Objective:To observe the law of changes in mechanical paw-withdrawal threshold(MPWT) and thermal paw-withdrawal latency(TPWL) on the modeled side of rats with pain transition(hyperalgesic priming[HP]),and the expressi...Objective:To observe the law of changes in mechanical paw-withdrawal threshold(MPWT) and thermal paw-withdrawal latency(TPWL) on the modeled side of rats with pain transition(hyperalgesic priming[HP]),and the expression of prostaglandin E receptor 1(EP1) and transient receptor potential vanilloid type 1(TRPV1) in the dorsal root ganglion(DRG) of the affected side of rats.To observe the effects of electroacupuncture(EA) on the TPWL on the modeled side of rats with pain transition and regulation of EP1 and TRPV1 expression in DRG.Methods:Part 1:Eighteen SD rats were randomly divided into control,sham HP,and HP groups,with 6rats in each group.The modeling comprised two injections.The rats in the HP group were subcutaneously injected with 1% carrageenan(100 μL) into the left hind paw in the first injection(those in the control and sham HP groups were injected with saline).The second injection was administered 8 days later by injecting prostaglandin E2(PGE2)(100 ng/25 μL) into the dorsum of the paw(the rats in the control group were administered with saline and those in the sham HP group PGE2),thereby developing a pain transition model.TPWL and MPWT were measured before rat modeling,4 h,and 1,2,3,and 7 days after the first injection,and 1,4,24,and 48 h after the second injection(8 days after the first injection).The expression rates of EP1-and TRPV1-positive cells in the affected DRG were measured by immunofluorescence(IF).Part 2:Eighteen SD rats were randomly divided into sham HP(6 rats),and HP groups(12rats),then the rats in HP group were randomly divided into HP(6 rats) and HP+EP1 antagonist(6 rats)groups for the detection of TPWL.Rats in the EP1 antagonist group were injected with EP1 antagonist5 min before PGE2 injection.The expression rate of TRPV1-positive cells in the affected DRG 48 h after PGE2 injection was detected using the IF method.Part 3:Twenty-four SD rats were randomly divided into sham HP(6 rats),and HP groups(18 rats),then the rats in HP group were randomly divided into HP(6 rats),sham EA(6 rats)展开更多
The Transantarctic Mountains are an important corridor between the East Antarctic Ice Sheet and the Western Ross Sea,where most current ice streams and outlet glaciers arise.We investigated Ricker Hills,the largest ex...The Transantarctic Mountains are an important corridor between the East Antarctic Ice Sheet and the Western Ross Sea,where most current ice streams and outlet glaciers arise.We investigated Ricker Hills,the largest exposed mountainous region between Southern Victoria Land and Terra Nova Bay,and dated the glacial landforms using in-situ cosmogenic-nuclide 10 Be surface exposure dating to reconstruct the paleo-glacial dynamics.The surface of the Hollingsworth glacier lowered since the middle of Marine Isotope Stage(MIS)2(22.1 ka);therefore,the Last Glacial Maximum(LGM)occurred before that period.Cosmogenic,geomorphic,and climatic records constrained the glacial surface slope to be between 5.4°and 6.8°.The ice was 270-320 m thicker at the LGM(MIS 2)than presently but did not override the top surface of the Benson Knob.Moreover,previous glacial periods such as the local LGM(MIS 4)or Penultimate Glacial Maximum(MIS 6)maintained thicker ice than the LGM(MIS 2).The Ross Ice Shelf opening during the mid-Holocene(~6 ka)caused the rapid collapse of the terminal outlet glaciers and supplied notable snow accumulation upstream,which stagnated lowering.The greatest lowering and retreat occurred during the late Holocene(2.3~0.8 ka),when elephant seal colonies thrived in the Ross Embayment.展开更多
目的探讨纤连蛋白(FN)基因和第10号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)在原发性肝癌患者肝组织中的表达及联合检测的价值。方法选取2013年1月-2015年10月衡水市人民医院肝胆外科手术切除的256例肝癌组织及癌旁组织,50例正常肝...目的探讨纤连蛋白(FN)基因和第10号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)在原发性肝癌患者肝组织中的表达及联合检测的价值。方法选取2013年1月-2015年10月衡水市人民医院肝胆外科手术切除的256例肝癌组织及癌旁组织,50例正常肝组织标本为同期非癌症患者。分别采用RT-PCR检测FN和PTEN mRNA的表达,采用Western blotting检测FN和PTEN蛋白的表达,比较肝癌组织和正常肝组织中FN和PTEN的表达差异。以FN mRNA表达、FN蛋白表达中任一为阳性,且同时PTEN mRNA表达、PTEN蛋白表达中任一为阴性,判定为联合检测肝癌阳性。采用ROC曲线评价单项检测和联合检测的诊断肝癌价值,分析联合检测与术后24个月预后的关系。结果肝癌组织FN mRNA和FN蛋白表达高于正常组织(P<0.05),肝癌组织的PTEN m RNA和PTEN蛋白表达低于癌旁组织和正常组织(P<0.05)。FN m RNA诊断肝癌的敏感性为74.22%、特异性为86.00%,FN蛋白检测的敏感性为76.92%、特异性为88.00%,PTEN m RNA检测的敏感性为72.65%、特异性为90.00%,PTEN蛋白检测的敏感性为75.00%、特异性为100.00%。联合检测的敏感性为86.72%,高于单项检测(P<0.05),特异性为82.00%,ROC曲线下面积为0.912,具有极高的诊断价值。联合检测诊断为阴性的肝癌患者术后24个月的复发率、转移率低于阳性患者(P<0.05),生存率高于阳性患者(P<0.05)。结论 FN与PTEN联合检测可用于原发性肝癌的诊断,具有极高的诊断价值,FN的高表达和PTEN的低表达可以作为评估肝癌患者预后的潜在指标。展开更多
