Problems continue to be encountered concerning the traditional vacuum preloading method in field during the treatment of newly deposited dredger fills.In this paper,an improved multiple-vacuum preloading method was de...Problems continue to be encountered concerning the traditional vacuum preloading method in field during the treatment of newly deposited dredger fills.In this paper,an improved multiple-vacuum preloading method was developed to consolidate newly dredger fills that are hydraulically placed in seawater for land reclamation in Lingang Industrial Zone of Tianjin City,China.With this multiplevacuum preloading method,the newly deposited dredger fills could be treated effectively by adopting a novel moisture separator and a rapid improvement technique without sand cushion.A series of model tests was conducted in the laboratory for comparing the results from the multiple-vacuum preloading method and the traditional one.Ten piezometers and settlement plates were installed to measure the variations in excess pore water pressures and moisture content,and vane shear strength was measured at different positions.The testing results indicate that water dischargeetime curves obtained by the traditional vacuum preloading method can be divided into three phases:rapid growth phase,slow growth phase,and steady phase.According to the process of fluid flow concentrated along tiny ripples and building of larger channels inside soils during the whole vacuum loading process,the fluctuations of pore water pressure during each loading step are divided into three phases:steady phase,rapid dissipation phase,and slow dissipation phase.An optimal loading pattern which could have a best treatment effect was proposed for calculating the water discharge and pore water pressure of soil using the improved multiple-vacuum preloading method.For the newly deposited dredger fills at Lingang Industrial Zone of Tianjin City,the best loading step was 20 kPa and the loading of 40-50 k Pa produced the highest drainage consolidation.The measured moisture content and vane shear strength were discussed in terms of the effect of reinforcement,both of which indicate that the multiple-vacuum preloading method has a better treatment effect not only in decreasing t展开更多
Protein-mediated chromatin interactions can be revealed by coupling proximity-based ligation with chromatin immunoprecipitation.However,these techniques require complex experimental procedures and millions of cells pe...Protein-mediated chromatin interactions can be revealed by coupling proximity-based ligation with chromatin immunoprecipitation.However,these techniques require complex experimental procedures and millions of cells per experiment,which limits their widespread application in life science research.Here,we develop a novel method,Hi-Tag,that identifies high-resolution,long-range chromatin interactions through transposase tagmentation and chromatin proximity ligation(with a phosphorothioate-modified linker).Hi-Tag can be implemented using as few as 100,000 cells,involving simple experimental procedures that can be completed within 1.5 days.Meanwhile,Hi-Tag is capable of using its own data to identify the binding sites of specific proteins,based on which,it can acquire accurate interaction information.Our results suggest that Hi-Tag has great potential for advancing chromatin interaction studies,particularly in the context of limited cell availability.展开更多
Objective:The objective of the study was to compare the application effects of in-vial exhaust method and conventional exhaust method in the process of coronavirus disease 2019 vaccine injection.Materials and Methods:...Objective:The objective of the study was to compare the application effects of in-vial exhaust method and conventional exhaust method in the process of coronavirus disease 2019 vaccine injection.Materials and Methods:Using convenient sampling method,102 vaccines were selected as experiment group during the process of vaccine injection,and the in-vial exhaust method was used.One hundred and five vaccines were selected as the control group and the conventional exhaust method was adopted.The incidence of vaccine solution spillage and exhausting time in the two groups during exhaust were observed.Results:The incidence of solution spillage in the experiment group was lower than that in the control group(0 vs.6.67%,P<0.05).The exhausting time of the experiment group was shorter than that of the control group([15.12±4.43]s vs.[22.74±6.53]s,P<0.05).Conclusion:Implementing the in-vial exhaust method in the vaccine injection can effectively reduce the incidence of solution spillage,reduce nucleic acid contamination,and ensure that the vaccine is injected at the prescribed dose.Moreover,the operation is simple and easy,which improves the nurse’s vaccination efficiency,and has a higher promotion and application value.展开更多

Luotian(herein, Luotian), a natural variety of C. lanceolata(Chinese-fir, herein Lanceolata) in China, were analyzed using trunk and br...The structural characteristics of the tree crowns of Cunninghamia lanceolata var. Luotian(herein, Luotian), a natural variety of C. lanceolata(Chinese-fir, herein Lanceolata) in China, were analyzed using trunk and branch measurements and biomass determinations. Samples from two typical cultivated varietal populations were collected, including twenty-six 15–23-year-old trees of Luotian from a plantation, and nine 16–23-year-old trees of Lanceolata. Our results show that Luotian and Lanceolata samples differed significantly in crown structure, morphological indices, and biomass:(1) the oldest live branches on Luotian trees were 5–6 years old and 8–11 years old on Lanceolata. The ages of the live branches were not affected by the ages of the Luotian trees, while live branch ages increased with ages of Lanceolata trees;(2) the maximumbranching order of Luotian was level two. Compared to Lanceolata, the average number of first-order lateral branches(i.e., branches emerging from the trunk) and the number of first-order lateral branch whorls per sample tree were 12.9% and 32.2% lower, respectively, in Luotian.However, the average number of branches within a single whorl was 21.8% greater in Luotian;the average number of branch whorls at crown height was 51.1% greater. Thus,the Luotian variety has thicker branches;(3) the average lateral branch angles in Luotian and Lanceolata sample trees were 105.2° and 61.4°, respectively. The branch angles in 53.0% of lateral branches on Luotian ranged from105° to 135°, but 30° to 90° in 96% of the lateral branches on Lanceolata. Within the same crown layer, the average branch angle was 1.6–2.2 times greater in Luotian, and the angle was directly proportional to crown thickness;(4) the average base diameter and branch length on Luotian were1.3 cm and 75.8 cm, respectively, and 1.6 cm and112.2 cm for Lanceolata.
