单因素结合响应面法优化魔芋多酚含量测定方法 被引量:1

Optimization of Content Determination Method of Polyphenols from Konjac by Single Factorand Response Surface Methodology

摘要 对Folin-Ciocalteu比色法测定魔芋中多酚含量的条件进行了优化,研究了5%碳酸钠用量、福林酚试剂用量、水浴温度及时间对魔芋多酚含量测定的影响,在单因素试验的基础上,进一步通过响应面设计优化魔芋多酚的检测条件。结果表明,魔芋多酚的最适检测条件为5%碳酸钠溶液用量为2.75 mL、福林酚试剂用量为1.20 mL、反应温度为62℃,待其反应60 min后在769 nm波长处测定吸光度。经验证该检测方法重复性、稳定性好,精密度、回收率高,检测结果准确可靠,该研究结果可为魔芋的开发利用提供实验参考。 In this research,the Folin-Ciocalteu colorimetric method was used to determine the content of konjac polyphenols.The effects of 5% Sodium carbonate addition amount,Folin-Ciocalteu addition amount,water bath temperature and time on the determination of konjac polyphenols were studied.On the basis of single factor experiment,the detection conditions of polyphenols from konjac were further optimized by Box-Behnken design.The results showed that the optimal conditions for the determination of polyphenol content were 2.75 mL of addition amount of Sodium carbonate,1.20 mL of Folin-Ciocalteu and 62 ℃ of reaction temperature,after 60 min of reaction,the absorbance value was measured at 769 nm wavelength.It was proved that the method had good repeatability,stability,precision and recovery,and the detection result was accurate and reliable.The research results could provide experimental reference for the development and utilization of konjac.
