This work aims to improve the breeding efficiency of special varieties with high carotene content by combining molecular markers and agronomic traits to evaluate and screen the parents. The cluster analysis was carried out to identify and screen promising parents by utilizing phenotypic results of the agronomic and characteristic traits, and RAPD and ISSR markers of 15 parents and their offspring by group crossing. Among different parental materials, greater variations were observed in two important traits, the carotene content and the storage root yield. Negative correlation was found between the carotene content and both fresh and dried root yields. The most significant positive correlation was between the carotene content of parents and that of their offspring, with the coefficient of correlation value of 0.7932**. The relationship based on the agronomic characters of the trial materials was not in agreement with the known genealogy, while that based on the molecular marker data showed better result. Each primer amplified 13.8 bands oflSSR markers on average, in which the rate of polymorphic loci was 89.6%. 9.4 bands of RAPD markers were arnplified per primer, in which the rate of polymorphic bands was 74.46%. Large genetic variation of carotene content was found among the parents. For analyzing the genetic diversity of sweetpotato, the molecular marker methods were better than morphological traits; ISSR markers were more efficient than RAPD markers. The combination of molecular markers and agronomic trait analysis may provide valuable theoretic basis for selection of parents to breed new varieties with high carotene content.展开更多

Up to now, many problems in the shape control domain have not been solved satisfactorily, and a review on the research progress in the shape control domain can help to seek new breakthrough directions. In the past 10 years, researches and applications of shape control models, shape control means, shape detection technology, and shape con- trol system have achieved significant progress. In the aspect of shape control models, the researches in the past improve the accuracy, speed and robustness of the models. The intelligentization of shape control models should be strengthened in the future. In the aspect of the shape control means, the researches in the past focus on the roll opti- mization, mill type selection, process optimization, local strip shape control, edge drop control, and so on. In the future, more attention should be paid to the coordination control of both strip shape and other quality indexes, and the refinement of control objective should be strengthened. In the aspects of shape detection technology and shape control system, some new types of shape detection meters and shape control systems are developed and have successfully indus- trial applications. In the future, the standardization of shape detection technology and shape control system should be promoted to solve the problem of compatibility. In general,the four expected development trends of shape control for cold roiling strip in the future are intelligentization, coordi- nation, refinement, and standardization. The nonlinear vibration model of moving strip between two mills with time-dependent tension was established. Approximate solutions were obtained using the method of multiple scales. Depending on the variation of the tension, three distinct cases arise: frequency away from zero or two times the natural frequency; frequency close to zero; frequency close to two times the natural frequency. For frequency close to zero and away from zero and two times the natural frequency, the system is always stable. For frequency close to two times the natural frequency, the stability is analyzed respectively when the trivial solution exists and the nontrivial solution exists. Numerical simulation was made on some 1660 mm tandem rolling mill, and the stable regions and unstable regions for parametric resonance are determined with different cases. The rolling speed and the thickness of strip have strong influences on the stability of principle parametric resonances. But the distance between two mills has little influence on the stability of principle parametric resonances.展开更多

This study was to investigate the association of TNF-α gene polymorphism with pulmonary tuberculosis (TB) among patients with coal worker's pneumoconiosis (CWP). Methods A case-control study was conducted in 113 patients with confirmed CWP complicated with pulmonary TB and 113 non-TB controls with CWP. They were matched in gender, age, job, and stage of pneumoconiosis. All participants were interviewed with questionnaires and their blood specimens were collected for genetic determination with informed consent. The TNF-α gene polymorphism was determined with polymerase chain reaction of restriction fragment length polymorphism (PCR-RFLP). Frequency of genotypes was assessed for Hardy-Weinberg equilibrium by chi-square test or Fisher's exact probability. Factors influencing the association of individual susceptibility with pulmonary TB were evaluated with logistic regression analysis. Gene-environment interaction was evaluated by a multiplieative model with combined OR. All data were analyzed using SAS version 8.2 software. Results No significant difference in frequency of the TNF-α-308 genotype was found between CWP complicated with pulmonary TB and non-TB controls (2,2=5.44, P=-0.07). But difference in frequency of the TNF-α-308 A allele was identified between them (2,2-5.14, P=0.02). No significant difference in frequencies of the TNF-α-238 genotype and allele (P=0.23 and P=0.09, respectively) was found between cases and controls either, with combined (GG and AA) OR of 3.96 (95% confidence interval of 1.30-12.09) at the -308 locus of the TNF-α gene, as compared to combination of the TNF-α-238 GG and TNF-α-308 GG genotypes. Multivariate-adjusted odds ratio of the TNF-α-238 GG and TNF-α-308 GA genotypes was 1.98 (95% CI of 1.06-3.71) for risk for pulmonary TB in patients with CWP. There was a synergic展开更多

A brief overview of the available approaches based on thermo- mechanical processing technology in producing bimodal microstructure for various metallic materials is given, along with a summary of unusual mechanical properties achievable by bimodality, where focus is placed on the microstructure-mechanical properties and relevant mechanisms. However, it is difficult to discover the eloquent areas intraoperativ...Background The goal of surgery in the treatment of intrinsic cerebral tumors is to resect the maximum tumor volume,and to spare the eloquent areas. However, it is difficult to discover the eloquent areas intraoperatively due to individual anatomo-functional variability both for sensori-motor and language functions. Consequently, the surgery of intrinsic cerebral tumors frequently results in poor extent of resection or permanent postoperative deficits, or both, and remains a difficult problem for neurosurgeons.Methods From January 2003 to January 2010, 112 patients with neuroepithelial tumors in/close to the eloquent areas were operated on under awake anesthesia with the intraoperative direct electrical stimulation for functional mapping of the eloquent areas. The extent of the tumors was verified by intraoperative ultrasonography. The maximal resection of the tumors and minimal damage of the eloquent areas were the surgical goal of all patients.Results Totally 356 cortical sites in 99 patients were detected for motor response by intraoperative direct electrical stimulation, 50 sites in 16 patients for sensory, 72 sites in 48 patients for language. Sixty-six patients (58.9%) achieved total resection, 34 (30.4%) subtotal and 12 (10.7%) partial. Fifty-eight patients (51.8%) had no postoperative deficit, while 37 patients (33.0%) had transitory postoperative paralysis, 26 patients (23.2%) with transitory postoperative language disturbance and 3 patients (2.7%) with permanent neurological deficits. No patient complained of pain recollection following operation.Conclusions Awake anesthesia, intraoperative direct electrical stimulation and ultrasonography are three core techniques for the resection of intrinsic cerebral tumors near the eloquent areas. This new concept allows an improvement in the quality of surgery for neuroepithelial tumors in/adjacent to eloquent areas.展开更多
Sepsis is defined as life-threatening organ dysfunction caused by a dysregulated host response to infection.The heart is one of the most important oxygen delivery organs,and dysfunction significantly increases the mor...Sepsis is defined as life-threatening organ dysfunction caused by a dysregulated host response to infection.The heart is one of the most important oxygen delivery organs,and dysfunction significantly increases the mortality of the body.Hence,the heart has been studied in sepsis for over half a century.However,the definition of sepsis-induced cardiomyopathy is not unified yet,and the conventional conception seems outdated:left ventricular systolic dysfunction(LVSD)along with enlargement of the left ventricle,recovering in 7 to 10 days.With the application of echocardiography in intensive care units,not only LVSD but also left ventricular diastolic dysfunction,right ventricular dysfunction,and even diffuse ventricular dysfunction have been seen.The recognition of sepsis-induced cardiomyopathy is gradually becoming complete,although our understanding of it is not deep,which has made the diagnosis and treatment stagnate.In this review,we summarize the research on sepsis-induced cardiomyopathy.Women and young people with septic cardiomyopathy are more likely to have LVSD,which may have the same mechanism as stress cardiomyopathy.Elderly people with ischemic cardiomyopathy and hypertension tend to have left ventricular diastolic dysfunction.Patients with mechanical ventilation,acute respiratory distress syndrome or other complications of increased right ventricular afterload mostly have right ventricular dysfunction.Diffuse cardiac dysfunction has also been shown in some studies;patients with mixed or co-existing cardiac dysfunction are more common,theoretically.Thus,understanding the pathophysiology of sepsis-induced cardiomyopathy from the perspective of critical care echocardiography is essential.展开更多

METHODS: Wistar rats were used in this study. A gastric mucosal injury model was established by giving the rats alcohol. Gross and microscopic appearance of gastric mucosa and ultrastructure of mitochondria were evaluated. Malondiadehyde (MDA) in gastric mucosa was measured with thiobarbituric acid. Expression of ATP synthase (ATPase) subunits 6 and 8 in mitochondrial DNA (mtDNA) was determined by reverse transcription polymerase chain reaction (RT- PCR). RESULTS:The gastric mucosal lesion index was correlated with the MDA content in gastric mucosa. As the concentration of ethanol was elevated and theexposure time to ethanol was extended, the content of MDA in gastric mucosa increased and the extent of damage aggravated. The ultrastructure of mitochondria was positively related to the ethanol concentration and exposure time. The expression of mtDNA ATPase subunits 6 and 8 mRNA declined with the increasing MDA content in gastric mucosa after gavage with ethanol. CONCLUSION: Ethanol-induced gastric mucosa injury is related to oxidative stress, which disturbs energy metabolism of mitochondria and plays a critical role in the pathogenesis of ethanol-induced gastric mucosa injury.展开更多

Results show that annual mean temperature increased at a significant rate of 0.35℃ per decade, most notably in the Lesser Khingan Mountains and in winter. Annual rainfall had no obvious linear trend, while rainy days had a significant decreasing trend. So, the rain intensity increased. High-temperature days had a weak increasing trend, and low-temperature days and cold wave showed significant decreasing trends with rates of 3.9 d per decade and -0.64 times per decade, respectively. Frequency and spatial scope of low-temperature hazard reduced significantly. Warm days and warm nights significantly increased at 1.0 and 2.4 d per decade, while cold days and cold nights decreased significantly at -1.8 and -4.1 d per decade, respectively. The nighttime warming rate was much higher than that for daytime, indicating that nighttime warming had a greater contribution to the overall warming trend than daytime warming. The annual mean wind speed, gale days, and sunshine duration had significant decreasing trends at rates of-0.21 m s-1 per decade, -4.0 d per decade and -43.3 h per decade, respectively. The snow cover onset dates postponed at a rate of 1.2 d per decade, and the snow cover end date advanced at 1.5 d per decade, which leads to shorter snow cover duration by -2.7 d per decade. Meanwhile, the maximum snow depth decreased at -0.52 cm per decade. In addition, the snow cover duration shows a higher correlation with precipitation than with temperature, which suggests that precipitation plays a more important role in maintaining snow cover duration than temperature.展开更多

Renal replacement therapy (RRT) is a treatment for severe AKI;however, the time of initiation of RRT and factors that affect the recovery of kidney function remains unclear. This study was to explore whether early initiation of RRT treatment for fluid management to reduce central venous pressure (CVP) can help to improve patients5 kidney function recovery. Methods: A retrospective analysis of septic patients who had received RRT treatment was conducted. Patients received RRT either within 12 h after they met the diagnostic criteria of renal failure (early initiation) or after a delay of 48 h if renal recovery had not occurred (delayed initiation). Parameters such as patients5 renal function recovery at discharge, fluid balance, and levels of CVP were assessed. Results: A total of 141 patients were eligible for enrolment: 40.4% of the patients were in the early initiation group (57 of 141 patients), and 59.6% were in the delayed initiation group (84 of 141 patients). There were no significant differences in the characteristics at baseline between the two groups, and there were no differences in 28-day mortality between the two groups (χ^2 = 2.142, P = 0.143);however, there was a significant difference in the recovery rate of renal function between the two groups at discharge (χ^2 = 4.730, P < 0.001). More importantly, early initiation of RRT treatment and dehydration to reduce CVP are more conducive to the recovery of renal function in patients with AKI. Conclusion: Compared with those who received delayed initiation RRT, patients who received early-initiation RRT for dehydration to reduce CVP have enhanced kidney function recovery.展开更多
AIM:To study the relation between higher-order aberrations(HOAs)and contrast sensitivity(CS)in normal eyes among a population of laser in situ keratomileusis(LASIK)candidates.METHODS:In 6629 eyes of 3315 LASIK...AIM:To study the relation between higher-order aberrations(HOAs)and contrast sensitivity(CS)in normal eyes among a population of laser in situ keratomileusis(LASIK)candidates.METHODS:In 6629 eyes of 3315 LASIK candidates,CS were measured under dark environment at the spatial frequencies of 1.5,3,6,12 and 18 cycles per degree(c/d),respectively,using an Optec 6500 visual function tester.Meanwhile,ocular HOAs were measured for a 6.0 mm pupil with a Hartmann-Shack wavefront analyzer.RESULTS:In the study,the subjects with an average spherical equivalent of-4.86±2.07 D were included.HOAs decreased from the third to the sixth order aberrations with predominant aberrations of third-order coma,trefoil and fourth-order spherical aberration.At low and moderate spatial frequencies,CS was negatively correlated with the third-order coma and trefoil aberrations,and decreased with increasing Z31,but increased with increasing Z3-3and Z5-1.At high spatial frequencies,CS decreased with increasing Z3-3and increased with increasing Z5-1.CONCLUSION:At a large pupil size of 6.0 mm,the thirdorder aberrations,but not the total aberrations,are the main factors affecting CS.Vertical coma is negatively correlated with CS.展开更多
Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Neverth...Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Nevertheless,many patients failed to respond to anti-PD-1/PD-L1 treatment,so it is necessary to seek an alternative strategy for traditional PD-1/PD-L1 targeting immunotherapy.Here with the data from The Cancer Genome Atlas(TCGA)and our in-house tissue library,PD-L1 expression was found to be positively correlated with the expression of ubiquitin-specific processing protease 7(USP7)in GC.Furthermore,USP7 directly interacted with PD-L1 in order to stabilize it,Gastric cancer;Immunosuppression;Cancer biologywhile abrogation of USP7 attenuated PD-L1/PD-1 interaction and sensitized cancer cells to T cell killing in vitro and in vivo.Besides,USP7 inhibitor suppressed GC cells proliferation by stabilizing P53 in vitro and in vivo.Collectively,our findings indicate that in addition to inhibiting cancer cells proliferation,USP7 inhibitor can also downregulate PD-L1 expression to enhance anti-tumor immune response simultaneously.Hence,these data posit USP7 inhibitor as an anti-proliferation agent as well as a novel therapeutic agent in PD-L1/PD-1 blockade strategy that can promote the immune response of the tumor.展开更多

There- fore, four-high rolling mill vibrations take place in six degree of freedom (DOF) leading to spatial behaviors invol- ving vertical, horizontal, axial, torsional, cross and swinging vibration modes resulting in complex relative motions between the rolls. Two numerical methods, modified Riccati-transfer matrix method (Riccati-TMM) and finite ele- ment method (FEM), are presented to analyze a spatial vibration characteristic of two four-high rolling mills with different stability. The natural frequency and mode shape of four-high rolling mills are obtained, and the clearance has a great effect on natural frequency and mode shape. In addition, field testing experiment is also conducted to measure natural frequency by power spectrum analysis of rolling mill vibration. Experimental results basically agree with those calculated by Riccati-TMM and FEM, which means that the Riccati-TMM and the FEM can be used for analysis of spatial vibration of four-high rolling mill. Meanwhile, the spatial vibration shows more compound vibra- tion behaviors and the negative effect of horizontal, vertical, cross and swinging vibration modes are effectively con- trolled after redesign of rolling mill. These advantages have a great significance for the rolling mill to be operated with a much higher rolling speed and improved yield of products.展开更多

文摘Sweetpotato varieties with high carotene content have high value for human health. This work aims to improve the breeding efficiency of special varieties with high carotene content by combining molecular markers and agronomic traits to evaluate and screen the parents. The cluster analysis was carried out to identify and screen promising parents by utilizing phenotypic results of the agronomic and characteristic traits, and RAPD and ISSR markers of 15 parents and their offspring by group crossing. Among different parental materials, greater variations were observed in two important traits, the carotene content and the storage root yield. Negative correlation was found between the carotene content and both fresh and dried root yields. The most significant positive correlation was between the carotene content of parents and that of their offspring, with the coefficient of correlation value of 0.7932**. The relationship based on the agronomic characters of the trial materials was not in agreement with the known genealogy, while that based on the molecular marker data showed better result. Each primer amplified 13.8 bands oflSSR markers on average, in which the rate of polymorphic loci was 89.6%. 9.4 bands of RAPD markers were arnplified per primer, in which the rate of polymorphic bands was 74.46%. Large genetic variation of carotene content was found among the parents. For analyzing the genetic diversity of sweetpotato, the molecular marker methods were better than morphological traits; ISSR markers were more efficient than RAPD markers. The combination of molecular markers and agronomic trait analysis may provide valuable theoretic basis for selection of parents to breed new varieties with high carotene content.

文摘The grain oriented silicon strip was rolled by cross shear rolling (CSR) and then annealed to manufacture non-oriented thin silicon strip of high quality. The recrystallization of rolled grain-oriented silicon steel into non-oriented silicon steel was studied. For this purpose, CSR is better than conventional rolling, and the higher the mismatched speed rate is, the better the properties of the non-oriented thin silicon strip are. The optimum annealing schedule is heating at 1 000 ℃ for 1 h in pure hydrogen atmosphere added with H 2S of 0.001 0 %.

文摘Shape is an important quality index of cold rolling strip. Up to now, many problems in the shape control domain have not been solved satisfactorily, and a review on the research progress in the shape control domain can help to seek new breakthrough directions. In the past 10 years, researches and applications of shape control models, shape control means, shape detection technology, and shape con- trol system have achieved significant progress. In the aspect of shape control models, the researches in the past improve the accuracy, speed and robustness of the models. The intelligentization of shape control models should be strengthened in the future. In the aspect of the shape control means, the researches in the past focus on the roll opti- mization, mill type selection, process optimization, local strip shape control, edge drop control, and so on. In the future, more attention should be paid to the coordination control of both strip shape and other quality indexes, and the refinement of control objective should be strengthened. In the aspects of shape detection technology and shape control system, some new types of shape detection meters and shape control systems are developed and have successfully indus- trial applications. In the future, the standardization of shape detection technology and shape control system should be promoted to solve the problem of compatibility. In general,the four expected development trends of shape control for cold roiling strip in the future are intelligentization, coordi- nation, refinement, and standardization. The proposed research provides new breakthrough directions for improv- ing shape quality.

文摘AIM:To investigate whether electroacupuncture(EA)at Zusanli(ST36)prevents intestinal barrier and remote organ dysfunction following prolonged hemorrhagic shock through a vagus anti-inflammatory mechanism.METHODS:Sprague-Dawley rats were subjected to about 45%of total blood volume loss followed by delayed fluid replacement(DFR)with Ringer lactate 3h after hemorrhage.In a first study,rats were randomly divided into six groups:(1)EAN:EA at non-channel acupoints followed by DFR;(2)EA:EA at ST36 after hemorrhage followed by DFR;(3)VGX/EA:vagotomy(VGX)before EA at ST36 and DFR;(4)VGX/EAN:VGX before EAN and DFR;(5)α-bungarotoxin(α-BGT)/EA:intraperitoneal injection ofα-BGT before hemorrhage,followed by EA at ST36 and DFR;and(6)α-BGT/EAN group:α-BGT injection before hemorrhage followed by EAN and DFR.Survival and mean arterial pressure(MAP)were monitored over the next 12 h.In a second study,with the same grouping and treatment,cytokine levels in plasma and intestine,organ parameters,gut injury score,gut permeability to 4 kDa FITC-dextran,and expression and distribution of tight junction protein ZO-1 were evaluated.RESULTS:MAP was significantly lowered after blood loss;EA at ST36 improved the blood pressure at corresponding time points 3 and 12 h after hemorrhage.EA at ST36 reduced tumor necrosis factor-αand interleukin(IL)-6 levels in both plasma and intestine homogenates after blood loss and DFR,while vagotomy or intraperitoneal injection ofα-BGT before EA at ST36reversed its anti-inflammatory effects,and EA at ST36did not influence IL-10 levels in plasma and intestine.EA at ST36 alleviated the injury of intestinal villus,the gut injury score being significantly lower than that of EAN group(1.85±0.33 vs 3.78±0.59,P<0.05).EA at ST36 decreased intestinal permeability to FITCdextran compared with EAN group(856.95 ng/mL±90.65 ng/mL vs 2305.62 ng/mL±278.32 ng/mL,P<0.05).EA at ST36 significantly preserved ZO-1 protein expression and localization at 12 h after hemorrhage.However,EA at non-channel acupoints had no suc

文摘AIM:To investigate whether electroacupuncture ST36 activates enteric glial cells,and alleviates gut inflammation and barrier dysfunction following hemorrhagic shock.METHODS:Sprague-Dawley rats were subjected to approximately 45% total blood loss and randomly divided into seven groups:(1) sham:cannulation,but no hemorrhage;(2) subjected to hemorrhagic shock(HS);(3) electroacupuncture(EA) ST36 after hemorrhage;(4) vagotomy(VGX)/EA:VGX before hemorrhage,then EA ST36;(5) VGX:VGX before hemorrhage;(6) a-bungarotoxin(BGT)/EA:intraperitoneal injection of a-BGT before hemorrhage,then EA ST36; and(7) a-BGT group:a-BGT injection before hemorrhage.Morphological changes in enteric glial cells(EGCs) were observed by immunofluorescence,and glial fibrillary acidic protein(GFAP; a protein marker of enteric glial activation) was evaluated using reverse transcriptase polymerase chain reaction and western blot analysis.Intestinal cytokine levels,gut permeability to 4-k Da fluorescein isothiocyanate(FITC)-dextran,and the expression and distribution of tight junction protein zona occludens(ZO)-1 were also determined.RESULTS:EGCs were distorted following hemorrhage and showed morphological abnormalities.EA ST36 attenuated the morphological changes in EGCs at 6 h,as compared with the VGX,a-BGT and HS groups.EA ST36 increased GFAP expression to a greater degree than in the other groups.EA ST36 decreased intestinal permeability to FITC-dextran(760.5 ± 96.43 ng/m L vs 2466.7 ± 131.60 ng/m L,P < 0.05) and preserved ZO-1 protein expression and localization at 6 h afterhemorrhage compared with the HS group.However,abdominal VGX and a-BGT treatment weakened or eliminated the effects of EA ST36.EA ST36 reduced tumor necrosis factor-a levels in intestinal homogenates after blood loss,while vagotomy or intraperitoneal injection of a-BGT before EA ST36 abolished its antiinflammatory effects.CONCLUSION:EA ST36 attenuates hemorrhageinduced intestinal inflammatory insult,and protects the intestinal barrier integrity,partly via activation of EGCs.

文摘The strip with a time-dependent tension moves, namely a harmonically varying tension about a constant initial tension. The nonlinear vibration model of moving strip between two mills with time-dependent tension was established. Approximate solutions were obtained using the method of multiple scales. Depending on the variation of the tension, three distinct cases arise: frequency away from zero or two times the natural frequency; frequency close to zero; frequency close to two times the natural frequency. For frequency close to zero and away from zero and two times the natural frequency, the system is always stable. For frequency close to two times the natural frequency, the stability is analyzed respectively when the trivial solution exists and the nontrivial solution exists. Numerical simulation was made on some 1660 mm tandem rolling mill, and the stable regions and unstable regions for parametric resonance are determined with different cases. The rolling speed and the thickness of strip have strong influences on the stability of principle parametric resonances. But the distance between two mills has little influence on the stability of principle parametric resonances.

文摘Some unavoidable factors in the process of cold strip shape measurement interfere with the shape meter, so the shape measuring results cannot reflect the true shape of the strip and the measuring precision is low. The influ- ences of the measuring error of the strip edges, the transverse temperature difference of the strip, the deflection of shape detection roller, and the shape of the strip coil on the shape measuring results were analyzed in detail, and the corresponding compensation models were established. The simulation calculation and analysis were carried out on a cold strip mill, and a number of disciplinarian cognitions were obtained.

文摘Objectives Tumor necrosis factor-α (TNF-α) may play an important role in host's immune response to mycobacterium tuberculosis (M. tuberculosis) infection. This study was to investigate the association of TNF-α gene polymorphism with pulmonary tuberculosis (TB) among patients with coal worker's pneumoconiosis (CWP). Methods A case-control study was conducted in 113 patients with confirmed CWP complicated with pulmonary TB and 113 non-TB controls with CWP. They were matched in gender, age, job, and stage of pneumoconiosis. All participants were interviewed with questionnaires and their blood specimens were collected for genetic determination with informed consent. The TNF-α gene polymorphism was determined with polymerase chain reaction of restriction fragment length polymorphism (PCR-RFLP). Frequency of genotypes was assessed for Hardy-Weinberg equilibrium by chi-square test or Fisher's exact probability. Factors influencing the association of individual susceptibility with pulmonary TB were evaluated with logistic regression analysis. Gene-environment interaction was evaluated by a multiplieative model with combined OR. All data were analyzed using SAS version 8.2 software. Results No significant difference in frequency of the TNF-α-308 genotype was found between CWP complicated with pulmonary TB and non-TB controls (2,2=5.44, P=-0.07). But difference in frequency of the TNF-α-308 A allele was identified between them (2,2-5.14, P=0.02). No significant difference in frequencies of the TNF-α-238 genotype and allele (P=0.23 and P=0.09, respectively) was found between cases and controls either, with combined (GG and AA) OR of 3.96 (95% confidence interval of 1.30-12.09) at the -308 locus of the TNF-α gene, as compared to combination of the TNF-α-238 GG and TNF-α-308 GG genotypes. Multivariate-adjusted odds ratio of the TNF-α-238 GG and TNF-α-308 GA genotypes was 1.98 (95% CI of 1.06-3.71) for risk for pulmonary TB in patients with CWP. There was a synergic

文摘Introducing a bimodal grain-size distribution has been demonstrated an efficient strategy for fabricating high-strength and ductile metallic materials, where fine grains provide strength, while coarse grains enable strain hardening and hence decent ductility. Over the last decades, research activities in this area have grown enormously, including interesting results onfcc Cu, Ni and Al-Mg alloys as well as steel and Fe alloys via various thermo-mechanical processing approaches. However, investigations on bimodal Mg and other hcp metals are relatively few. A brief overview of the available approaches based on thermo- mechanical processing technology in producing bimodal microstructure for various metallic materials is given, along with a summary of unusual mechanical properties achievable by bimodality, where focus is placed on the microstructure-mechanical properties and relevant mechanisms. In addition, key factors that influencing bimodal strategies, such as compositions of starting materials and processing parameters, together with the challenges this research area facing, are identified and discussed briefly.

文摘N6-Methyladenosine(m^(6)A) is the most abundant internal modification in eukaryotic mRNA,playing critical role in various bioprocesses. Like other epigenetic modifications, m^(6)A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m^(6)A demethylases have been reported, fat mass and obesity-associated protein(FTO)and alkylation protein AlkB homolog 5(ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. In this review, we focus on the studies about structure, inhibitors development and biological function of FTO and ALKBH5.
文摘Background The goal of surgery in the treatment of intrinsic cerebral tumors is to resect the maximum tumor volume,and to spare the eloquent areas. However, it is difficult to discover the eloquent areas intraoperatively due to individual anatomo-functional variability both for sensori-motor and language functions. Consequently, the surgery of intrinsic cerebral tumors frequently results in poor extent of resection or permanent postoperative deficits, or both, and remains a difficult problem for neurosurgeons.Methods From January 2003 to January 2010, 112 patients with neuroepithelial tumors in/close to the eloquent areas were operated on under awake anesthesia with the intraoperative direct electrical stimulation for functional mapping of the eloquent areas. The extent of the tumors was verified by intraoperative ultrasonography. The maximal resection of the tumors and minimal damage of the eloquent areas were the surgical goal of all patients.Results Totally 356 cortical sites in 99 patients were detected for motor response by intraoperative direct electrical stimulation, 50 sites in 16 patients for sensory, 72 sites in 48 patients for language. Sixty-six patients (58.9%) achieved total resection, 34 (30.4%) subtotal and 12 (10.7%) partial. Fifty-eight patients (51.8%) had no postoperative deficit, while 37 patients (33.0%) had transitory postoperative paralysis, 26 patients (23.2%) with transitory postoperative language disturbance and 3 patients (2.7%) with permanent neurological deficits. No patient complained of pain recollection following operation.Conclusions Awake anesthesia, intraoperative direct electrical stimulation and ultrasonography are three core techniques for the resection of intrinsic cerebral tumors near the eloquent areas. This new concept allows an improvement in the quality of surgery for neuroepithelial tumors in/adjacent to eloquent areas.
文摘Sepsis is defined as life-threatening organ dysfunction caused by a dysregulated host response to infection.The heart is one of the most important oxygen delivery organs,and dysfunction significantly increases the mortality of the body.Hence,the heart has been studied in sepsis for over half a century.However,the definition of sepsis-induced cardiomyopathy is not unified yet,and the conventional conception seems outdated:left ventricular systolic dysfunction(LVSD)along with enlargement of the left ventricle,recovering in 7 to 10 days.With the application of echocardiography in intensive care units,not only LVSD but also left ventricular diastolic dysfunction,right ventricular dysfunction,and even diffuse ventricular dysfunction have been seen.The recognition of sepsis-induced cardiomyopathy is gradually becoming complete,although our understanding of it is not deep,which has made the diagnosis and treatment stagnate.In this review,we summarize the research on sepsis-induced cardiomyopathy.Women and young people with septic cardiomyopathy are more likely to have LVSD,which may have the same mechanism as stress cardiomyopathy.Elderly people with ischemic cardiomyopathy and hypertension tend to have left ventricular diastolic dysfunction.Patients with mechanical ventilation,acute respiratory distress syndrome or other complications of increased right ventricular afterload mostly have right ventricular dysfunction.Diffuse cardiac dysfunction has also been shown in some studies;patients with mixed or co-existing cardiac dysfunction are more common,theoretically.Thus,understanding the pathophysiology of sepsis-induced cardiomyopathy from the perspective of critical care echocardiography is essential.

文摘For the problems occurring in a least square method model, a fuzzy model, and a neural network model for flatness pattern recognition, a fuzzy neural network model for flatness pattern recognition with only three-input and three output signals was proposed with Legendre orthodoxy polynomial as basic pattern, based on fuzzy logic expert experiential knowledge and genetic-BP hybrid optimization algorithm. The model not only had definite physical meanings in its inner nodes, but also had strong self-adaptability, anti interference ability, high recognition precision, and high velocity, thereby meeting the demand of high-precision flatness control for cold strip mill and providing a convenient, practical, and novel method for flatness pattern recognition.

文摘AIM: To study the role of mitochondrial energy disorder in the pathogenesis of ethanol-induced gastric mucosa injury. METHODS: Wistar rats were used in this study. A gastric mucosal injury model was established by giving the rats alcohol. Gross and microscopic appearance of gastric mucosa and ultrastructure of mitochondria were evaluated. Malondiadehyde (MDA) in gastric mucosa was measured with thiobarbituric acid. Expression of ATP synthase (ATPase) subunits 6 and 8 in mitochondrial DNA (mtDNA) was determined by reverse transcription polymerase chain reaction (RT- PCR). RESULTS:The gastric mucosal lesion index was correlated with the MDA content in gastric mucosa. As the concentration of ethanol was elevated and theexposure time to ethanol was extended, the content of MDA in gastric mucosa increased and the extent of damage aggravated. The ultrastructure of mitochondria was positively related to the ethanol concentration and exposure time. The expression of mtDNA ATPase subunits 6 and 8 mRNA declined with the increasing MDA content in gastric mucosa after gavage with ethanol. CONCLUSION: Ethanol-induced gastric mucosa injury is related to oxidative stress, which disturbs energy metabolism of mitochondria and plays a critical role in the pathogenesis of ethanol-induced gastric mucosa injury.

文摘This study focuses on examining the characteristics of climate factors and extreme climate events in Northeast China during 1961- 2010 by using daily data from 104 stations, including surface air temperature, precipitation, wind speed, sunshine duration, and snow depth. Results show that annual mean temperature increased at a significant rate of 0.35℃ per decade, most notably in the Lesser Khingan Mountains and in winter. Annual rainfall had no obvious linear trend, while rainy days had a significant decreasing trend. So, the rain intensity increased. High-temperature days had a weak increasing trend, and low-temperature days and cold wave showed significant decreasing trends with rates of 3.9 d per decade and -0.64 times per decade, respectively. Frequency and spatial scope of low-temperature hazard reduced significantly. Warm days and warm nights significantly increased at 1.0 and 2.4 d per decade, while cold days and cold nights decreased significantly at -1.8 and -4.1 d per decade, respectively. The nighttime warming rate was much higher than that for daytime, indicating that nighttime warming had a greater contribution to the overall warming trend than daytime warming. The annual mean wind speed, gale days, and sunshine duration had significant decreasing trends at rates of-0.21 m s-1 per decade, -4.0 d per decade and -43.3 h per decade, respectively. The snow cover onset dates postponed at a rate of 1.2 d per decade, and the snow cover end date advanced at 1.5 d per decade, which leads to shorter snow cover duration by -2.7 d per decade. Meanwhile, the maximum snow depth decreased at -0.52 cm per decade. In addition, the snow cover duration shows a higher correlation with precipitation than with temperature, which suggests that precipitation plays a more important role in maintaining snow cover duration than temperature.

文摘The existing research of the flatness control for strip cold rolling mainly focuses on the calculation of the optimum adjustment of individual flatness actuator in accordance with the flatness deviation , which is used for general flatness control.As the basis of flatness control system , the efficiencies of flatness actuators provide a quantitative description to the law of flatness control.Therefore , the determination of actuator efficiency factors is crucial in flatness control.The strategies of closed loop feedback flatness control and rolling force feed-forward control were established respectively based on actuator efficiency factors.For the purpose of obtaining accurate efficiency factors matrixes of flatness actuators , a self-learning model of actuator efficiency factors was established.The precision of actuator efficiency factors can be improved continuously by the input of correlative measured flatness data.Meanwhile , the self-learning model of actuator efficiency factors permits the application of this flatness control for all possible types of actuators and every stand type.The application results show that the self-learning model is capable of obtaining good flatness.
文摘Background:Acute kidney injury (AKI) is a serious complication in critically ill patients with septic shock treated in the intensive care unit. Renal replacement therapy (RRT) is a treatment for severe AKI;however, the time of initiation of RRT and factors that affect the recovery of kidney function remains unclear. This study was to explore whether early initiation of RRT treatment for fluid management to reduce central venous pressure (CVP) can help to improve patients5 kidney function recovery. Methods: A retrospective analysis of septic patients who had received RRT treatment was conducted. Patients received RRT either within 12 h after they met the diagnostic criteria of renal failure (early initiation) or after a delay of 48 h if renal recovery had not occurred (delayed initiation). Parameters such as patients5 renal function recovery at discharge, fluid balance, and levels of CVP were assessed. Results: A total of 141 patients were eligible for enrolment: 40.4% of the patients were in the early initiation group (57 of 141 patients), and 59.6% were in the delayed initiation group (84 of 141 patients). There were no significant differences in the characteristics at baseline between the two groups, and there were no differences in 28-day mortality between the two groups (χ^2 = 2.142, P = 0.143);however, there was a significant difference in the recovery rate of renal function between the two groups at discharge (χ^2 = 4.730, P < 0.001). More importantly, early initiation of RRT treatment and dehydration to reduce CVP are more conducive to the recovery of renal function in patients with AKI. Conclusion: Compared with those who received delayed initiation RRT, patients who received early-initiation RRT for dehydration to reduce CVP have enhanced kidney function recovery.
文摘AIM:To study the relation between higher-order aberrations(HOAs)and contrast sensitivity(CS)in normal eyes among a population of laser in situ keratomileusis(LASIK)candidates.METHODS:In 6629 eyes of 3315 LASIK candidates,CS were measured under dark environment at the spatial frequencies of 1.5,3,6,12 and 18 cycles per degree(c/d),respectively,using an Optec 6500 visual function tester.Meanwhile,ocular HOAs were measured for a 6.0 mm pupil with a Hartmann-Shack wavefront analyzer.RESULTS:In the study,the subjects with an average spherical equivalent of-4.86±2.07 D were included.HOAs decreased from the third to the sixth order aberrations with predominant aberrations of third-order coma,trefoil and fourth-order spherical aberration.At low and moderate spatial frequencies,CS was negatively correlated with the third-order coma and trefoil aberrations,and decreased with increasing Z31,but increased with increasing Z3-3and Z5-1.At high spatial frequencies,CS decreased with increasing Z3-3and increased with increasing Z5-1.CONCLUSION:At a large pupil size of 6.0 mm,the thirdorder aberrations,but not the total aberrations,are the main factors affecting CS.Vertical coma is negatively correlated with CS.

文摘Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Nevertheless,many patients failed to respond to anti-PD-1/PD-L1 treatment,so it is necessary to seek an alternative strategy for traditional PD-1/PD-L1 targeting immunotherapy.Here with the data from The Cancer Genome Atlas(TCGA)and our in-house tissue library,PD-L1 expression was found to be positively correlated with the expression of ubiquitin-specific processing protease 7(USP7)in GC.Furthermore,USP7 directly interacted with PD-L1 in order to stabilize it,Gastric cancer;Immunosuppression;Cancer biologywhile abrogation of USP7 attenuated PD-L1/PD-1 interaction and sensitized cancer cells to T cell killing in vitro and in vivo.Besides,USP7 inhibitor suppressed GC cells proliferation by stabilizing P53 in vitro and in vivo.Collectively,our findings indicate that in addition to inhibiting cancer cells proliferation,USP7 inhibitor can also downregulate PD-L1 expression to enhance anti-tumor immune response simultaneously.Hence,these data posit USP7 inhibitor as an anti-proliferation agent as well as a novel therapeutic agent in PD-L1/PD-1 blockade strategy that can promote the immune response of the tumor.

文摘The rolls in contemporary four-high mills cannot be maintained parallel during the rolling process. There- fore, four-high rolling mill vibrations take place in six degree of freedom (DOF) leading to spatial behaviors invol- ving vertical, horizontal, axial, torsional, cross and swinging vibration modes resulting in complex relative motions between the rolls. Two numerical methods, modified Riccati-transfer matrix method (Riccati-TMM) and finite ele- ment method (FEM), are presented to analyze a spatial vibration characteristic of two four-high rolling mills with different stability. The natural frequency and mode shape of four-high rolling mills are obtained, and the clearance has a great effect on natural frequency and mode shape. In addition, field testing experiment is also conducted to measure natural frequency by power spectrum analysis of rolling mill vibration. Experimental results basically agree with those calculated by Riccati-TMM and FEM, which means that the Riccati-TMM and the FEM can be used for analysis of spatial vibration of four-high rolling mill. Meanwhile, the spatial vibration shows more compound vibra- tion behaviors and the negative effect of horizontal, vertical, cross and swinging vibration modes are effectively con- trolled after redesign of rolling mill. These advantages have a great significance for the rolling mill to be operated with a much higher rolling speed and improved yield of products.
