性肿瘤占全部恶性肿瘤死亡的78.07%。截至2016年12月31日,上海市共有399 027例现患肿瘤病例,现患率为2.77%。市区现患率为3.07%,郊区为2.55%。乳腺癌是现患病例中最常见的恶性肿瘤,占15.33%�展开更多

文摘Background: A comprehensive metabolism network of engineered E. coli is very important in systems biology and metabolomics studies. Many tools focus on two-dimensional space to display pathways in metabolic network. However, the usage of three-dimensional visualization may help to understand better the intricate topology of metabolic and regulatory networks. Methods: We manually curated large amount of experimental data (including pathways, reactions and metabolites) from literature related with different types of engineered E. coli and then utilized a novel technology of three dimensional visualization to develop a comprehensive metabolic network named SynBioEcoll. Results: SynBioEoli contains 740 biosynthetic pathways, 3,889 metabolic reactions, 2,255 chemical compounds manually curated from about 11,000 metabolism publications related with different types of engineered E. coil Furthermore, SynBioEcoli integrates with various informatics techniques. Conclusions: SynBioEcoli could be regarded as a comprehensive knowledgebase of engineered E. coli and represents the next generation cellular metabolism network visualization technology. It could be accessed via web browsers (such as Google Chrome) supporting WebGL, at http://www.rxnfinder.org/synbioecoli/.

文摘To investigate the feasibility and safety of alginate fibers loaded with chlorhexidine gluconate(CHG)for biomedical applications,vitro cytotoxic test and drug release pattern were studied.Discovery Studio(DS),a computer aided software,was utilized to briefly investigate the energy state and solvation at the molecular level.Alginate fiber loaded with CHG presented different drug release patterns in PBS and A solution.Cytotoxicity results indicated that CHG-loaded alginate fibers can promote the growth of L-929 mouse fibroblast cells with better biocompatibility.Through the simulation,the lowest energy conformation of three representative molecular bond regions was optimized with stronger stability.As the torsional energy increased,the bond energy of the target regions decreased.Besides,the transition energy of the target regions was greatly reduced by solvation.In a word,alginate fibers loaded with CHG expressed better biocompatibility and drug sustained-release ability,which indicates that CHG-loaded alginate fibers has a broad application prospect in medical dressing.
文摘Ins paper describes the development and validation of a liquid chromatography mass spectrometric assay for propafenone and its application to a pharmacokinetic study of propafenone administered as a new propafenone hydrochloride sustained-release capsule (SR-test), as an instant-release tablet (IR-reference) and as the market leader sustained-release capsule (Rythmod SR-reference) in male beagle dogs (n= 8). In Study A comparing SR-test with IR-reference in a crossover design T-max and t(1/2) of propafenone for SR-test were significantly higher than those for IR-reference while C-max and AUC were lower demonstrating the sustained release properties of the new formulation. In Study B comparing SR-test with SR-reference the observed C and A TIC of propafenone for SR test (124.5 +/- 140.0 ng/mL and 612.0 +/- 699.2 ng.h/mL, respectively) were higher than for SR-reference (78.52 +/- 72.92 ng/mL and 423.6 +/- 431.6 ng.h/mL, respectively) although the differences were not significant. Overall, the new formulation has as good if not better sustained release characteristics to the market leader formulation. 文摘背景与目的:恶性肿瘤严重威胁着居民健康,已成为重大的公共卫生问题。本研究旨在描述和分析2015年上海市恶性肿瘤流行特征。方法:根据上海市恶性肿瘤病例报告登记系统收集的恶性肿瘤发病和死亡资料,按地区、性别分层,分别计算恶性肿瘤发病与死亡粗率、标化率、前10位恶性肿瘤发病与死亡顺位和构成等,并应用Joinpoint统计软件分析2002—2015年上海市肺癌发病和死亡趋势,估算总体和分阶段的年度变化百分比(annual percent change,APC)。采用Segi’s世界标准人口年龄构成计算标化率。结果:2015年上海市共报告恶性肿瘤新发病例71 610例,死亡病例38 445例。病理学诊断比例(percentage of morphologically verified cases,MV%)为78.42%,只有死亡医学证明书比例(percentage of death certifications only,DCO%)为0.21%,死亡发病比(mortality to incidence ratio,M/I)为0.55。上海市恶性肿瘤粗发病率为497.33/10万,标化发病率为228.82/10万,男性标化发病率低于女性,市区低于郊区。恶性肿瘤发病在40岁以后快速上升,在80~84岁年龄组达到高峰。全市发病前10位恶性肿瘤依次为肺癌、结直肠癌、甲状腺癌、胃癌、乳腺癌、肝癌、前列腺癌、胰腺癌、脑和中枢神经系统肿瘤以及膀胱癌,前10位恶性肿瘤占全部恶性肿瘤发病的76.59%。全市恶性肿瘤粗死亡率为267.00/10万,标化死亡率为95.99/10万,男性标化死亡率高于女性,市区和郊区基本持平。死亡率在45岁以后快速上升,在≥85岁年龄组达到高峰。死亡前10位恶性肿瘤依次为肺癌、结直肠癌、胃癌、肝癌、胰腺癌、乳腺癌、食管癌、胆囊癌、前列腺癌以及脑和中枢神经系统肿瘤,前10位恶性肿瘤占全部恶性肿瘤死亡的78.07%。截至2016年12月31日,上海市共有399 027例现患肿瘤病例,现患率为2.77%。市区现患率为3.07%,郊区为2.55%。乳腺癌是现患病例中最常见的恶性肿瘤,占15.33%�
文摘目的 :分析2003—2012年上海市胃癌病例发病特征,并与国际数据进行比较。方法 :选取上海市恶性肿瘤病例报告登记管理系统中记录的2003—2012年诊断的所有胃癌新发病例。国际恶性肿瘤发病数据来自WHO国际癌症研究机构(International Agency for Research on Cancer,IARC)的GLOBOCAN 2012数据库和美国国立癌症研究所(Nation Cancer Institute,NCI)监测、流行和终点项目(Surveillance,Epidemiology and End Result,SEER)18数据库。计算上海市胃癌年平均发病例数、平均年龄、构成比、发病率、年龄别发病率和标化发病率。采用Pearsonχ2检验比较不同时间阶段病理诊断依据、病理类型及诊断时期别构成比间的差异;采用Cochranχ2检验比较不同时间阶段以及地区间的标化发病率。结果 :2003—2012年,上海市胃癌新发病例57 812例,年平均5 781例,占全部恶性肿瘤的10.89%,发病率为41.88/10万,标化发病率为19.92/10万,位居上海市恶性肿瘤发病的第3位。2008—2012年,上海市胃癌新发病例29 373例,年平均5 875例,占全部恶性肿瘤的10.19%,发病中位年龄为69岁,平均年龄为67.70岁,标化发病率为18.64/10万;2003—2007年,上海市胃癌新发病例28 439例,年平均5 688例,占全部恶性肿瘤的11.74%,发病中位年龄为70岁,平均年龄为66.93岁,标化发病率为21.40/10万;2008—2012年标化发病率较2003—2007年显著降低(P<0.001)。2008—2012年上海市胃癌标化发病率高于世界平均水平(12.1/10万)和发达国家(10.6/10万),低于东亚地区(24.2/10万)和中国(22.7/10万)。2003—2007年和2008—2012年这2个阶段胃癌年龄别发病率随年龄的增长而上升,在80~84岁达到高峰。与2003—2007年相比,2008—2012年胃癌患者诊断时期别为Ⅰ期、有明确临床分期、有明确病理学分型和最高诊断依据为病理学诊断的患者所占百分比均明显增加(P值均<0.001)。结论 :与2003—2007年相比,2008—2012年上海市胃癌发
