Nd^(3+)-doped tellurite glasses are promising materials for thermometers based on the fluorescence intensity ratio(FIR)technique.Nevertheless,at high Nd^(3+)concentrations,energy transfer(ET)processes such as optical reabsorption and cross-relaxation can affect the Nd^(3+)emission,which has been little explored in the literature.Therefore,the present work investigated the use of Nd^(3+)-doped tellurite glass(samples doped with Nd^(3+)at 0.2 mol%,0.5 mol%,2.0 mol%,and 4.0 mol%)in fluorescence thermometers,in the temperature range from 299 to 371 K.The results indicate a strong dependence of the FIR parameters on the Nd^(3+)concentration,due to changes in the emission band profiles caused by optical reabsorption of the Nd^(3+)emissions and cross-relaxation processes.A decrease of the relative sensitivity of the ratio^(4)F_(5/2)→^(4)I_(9/2)/^(4)F_(3/2)→^(4)I_(9/2)is observed for samples doped with higher amounts of Nd^(3+).The maximum relative sensitivity at 299 K is 3.00%/K,which is the highest value among the reported Nd^(3+)ions. 展开更多

Inhibition of guanosine triphosphate(GTP) and cytidine triphosphate(CTP) biosynthetic pathways induces cells to assemble rod/ring(RR) structures,also named cytoophidia,which consist of the enzymes cytidine triphosphate synthase(CTPS) and inosine-50-monophosphate dehydrogenase 2(IMPDH2).We aim to explore the interaction of CTPS and IMPDH2 in the generation of RR structures.He La and COS-7 cells were cultured in normal conditions or in the presence of 6-diazo-5-oxo-L-norleucine(DON),ribavirin,or mycophenolic acid(MPA).Over 90% of DON-treated cells presented RR structures.In He La cells,35% of the RR structures were positive for IMPDH2 alone,26% were CTPS alone,and 31% were IMPDH2/CTPS mixed,while in COS-7 cells,42% of RR were IMPDH2 alone,41% were CTPS alone,and 10% were IMPDH2/CTPS mixed.Ribavirin and MPA treatments induced only IMPDH2-based RR.Cells were also transfected with an N-terminal hemagglutinin(NHA)-tagged CTPS1 construct.Over 95% of NHA-CTPS1 transfected cells with DON treatment presented IMPDH2-based RR and almost 100% presented CTPS1-based RR;when treated with ribavirin,over 94% of transfected cells presented IMPDH2-based RR and 37% presented CTPS1-based RR,whereas 2% of untreated transfected cells presented IMPDH2-based RR and 28% presented CTPS1-based RR.These results may help in understanding the relationship between CTP and GTP biosynthetic pathways,especially concerning the formation of filamentous RR structures. 展开更多

The present study aims to demonstrate the chemical composition of the ethanolic extract of Sarcomphalus joazeiro bark and to evaluate its antibacterial and antibiofilm activity.For that,the ethanolic extract of the stem bark was prepared,and chemical characterization was carried out in UPLC-ESI-QToF-MSE.Minimum inhibitory concentration and antibiotic modification test were performed by the microdilution method.Formation and antiformation assays were evaluated by the crystal violet method.In the chemical characterization,the presence of 24 peaks was observed.Antibacterial activity showed minimum inhibitory concentration≥2048μg/mL.In the evaluations of the potentiating activity,synergism was the most evident.In the formation analysis,it was observed that most strains showed moderate biofilm formation.It was concluded that saponin derivatives are the major secondary metabolites in the analyzed extract and that it presents relevant synergism with the antibiotics studied and a significant ability to inhibit bacterial biofilm formation. 展开更多

Ten different casing soils were collected from two soils at two depths (0.2 and 2.0 m below soil surface) to examine the relationships between the physical properties of non-axenic casing soil and yield, number and weight of the medicinal mushroom Agamcua blaaei ss. Heinemann. The results showed that soil clay content and bulk density were negatively correlated with the mushroom yield, respectively, but soil silt content and water-holding capacity were found to be positively correlated with the yield. The number of mushrooms was negatively correlated with soil water-holding capacity but positively correlated with soil clay, bulk density and porosity. The weight of mushroom was positively correlated with the content of soil fine sand and negatively correlated with the contents of soil coarse sand, total sand and clay. Neither soil depth nor different soil combinations affected the yield and number of mushrooms, but the mushroom weight was affected by the soil combinations and soil depth, so interplay in the fructification process with the physical characteristics of casing is complicated. 展开更多
