诸多研究表明,在生物大分子的相互作用中,很多过程都伴随着体积的变化,如蛋白亚基结合成寡聚蛋白、蛋白和核酸结合成复合物均是一个体积增大的过程.体积增大主要源于:(1)在上述过程中,寡聚蛋白或复合物的亚基间形成dead space.(2)形成盐... 诸多研究表明,在生物大分子的相互作用中,很多过程都伴随着体积的变化,如蛋白亚基结合成寡聚蛋白、蛋白和核酸结合成复合物均是一个体积增大的过程.体积增大主要源于:(1)在上述过程中,寡聚蛋白或复合物的亚基间形成dead space.(2)形成盐键.(3)疏水基团之间的相互作用.从勒夏特利原理可推知,若对上述平衡体系施加一定的压力(hydrostatic pressure),就可改变原有平衡状态,使上述“结合”向相反方向进行——解离,而且实验已证明,在压力小于3000bar时(1bar=1.02kg/cm^2)。 展开更多

Studies so far indicate that many processes in the interactions between biomacromole-cules are accompanied by a change in occupied volume. For instance, the association ofprotein subunits into oligomer and the formation of protein-nucleic acid complex give anincrease in volume. The increase in volume in these processes mainly results from: (i) theformation of the dead space between the subunits of the oligomeric protein or the com-plexes in the processes mentioned above; (ii) the formation of salt bonds and (iii) theinteractions between hydrophobic groups. A thirty-nine nucleotide fragment (nucleotides 38—76) of yeast alanyl tRNA was prepared by using calf spleen phosphodiesterase to delete C_(36)m^1I_(37) from the 3′-half molecule of this tRNA under controlled condit... A thirty-nine nucleotide fragment (nucleotides 38—76) of yeast alanyl tRNA was prepared by using calf spleen phosphodiesterase to delete C_(36)m^1I_(37) from the 3′-half molecule of this tRNA under controlled conditions. Analogs of yeast alanyl tRNA with A, G or C instead of m^1I_(37) were synthesized and their biological activities determined. The results indicated that the aminoacylation activity of these analogs was not affected in the rat liver aminoacyl-tRNA synthetase system, compared with natural yeast alanyl tRNA. However, the incorporation activity (i. e. the activity of transferring alanine into proteins) of these analogs was significantly reduced to 20—30% of that of natural yeast alanyl tRNA in the rabbit reticulocyte lysate cell-free protein synthesizing system. 展开更多
