摘要
为建立同时测定地顶孢霉培养物中腺苷、虫草素和麦角甾醇含量的高效液相色谱法,选择色谱柱为Waters C18柱(4.6 mm×250 mm,5μm),流动相为水-乙腈,梯度洗脱,检测波长270 nm,流速1 mL·min^(-1),柱温30℃,进样量20μL。结果表明:腺苷、虫草素和麦角甾醇在浓度20~640μg·mL-1(X)范围内与峰面积(Y)呈良好的线性关系,腺苷、虫草素和麦角甾醇的回归方程分别为Y=98.53X+586.26(R^(2)=0.9993)、Y=124.16X+277.69(R^(2)=0.9997)、Y=57.652X+107.47(R^(2)=0.9996)。腺苷、虫草素和麦角甾醇的加标回收率分别为95.96%~99.95%、97.43%~99.94%和96.31%~99.95%。该方法准确度高、重复性好、灵敏度高,适用于地顶孢霉培养物中腺苷、虫草素和麦角甾醇含量的测定,为其质量监控提供了技术支撑。
A high performance liquid chromatography(HPLC)method was established for the simultaneous determination of adenosine,cordycepin and ergosterol in Acremoniumterricola culture.Separation was completed in Waters C18 column(4.6 mm×250 mm,5μm).Mobile phase was water-acetonitrile.Detection wavelength was set at 270 nm.The flow rate was 1 mL·min^(-1).The column temperature was controlled at 30℃,and the injection volume was 20μL.The calibration curve was good linear between the ratio of the peak areas(Y)and concentrations(X)of adenosine,cordycepin and ergosterol from 20 to 640 ng·mL-1.The regression equations of adenosine,cordycepin and ergosterol were Y=98.53X+586.26,Y=124.16X+277.69,Y=57.652X+107.47,respectively.The R^(2)values were higher than 0.9993.The average recovery rate of adenosine,cordycepin and ergosterol was 95.96%-99.95%,97.43%-99.94%and 96.31%-99.95%,respectively.This method had characteristics of high accuracy,good repeatability and sensitivity,which was suitable for the determination of adenosine,cordycepin and ergosterolin Acremoniumterricolaculture and provide technical support for its quality control.
