It has been studied as a potential anticoagulative agent since it was reported to protect against the myocardial ischemic injury by inhibition of platelet function. Studies in hemostasis revealed that IHC-65 acts by its effect on platelet aggregation. However, the underlying mechanisms of its antiplatelet action are essentially unknown. We studied the effect of IHC-65 on: (1) the generation of TX A2and cAMP in rabbit platelets;(2) the generation of PG I2 from isolated perfused rabbit aorta; and (3) Ca2+ influx and intracellular Ca2+ release in rabbit platelets using radioimmunoassay and fluorescent-dye (Quin-2/AM) techniques. To study the effect of IHC-65 on intraplatelet Ca2+ release, EGTA was used to deplete extracellular Ca2+, and the net increase of free Ca2+ in the platelet was used as an index of the Ca2+ release in the platelet.展开更多
