METHODS: We used flow cytometry to analyze and sort the SP and non-SP cells in established HCC lines MHCC97 and hHCC. We evaluated cell proliferation by methyl thiazolyl tetrazolium (MTT) assay and investigated the expression of p53, bcl-2 and bax genes during denutrition, by RT-PCR and immunofluorescence staining. RESULTS: The percentage of SP cells in the two established HCC lines was 0.25% and 0.5%, respectively. SP cells had greater anti-apoptosis and proliferation ability than non-SP cells. Expression of Bcl-2 and Bax in SP and non-SP cells differed during denutrition. The former was up-regulated in SP cells, and the latter was up-regulated in non-SP cells. CONCLUSION: It may be that different upstream molecules acted and led to different expression levels of Bcl-2 and Bax in these two cell lines. There was a direct relationship between up-regulation of Bcl-2 and down-regulation of Bax and higher anti-apoptosis ability in SP cells. It may be that the existence and activity of SP cells are partly responsible for some of the clinical phenomena which are seen in HCC, such as relapse or metastasis. Further research on SP cells may have potential applications in the field of anticancer therapy.展开更多

A goal of an ecological water conveyance project is to protect and restore the natural vegetation in the lower reaches of Tadm River by transferring water from Bosten Lake, through the river channel, to the lower reaches. This study describes the changes in groundwater depth during the water transfer and the respondence of riparian vegetation to alterations in groundwater levels. The results indicate that groundwater depth along the Tarim River channel has a significant spatial-temporal component. Groundwater levels closest to the river channel show the most immediate and pronounced changes as a response to water transfer while those further away respond more slowly, although the observed change appears to be longer in duration. With a rise in the groundwater level, natural vegetation responded with higher growth rates, biomass and biodiversity. These favorable changes show that it is feasible to protect and restore the degraded natural vegetation by raising the groundwater depth. Plant communities are likely to reflect the hysteresis phenomenon, requiting higher water levels to initiate and stimulate desired growth than what may be needed to maintain the plant community. Because different species have different ecologies, including different root depths and densities and water needs, their response to increasing water availability will be spatially and temporally heterogenous. The response of vegetation is also influenced by microtopography and watering style. This paper discusses strategies for the protection and restoration of the degraded vegetation in the lower reaches of the Tarim River and provides information to complement ongoing theoretical research into ecological restoration in add or semi-arid ecosystems.展开更多

The optimal L^2 time convergence rate for the global classical solution is obtained for a small initial perturbation of the constant equilibrium state. It is shown that due to the electric field, the difference of the charge densities tend to the equilibrium states at the optimal rate (1 + t)^-3/4 in L^2-norm, while the individual momentum of the charged particles converges at the optimal rate (1 + t)^-1/4 which is slower than the rate (1 + t)^-3/4 for the compressible Navier-Stokes equations (NS). In addition, a new phenomenon on the charge transport is observed regarding the interplay between the two carriers that almost counteracts the influence of the electric field so that the total density and momentum of the two carriers converges at a faster rate (1 + t)^-3/4+ε for any small constant ε 〉 0. The above estimates reveal the essential difference between the unipolar and the bipolar Navier-Stokes-Poisson systems.展开更多

The variations in the surface temperature of the work roll during hot strip rolling were described, and the thermal stress field of the work roll was also analyzed. The results showed that the highest roll surface temperature is 593 ℃, and the difference between the minimum and maximum values of thermal stress of the work roll surface is 145.7 MPa. Furthermore, the results of this analysis indicate that temperature and thermal stress are useful parameters for the investigation of roll thermal fatigue and also for improving the quality of strip during rolling.展开更多

Ninety-two male bladder cancer patients who underwent radical cystoprostatectomy at our center between January 2003 and January 2008 were included in this study. The mean age of patients was 67.1 years (range: 32-75 years). Prostate-specific antigen (PSA) levels and digital rectal examination (DRE) results before surgery were obtained retrospectively. Prostates of all patients were embedded and sectioned at 5-mm intervals. The same pathologist examined the prostatic tissues from radical cystoprostatectomy specimens. Finally, a structured literature review was performed using MEDLINE and PUBMED to estimate the occurrence of incidental Pca in Asia. Of the 92 patients, 3 (3.3%) were found to have Pca; in one out of three (33.3%) patients the disease was clinically significant due to a Gleason grade 4 carcinoma. Eight articles were included in our review. The overall incidence of Pca discovered incidentally in radical cystoprostatectomy specimens in Asia was 9.9% (64/642). When age was restricted to 〈 60 years, only 7 out of 222 (3.2%) patients were found to have synchronous Pca, and none of the cases was clinically significant. The occurrence of Pca in radical cystoprostatectorny specimens in Asia is much lower than that in Western countries. PSC might be feasible for Asian patients under a strict preoperative selection.展开更多

This system enables the neurosurgeon to interact intuitively with the three-dimensional graphics in a direct manner resembling the way one communicates with the real objects. In the paper, we explored its values in pre-operation surgical planning for intracranial meningiomas resection. Methods Brain computed tomography (CT), magnetic resonance imaging (MRI), and magnetic resonance venography (MRV) were performed on 10 patients with parasagittal and falcine meningiomas located on central groove area; brain CT, MRI and magnetic resonance angiography (MIRA) were performed on 10 patients with anterior skull base meningiomas and 10 patients with sphenoid ridge meningiomas. All these data were transferred to Dextroscope virtual reality system, and reconstructed. Then meningiomas, skull base, brain tissue, drainage vein and cerebral arteries were displayed within the system, and their anatomic relationships were evaluated. Also, the simulation operations were performed. Results For parasagittal and falcine meningiomas, the relationships of tumor with drainage vein and superior sagittal sinus were clearly displayed in the Dextroscope system. For anterior skull base and sphenoid ridge meningiomas, the relationships of tumor with bilateral internal carotid arteries, anterior cerebral arteries, middle cerebral arteries and skull base were vividly displayed within the virtual reality system. Surgical planning and simulation operation of all cases were performed as well. The real operations of all patients were conducted according to the simulation with well outcomes. Conclusions According to the virtual reality planning, neurosurgeons could get more anatomic information about meningioma and its surrounding structures, especially important vessels, and choose the best approach for tumor resection, which would lead to better prognosis for patients.展开更多

The governing equations are derived by circumferentially averaging the three-dimensional Navier-Stokes equations neglecting the circumferentially non-uniform and viscous terms. An inviscid blade force model similar to the Large-particle method is derived. The viscous blade force has been modeled by the distributed loss model. The convective fluxes of the governing equation are discretized with the Edward’s low-diffusion flux-splitting (LDFSS) scheme. And a point-iterative Symmetric Gauss-Seidel (SGS) scheme is used in the temporal discretization. The throughflow model has been applied to the NASA Rotor 67 and a high-load transonic fan stage ATS-2. The reasonable good agreements with the experiments and the 3D viscous computations show the potential of the method.展开更多

We have found that short T1 inversion recovery long time echo (STIR LONG TE) was better in showing radicular vein. To further study the methods and character of the 3D-MRI of lumbar nerve root, ganglions and radicular vein in normal and lumbar disc herniation (LDH) adults, in the present study, we evaluated two 3D-MRI techniques, STIR TSE and STIR LONG TE for demonstrating lumbar nerve roots and especial radicular vein in normal and LDH adults. Methods Twelve normal adult persons and 19 LDH patients were included in this study; special oblique coronal location was adopted. STIR TSE and STIR LONG TE were performed in all participants, and the detection rates of radicular vein and nerve root of L3, L4, L5,S1 were calculated. The grading system was used in grading compromise of the intraspinal extradural lumbar nerve root, ganglion and radicular vein in LDH patients, and all the grading injury of nerve root, ganglion and radicular vein had been evaluated. Results The definite rate of radicular vein (DRRV) of 3D-MRI of STIR LONG TE was significantly different from that of STIR TSE. DRRV of STIR LONG TE was L3, 91.7%, L4, 100%, L5, 100%, $1, 100% and that of STIR TSE was L3, 33.3%, L4, 37.5%, L5, 58.3%, $1,45.8% in normal adults. It showed no difference between STIR LONG TE and STIR TSE in the detection rate of nerve root. The different patho-injuries of nerve root, ganglion and radicular vein could be seen in all LDH patients by the 3D-MRI of STIR LONG TE. The mean score of Japanese Orthopaedic Association (JOA) was 16.16, and 29 nerve roots were calculated in all the study. The mean grading injury of nerve root was 2.17, that of nerve root ganglion was 1.28, that of radicular vein was 1.83, and the mean co-grading in展开更多

Berbamine(BM), a natural product mainly derived from Berberis vulgaris L, possesses multiple bioactivities as a traditional medicine. However, the protective effect of BM on ALD remains unknown. In this study, we investigated the effect of BM on ethanol-induced hepatic injury in mice and its underlying mechanism. It was shown that BM at 0.3125-40 μmol·L^-1 had no effect on macrophages and hepatocytes proliferation. BM at 5-20 μmol·L^-1 significantly inhibited lipopolysaccharide(LPS) or acetate-induced IL-1β and IL-6 m RNA expression in RAW264.7 cells. Moreover, BM treatment significantly inhibited LPS-induced p65 and STAT3 phosphorylation in RAW264.7 cells. Hepatic histopathology analysis showed that inflammatory cells infiltration and lipid accumulation were suppressed by 25 and 50 mg·kg^-1 BM administration in ethanol-induced hepatic injury mouse model. Meanwhile, BM treatment significantly inhibited serum ALT and AST levels in ethanol-fed mice. Oil red O staining results showed that BM administration ameliorated hepatic lipid accumulation in ethanolfed mice. Preventions of ethanol-induced hepatic injury by BM were reflected by markedly decreased serum and hepatic triglyceride(TG) and total cholesterol(TC) contents. Real-time PCR results showed that BM treatment significantly inhibited pro-inflammatory cytokines m RNA expression in ethanol-fed mouse liver. Remarkably, the mechanism of action of BM was related to the reduction of ethanol-induced NF-κB and STAT3 phosphorylation levels in liver. In addition, BM treatment significantly inhibited ERK phosphorylation but not JNK and p38 of MAPK pathway. Taken together, our results demonstrate a beneficial effect of BM on ethanol-induced liver injury via a mechanism associated with inactivation of NF-κB, STAT3 and ERK pathway, which gives insight into the further evaluation of the therapeutic potential of BM for ALD.展开更多

A total of 115 patients with mCRPC undergoing a docetaxel q3w regimen were enrolled in this study. A survival analysis was performed using the Kaplan-Meier method. Cox proportional hazards models were used to evaluate the prognostic value of all covariates for OS. OS was also analysed after stratifying patients according to the results of multivariate analysis. The median OS for the entire cohort was 17.0 months. The multivariate analysis showed that the prostate-specific antigen doubling time (PSADT), baseline haemoglobin (Hb) concentration, alkaline phosphatase (ALP) concentration, cycles of chemotherapy and time to castration resistance were independent prognostic factors of OS. According to the presence of PSADT 〈46.3 days and baseline ALP/〉 110 IU 1-1, all patients were divided into three risk groups: low-risk group (no risk factors), intermediate-risk group (one risk factor) and high-risk group (two risk factors). Median OSs for patients in low-, intermediate- and high-risk groups were 28.0 months (95% Ch 23.8-32.2), 21.0 months (95% Ch 18.9-23.1) and 11.0 months (95% Ch 7.6-14.4), respectively (P〈O.O01). In conclusion, PSADT, baseline Hb concentration, ALP concentration, cycles of chemotherapy and time to castration resistance were independent prognostic factors of OS in Chinese patients with mCRPC treated with docetaxel. PSADT combined with the baseline ALP concentration could be a useful risk stratification parameter for evaluating survival outcomes.展开更多

The present study was designed to investigate the in vitro effects of estrogen-like activities of two widespread coumarins, osthole and imperatorin, using the MCF-7 cell proliferation assay and their alkaline phosphatase(ALP) activities in osteoblasts Saos-2 cells. The two compounds were found to strongly stimulate the proliferation of MCF-7 cells. The estrogen receptor-regulated ERα, progesterone receptor(PR) and PS2 m RNA levels were increased by treatment with osthole and imperatorin. All these effects were significantly inhibited by the specific estrogen receptor antagonist ICI182, 780. Cell cycle analysis revealed that their proliferation stimulatory effect was associated with a marked increase in the number of MCF-7 cells in S phase, which was similar to that observed with estradiol. It was also observed that they significantly increased ALP activity, which was reversed by ICI182,780. These results suggested that osthole and imperatorin could stimulate osteoblastic activity by displaying estrogenic properties or through the ER pathway. In conclusion, osthole and imperatorin may represent new pharmacological tools for the treatment of osteoporosis.展开更多

文摘AIM: To understand the role and significance of side population (SP) cells from hepatocellular carcinoma (HCC) in hepatocarcinogenesis, development, relapse and metastasis, we simulated the denutrition conditions that cancer cells experience in clinical therapy, observed the different anti-apoptosis ability of SP cells and non-SP cells under such conditions, and established the possible effects of P53, Bcl-2 and Bax on survival of SP cells. Further research on SP cells may have potential applications in the field of anticancer therapy.

文摘Restoration and reconstruction of the degraded Tarim River ecosystem is an important challenge. This paper discusses strategies for the protection and restoration of the degraded vegetation in the lower reaches of the Tarim River and provides information to complement ongoing theoretical research into ecological restoration in add or semi-arid ecosystems.

文摘Magnesium(Mg)alloys have been extensively used in various fields, such as aerospace, automobile, electronics, and biomedical industries, due to their high specific strength and stiff ness, excellent vibration absorption, electromagnetic shielding eff ect, good machinability, and recyclability.Friction stir processing(FSP)is a severe plastic deformation technique, based on the principle of friction stir welding. In addition to introducing the basic principle and advantages of FSP,this paper reviews the studies of FSP in the modification of the cast structure, superplastic deformation behavior, preparation of finegrained Mg alloys and Mg-based surface composites, and additive manufacturing.FSP not only refines, homogenizes, and densifies the microstructure, but also eliminates the cast microstructure defects, breaks up the brittle and network-like phases, and prepares fine-grained,ultrafine-,and nano-grained Mg alloys.Indeed,FSP significantly improves the comprehensive mechanical properties of the alloys and achieves low-temperature and/or high strain rate superplasticity.Furthermore,FSP can produce particle-and fiber-reinforced Mg-based surface composites. As a promising additive manufacturing technique of light metals,FSP enables the additive manufacturing of Mg alloys. Finally,we prospect the future research direction and application with friction stir processed Mg alloys.

文摘Silicon can improve drought tolerance of plants, but the mechanism still remains unclear.Previous studies have mainly concentrated on silicon-accumulating plants, whereas less work has been conducted in silicon-excluding plants, such as tomato(Solanum lycopersicum L.).In this study,we investigated the effects of exogenous silicon(2.5 mmol L^(-1))on the chlorophyll fluorescence and expression of photosynthesis-related genes in tomato seedlings(Zhongza 9)under water stress induced by 10%(w/v)polyethylene glycol(PEG-6000).The results showed that under water stress, the growth of shoot and root was inhibited, and the chlorophyll and carotenoid concentrations were decreased, while silicon addition improved the plant growth and increased the concentrations of chlorophyll and carotenoid.Under water sterss, chlorophyll fluorescence parameters such as PSII maximum photochemical efficiency(F_v/F_m),effective quantum efficiency, actual photochemical quantum efficiency(Ф_(PSII)),photosynthetic electron transport rate(ETR),and photochemical quenching coefficient(q_P)were decreased;while these changes were reversed in the presence of added silicon. The expressions of some photosynthesis-related genes including PetE,PetF,PsbP,PsbQ,PsbW,and Psb28 were down-regulated under water stress, and exogenous Si could partially up-regulate their expressions. These results suggest that silicon plays a role in the alleviation of water stress by modulating some photosynthesis-related genes and regulating the photochemical process, and thus promoting photosynthesis.

文摘The bipolar Navier-Stokes-Poisson system (BNSP) has been used to simulate the transport of charged particles (ions and electrons for instance) under the influence of electrostatic force governed by the self-consistent Poisson equation. The above estimates reveal the essential difference between the unipolar and the bipolar Navier-Stokes-Poisson systems.

文摘Cu/Al multilayers were produced by high-temperature accumulative roll bonding(ARB)methods up to three passes. To achieve a high bonding strength, prior to ARB processing, the Cu and Al sheets were heated to 350,400,450 and 500 ℃,respectively. The mechanical properties were evaluated by tensile tests. The microstructure was examined by optical microscopy and scanning electron microscopy equipped with energy dispersive spectrometry. The ultimate tensile stress, the grain size and the thickness of diffusion layer of lamellar composites increase with rolling temperature. When the rolling temperature is 400 ℃,the laminates show the highest ductility, but the yield stress is the lowest. As the rolling temperature further increases, both the yield stress and the ultimate tensile stress increase and the ductility decreases slightly. The mechanical properties of lamellar composites processed by low and high temperature ARB are determined by grain size and the thickness of diffusion layer, respectively.

文摘Based on the thermal conduction equations, the three-dimensional (3D) temperature field of a work roll was investigated using finite element method (FEM). Furthermore, the results of this analysis indicate that temperature and thermal stress are useful parameters for the investigation of roll thermal fatigue and also for improving the quality of strip during rolling.

文摘The purpose of this study is to investigate the frequency of prostate cancer (Pca) discovered incidentally in radical cystoprostatectomy specimens in Asia and to determine the feasibility of prostate-sparing cystectomy (PSC) for Asian patients. PSC might be feasible for Asian patients under a strict preoperative selection.

文摘Background The Dextroscope system by Volume Interactions (Singapore) had been applied to minimally invasive neurosurgery in many units. Conclusions According to the virtual reality planning, neurosurgeons could get more anatomic information about meningioma and its surrounding structures, especially important vessels, and choose the best approach for tumor resection, which would lead to better prognosis for patients.

文摘An Al−3.6Cu−1Li alloy was subjected to room temperature rolling and cryorolling to investigate their effects on microstructure evolution and mechanical properties. The microstructure and aging characteristics of the room temperature-rolled and the cryorolled alloys with 70%and 90%of thickness reductions were studied by microstructure analysis and mechanical tests. The samples subjected to cryorolling with 90%of thickness reduction have high strength and good toughness. This is mainly due to the inhibition of dynamic recovery and the accumulation of high-density dislocations in cryorolled samples. In addition, the artificial aging reveals that the temperature at which peak hardness is attained is inversely proportional to the deformation amount and directly proportional to the rolling temperature. Moreover, bright field images of cryorolled samples after aging indicate the existence of T1(Al2CuLi)precipitates.This suggests that the high stored strain energy enhances the aging kinetics of the alloy, which further promotes the nucleation of T1 phases.

文摘A throughflow model based on the time-marching finite volume approach is described in this paper. The reasonable good agreements with the experiments and the 3D viscous computations show the potential of the method.
文摘Background Although three-dimensional MRI (3D-MRI) of short T1 inversion recovery turbo spin-echo (STIR TSE) has showed superior to two-dimensional MRI (2D-MRI) in showing the spinal cord and lumbar nerve roots, it remains difficult in demonstrating radicular vein at present. The mean grading injury of nerve root was 2.17, that of nerve root ganglion was 1.28, that of radicular vein was 1.83, and the mean co-grading in

文摘Alcoholic liver disease(ALD) has become one of the leading causes of death in the world. Taken together, our results demonstrate a beneficial effect of BM on ethanol-induced liver injury via a mechanism associated with inactivation of NF-κB, STAT3 and ERK pathway, which gives insight into the further evaluation of the therapeutic potential of BM for ALD.
文摘This study aims to evaluate the potential value of patient characteristics in predicting overall survival (OS) in patients with metastatic castration-resistant prostate cancer (mCRPC) treated with docetaxel-based thermotherapy. PSADT combined with the baseline ALP concentration could be a useful risk stratification parameter for evaluating survival outcomes.

文摘Ultrafine-grained(UFG)AA1060 sheets were fabricated via five-cycle accumulative roll bonding(ARB)and subsequent three-pass cold rolling(298 K),or cryorolling(83 K and 173 K).Microstructures of the aluminum samples were examined via transmission electron microscopy, and their mechanical properties were measured via tensile and microhardness testing. Results indicate that ultrafine grains in ARB-processed sheets were further refined by subsequent rolling, and the grain size became finer with reducing rolling temperature. The mean grain size of 666 nm in the sheets subjected to ARB was refined to 346 or 266 nm,respectively, via subsequent cold rolling or cryorolling(83 K).Subsequent cryorolling resulted in ultrafine-grained sheets of higher strength and ductility than those of the sheets subjected to cold rolling.

文摘There is an increasing interest in phytoestrogens due to their potential medical usage in hormone replacement therapy(HRT). In conclusion, osthole and imperatorin may represent new pharmacological tools for the treatment of osteoporosis.
