目的观察川崎病患儿冠状动脉病变的发生机理及丙种球蛋白对其防治作用。方法应用细胞培养、免疫细胞化学染色、核酸杂交和流式细胞分析方法。结果川崎病患儿血清能够强烈诱导人血管内皮细胞表达血小板源生长因子(PDGF)B链蛋白;... 目的观察川崎病患儿冠状动脉病变的发生机理及丙种球蛋白对其防治作用。方法应用细胞培养、免疫细胞化学染色、核酸杂交和流式细胞分析方法。结果川崎病患儿血清能够强烈诱导人血管内皮细胞表达血小板源生长因子(PDGF)B链蛋白;经川崎病血清作用后的内皮细胞条件培养基(ECM)亦能显著促进血管平滑肌细胞表达PDGF受体mRNA,并强烈诱导平滑肌细胞增殖。丙种球蛋白则能显著抑制川崎病血清的上述诱导作用。结论PDGF-PDGF受体途径激活参与了川崎病冠状动脉病变的形成; 展开更多

目的:以柯萨奇病毒B3(CVB3)感染体外培养的小鼠心肌细胞,探讨CVB3感染心肌细胞后对心肌细胞周期、心肌细胞凋亡和心肌细胞浆游离钙的影响。    方法:制备心肌细胞样本,设正常对照组及实验组(CVB3感染心肌细胞... 目的:以柯萨奇病毒B3(CVB3)感染体外培养的小鼠心肌细胞,探讨CVB3感染心肌细胞后对心肌细胞周期、心肌细胞凋亡和心肌细胞浆游离钙的影响。    方法:制备心肌细胞样本,设正常对照组及实验组(CVB3感染心肌细胞),流式细胞仪、钙荧光探针标记分别检测细胞周期、细胞凋亡及心肌细胞浆内游离钙。    结果:CVB3感染心肌细胞后,可显著抑制心肌细胞的分裂增殖,诱导心肌细胞凋亡;显著增高心肌细胞浆内游离钙浓度和减少细胞外钙含量。    结论:CVB3感染体外培养的心肌细胞,可能引起心肌细胞凋亡和心肌细胞浆内钙超载,初步实验提示此种变化可能是病毒性心肌炎心脏损伤的重要机制。 展开更多

目的 为探索肠道病毒性脑炎的发病机制 ,以柯萨奇病毒 (CV)B3 感染小鼠 ,观察其神经细胞内钙离子 [Ca2 +]i及脑组织N 甲基 D 天冬氨酸受体I型 (NMDAR1)mRNA表达的变化。方法(1)取新生Balb/c小鼠大脑皮质神经细胞 ,培养 12d后 ,于CVB3 ... 目的 为探索肠道病毒性脑炎的发病机制 ,以柯萨奇病毒 (CV)B3 感染小鼠 ,观察其神经细胞内钙离子 [Ca2 +]i及脑组织N 甲基 D 天冬氨酸受体I型 (NMDAR1)mRNA表达的变化。方法(1)取新生Balb/c小鼠大脑皮质神经细胞 ,培养 12d后 ,于CVB3 感染 12h(12份 )用流式细胞仪检测细胞内 [Ca2 +]i,并用原子吸收光谱法检测神经细胞培养上清液中游离钙Ca2 +含量 ;(2 )以CVB3 颅内接种小鼠 ,用NMDAR1的cDNA地高辛标记探针对感染 3d后处死的石蜡包埋脑组织切片作原位杂交检测。结果  (1)感染 12h小鼠脑神经细胞内 [Ca2 +]i值较对照组明显增高 (P <0 0 1)。感染组神经细胞培养上清液中Ca2 +含量低于对照组 (P <0 0 1)。 (2 )感染组脑组织中NMDAR1mRNA表达明显高于对照组 (P <0 0 1)。结论 CVB3 感染小鼠脑神经细胞可导致细胞内 [Ca2 +]i超载 ,且与Ca2 +内流有关。CVB3 感染又可引起小鼠脑组织NMDAR1mRNA表达增高 ,其结果可能进一步引起Ca2 展开更多

The isolated cardiac myocytes of rats were immediately infected by cosackievirus B3 (CVB3) to investigate the effects of such procedure on the cell cycle, apoptosis and intracellular ionized calcium (Ca 2+ i) of cardiac myocytes. Newborn Balb/c murine cardiac myocytes were cultivated, then infected by CVB3. Intracellular Ca 2+ i was measured by flow cytometer. The calcium in the medium for culturing cardiac myocytes was detected by using atom absorb spectrum test. It was found that CVB3 could markedly inhibit the differentiation and proliferation of the infected cardiac myocytes and induce the apoptosis. The intracellular Ca 2+ i level in the infected group was significantly higher than in the control group (P<0.01). The calcium concentration in the medium for culturing cardiac myocytes in the infected group was significantly lower than in the control group (P<0.05). It was suggested that the apoptosis and intracellular calcium overload of the CVB3-affected cardiac myocytes are likely to play an important role in the pathogenesis of viral myocarditis. 展开更多

The two groups were controlled for age. Determination of HSP(60), HSP(70) was conducted in lymphocytes of 14 cases of KD and 26 healthy children. The results were compared with those of 12 patients with febrile diseases and 10 patients with tuberculosis. Our results showed that except a significant difference in plasma HSP(70) found between acute phase and convalescent phase of KD (P<0. 01), no significant difference was found in HSP(60), HSP(70)among all groups (P>0. 05). The differences in HSP(60), HS(70), in lymphocytes were relatively obvious among all groups. The levels of HSP(60), HSP(70) in acute phase of KD were significantly higher than those in convalescent phase or in healthy controls (P<0. 01). The levels of HSP(60) in KD were significantly higher than those of patients with febrile diseases. HSP(60) of KD children was significantly lower than those of children with tuberculosis (P<0.01). The findings showed that HSP(60), HSP(70) might contribute to the pathogenesis of KD. Determination of HSP(60), HSP(70) in lymphocytes is of help in the diagnosis of KD. 展开更多
