However, the function of DEAD-box proteins in aquaculture specie...RNA helicases of the DEAD-box and related families are involved in various cellular processes including DNA replication, DNA repair, and RNA processing. However, the function of DEAD-box proteins in aquaculture species is poorly understood at molecular level. We obtained the full-length cDNA sequences of two genes encoding helicase-related proteins, Fc-vasa and Fc-PL10a, from the testes of Chinese shrimp, Fenneropenaeus chinensis. The two predicted amino acid sequences contain all the conserved motifs characterized by the DEAD-box family and several RGG repeats in the N-terminal regions. Homology and phylogenetic analyses indicate that they belong to the vasa and PLIO subfamilies. The three-dimensional structures of the two proteins were predicted with a homology modeling approach. Both core proteins consist of two tandem RecA-like domains similar to those of the DEAD-box RNA helicase. Using reverse transcription-polymerase chain reaction (RT-PCR) and real-time PCR we found that Fc-vasa was expressed specifically in the adult gonads. Transcription decreased in the ovary but increased in the testis during gonadal development. Fc-PL10a expression was widely distributed in the tissues we examined. Using in situ hybridization, we demonstrated that the Fc-vasa transcript is localized to the cytoplasm of the spermatogonia and oocytes. Thus, our results suggest that Fc-wasa plays an important role in germ-line development, and has utility as a germ cell lineage marker which will help to generate new insight into the origin and differentiation of germ cells as well as the regulation of reproduction in F. chinensis.展开更多

As an important economic macroalga, Gracilariopsis lemaneiformis has the potential to significantly affect carbon absorption and storage in wave-sheltered intertidal reef systems. However, detailed knowledge on seasonal biomass changes and carbon storage of G. lemaneiformis is lacking, especially in many small and scattered ecosystems. Considering the influence of human activities on wild distribution of G. lemaneiformis, the understanding of seasonal dynamics of an economically important species in nature is necessary. In this study, we first investigated seasonal variations in biomass, coverage area, and carbon storage during low tide from August 2011 to July 2012 in Zhanshan Bay, Qingdao, China. Furthermore, we estimated the carbon storage potential of wild G. lemaneiformis using light use efficiency (LUE). The results show that the standing biomass and coverage area changed significantly with season. However, seasonal variations in carbon content and water content were not obvious, with an average content of 35.1% and 83.64%, respectively. Moreover, carbon storage in individual months varied between 0.67 and 47.03 g C/mE, and the value of carbon storage was the highest in August and June and the lowest in February. In Zhanshan Bay, LUE of G. lemaneiformis was only 0.23%. If it is increased to the theoretical maximum (5%-6%), the carbon storage will have an increase of at least 21 times compared with the current, which suggested that carbon storage of wild G. lemaneiformis had a high enhancement potential. The results show that the concentration of t...In this study, the ability of macroalgae Gracilaria sp. of removing eutrophication factors and toxic heavy metals A1, Cr, and Zn in a closed cultivation system is reported. The results show that the concentration of the three heavy metals decreased significantly during the experimental period in an algal biomass dependent manner. The biofiltration capacity of the alga for A1, Cr, and Zn is 10.1%-72.6%, 52.5%-83.4% and 36.5%,91.7%, respectively. Using more materials resulted in stronger heavy metal removal. Additionally, the concentration of chl-a, TN, TP and DIN of water samples fxom aquariums involving large, medium, and small algal biomass cultivation increased first and then decreased during the experiment. COD value of all three groups decreased with time and displayed algal biomass dependency: more algae resulting in a greater COD value than those of less biomass. Furthermore, changes in COD reflect an obvious organic particles deprivation process of algae. This is the first report on heavy metal removal effect by Gracilaria species. The results suggest that macroalgae can be used as a biofilter for the treatment of nutrient-enriched or heavy-metal polluted water, to which an appropriate time range should be carefully determined.展开更多

Endosulfan is an organochlorine pesticide used globally to control insects and mites; however, it is a source of pollution in aquaculture as a result of runoff or accidental release. In this study, we evaluated the toxicity of endosulfan to polychaeta and its ability to improve polluted sediment. Specifically, the effects of a series of endosulfan concentrations (0, 1.25, 2.5, 5, 10, 15, and 20 mg/L) were investigated, and the results indicated that the 24-h median lethal concentration (24-h LCs0) was 55.57 mg/L, while the 48-h median lethal concentration (48-h LCs0) was 15.56 mg/L, and the safe concentration was about 1.556 mg/L. In a 30-d exposure experiment, the animal specimen could decompose endosulfan effectively while improving endosulfan-polluted aquatic sediment.展开更多

A rapid and accurate detection process for HAB algae has yet to be developed. Heterosigma akashiwo is one of the most important HABs species in China. The objective of this study was to develop an immunologic technique that can rapidly and sensitively count H. akashiwo cells. Five HABs species (Alexandrium catenella, Thalassiosira sp., Cryptomonas sp., Alexandrium tamarense and Symbiodinium sp.,) were used in this study to evaluate the analysis process we developed. A polyclonal antibody with high titers against H. akashiwo was obtained by injecting H. akashiwo cells into rabbits. Immuno-magnetic beads (IMB) were produced via conjugated polyclonal antibodies with magnetic beads and applied to isolate and count H. akashiwo cells from the culture. Results show that 66.7%-91.6% of the cells were captured from unialgal culture by IMBs, and only 5.3%-12.5% of the four other HAB microalgae species were captured, indicating that the constructed IMBs combined specifically with the H. akashiwo cells. At the same time, flow cytometry (FCM) sorting was exploited to screen H. akashiwo cells after labeling with FITC conjugated polyclonal antibodies. Using the FCM technique, 91.7% of the targeted cells were sorted out from mixed microalgae samples in just a few minutes. These results indicate that both antibody-involved IMB and antibody-based FCM techniques are highly effective at detecting and quantifying HAB species. These techniques, especially immuno-magnetic separation, have low associated cost, and are fast and simple processes compared with other techniques currently in use.展开更多
