Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds(BRSs).In particular,it is urgent to establish a nondestructive approach to e...Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds(BRSs).In particular,it is urgent to establish a nondestructive approach to examine in vivo degradation of a new-generation coronary stent for interventional treatment based on mammal experiments;otherwise it is not available to semi-quantitatively monitor biodegradation in any clinical trial.Herein,we put forward a semi-quantitative approach to measure degradation of a sirolimus-eluting iron bioresorbable scaffold(IBS)based on optical coherence tomography(OCT)images;this approach was confirmed to be consistent with the present weight-loss measurements,which is,however,a destructive approach.The IBS was fabricated by a metal-polymer composite technique with a polylactide coating on an iron stent.The efficacy as a coronary stent of this new bioresorbable scaffold was compared with that of a permanent metal stent with the name of trade mark Xience,which has been widely used in clinic.The endothelial coverage on IBS was found to be greater than on Xience after implantation in a rabbit model;and our well-designed ultrathin stent exhibited less individual variation.We further examined degradation of the IBSs in both minipig coronary artery and rabbit abdominal aorta models.The present result indicated much faster iron degradation of IBS in the rabbit model than in the porcine model.The semi-quantitative approach to detect biodegradation of IBS and the finding of the species difference might be stimulating for fundamental investigation of biodegradable implants and clinical translation of the next-generation coronary stents.展开更多
T cells secrete bioactive exosomes(EXO),but the potential immunoregulatory effect of T-cell EXO is largely unknown.In this study,we generated activated ovalbumin(OVA)-specific CD4^(+) T cells in vitro via coculture of...T cells secrete bioactive exosomes(EXO),but the potential immunoregulatory effect of T-cell EXO is largely unknown.In this study,we generated activated ovalbumin(OVA)-specific CD4^(+) T cells in vitro via coculture of OVA-pulsed dendritic cells(DC_(OVA))with naive CD4^(+) T cells derived from OVA-specific T-cell receptor(TCR)transgenic OTII mice.CD4^(+) T-cell EXO were then purified from the CD4^(+) T-cell culture supernatants by differential ultracentrifugation.CD4^(+) T-cell EXO exhibited the‘saucer’shape that is characteristic of EXO with a diameter between 50 and 100 nm,as assessed by electron microscopy,and contained the EXO-associated proteins LAMP-1,TCR and lymphocyte function associated antigen-1(LFA-1),as determined by western blot.Flow cytometric analysis showed that CD4^(+) T-cell EXO expressed CD4^(+) T-cell markers(CD4,TCR,LFA-1,CD25 and Fas ligand),but to a lesser extent than CD4^(+) T cells.We demonstrated that DC_(OVA) took up CD4^(+) T-cell EXO via peptide/major histocompatibility complex(pMHC)II/TCR and CD54/LFA-1 interactions.OVA-specific CD4^(+) T-cell EXO from OTII mice,but not ConA-stimulated polyclonal CD4^(+) T-cell EXO from wild-type C57BL/6 mice inhibited DC_(OVA)-stimulated in vitro CD4^(+) T-cell proliferation and in vivo CD81 cytotoxic T lymphocyte(CTL)responses and antitumor immunity against OVA-expressing B16 melanoma BL6-10OVA cells.In addition,EXO derived from a T-cell hybridoma cell line,MF72.2D9,expressing an OVA-specific CD4^(+) TCR,had a similar inhibitory effect as OTII CD4^(+) T-cell EXO on CTL-mediated antitumor immunity.Taken together,our data indicate that antigen-specific T-cell EXO may serve as a new type of immunosuppressive reagent for use in transplant rejection and treatment of autoimmune diseases.展开更多
The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by China,fostered through extensive collaboration with international partners.The complex comprises four accelerators:a 3...The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by China,fostered through extensive collaboration with international partners.The complex comprises four accelerators:a 30 GeV Linac,a 1.1 GeV Damping Ring,a Booster capable of achieving energies up to 180 GeV,and a Collider operating at varying energy modes(Z,W,H,and tt).The Linac and Damping Ring are situated on the surface,while the subterranean Booster and Collider are housed in a 100 km circumference underground tunnel,strategically accommodating future expansion with provisions for a potential Super Proton Proton Collider(SPPC).The CEPC primarily serves as a Higgs factory.In its baseline design with synchrotron radiation(SR)power of 30 MW per beam,it can achieve a luminosity of 5×10^(34)cm^(-2)s^(-1)per interaction point(IP),resulting in an integrated luminosity of 13 ab^(-1)for two IPs over a decade,producing 2.6 million Higgs bosons.Increasing the SR power to 50 MW per beam expands the CEPC's capability to generate 4.3 million Higgs bosons,facilitating precise measurements of Higgs coupling at sub-percent levels,exceeding the precision expected from the HL-LHC by an order of magnitude.This Technical Design Report(TDR)follows the Preliminary Conceptual Design Report(Pre-CDR,2015)and the Conceptual Design Report(CDR,2018),comprehensively detailing the machine's layout,performance metrics,physical design and analysis,technical systems design,R&D and prototyping efforts,and associated civil engineering aspects.Additionally,it includes a cost estimate and a preliminary construction timeline,establishing a framework for forthcoming engineering design phase and site selection procedures.Construction is anticipated to begin around 2027-2028,pending government approval,with an estimated duration of 8 years.The commencement of experiments and data collection could potentially be initiated in the mid-2030s.展开更多

To understand post-orogenic uplift history and mechanism of the Dabie orogenic belt, many samples of granite, granodiorite, diorite, and alkali feldspar granite with precise zircon U-Pb ages were collected for electron microprobe analysis of hornblende. Emplacement pressure values were then cal- culated by using an Al-in-hornblende geobarometer. The result shows that total A1 (A1T) values in hornblende range from 1.17 to 1.97 p.f.u., XFe(=Fe/(Fe+Mg)) from 0.4 to 0.65, and corresponding pressure values from 256.8 to 676.2 MPa for A1T greater than 1.17. It is suggested that although the main mechanism for the uplift is tectonic extension, inhomogeneous uplift related to the pluton emplacement is also important during post-orogenic evolution of the Dabie orogenic belt. Emplacement depth dis- tribution of the plutons shows that uplift in the NDC is affected by horst-graben structures since 120 Ma.展开更多
The Jiangmen Underground Neutrino Observatory(JUNO)is a large liquid scintillator detector designed to explore many topics in fundamental physics.In this study,the potential of searching for proton decay in the p→νK...The Jiangmen Underground Neutrino Observatory(JUNO)is a large liquid scintillator detector designed to explore many topics in fundamental physics.In this study,the potential of searching for proton decay in the p→νK^(+)mode with JUNO is investigated.The kaon and its decay particles feature a clear three-fold coincidence signature that results in a high efficiency for identification.Moreover,the excellent energy resolution of JUNO permits suppression of the sizable background caused by other delayed signals.Based on these advantages,the detection efficiency for the proton decay via p→νK^(+)is 36.9%±4.9%with a background level of 0.2±0.05(syst)±0.2(stat)events after 10 years of data collection.The estimated sensitivity based on 200 kton-years of exposure is 9.6×1033 years,which is competitive with the current best limits on the proton lifetime in this channel and complements the use of different detection technologies.展开更多
