AIM:To analyze the differences and relevance of Yes-associated protein (YAP) and survivin, and to explore the correlation and signifi cance of their expression in gastric carcinoma and precancerous lesions.METHODS: Th...AIM:To analyze the differences and relevance of Yes-associated protein (YAP) and survivin, and to explore the correlation and signifi cance of their expression in gastric carcinoma and precancerous lesions.METHODS: The PV9000 immunohistochemical method was used to detect the expression of YAP and survivin in 98 cases of normal gastric mucosa, 58 intestinal metaplasia (IM), 32 dysplasia and 98 gastric carcinoma.RESULTS: The positive rates of YAP in dysplasia (37.5%) and gastric carcinoma (48.0%) were significantly higher than that in normal gastric mucosa (13.3%), P<0.01. The positive rates of survivin in IM (53.4%), dysplasia (59.4%) and gastric carcinoma (65.3%) were significantly higher than in normal gastric mucosa (11.2%), P<0.01. Survivin expression gradually increased from 41.7% in well differentiated adenocarcinoma through 58.3% in moderately differentiated adenocarcinoma to 75.6% in poorly differentiated adenocarcinoma, with significant Rank correlation, rk=0.279, P<0.01. The positive rate of survivin in gastric carcinoma of diffused type (74.6%) was significantly higher than that in intestinal type (51.3%), P<0.05. In gastric carcinoma with lymph node metastasis (76.9%), the positive rate of survivin was signifi cantly higher than that in the group without lymph node metastasis (41.2%), P<0.01. In 98 cases of gastric carcinoma, the expression of YAP and of survivin were positively correlated, rk=0.246, P<0.01.CONCLUSION: YAP may play an important role as a carcinogenic factor and may induce survivin expression. Detecting both markers together may help in early diagnosis of gastric carcinoma.展开更多
Background The new Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation was developed to address the systematic underestimation of glomerular filtration rate (GFR) by the Modification of Diet in Re...Background The new Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation was developed to address the systematic underestimation of glomerular filtration rate (GFR) by the Modification of Diet in Renal Disease (MDRD) Study equation in patients with relatively well-preserved kidney function.Performance of the new equation in the Chinese population is unknown.The goal of the present study was to compare performance of these two equations in Chinese patients with chronic kidney disease (CKD).Methods We enrolled 450 Chinese patients (239 women and 211 men) with CKD in the present study.The renal dynamic imaging method was used to measure the referenced standard GFR (rGFR) for comparison with estimations using the two equations.Their overall performance was assessed with the Bland-Altman method and receiver-operating characteristics (ROC) analysis.Performance of the two equations in lower and higher estimated GFR (eGFR) subgroups was further investigated.Results Both eGFRs correlated well with rGFR (r=0.88,0.81,P〈0.05).In overall performance,the CKD-EPI equation showed less bias,higher precision and improved accuracy,and was better for detecting CKD.In the higher-eGFR subgroup,the CKD-EPI equation corrected the underestimation of GFR by the abbreviated MDRD equation.Conclusions The CKD-EPI equation outperformed the abbreviated MDRD equation not only in overall performance but also in the subgroups studied.For the present,the CKD-EPI equation appears to be the first-choice prediction equation for estimating GFR.展开更多
Background:The number of pulmonary tuberculosis(PTB)cases in China ranks third in the world.A continuous increase in cases has recently been recorded in Zhaotong prefecture-level city,which is located in the northeast...Background:The number of pulmonary tuberculosis(PTB)cases in China ranks third in the world.A continuous increase in cases has recently been recorded in Zhaotong prefecture-level city,which is located in the northeastern part of Yunnan province.This study explored the space-time dynamics of PTB cases in Zhaotong to provide useful information that will help guide policymakers to formulate effective regional prevention and control strategies.Methods:The data on PTB cases were extracted from the nationwide tuberculosis online registration system.Time series and spatial cluster analyses were applied to detect PTB temporal trends and spatial patterns at the town level between 2011 and 2015 in Zhaotong.Three indicators of PTB treatment registration history were used:initial treatment registration rate,re-treatment registration rate,and total PTB registration rate.Results:Seasonal trends were detected with an apparent symptom onset peak during the winter season and a registration peak during the spring season.A most likely cluster and six secondary clusters were identified for the total PTB registration rate,one most likely cluster and five secondary clusters for the initial treatment registration rate,and one most likely cluster for the re-treatment registration rate.The most likely cluster of the three indicators had a similar spatial distribution and size in Zhenxiong County,which is characterised by a poor socio-economic level and the largest population in Yunnan.Conclusion:This study identified temporal and spatial distribution of PTB in a high PTB burden area using existing health data.The results of the study provide useful information on the prevailing epidemiological situation of PTB in Zhaotong and could be used to develop strategies for more effective PTB control at the town level.The cluster that overlapped the three PTB indicators falls within the geographic areas where PTB control efforts should be prioritised.展开更多

Adipocytokines might mediate the relationship. Our study was aimed to investigate the relationship between serum levels of resistin, adiponectin and leptin and the onset, invasion and metastasis of breast cancer. Methods Blood samples were collected from 80 newly diagnosed, histologically confirmed breast cancer patients and 50 age-matched healthy controls. Serum levels of resistin, adiponectin and leptin were determined by enzyme-linked immunosorbent assays (ELISA); fasting blood glucose (FBG), lipids, body mass index (BMI), and waist circumference (WC) were assayed simultaneously. Results Serum levels of adiponectin ((8.60±2.92) mg/L vs (10.37±2.81) mg/L, P=-0.001) and HDL-c were significantly decreased in breast cancer patients in comparison to controls. Serum levels of resistin ((26.35±5.36) μg/L vs (23.32±4.75) μg/L, P=-0.000), leptin ((1.35±0.42) μg/L vs (1.06±0.39) μg/L, P=0.003), FBG and triglyceride (TG) in breast cancer patients were increased in contrast to controls, respectively. However, we did not find the significant difference of the serum levels of resistin, adiponectin and leptin between premenopausal breast cancer patients and healthy controls (P= 0.091, 0.109 and 0.084, respectively). The serum levels of resistin, adiponectin and leptin were significantly different between patients with lymph node metastasis (LNM) and those without LNM (P=-0.001, 0.000 and 0.006, respectively). The stepwise regression analysis indicated that the tumor size had the close correlation with leptin (R^2=0.414, P=-0.000) and FBG (R^2=0.602, P=0.000). Simultane...The present study was designed to investigate the bioactive constituents ofXanthoceras sorbifolia in terms of amounts and their antioxidant, DNA scissiou protection, and ct-glucosidase inhibitory activities. Simultaneous quantification of 10 X. sorbifolia constituents was carried out by a newly established ultra-high performance liquid chromatography-quadrupole mass spectrometry method (UHPLC-MS), The antioxidant activities were evaluated by measuring DPPH radical scavenging and DNA scission protective activities. The a-glucosidase inhibitory activities were investigated by using an assay with a-glucosidase from Bacillus Stearothermophilus and disaccharidases from mouse intestine. We found that the wood ofX. sorbifolia was rich in phenolic compounds with the contents of catechin, epicatechin, myricetin, and dihydromyricetin being 0.12-0.19, 1.94-2.16, 0.77-0.91, and 6.76-7.89 mg.g-1, respectively. The four constituents strongly scavenged DPPH radicals (with ECs0 being 4.2, 3.8 and 5.7 μg-mL-1, respectively) and remarkably protected peroxyl radical-induced DNA strand scission (92.10%, 94.66%, 75.44% and 89.95% of protection, respectively, at a concentration of 10 μmol.L-1). A dimeric flavan 3-ol, epigallocatechin-(4β→8, 2β→O-7)-epicatechin potently inhibited a-glucosidase with an IC50 value being as low as 1.2 μg.mL1. The established UHPLC-MS method could serve as a quality control tool for X. sorbifolia. In conclusion, the high contents of antioxidant and α-glucosidase inhibitory constituents in X. sorbifolia support its use as complementation of other therapeutic agents for metabolic disorders, such as diabetes and hypertension.展开更多

In this study, ...Objective The results of studies on association between -148C/T polymorphism in promoter region of β3-fibrinogen gene and susceptibility to cerebral infarction in Chinese population are controversial. In this study, we summarize the results of published works in this field by a meta-analysis. Data sources Genetic association studies evaluating the β-fibdnogen gene -148C/T polymorphisms and cerebral infarction involving Chinese population published before December 2005 were collected from PubMed, EMBASE and CNKI. Study selection Case control studies involving unrelated, Han subjects aged from 18 to 80 years, and the internationally recognized diagnostic standard of cerebral infarction and genotype frequencies in control group consistent with Hardy-Weinberg equilibrium were used. Publication bias was tested by funnel plot and the odds ratios of all studies were combined dependent on the result of heterogeneity test among the individual studies. The software Review Manager (Version 4.2) was used for meta-analysis. Results Eleven studies including 1223 patients and 1433 controls met the selection criteria. There was no heterogeneity among the odds ratios (ORs) of individual studies (Х^2=17.82, P=0.06). The combined OFt of susceptibility to cerebral infarction in -148T allele carriers compared to the wild homozygote was 1.32 (95%CI 1.12 to 1.55, P=-0.0008). In the patients with cerebral infarction, the average plasma fibrinogen level of allele T carrier was 0.42 g/L (95% CI 0.29 to 0.54, P〈0.001), higher than that of -148C/C homozygous ones. Conclusions β3-fibrinogen gene -148C/T polymorphism might contribute to susceptibility to cerebral infarction in Han Chinese. To reach a definitive conclusion, further gene to gene and gene to environment interactions studies on β3-fibrinogen polymorphisms and cerebral infarction with large sample size are required.展开更多

This study aimed to evaluate the dynamic influence of hyperglycemia on the expression of these GLUTs by measuring their expression in the brain at different blood glucose levels in e rat model of diabetes. This might help to determine the proper blood glucose threshold level in the treatment of diabetic apoplexy. Methods Diabetes mellitus was induced with streptozotocin (STZ) in 30 rats. The rats were randomly divided into 3 groups: diabetic group without blood glucose control (group DM1), diabetic rats treated with low dose insulin (group DM2) end diabetic rats treated with high dose insulin (group DM3). The mRNA end protein levels of GLUT1 end GLUT3 were essayed by reverse trenscriptese-polymerese chain reaction (RT-PCR) end immunohistochemistry, respectively. Results Compared with normal control rats, the G/UT1 mRNA was reduced by 46.08%, 29.80%, 19.22% (P〈0.01) in DM1, DM2, end DM3 group, respectively; end the GLUT3 mRNA was reduced by 75.00%, 46.75%, end 17.89% (P〈0.01) in DM1, DM2, end DM3 group, respectively. The abundance of GLUT1 end GLUT3 proteins had negative correlation with the blood glucose level (P〈0.01). The density of microvessels in the brain of diabetic rats did not change significantly compared with normal rats. Conclusions Chronic hyperglycemia downreguletes G/UT1 end GLUT3 expression at both mRNA end protein levels in the rat brain, which is not due to the decrease of the density of microvessels. The downreguletion of G/UT1 end GLUT3 expression might be the adaptive reaction of the body to prevent excessive glucose entering the cell that may lead to cell damage.展开更多

Rich low-grade diatomite mineral, as raw material, was modified with calcium carbonate to improve the adsorption properties of diatomite for heavy metal ions. The effects of dosage(m), pH, adsorbing time(t), temperature(T) and concentration of adsorbent(c) on the removal rate of heavy metal ions(Cu2+, Pb2+, Zn2+ and Cd2+) were studied. The results show that under the suitable conditions, T=15 ℃, c=1.5 mmol/L, m=10 g/L, t=60 min, in a weak acid environment, the removal effect of the four kinds of metal ions by modified diatomite is the best.展开更多

Doublelabeling staining was used to display the distribution of Bcl-2^+/ki-67 cells in 162 cases of GC and its matched normal mucosa and precancerous lesion.RESULTS: The positive rate of Bmi-1 expression in GC(52.5%) was significantly higher than that in normal gastric mucosa (21.6%, X^2 = 33.088, P 〈 0.05). The Bmi-1 expression in GC was closely related with the Lauren's and Borrmann's classification and clinicalstage (X^2 = 4.400, 6.122 and 11.190, respectively, P〈 0.05). The expression of ki-67 was related to the Borrmann's classification (X^2 = 13.380, P 〈 0.05).Bcl-2 expression was correlated with the Lauren's classification (Z2 = 4.725, P 〈 0.05), and the Bmi-1 expression both in GC (rk = 0.157, P 〈 0.05) and inintestinal metaplasia (rk = 0.270, P 〈 0.05).CONCLUSION: Abnormal Bmi-1 expression in GCmay be involved in cell proliferation, apoptosis andcancerization. However, whether acute administration of ethanol in the m PFC can modulate pain perception is unknown.Here we showed that bilateral microinjections of ethanol into the prelimbic and infralimbic areas of the m PFC lowered the bilateral mechanical pain threshold for 48 h without influencing thermal pain sensitivity in adult rats.However, bilateral microinjections of artificial cerebrospinal fluid into the m PFC or bilateral microinjections of ethanol into the dorsolateral PFC(also termed as motor cortex area 1 in Paxinos and Watson's atlas of The Rat Brain. Elsevier Academic Press, Amsterdam, 2005) failed to do so, suggesting regional selectivity of the effects of ethanol. Moreover, bilateral microinjections of ethanol didnot change the expression of either pro-apoptotic(caspase-3 and Bax) or anti-apoptotic(Bcl-2) proteins, suggesting that the dose was safe and validating the method used in the current study. To determine whether c-aminobutyric acid A(GABA_A) receptors are involved in mediating the ethanol effects, muscimol, a selective GABA_Areceptor agonist, or bicuculline, a selective GABA_A receptor antagonist, was administered alone or co-administered with ethanol through the same route into the bilateral m PFC. The results showed that muscimol mimicked the effects of ethanol while bicuculline completely reversed the effects of ethanol and muscimol. In conclusion, ethanol increases mechanical pain sensitivity through activation of GABA_A receptors in the m PFC of rats.展开更多

METHODS:We cloned Tip-αfrom the H.pylori strain 26695,transformed Escherichia coli with an expression plasmid,and then confirmed the expression product by Western blotting.Using different concentrations of Tip-αthat affected SGC7901 and GES-1 cells at different times,we assessed cytokine levels using enzyme-linked immunosorbent assay.We blocked SGC7901 cells with pyrrolidine dithiocarbamate(PDTC),a specific inhibitor of nuclear factorκB(NF-κB).We then detected interleukin(IL)-1βand TNF-αlevels in SGC7901 cells. RESULTS:Western blot analysis using an anti-Tip-α antibody revealed a 23-kDa protein,which indicated that recombinant Tip-αprotein was recombined successfully.The levels of IL-1β,IL-8 and TNF-αwere sig-nificantly higher following Tip-αinterference,whether GES-1 cells or SGC-7901 cells were used(P<0.05).However,the levels of cytokines(including IL-1β,IL-8 and TNF-α)secreted by SGC-7901 cells were greater than those secreted by GES-1 cells following treatment with Tip-αat the same concentration and for the same duration(P<0.05).After blocking NF-κB with PDTC, the cells(GES-1 cells and SGC-7901 cells)underwent interference with Tip-α.We found that IL-1βand TNF-αlevels were significantly decreased compared to cells that only underwent Tip-αinterference(P<0.05). CONCLUSION:Tip-αplays an important role in cyto-kine expression through NF-κB.展开更多

Methods: The tRRL sequence motif was identified as a tumor molecular marker specifically binding to TDECs. Tyrosine was conjugated to the amino terminal of RRL (Cys-Gly-Gly-Arg-Arg-Leu-Gly-Gly-Cys) for labeling with radionuclide iodine-131 (1311-tRRL). The uptake ability and molecular binding of tRRL to tumor cells and angiogenic endothelium were studied using flow cytometry and radioactivity counter in vitro. Whether VEGFR-2 is the binging site of tRRL was investigated. Biodistribution and single-photon emission computed tomography (SPECT) imaging of 131-tRRL were used to evaluate the effectiveness of this new imaging agent to visualize varied tumor xenografts in nude mice. Results: In vitro cellular uptake experiments revealed that tRRL could not only adhere to tumor angiogenic endothelial cells but also largely accumulate in malignant tumor cells. VEGFR-2, which is highly expressed on TDECs, was probably not the solely binding ligand for tRRL targeted to tumor angiogenic endothelium, 131-tRRL mainly accumulated in tumors in vivo, not other organs at 24 h after injection. SPECT imaging with 131-tRRL clearly visualized tumors in nude mice, especially at 24 h. Heavy metal containing pickling sludge(PS) is one of the by-products of the stainless-steel-making industry,which has been considered hazardous due to contained chromium and nickel.Traditional methods of PS disposin...Heavy metal containing pickling sludge(PS) is one of the by-products of the stainless-steel-making industry,which has been considered hazardous due to contained chromium and nickel.Traditional methods of PS disposing are landfill and cement solidification.This research is aimed at disposing PS by solidification/stabilization and reusing it as a nucleation agent of glass–ceramics.The crystallization behavior and the properties of a glass in the CaO–MgO–SiO2–Al2O3 system were studied by considering PS as the nucleation agent.Experimental results confirm that introducing 14 wt% PS as the nucleation agent of glass–ceramics can decrease crystallization temperature by 110.8 °C,refine the grain size by forming isometric crystals with size of 2 lm,enhance Vickers hardness by 2690 MPa and decrease water absorption from(1.21 ± 0.10) wt% to(0.04 ± 0.01) wt%.Therefore,it is reasonable to conclude that PS can be utilized as a nucleation agent to improve the crystallization and mechanical properties of the glass–ceramics.The testing results of US EPA toxicity characteristic leaching procedure(TCLP)confirm the safety of this reusing method.展开更多
Background:Pulmonary tuberculosis(PTB,both smear positive and smear negative)is an airborne infectious disease of major public health concern in China and other parts of the world where PTB endemicity is reported.This...Background:Pulmonary tuberculosis(PTB,both smear positive and smear negative)is an airborne infectious disease of major public health concern in China and other parts of the world where PTB endemicity is reported.This study aims at identifying PTB spatio-temporal clusters and associated risk factors in Zhaotong prefecture-level city,located in southwest China,where the PTB notification rate was higher than the average rate in the entire country.Methods:Space-time scan statistics were carried out using PTB registered data in the nationwide TB online registration system from 2011 to 2015,to identify spatial clusters.PTB patients diagnosed between October 2015 and February 2016 were selected and a structured questionnaire was administered to collect a set of variables that includes socio-economic status,behavioural characteristics,local environmental and biological characteristics.Based on the discovery of detailed town-level spatio-temporal PTB clusters,we divided selected subjects into two groups including the cases that resides within and outside identified clusters.Then,logistic regression analysis was applied comparing the results of variables between the two groups.Results:A total of 1508 subjects consented and participated in the survey.Clusters for PTB cases were identified in 38 towns distributed over south-western Zhaotong.Logistic regression analysis showed that history of chronic bronchitis(OR=3.683,95%CI:2.180-6.223),living in an urban area(OR=5.876,95%CI:2.381-14.502)and using coal as the main fuel(OR=9.356,95%CI:5.620-15.576)were independently associated with clustering.While,not smoking(OR=0.340,95%CI:0.137-0.843)is the protection factor of spatial clustering.Conclusions:We found PTB specially clustered in south-western Zhaotong.The strong associated factors influencing the PTB spatial cluster including:the history of chronic bronchitis,living in the urban area,smoking and the use of coal as the main fuel for cooking and heating.Therefore,efforts should be made to curtail these associated factors.展开更多
Objective: To ascertain the influence of light intensity and water content of medium on the total dendrobine of Dendrobium nobile(D. nobile).Method: The principal component analysis combined with total dendrobine accu...Objective: To ascertain the influence of light intensity and water content of medium on the total dendrobine of Dendrobium nobile(D. nobile).Method: The principal component analysis combined with total dendrobine accumulation was conducted to assess the yield and quality of D. nobile in all treatments. In the experiment, D. nobile plants were cultivated in greenhouse as tested materials, and complete test of 9 treatments was adopted with relative light intensities 75.02%, 39.74%,29.93% and relative water content of medium 50%, 65%, 80%. The plants were treated in June and harvested till December. Indexes including agronomic traits, fresh weight and dry weight of stem and leaf, ash content, extract, and dendrobine were measured.Results: Under the light intensity treatments of 75.02% with 50%, 65%, 80% water content of medium, the basal stems of plants were comparatively thicker with more leaves, and the fresh weight and dry weight of stems and leaves were significantly higher than other 6 treatments.Leaves in all treatments contained dendrobine. Under the light intensity treatments of 75.02%with 50%, 65%, 80% water content of medium, dendrobine content of leaves was lower while dendrobine contents of other treatments were more than 0.60%. After comprehensive assessment through the principal component analysis and total dendrobine accumulation, the results showed that 3 treatments with relative light intensity of 75.02% ranked the top three.Conclusions: In brief, the moderately strong light intensity and water content of medium from low to medium can facilitate the growth and yield of D. nobile plants, while light intensity from moderately weak to weak can enhance the dendrobine content.展开更多
Background:Acute myeloid leukemia(AML)is a common type of hematological malignancy in elderly people.Geriatricians developed comprehensive geriatric assessment(CGA)methods for elderly patients;however,the tools used f...Background:Acute myeloid leukemia(AML)is a common type of hematological malignancy in elderly people.Geriatricians developed comprehensive geriatric assessment(CGA)methods for elderly patients;however,the tools used for CGA in AML are not uniform.Thus,we aimed to validate the instrumental activities of daily living(IADL)scales,age,comorbidities(Charlson Comorbidity Index),and albumin(IACA)index,which is a new tool for CGA,in elderly patients with AML.Methods:Patients aged>60 years who had been diagnosed with AML were screened for eligibility.Among the IACA low-,intermediate-,and high-risk groups,continuous variables were compared using the Mann-Whitney U test,and categorical variables were compared using/2 and Fisher exact tests.In addition,probabilities of overall survival(OS)were estimated using the Kaplan-Meier method.Results:A total of 21,34,and 6 patients were categorized into IACA low-risk(0 point),intermediate-risk(1-2 points),and high-risk(>3 points)groups,respectively.The rates of relapse/progression-related mortality were 23.8%,58.8%,and 100.0%in the IACA low-,intermediate-,and high-risk groups,respectively(χ^2=12.81,P<0.001).The 2-year probabilities of OS were 47.7%(95%confidence interval[CI]22.8%-72.6%)and 20.2%(95%CI 5.9%-34.5%)in the IACA low-and intermediate-risk groups,respectively(χ^2=5.99,P=0.014),which were significantly higher than those in the high-risk group(low-risk[47.7%(95%CI 22.8%-72.6%)]vs.high-risk[0],/=20.80,P<0.001;intermediate-risk[20.2%(95%CI 5.9%-34.5%)]vs.high-risk[0],χ^2=7.56,P=0.006;respectively).In the IACA low-risk group,the 2-year probability of OS in patients receiving induction chemotherapy(50.8%[95%CI 24.1%-77.5%])was significantly higher than that in those receiving best supportive care(0,χ^2=25.74,P<0.001).Conclusion:We suggest that the IACA index might be a simple and effective tool for comprehensive geriatric assessment in elderly AML patients.展开更多

文摘AIM:To analyze the differences and relevance of Yes-associated protein (YAP) and survivin, and to explore the correlation and signifi cance of their expression in gastric carcinoma and precancerous lesions.METHODS: The PV9000 immunohistochemical method was used to detect the expression of YAP and survivin in 98 cases of normal gastric mucosa, 58 intestinal metaplasia (IM), 32 dysplasia and 98 gastric carcinoma.RESULTS: The positive rates of YAP in dysplasia (37.5%) and gastric carcinoma (48.0%) were significantly higher than that in normal gastric mucosa (13.3%), P<0.01. The positive rates of survivin in IM (53.4%), dysplasia (59.4%) and gastric carcinoma (65.3%) were significantly higher than in normal gastric mucosa (11.2%), P<0.01. Survivin expression gradually increased from 41.7% in well differentiated adenocarcinoma through 58.3% in moderately differentiated adenocarcinoma to 75.6% in poorly differentiated adenocarcinoma, with significant Rank correlation, rk=0.279, P<0.01. The positive rate of survivin in gastric carcinoma of diffused type (74.6%) was significantly higher than that in intestinal type (51.3%), P<0.05. In gastric carcinoma with lymph node metastasis (76.9%), the positive rate of survivin was signifi cantly higher than that in the group without lymph node metastasis (41.2%), P<0.01. In 98 cases of gastric carcinoma, the expression of YAP and of survivin were positively correlated, rk=0.246, P<0.01.CONCLUSION: YAP may play an important role as a carcinogenic factor and may induce survivin expression. Detecting both markers together may help in early diagnosis of gastric carcinoma.
文摘Background The new Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation was developed to address the systematic underestimation of glomerular filtration rate (GFR) by the Modification of Diet in Renal Disease (MDRD) Study equation in patients with relatively well-preserved kidney function.Performance of the new equation in the Chinese population is unknown.The goal of the present study was to compare performance of these two equations in Chinese patients with chronic kidney disease (CKD).Methods We enrolled 450 Chinese patients (239 women and 211 men) with CKD in the present study.The renal dynamic imaging method was used to measure the referenced standard GFR (rGFR) for comparison with estimations using the two equations.Their overall performance was assessed with the Bland-Altman method and receiver-operating characteristics (ROC) analysis.Performance of the two equations in lower and higher estimated GFR (eGFR) subgroups was further investigated.Results Both eGFRs correlated well with rGFR (r=0.88,0.81,P〈0.05).In overall performance,the CKD-EPI equation showed less bias,higher precision and improved accuracy,and was better for detecting CKD.In the higher-eGFR subgroup,the CKD-EPI equation corrected the underestimation of GFR by the abbreviated MDRD equation.Conclusions The CKD-EPI equation outperformed the abbreviated MDRD equation not only in overall performance but also in the subgroups studied.For the present,the CKD-EPI equation appears to be the first-choice prediction equation for estimating GFR.

文摘Background:The number of pulmonary tuberculosis(PTB)cases in China ranks third in the world.A continuous increase in cases has recently been recorded in Zhaotong prefecture-level city,which is located in the northeastern part of Yunnan province.This study explored the space-time dynamics of PTB cases in Zhaotong to provide useful information that will help guide policymakers to formulate effective regional prevention and control strategies.Methods:The data on PTB cases were extracted from the nationwide tuberculosis online registration system.Time series and spatial cluster analyses were applied to detect PTB temporal trends and spatial patterns at the town level between 2011 and 2015 in Zhaotong.Three indicators of PTB treatment registration history were used:initial treatment registration rate,re-treatment registration rate,and total PTB registration rate.Results:Seasonal trends were detected with an apparent symptom onset peak during the winter season and a registration peak during the spring season.A most likely cluster and six secondary clusters were identified for the total PTB registration rate,one most likely cluster and five secondary clusters for the initial treatment registration rate,and one most likely cluster for the re-treatment registration rate.The most likely cluster of the three indicators had a similar spatial distribution and size in Zhenxiong County,which is characterised by a poor socio-economic level and the largest population in Yunnan.Conclusion:This study identified temporal and spatial distribution of PTB in a high PTB burden area using existing health data.The results of the study provide useful information on the prevailing epidemiological situation of PTB in Zhaotong and could be used to develop strategies for more effective PTB control at the town level.The cluster that overlapped the three PTB indicators falls within the geographic areas where PTB control efforts should be prioritised.
文摘Background Many researches suggested that obesity increased the risk of breast cancer, but the mechanism was currently unknown. Adipocytokines might mediate the relationship. Our study was aimed to investigate the relationship between serum levels of resistin, adiponectin and leptin and the onset, invasion and metastasis of breast cancer. Methods Blood samples were collected from 80 newly diagnosed, histologically confirmed breast cancer patients and 50 age-matched healthy controls. Serum levels of resistin, adiponectin and leptin were determined by enzyme-linked immunosorbent assays (ELISA); fasting blood glucose (FBG), lipids, body mass index (BMI), and waist circumference (WC) were assayed simultaneously. Results Serum levels of adiponectin ((8.60±2.92) mg/L vs (10.37±2.81) mg/L, P=-0.001) and HDL-c were significantly decreased in breast cancer patients in comparison to controls. Serum levels of resistin ((26.35±5.36) μg/L vs (23.32±4.75) μg/L, P=-0.000), leptin ((1.35±0.42) μg/L vs (1.06±0.39) μg/L, P=0.003), FBG and triglyceride (TG) in breast cancer patients were increased in contrast to controls, respectively. However, we did not find the significant difference of the serum levels of resistin, adiponectin and leptin between premenopausal breast cancer patients and healthy controls (P= 0.091, 0.109 and 0.084, respectively). The serum levels of resistin, adiponectin and leptin were significantly different between patients with lymph node metastasis (LNM) and those without LNM (P=-0.001, 0.000 and 0.006, respectively). The stepwise regression analysis indicated that the tumor size had the close correlation with leptin (R^2=0.414, P=-0.000) and FBG (R^2=0.602, P=0.000). Logistic regression analysis showed that reduced serum levels of adiponectin (OR:. 0.805; 95%CI: 0.704--0.921; P=0.001), HDL (OR: 0.087; 95%C/: 0.011-0.691, P=0.021), elevated leptin (OFt: 2.235; 95%C/:1.898--4.526; P=0.004) and resisti

文摘The present study was designed to investigate the bioactive constituents ofXanthoceras sorbifolia in terms of amounts and their antioxidant, DNA scissiou protection, and ct-glucosidase inhibitory activities. Simultaneous quantification of 10 X. sorbifolia constituents was carried out by a newly established ultra-high performance liquid chromatography-quadrupole mass spectrometry method (UHPLC-MS), The antioxidant activities were evaluated by measuring DPPH radical scavenging and DNA scission protective activities. The a-glucosidase inhibitory activities were investigated by using an assay with a-glucosidase from Bacillus Stearothermophilus and disaccharidases from mouse intestine. We found that the wood ofX. sorbifolia was rich in phenolic compounds with the contents of catechin, epicatechin, myricetin, and dihydromyricetin being 0.12-0.19, 1.94-2.16, 0.77-0.91, and 6.76-7.89 mg.g-1, respectively. The four constituents strongly scavenged DPPH radicals (with ECs0 being 4.2, 3.8 and 5.7 μg-mL-1, respectively) and remarkably protected peroxyl radical-induced DNA strand scission (92.10%, 94.66%, 75.44% and 89.95% of protection, respectively, at a concentration of 10 μmol.L-1). A dimeric flavan 3-ol, epigallocatechin-(4β→8, 2β→O-7)-epicatechin potently inhibited a-glucosidase with an IC50 value being as low as 1.2 μg.mL1. The established UHPLC-MS method could serve as a quality control tool for X. sorbifolia. In conclusion, the high contents of antioxidant and α-glucosidase inhibitory constituents in X. sorbifolia support its use as complementation of other therapeutic agents for metabolic disorders, such as diabetes and hypertension.

文摘Objective The results of studies on association between -148C/T polymorphism in promoter region of β3-fibrinogen gene and susceptibility to cerebral infarction in Chinese population are controversial. In this study, we summarize the results of published works in this field by a meta-analysis. Data sources Genetic association studies evaluating the β-fibdnogen gene -148C/T polymorphisms and cerebral infarction involving Chinese population published before December 2005 were collected from PubMed, EMBASE and CNKI. Study selection Case control studies involving unrelated, Han subjects aged from 18 to 80 years, and the internationally recognized diagnostic standard of cerebral infarction and genotype frequencies in control group consistent with Hardy-Weinberg equilibrium were used. Publication bias was tested by funnel plot and the odds ratios of all studies were combined dependent on the result of heterogeneity test among the individual studies. The software Review Manager (Version 4.2) was used for meta-analysis. Results Eleven studies including 1223 patients and 1433 controls met the selection criteria. There was no heterogeneity among the odds ratios (ORs) of individual studies (Х^2=17.82, P=0.06). The combined OFt of susceptibility to cerebral infarction in -148T allele carriers compared to the wild homozygote was 1.32 (95%CI 1.12 to 1.55, P=-0.0008). In the patients with cerebral infarction, the average plasma fibrinogen level of allele T carrier was 0.42 g/L (95% CI 0.29 to 0.54, P〈0.001), higher than that of -148C/C homozygous ones. Conclusions β3-fibrinogen gene -148C/T polymorphism might contribute to susceptibility to cerebral infarction in Han Chinese. To reach a definitive conclusion, further gene to gene and gene to environment interactions studies on β3-fibrinogen polymorphisms and cerebral infarction with large sample size are required.
文摘Background The delivery of glucose from the blood to the brain involves its passage across the endothelial cells of the blood-brain barrier (BBB), which is mediated by the facilitative glucose transporter protein 1 (GLUT1), end then across the neural cell membranes, which is mediated by GLUT3. This study aimed to evaluate the dynamic influence of hyperglycemia on the expression of these GLUTs by measuring their expression in the brain at different blood glucose levels in e rat model of diabetes. This might help to determine the proper blood glucose threshold level in the treatment of diabetic apoplexy. Methods Diabetes mellitus was induced with streptozotocin (STZ) in 30 rats. The rats were randomly divided into 3 groups: diabetic group without blood glucose control (group DM1), diabetic rats treated with low dose insulin (group DM2) end diabetic rats treated with high dose insulin (group DM3). The mRNA end protein levels of GLUT1 end GLUT3 were essayed by reverse trenscriptese-polymerese chain reaction (RT-PCR) end immunohistochemistry, respectively. Results Compared with normal control rats, the G/UT1 mRNA was reduced by 46.08%, 29.80%, 19.22% (P〈0.01) in DM1, DM2, end DM3 group, respectively; end the GLUT3 mRNA was reduced by 75.00%, 46.75%, end 17.89% (P〈0.01) in DM1, DM2, end DM3 group, respectively. The abundance of GLUT1 end GLUT3 proteins had negative correlation with the blood glucose level (P〈0.01). The density of microvessels in the brain of diabetic rats did not change significantly compared with normal rats. Conclusions Chronic hyperglycemia downreguletes G/UT1 end GLUT3 expression at both mRNA end protein levels in the rat brain, which is not due to the decrease of the density of microvessels. The downreguletion of G/UT1 end GLUT3 expression might be the adaptive reaction of the body to prevent excessive glucose entering the cell that may lead to cell damage.

文摘Remyelination plays a key role in functional recovery of axons after spinal cord injury.Glial cells are the most abundant cells in the central nervous system.When spinal cord injury occurs,many glial cells at the lesion site are immediately activated,and different cells differentially affect inflammatory reactions after injury.In this review,we aim to discuss the core role of oligodendrocyte precursor cells and crosstalk with the rest of glia and their subcategories in the remyelination process.Activated astrocytes influence proliferation,differentiation,and maturation of oligodendrocyte precursor cells,while activated microglia alter remyelination by regulating the inflammatory reaction after spinal cord injury.Understanding the interaction between oligodendrocyte precursor cells and the rest of glia is necessary when designing a therapeutic plan of remyelination after spinal cord injury.
文摘At present, heavy metal pollution has become a major environmental problem, influencing the survival of human and other creatures. Developing heavy metal ion adsorbents, which are environment friendly, of low cost and good performance is one of the effective means to solve heavy metal pollution. Rich low-grade diatomite mineral, as raw material, was modified with calcium carbonate to improve the adsorption properties of diatomite for heavy metal ions. The effects of dosage(m), pH, adsorbing time(t), temperature(T) and concentration of adsorbent(c) on the removal rate of heavy metal ions(Cu2+, Pb2+, Zn2+ and Cd2+) were studied. The results show that under the suitable conditions, T=15 ℃, c=1.5 mmol/L, m=10 g/L, t=60 min, in a weak acid environment, the removal effect of the four kinds of metal ions by modified diatomite is the best.

文摘AIM: To explore the relation between B-cell-specific Moloney murine leukemia virus insertion site 1 (Bmi-1) expression and the clinicopathological features of gastric carcinoma (GC).METHODS: Immunohistochemistry was used to detect the expression of Bmi-1 and ki-67. Doublelabeling staining was used to display the distribution of Bcl-2^+/ki-67 cells in 162 cases of GC and its matched normal mucosa and precancerous lesion.RESULTS: The positive rate of Bmi-1 expression in GC(52.5%) was significantly higher than that in normal gastric mucosa (21.6%, X^2 = 33.088, P 〈 0.05). The Bmi-1 expression in GC was closely related with the Lauren's and Borrmann's classification and clinicalstage (X^2 = 4.400, 6.122 and 11.190, respectively, P〈 0.05). The expression of ki-67 was related to the Borrmann's classification (X^2 = 13.380, P 〈 0.05).Bcl-2 expression was correlated with the Lauren's classification (Z2 = 4.725, P 〈 0.05), and the Bmi-1 expression both in GC (rk = 0.157, P 〈 0.05) and inintestinal metaplasia (rk = 0.270, P 〈 0.05).CONCLUSION: Abnormal Bmi-1 expression in GCmay be involved in cell proliferation, apoptosis andcancerization. This marker can objectively indicate theclinicopathological characteristics of GC.

文摘Ethanol is widely known for its ability to cause dramatic changes in emotion, social cognition, and behavior following systemic administration in humans.Human neuroimaging studies suggest that alcohol dependence and chronic pain may share common mechanisms through amygdala-medial prefrontal cortex(m PFC) interactions. However, whether acute administration of ethanol in the m PFC can modulate pain perception is unknown.Here we showed that bilateral microinjections of ethanol into the prelimbic and infralimbic areas of the m PFC lowered the bilateral mechanical pain threshold for 48 h without influencing thermal pain sensitivity in adult rats.However, bilateral microinjections of artificial cerebrospinal fluid into the m PFC or bilateral microinjections of ethanol into the dorsolateral PFC(also termed as motor cortex area 1 in Paxinos and Watson's atlas of The Rat Brain. Elsevier Academic Press, Amsterdam, 2005) failed to do so, suggesting regional selectivity of the effects of ethanol. Moreover, bilateral microinjections of ethanol didnot change the expression of either pro-apoptotic(caspase-3 and Bax) or anti-apoptotic(Bcl-2) proteins, suggesting that the dose was safe and validating the method used in the current study. To determine whether c-aminobutyric acid A(GABA_A) receptors are involved in mediating the ethanol effects, muscimol, a selective GABA_Areceptor agonist, or bicuculline, a selective GABA_A receptor antagonist, was administered alone or co-administered with ethanol through the same route into the bilateral m PFC. The results showed that muscimol mimicked the effects of ethanol while bicuculline completely reversed the effects of ethanol and muscimol. In conclusion, ethanol increases mechanical pain sensitivity through activation of GABA_A receptors in the m PFC of rats.
文摘AIM:To study the effects of Helicobacter pylori(H. pylori)tumor necrosis factor-α(TNF)inducing protein (Tip-α)on cytokine expression and its mechanism. METHODS:We cloned Tip-αfrom the H.pylori strain 26695,transformed Escherichia coli with an expression plasmid,and then confirmed the expression product by Western blotting.Using different concentrations of Tip-αthat affected SGC7901 and GES-1 cells at different times,we assessed cytokine levels using enzyme-linked immunosorbent assay.We blocked SGC7901 cells with pyrrolidine dithiocarbamate(PDTC),a specific inhibitor of nuclear factorκB(NF-κB).We then detected interleukin(IL)-1βand TNF-αlevels in SGC7901 cells. RESULTS:Western blot analysis using an anti-Tip-α antibody revealed a 23-kDa protein,which indicated that recombinant Tip-αprotein was recombined successfully.The levels of IL-1β,IL-8 and TNF-αwere sig-nificantly higher following Tip-αinterference,whether GES-1 cells or SGC-7901 cells were used(P<0.05).However,the levels of cytokines(including IL-1β,IL-8 and TNF-α)secreted by SGC-7901 cells were greater than those secreted by GES-1 cells following treatment with Tip-αat the same concentration and for the same duration(P<0.05).After blocking NF-κB with PDTC, the cells(GES-1 cells and SGC-7901 cells)underwent interference with Tip-α.We found that IL-1βand TNF-αlevels were significantly decreased compared to cells that only underwent Tip-αinterference(P<0.05). CONCLUSION:Tip-αplays an important role in cyto-kine expression through NF-κB.

文摘One-step crystallization is one of the most energy conserving methods for glass-ceramics preparation.However,only a few kinetics studies focused on the glass-ceramics prepared by the one-step crystallization.The onestep crystallization kinetic parameters were studied using differential scanning calorimetry.The activation energy(Ea)and the Avrami parameter(n)were calculated as 152.79kJ·mol-1 and 4.39,respectively.These parameters indicate that continuous nucleation and three-dimensional crystal growth are the dominating mechanisms in the one-step crystallization process of the parent glass.The properties of the obtained glass-ceramics can be compared to the glass-ceramics prepared by the two-stage heat treatment and sintering method.This crystallization kinetics research can be used to evaluate the one-step crystallization potential of a parent glass.

文摘Objective: To explore a tumor peptide imaging agent Arginine-Arginine-Leucine (Tyr-Cys-Gly-Gly-Arg-Arg- Leu-Gly-Gly-Cys, tripeptide RRL [tRRL]) that targeted to tumor cells and tumor-derived endothelial cells (TDECs) and primarily investigate the possible relationship between tRRL and vascular endothelial growth factor receptor 2 (VEGFR-2). Methods: The tRRL sequence motif was identified as a tumor molecular marker specifically binding to TDECs. Tyrosine was conjugated to the amino terminal of RRL (Cys-Gly-Gly-Arg-Arg-Leu-Gly-Gly-Cys) for labeling with radionuclide iodine-131 (1311-tRRL). The uptake ability and molecular binding of tRRL to tumor cells and angiogenic endothelium were studied using flow cytometry and radioactivity counter in vitro. Whether VEGFR-2 is the binging site of tRRL was investigated. Biodistribution and single-photon emission computed tomography (SPECT) imaging of 131-tRRL were used to evaluate the effectiveness of this new imaging agent to visualize varied tumor xenografts in nude mice. Results: In vitro cellular uptake experiments revealed that tRRL could not only adhere to tumor angiogenic endothelial cells but also largely accumulate in malignant tumor cells. VEGFR-2, which is highly expressed on TDECs, was probably not the solely binding ligand for tRRL targeted to tumor angiogenic endothelium, 131-tRRL mainly accumulated in tumors in vivo, not other organs at 24 h after injection. SPECT imaging with 131-tRRL clearly visualized tumors in nude mice, especially at 24 h. 文摘Heavy metal containing pickling sludge(PS) is one of the by-products of the stainless-steel-making industry,which has been considered hazardous due to contained chromium and nickel.Traditional methods of PS disposing are landfill and cement solidification.This research is aimed at disposing PS by solidification/stabilization and reusing it as a nucleation agent of glass–ceramics.The crystallization behavior and the properties of a glass in the CaO–MgO–SiO2–Al2O3 system were studied by considering PS as the nucleation agent.Experimental results confirm that introducing 14 wt% PS as the nucleation agent of glass–ceramics can decrease crystallization temperature by 110.8 °C,refine the grain size by forming isometric crystals with size of 2 lm,enhance Vickers hardness by 2690 MPa and decrease water absorption from(1.21 ± 0.10) wt% to(0.04 ± 0.01) wt%.Therefore,it is reasonable to conclude that PS can be utilized as a nucleation agent to improve the crystallization and mechanical properties of the glass–ceramics.The testing results of US EPA toxicity characteristic leaching procedure(TCLP)confirm the safety of this reusing method.

文摘Background:Pulmonary tuberculosis(PTB,both smear positive and smear negative)is an airborne infectious disease of major public health concern in China and other parts of the world where PTB endemicity is reported.This study aims at identifying PTB spatio-temporal clusters and associated risk factors in Zhaotong prefecture-level city,located in southwest China,where the PTB notification rate was higher than the average rate in the entire country.Methods:Space-time scan statistics were carried out using PTB registered data in the nationwide TB online registration system from 2011 to 2015,to identify spatial clusters.PTB patients diagnosed between October 2015 and February 2016 were selected and a structured questionnaire was administered to collect a set of variables that includes socio-economic status,behavioural characteristics,local environmental and biological characteristics.Based on the discovery of detailed town-level spatio-temporal PTB clusters,we divided selected subjects into two groups including the cases that resides within and outside identified clusters.Then,logistic regression analysis was applied comparing the results of variables between the two groups.Results:A total of 1508 subjects consented and participated in the survey.Clusters for PTB cases were identified in 38 towns distributed over south-western Zhaotong.Logistic regression analysis showed that history of chronic bronchitis(OR=3.683,95%CI:2.180-6.223),living in an urban area(OR=5.876,95%CI:2.381-14.502)and using coal as the main fuel(OR=9.356,95%CI:5.620-15.576)were independently associated with clustering.While,not smoking(OR=0.340,95%CI:0.137-0.843)is the protection factor of spatial clustering.Conclusions:We found PTB specially clustered in south-western Zhaotong.The strong associated factors influencing the PTB spatial cluster including:the history of chronic bronchitis,living in the urban area,smoking and the use of coal as the main fuel for cooking and heating.Therefore,efforts should be made to curtail these associated factors.

文摘Objective: To ascertain the influence of light intensity and water content of medium on the total dendrobine of Dendrobium nobile(D. nobile).Method: The principal component analysis combined with total dendrobine accumulation was conducted to assess the yield and quality of D. nobile in all treatments. In the experiment, D. nobile plants were cultivated in greenhouse as tested materials, and complete test of 9 treatments was adopted with relative light intensities 75.02%, 39.74%,29.93% and relative water content of medium 50%, 65%, 80%. The plants were treated in June and harvested till December. Indexes including agronomic traits, fresh weight and dry weight of stem and leaf, ash content, extract, and dendrobine were measured.Results: Under the light intensity treatments of 75.02% with 50%, 65%, 80% water content of medium, the basal stems of plants were comparatively thicker with more leaves, and the fresh weight and dry weight of stems and leaves were significantly higher than other 6 treatments.Leaves in all treatments contained dendrobine. Under the light intensity treatments of 75.02%with 50%, 65%, 80% water content of medium, dendrobine content of leaves was lower while dendrobine contents of other treatments were more than 0.60%. After comprehensive assessment through the principal component analysis and total dendrobine accumulation, the results showed that 3 treatments with relative light intensity of 75.02% ranked the top three.Conclusions: In brief, the moderately strong light intensity and water content of medium from low to medium can facilitate the growth and yield of D. nobile plants, while light intensity from moderately weak to weak can enhance the dendrobine content.

文摘Background:Acute myeloid leukemia(AML)is a common type of hematological malignancy in elderly people.Geriatricians developed comprehensive geriatric assessment(CGA)methods for elderly patients;however,the tools used for CGA in AML are not uniform.Thus,we aimed to validate the instrumental activities of daily living(IADL)scales,age,comorbidities(Charlson Comorbidity Index),and albumin(IACA)index,which is a new tool for CGA,in elderly patients with AML.Methods:Patients aged>60 years who had been diagnosed with AML were screened for eligibility.Among the IACA low-,intermediate-,and high-risk groups,continuous variables were compared using the Mann-Whitney U test,and categorical variables were compared using/2 and Fisher exact tests.In addition,probabilities of overall survival(OS)were estimated using the Kaplan-Meier method.Results:A total of 21,34,and 6 patients were categorized into IACA low-risk(0 point),intermediate-risk(1-2 points),and high-risk(>3 points)groups,respectively.The rates of relapse/progression-related mortality were 23.8%,58.8%,and 100.0%in the IACA low-,intermediate-,and high-risk groups,respectively(χ^2=12.81,P<0.001).The 2-year probabilities of OS were 47.7%(95%confidence interval[CI]22.8%-72.6%)and 20.2%(95%CI 5.9%-34.5%)in the IACA low-and intermediate-risk groups,respectively(χ^2=5.99,P=0.014),which were significantly higher than those in the high-risk group(low-risk[47.7%(95%CI 22.8%-72.6%)]vs.high-risk[0],/=20.80,P<0.001;intermediate-risk[20.2%(95%CI 5.9%-34.5%)]vs.high-risk[0],χ^2=7.56,P=0.006;respectively).In the IACA low-risk group,the 2-year probability of OS in patients receiving induction chemotherapy(50.8%[95%CI 24.1%-77.5%])was significantly higher than that in those receiving best supportive care(0,χ^2=25.74,P<0.001).Conclusion:We suggest that the IACA index might be a simple and effective tool for comprehensive geriatric assessment in elderly AML patients.
