目的了解近两年北京市朝阳区体检人群维生素D水平的现状,探讨影响维生素D缺乏的因素及与血脂的相关性。方法采用横断面研究纳入于北京市第一中西医结合医院体检的受试者3905例,抽取静脉血检测其25-羟基维生素D[25(OH)D]和血生化指标,采... 目的了解近两年北京市朝阳区体检人群维生素D水平的现状,探讨影响维生素D缺乏的因素及与血脂的相关性。方法采用横断面研究纳入于北京市第一中西医结合医院体检的受试者3905例,抽取静脉血检测其25-羟基维生素D[25(OH)D]和血生化指标,采用多元线性回归分析评估25(OH)D与血脂的相关性。结果所有受试者的血清25(OH)D平均水平为(16.22±6.95)ng/ml,其中25(OH)D严重缺乏636例(16.29%),缺乏2372例(60.74%),不足713例(18.26%),充足184例(4.71%)。不同25(OH)D水平受试者的性别、年龄、体重、BMI、腰围、腰臀比、收缩压、舒张压、空腹血糖(FPG)、尿酸(UA)、肌酐(Cr)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)比较差异均有统计学意义(P<0.05)。不同性别(P=0.010)、不同BMI和性别(P=0.030)、不同季节体检(P<0.001)的受试者25(OH)D水平比较差异均有统计学意义。不同25(OH)D水平受试者的年龄分布比较差异有统计学意义(P<0.001)。多元线性回归分析结果显示25(OH)D水平分别与TC(r=-0.35,P=0.016)、TG(r=-0.28,P=0.046)、LDL-C(r=-0.33,P=0.022)、腰围(r=-0.30,P=0.035)和腰臀比(r=-0.30,P=0.032)呈负相关,与HDL-C(r=0.27,P=0.049)呈正相关。结论北京市朝阳区维生素D水平缺乏状况较为普遍,需引起重视;维生素D水平缺乏与年龄、性别、季节变化、腰围、腰臀比、BMI及血脂相关。 展开更多

An agrobacterium tumefaciens-mediated co-tr- ansformation method was established and successfully used to transfer an antisense waxy gene into rice. The binary vec- tor p13W4 contains an antisense waxy gene, a hygromycin resistance gene (hpt) and a report gene (gus). The other bi- nary vector p13W8 contains only an antisense waxy gene in the T-DNA region. Two separate strains of A. tumefaciens, containing p13W4 and p13W8 mixed at a ratio of 1︰9, were used to transform a high-yield rice strain Chao 2-10. A total of 34 transgenic plants were obtained. PCR analysis showed that 15 of them were co-transformed. After GUS staining, the T1seeds with GUS positive were eliminated. The GUS nega- tive T1 seeds were germinated, and the seedlings were ana- lyzed for the existence of antisense waxy gene. The transgenic T1 plants with only antisense waxy gene further were con- firmed through Southern blot analysis. The T2 seeds collected from marker-free T1 plants were used for amylose content analysis. The results showed that the amylose content in marker-free transgenic seeds reduced by up to 28.61% over the wild type rice. This marker-free transgenic rice with low amylose contents obtained in this project could be used to screen soft grain, and could also be used as a new rice re- source for eating quality improvement. 展开更多

During spaceflight,the cardiovascular system undergoes remarkable adaptation to microgravity and faces the risk of cardiac remodeling.Therefore,the effects and mechanisms of microgravity on cardiac morphology,physiology,metabolism,and cellular biology need to be further investigated.Since China started constructing the China Space Station(CSS)in 2021,we have taken advantage of the Shenzhou-13 capsule to send human pluripotent stem cell-derived cardiomyocytes(hPSC-CMs)to the Tianhe core module of the CSS.In this study,hPSC-CMs subjected to space microgravity showed decreased beating rate and abnormal intracellular calcium cycling.Metabolomic and transcriptomic analyses revealed a battery of metabolic remodeling of hPSC-CMs in spaceflight,especially thiamine metabolism.The microgravity condition blocked the thiamine intake in hPSC-CMs.The decline of thiamine utilization under microgravity or by its antagonistic analog amprolium affected the process of the tricarboxylic acid cycle.It decreased ATP production,which led to cytoskeletal remodeling and calcium homeostasis imbalance in hPSC-CMs.More importantly,in vitro and in vivo studies suggest that thiamine supplementation could reverse the adaptive changes induced by simulated microgravity.This study represents the first astrobiological study on the China Space Station and lays a solid foundation for further aerospace biomedical research.These data indicate that intervention of thiamine-modified metabolic reprogramming in human cardiomyocytes during spaceflight might be a feasible countermeasure against microgravity. 展开更多

目的 研究淫羊藿治疗化疗致周围神经病变(chemotherapy-induced peripheral neuropathy,CIPN)的有效部位,并制备磷脂复合物以提高其渗透性,为制剂设计提供实验依据。方法 以复合率为评价指标,采用单因素试验结合Box-Behnken试验设计考... 目的 研究淫羊藿治疗化疗致周围神经病变(chemotherapy-induced peripheral neuropathy,CIPN)的有效部位,并制备磷脂复合物以提高其渗透性,为制剂设计提供实验依据。方法 以复合率为评价指标,采用单因素试验结合Box-Behnken试验设计考察有效部位磷脂复合物的制备工艺,对磷脂复合物的形成进行表征,以朝藿定A、B、C及淫羊藿苷为评价指标测定其平衡溶解度与油水分配系数(lg P),并考察其体外释放行为。结果 确定磷脂复合物的最佳工艺条件为无水乙醇为反应溶剂,有效部位与大豆卵磷脂投料比为1∶2.7,药物质量浓度为13.5 mg/mL,反应温度为37℃,反应时间为2 h,复合率达到(96.21±0.22)%。紫外光谱、红外光谱、差示扫描量热分析表征验证了淫羊藿有效部位磷脂复合物的形成。有效部位通过磷脂复合物改性后,4种成分的溶解度提高2~3倍,lg P>1,药物的溶解性和渗透性得到显著提高。体外释放结果显示,4种成分的24 h体外累积释放率处于48%~74%,释放过程以一级动力学为主,释放机制为溶蚀和扩散的共同作用。结论 采用磷脂复合物技术对有效部位进行了改性,为淫羊藿治疗CIPN的进一步开发与利用奠定了基础。 展开更多

目的探讨脊髓神经运动功能标准化评估流程在胸腰椎疾病患者护理中的应用效果。方法选择河南省人民医院骨科病区2016年4-6月收治的60例胸腰椎疾病患者作为试验组,采用脊髓神经运动功能标准化评估流程。选择2016年1-3月收治的60例采用常... 目的探讨脊髓神经运动功能标准化评估流程在胸腰椎疾病患者护理中的应用效果。方法选择河南省人民医院骨科病区2016年4-6月收治的60例胸腰椎疾病患者作为试验组,采用脊髓神经运动功能标准化评估流程。选择2016年1-3月收治的60例采用常规护理评估的胸腰椎疾病患者作为对照组。比较脊髓神经运动功能标准化评估流程实施前后护士专科理论及操作成绩、2组患者平均住院日、次均费用、医生对护士专科能力满意度。结果脊髓神经运动功能标准化评估流程的制定和实施,经过培训后护士的理论考核得分为(94.70±1.18)分,操作考核得分为(93.50±2.34)分;平均住院日为(6.10±1.98)d,次均住院费用为(29088±341)元;与对照组的(42.30±12.95)分、(46.80±15.31)分、(7.80±2.37)d、(37566±592)元相比,差异有统计学意义(t=4.76~8.31,均P=0.00)。医生对护士总体满意度从11/15提升至14/15。结论胸腰椎疾病患者脊髓神经运动功能标准化评估流程规范了护士的工作流程和操作标准,使护士的工作更加全面、及时、专业,对临床有较好的指导作用。 展开更多

Polymer nanowire(NW)organic field-effect transistors(OFETs)integrated on highly aligned large-area flexible substrates are candidate structures for the development of high-performance flexible electronics.This work presents a universal technique,coaxial focused electrohydrodynamic jet(CFEJ)printing technology,to fabricate highly aligned 90-nm-diameter polymer arrays.This method allows for the preparation of uniformly shaped and precisely positioned nanowires directly on flexible substrates without transfer,thus ensuring their electrical properties.Using indacenodithiophene-co-benzothiadiazole(IDT-BT)and poly(9,9-dioctylfluorene-co-benzothiadiazole)(F8-BT)as example materials,5 cm^(2) arrays were prepared with only minute size variations,which is extremely difficult to do using previously reported methods.According to 2D-GIXRD analysis,the molecules inside the nanowires mainly adopted face-onπ-stacking crystallite arrangements.This is quite different from the mixed arrangement of thin films.Nanowire-based OFETs exhibited a high average hole mobility of 1.1 cm^(2) V^(−1) s^(−1) and good device uniformity,indicating the applicability of CFEJ printing as a potential batch manufacturing and integration process for high-performance,scalable polymer nanowire-based OFET circuits.This technique can be used to fabricate various polymer arrays,enabling the use of organic polymer semiconductors in large-area,high-performance electronic devices and providing a new path for the fabrication of flexible displays and wearable electronics in the future. 展开更多

The functionality of aut... Traffic congestion is a growing problem in urban areas all over the world. The transport sector has been in full swing event study on intelligent transportation system for automatic detection. The functionality of automatic incident detection on expressways is a primary objective of advanced traffic management system. In order to save lives and prevent secondary incidents, accurate and prompt incident detection is necessary. This paper presents a methodology that integrates moving average (MA) model with stationary wavelet decomposition for automatic incident detection, in which parameters of layer coefficient are extracted from the difference between the upstream and downstream occupancy. Unlike other wavelet-based method presented before, firstly it smooths the raw data with MA model. Then it uses stationary wavelet to decompose, which can achieve accurate reconstruction of the signal, and does not shift the signal transfer coefficients. Thus, it can detect the incidents more accurately. The threshold to trigger incident alarm is also adjusted according to normal traffic condition with con- gestion. The methodology is validated with real data from Tokyo Expressway ultrasonic sensors. Ex- perimental results show that it is accurate and effective, and that it can differentiate traffic accident from other condition such as recurring traffic congestion. 展开更多

目的:探讨下调生长阻滞和DNA损伤诱导蛋白45β(growth arrest and DNA damage inducible protein 45β,GADD45β)表达对PC9肺腺癌细胞及吉非替尼敏感性的影响。方法:设计并合成GADD45β基因小干扰RNA(GADD45β-small interfering RNA,GA... 目的:探讨下调生长阻滞和DNA损伤诱导蛋白45β(growth arrest and DNA damage inducible protein 45β,GADD45β)表达对PC9肺腺癌细胞及吉非替尼敏感性的影响。方法:设计并合成GADD45β基因小干扰RNA(GADD45β-small interfering RNA,GADD45β-siRNA)序列,通过慢病毒介导将GADD45β-siRNA转入PC9肺腺癌细胞中,通过实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)和Western印迹检测转染前后PC9肺腺癌细胞GADD45β的mRNA及蛋白水平,采用膜联蛋白V(annexin V)-别藻蓝蛋白(allophycocyanin,APC)双染流式细胞法检测转染后细胞凋亡水平;通过流式细胞术检测转染后细胞内DNA含量,计算转染后细胞各周期时相百分率,分析转染对细胞生长周期的影响;通过计数克隆形成数检测RNA干扰对细胞成瘤能力的影响;采用MTT法检测PC9肺腺癌细胞的吉非替尼半数抑制浓度IC50。结果:筛选出5'-AAATCCACTTCACGCTCAT-3'为GADD45β基因RNA干扰的有效序列。转染GADD45β-siRNA 48 h后,qRT-PCR和Western印迹结果显示PC9肺腺癌细胞GADD45β的mRNA和蛋白表达水平明显下调(均P<0.05),细胞凋亡率明显增加(P<0.05),且成瘤克隆数明显减少(P<0.05);PC9肺腺癌细胞位于S期及G2/M期细胞增多(P<0.05),吉非替尼的IC50明显下降(P<0.05)。结论:PC9肺腺癌细胞转染GADD45β-siRNA后,能成功下调GADD45β基因的mRNA和蛋白表达;下调GADD45β表达可降低PC9肺腺癌细胞的克隆形成能力,促进细胞凋亡;下调GADD45β表达可明显提高PC9肺腺癌细胞对吉非替尼的敏感性。 展开更多

Acetylcholinesterase(AChE) is a key enzyme used to detect organophosphorus pesticide residues by the enzyme inhibition method.An accidental discovery of a mutant strain with AChE activity was made in our laboratory during the process of AChE expression by Pichia pastoris.The pPIC9 K-Drosophila melanogaster acetylcholinesterase(DmAChE)-like expression vector was constructed by codon optimization of this mutant strain,which was transformed into P.pastoris GS115,and positive clones were selected on yeast peptone dextrose(YPD) plate with G418 at 4.0 mg/mL.The GS115-pPIC9 K-DmAChE-like strain was subjected to 0.5% methanol induction expression for 120 h,with a protein band at 4.3 kDa found by the tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) pattern of the fermentation supernatant.After preliminary purification by ammonium sulfate precipitation,the enzyme activity was detected to be 76.9 U/(mL·min).In addition,the pesticide sensitivity test proved that DmAChE-like is selective and sensitive to organophosphorus pesticides. 展开更多

The results show that sorption degree of vanadium(V) increases with a decrease ofpH values, and V(V) ions are easier sorbed than Si(IV) and AI(III) ions under the same conditions. The sorption degree ofV(V), Si(IV), and AI(III) at pH 9.14 for 15 man are 90.6%, 33.5%, and 21.6%, respectively. Si(IV), AI(III), and V(V) ions sorbed on 717 resin were eluted by use of 2 mol·L^-1 NaOH solution; the desorption degree ofV(V), Si(IV), and AI(III) for 5 min are 81.7 %, 99.1%, and 99.3%, respectively. 展开更多

In multi-cell cooperative multi-input multi-output (MIMO) systems, base station (BS) can exchange and utilize channel state information (CSI) of adjacent cell users to manage co-channel interference. Users quantize the CSIs of desired channel and interference channels using finite-rate feedback links, then BS can generate cooperative block diagonalization (BD) precoding matrices using the obtained quantized CSI at transmitter to supress co-channel interference. In this paper, a novel adaptive bit allocation scheme is proposed to minimize the rate loss due to imperfect CSI. We derive the closed-form expression of rate loss caused by both channel delay and limited feedback. Based on the derived rate loss expression, the proposed scheme can adaptively allocate more bits to quantize the better channels with smaller delays and fewer bits to worse channels with larger delays. Simulation results show that the proposed scheme yields higher performance than other allocation schemes. 展开更多

目的探讨尼莫地平联合依达拉奉治疗蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)的疗效。方法将80例SAH后CVS患者随机分为观察组与对照组,每组40例。2组均给予常规治疗,对照组应用依达拉奉静脉滴注,观察组联合尼莫地平静脉滴注。比较2组总有... 目的探讨尼莫地平联合依达拉奉治疗蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)的疗效。方法将80例SAH后CVS患者随机分为观察组与对照组,每组40例。2组均给予常规治疗,对照组应用依达拉奉静脉滴注,观察组联合尼莫地平静脉滴注。比较2组总有效率及治疗前后炎性因子水平、血清S100β蛋白、内皮素-1(ET-1)水平、大脑中动脉(MCA)血流速度、格拉斯哥昏迷评分(GCS)、美国国立卫生研究院卒中量表(NIHSS)评分、Barthel指数(BI)及不良反应。结果观察组总有效率97.50%,显著高于对照组的85.00%(P<0.05);治疗后,观察组炎性因子水平及血清S100β蛋白、ET-1水平、MCA血流速度均显著低于对照组(P<0.05);治疗后,观察组GCS评分、BI显著高于对照组,NIHSS评分显著低于对照组(P<0.05);2组不良反应发生率比较无显著差异(P>0.05)。结论联合应用尼莫地平与依达拉奉治疗SAH后CVS疗效显著,可有效抑制炎性反应,减轻CVS严重程度。 展开更多
