引力波是爱因斯坦最重要的预言之一,是检验广义相对论正确性的重要工具.引力波理论及其探测一直以来都是理论物理学家和实验物理学家感兴趣的研究领域.事实上,随着科学技术的发展,人类已经具备了建造极度灵敏的地面探测器以及空间探测...引力波是爱因斯坦最重要的预言之一,是检验广义相对论正确性的重要工具.引力波理论及其探测一直以来都是理论物理学家和实验物理学家感兴趣的研究领域.事实上,随着科学技术的发展,人类已经具备了建造极度灵敏的地面探测器以及空间探测器的能力,直接探测引力波已经成为现实.2015年9月14日,LIGO首次直接探测到引力波,该信号源自一次双黑洞并合事件,自此人类进入引力波常规化探测阶段,终于拉开了引力波天文学时代的序幕.地面引力波探测器最主要的波源是处于旋近和并合阶段的致密双星.如果在探测到这些波源所辐射出的引力波信号的同时,又能观测到波源对应的电磁波信号,那么引力波信号和电磁波信号可以相互补充,形成新的观测模式.然而单个地面引力波探测器很难准确地探测引力波信号,也不能进行波源精确定位,将多个探测器联网组合,这样既能准确探测引力波信号,又能大幅提高引力波波源的定位精度.本文中,首先介绍探测器联网对引力波信号GW150914源的定位情况,然后介绍了两种最常用的估计定位精度的方法,即马尔科夫链蒙特卡罗(Markov Chain Monte Carlo,MCMC)的方法和解析的方法.最后,选用一种解析的方法讨论未来中国引力波探测器与日本及澳大利亚所组成的探测器网络的定位精度,并给出了中国的较优台址.最后,还讨论了中国加入世界引力波探测器网络行列对引力波波源定位的贡献.展开更多
Atherosclerosis(AS) is a chronic inflammatory disease associated with high morbidity and mortality. The incidence of AS is increasing in the last decades. So development of safe and effective therapeutics for treating...Atherosclerosis(AS) is a chronic inflammatory disease associated with high morbidity and mortality. The incidence of AS is increasing in the last decades. So development of safe and effective therapeutics for treating AS has become prominently important. Although there are numerous chemical drugs available for treating AS, some drugs are not effective and some have serious side effects. Traditional Chinese medicine(TCM) has a long history for the prevention and treatment of AS due to its less side effects and superior efficacy. This paper describes the effectiveness and underlying mechanisms for prevention and treatment of AS by TCM or its active components. Some TCM, e.g. XuemaiNing, Tongxinluo and Salvia miltiorrhiza have been reported to have cardio-protective effect. Some active components of TCM, e.g. saikosaponin-A, kuwanon G, luteolin and β-elemene have been isolated from various TCM and demonstrated to have beneficial effects on prevention and treatment of AS.展开更多

A novel schinortriterpenoid(SNT),schinensilactone A(1),characterized by a unique 7,8-seco-1,8-cyclo-schisanartane scaffold,was isolated from the leaves of Schisandra chinensis(Turcz.)Baill,together with a new SNT(schi...A novel schinortriterpenoid(SNT),schinensilactone A(1),characterized by a unique 7,8-seco-1,8-cyclo-schisanartane scaffold,was isolated from the leaves of Schisandra chinensis(Turcz.)Baill,together with a new SNT(schinensilactone B,2)and two known(3,4).Their structures were elucidated using spectroscopy and X-ray diffraction.Furthermore,a hypothetical biosynthetic pathway for 1 was postulated due to its novel carbon skeleton.In addition,1 exhibited significant anti-proliferative activity against Caco-2 cells originat-ing from five different tumor cell lines,and its preliminary mechanism of action was investigated with respect to the expression of apoptosis-related proteins,including P53,Caspase 3,Bax,PUMA,and Bcl2.These biological activities would further our understanding of the food function of Schisandra chinensis leaves.展开更多
A rapid and high selective ultra-performance liquid chromatography(UPLC)with tandem mass spectrometry method for simultaneous determination of six compounds including albiflorin,paeoniflorin,picroside I,picroside II,s...A rapid and high selective ultra-performance liquid chromatography(UPLC)with tandem mass spectrometry method for simultaneous determination of six compounds including albiflorin,paeoniflorin,picroside I,picroside II,saikosaponin A,and saikosaponin D in rat plasma was developed and validated using butyl p-hydroxybenzoate as an internal standard.One-step direct protein precipitation with acetonitrile was used to extract the compounds from the rat plasma samples.Chromatographic separation was achieved using an ACQUITY UPLC BEH C18 column(100 mm×2.1 mm,1.7μm)at a flow rate of 0.4 m L/min,using gradient mode containing 0.1%formic acid in water and acetonitrile were used as the Mobile phase A and B.Electrospray ionization in negative ion mode and multiple reaction monitoring were used to identify and quantify active components.Calibration curves showed good linearity(R^2>0.9908)over a wide concentration range for all compounds.The intra-and interday precision(relative standard deviation)ranged 2.4%–7.0%and 2.6%–8.0%,respectively.The accuracy(relative error)was from-13.0%to 13.2%at all quality control levels.The recovery ranged from 81.1%to 92.5%.The validated method was successfully applied to pharmacokinetic study in rats after oral administration of Qing Gan-Shu Yu-Fang.The results show that one can draw a conclusion that these six active ingredients can be quickly absorbed and play a pharmacodynamic role rapidly in vivo.展开更多

PCR products of about 0.5 kb were s...Given the sequence of Chs gene promoter from Arabidopsis thaliana reported in GenBank (AF248988), a pair of specific PCR primers was designed with the Primer Premier 5.0 software. PCR products of about 0.5 kb were successfully amplified with the genome DNA of A. thaliana as a DNA template and Taq polymerase as DNA polymerase. The purified PCR products were ligated to the pMD18-T vector. The sequencing result showed that the Chs promoter from A. thaliana was 531 bp long. Sequence alignment analysis based on the DNAMAN software revealed that the sequence similarity between the cloned promoter and target promoter (AF248988) was up to 100%. Online PLACE analysis indicated that the Chs promoter contained cis-elements such as TATA-box, CAAT-box, pollen-box, G-box, ACGT-containing element, R response element, Myb recognition element and TACPyAT-box. In recent years,drug development and research have gradually shifted from chemical synthesis to biopharmaceutical and natural drugs.Natural medicines,such as traditional Chinese medicine,have been among the first stud...In recent years,drug development and research have gradually shifted from chemical synthesis to biopharmaceutical and natural drugs.Natural medicines,such as traditional Chinese medicine,have been among the first studied because of their long medicinal history,simplicity,and the relatively low cost of research.Among them,Xanthii Fructus(XF)is famous for the treatment of sinusitis.In this article,the achievements of research on XF from 1953 to 2020 are systematically reviewed,focusing on the aspects of chemical constituents,pharmacological effects,clinical applications,toxicity and side effects,and processing methods.To date,there have been significant advances in both the phytochemistry and pharmacology of XF.Some traditional uses have been validated and clarified in modern pharmacological studies.However,its mechanism of action in the treatment of allergic diseases has not been satisfactorily explained.Further in vitro and in vivo studies are required to rationally develop new drugs and to elucidate the therapeutic potential of XF.A comprehensive evaluation of XF and an understanding of network pharmacology are also needed.展开更多

In our investigation, a cross-introns genomic DNA gene encoding A...The last step of ethylene biosynthesis in higher plants is to convert 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene by ACC oxidase (ACO). In our investigation, a cross-introns genomic DNA gene encoding ACC oxidase, PsgACO, was isolated from Paeonia suffruticosa. It revealed that PsgACO (FJ855434) has 1281 bases, containing four exons and three introns, en- coding 312 amino acids. The four exons stretched from 1 to 105, 217 to 434, 592 to 925 and 1000 to 1281 bases. A splicing junction sequence of all the exon-introns conformed to the GT-AG rule in the cross-intron genomic DNA sequence of the ACO gene. PsgACO showed high homology to many characterized ACC oxidases both at the nucleic acid and amino acid levels. As well, twelve amino acid residues were conserved among many ACOs from other species. A phylogenetic tree analysis indicated that the amino acid of ACOs is quite conserved among the different eudicots. The phylogenetic tree showed that both the tree peony and herbaceous peony are quite isolated taxa. Bioinformatic analysis showed that the molecular weight of ACO is 35.29 kD, with a theoretical pI of 5.25. It is a non-secrete, stable hydrophilic protein, located in the cytoplasm.展开更多
