The dissected retroperitoneal area t...AIM: The study of immune response of open versus laparoscopical total mesorectal excision with anal sphincter preservation in patients with rectal cancer has not been reported yet. The dissected retroperitoneal area that contacts directly with carbon dioxide is extensive in laparoscopic total mesorectal excision with anal sphincter preservation surgery. Tt is important to clarify whether the immune response of laparoscopic total mesorectal excision with anal sphincter preservation (LTME with ASP) in patients with rectal cancer is suppressed more severely than that of open surgery (OTME with ASP). This study was designed to compare the immune functions after laparoscopic and open total mesorectal excision with anal sphincter preservation for rectal cancer.METHODS: This study involved 45 patients undergoing laparoscopic (n=20) and open (n=25) total mesorectal excisions with anal sphincter preservation for rectal cancer.Serum interleukin-2 (IL-2), interleukin-6 (IL-6), tumor necrosis factor α (TNFα) were assayed preoperatively and on days 1 and 5 postoperatively. CD3+ and CD56+ T lymphocyte count, CD3- and CD56+ natural killer cell (NK)count and immunoglobulin (IgG/IgM/IgA) were assayed preoperatively and on day 5 postoperatively. The numbers of CD3+ and CD56+ T lymphocytes and CD3- and CD56+ NK cells were counted using flow cytometry. An enzyme-linked immunosorbent assay (ELISA) was used for IL-2, TL-6 and TNFα determination. And IgG, IgM, and IgA were assayed using immunonephelometry.RESULTS: The demographic data of the two groups had no difference. The preoperative levels of CD3+ and CD56+ T lymphocyte count, CD3- and CD56+ NK count, serum IgG,IgM, IgA, IL-2, IL-6 and TNFα also had no significant difference in the two groups (P>0.05). The CD3+ and CD56+ T lymphocyte counts had no obvious changes after surgery in laparoscopic (d=-0.79±3.83 %) and open (d=0.42±2.09 %)groups. Background:Soil aggregates are the basic units of soil structure,and their stability is a key indicator of soil quality and capacity to support ecosystem functions.The impacts of various environmental factors on soil ...Background:Soil aggregates are the basic units of soil structure,and their stability is a key indicator of soil quality and capacity to support ecosystem functions.The impacts of various environmental factors on soil aggregates have been widely studied.However,there remains elusive knowledge on the synergistic effects of changing forest stand structure on soil aggregate stability(SAS),particularly in subtropical China where soil erosion remains a critical issue.Methods:We investigated variations in the components of soil humus(HS),including humic acids(HAs),fulvic acids(FAs),and humins(HMs),under pure Chinese fir(Cunninghamia lanceolata)plantation(PP)and multilayered mixed plantation(MP)comprising C.lanceolata,Castanopsis hystrix,and Michelia hedyosperma.The state of soil aggregate stability,was determined by three separate methods,i.e.,dry-sieving,wet-sieving,and the Le Bissonnais.High-throughput sequencing was used to determine the diversity and composition of microbial communities under PP and MP.We then built partial least squares path models(PLS-PM)for assessing the responses of SAS to the variations in soil microorganisms and HS components.Results:The MP stands had significantly greater SAS(P<0.05),higher content of HAs and more rapid organic matter humification within aggregates,than the PP stands.High-throughput sequencing confirmed that the Pielou andα-diversity index values(Chao1 and Shannon)for fungi were all significantly higher under MP than under PP,while no marked difference was found in bacterialα-diversity between the two plantation types.Moreover,there were markedly greater abundance of three bacterial phyla(Verrucomicrobia,Chloroflexi,and Gemmatimonadetes)and three fungal phyla(Ascomycota,Kickxellomycota,and Glomeromycota),and significantly less abundance of two bacterial phyla(Planctomycetes and Firmicutes)and four fungal phyla(Basidiomycota,Mortierellomycota,Mucoromycota,and Rozellomycota)under MP than under PP.The Chloroflexi and Ascomycota phyla appeared to be the primary drivers of soil 展开更多
AIM:To examine the role of nucleostemin in the growth regulation of gastric cancer,liver cancer and other cancers.METHODS:RT-PCR was used to clone the fragment of nucleostemin cDNA from HEK 293 cells.Eighteen kinds of...AIM:To examine the role of nucleostemin in the growth regulation of gastric cancer,liver cancer and other cancers.METHODS:RT-PCR was used to clone the fragment of nucleostemin cDNA from HEK 293 cells.Eighteen kinds of malignant tumor tissues including gastric adenocarcinoma and liver cancer tissues,3 kinds of benign tumor tissues,3 kinds of benign hyperplastic tissues and normal tissues were employed to examine nucleostemin gene expression by RT-PCR,Slot blot,Northern blot and in situ hybridization.RESULTS:We successfully cloned a 570 bp fragment of nucleostemin-cDNA from HEK-293 cells.All edtected malignant tumor tissues,benign tumor tissues,and benign hyperplastic tissues had hign levels of nucleostemin expression.Nucleostemin was also expressed in human placenta tissue at a high levle.In terminally differentiated normal human adult kidney and mammary gland tissues,no nucleostemin expression could be detected.CONCLUSION:Nucleostemin can help regulate the proliferation of both cancer cells and stem cells.It might play an important role in the growth regulation of gastric cancer,liver cancer and other cancers.展开更多
