摘要
目的 通过观察加味九分散对痛风性关节炎大鼠外周血肿瘤坏死因子-α(TNF-α)及关节骨病理的影响,探讨加味九分散的抗炎作用机制。方法 将SD雄性大鼠随机分为4组:空白对照组、模型组、扶他林乳胶剂组、加味九分散外敷组。模型组、扶他林乳胶剂组和加味九分散外敷组造痛风性关节炎模型。扶他林乳胶剂组和加味九分散组给药48 h后处死所有大鼠,取大鼠外周血,应用ELISA方法检测血清TNF-α水平,并应用SPSS 23.0软件统计分析;同时取关节骨,病理切片、染色,比较骨病理形态学改变。结果 造模组较空白组外周血TNF-α明显增高,差异具有统计学意义,P<0.01。扶他林乳胶剂组和加味九分散外敷组较造模组外周血TNF-α明显减少,差异具有统计学意义,P<0.01。造模组、扶他林乳胶剂组、味九分散外敷组均较空白对照组关节骨出现骨小梁减少、骨髓腔增大和脂肪细胞增多表现。模型组较扶他林乳胶剂组和加味九分散外敷组关节骨小梁减少、骨髓腔增大和脂肪细胞增多现象更加明显。结论 加味九分散通过降低痛风关节炎大鼠血清TNF-α水平起到抗炎止痛作用,从而减少关节骨炎症反应。
Objective To investigate the anti-inflammatory mechanism of Modified Jiufen Powder(加味九分散)by observing on the level of TNF-αin serum and pathology of bone in rats with gouty arthritis.Methods SD male rats were randomly divided into four groups:blank control group,model group,Futalin emulsion group and Modified Jiufen Powder group.Model group,Futalin emulsion group and Modified Jiufen Powder group rats suffer from gouty arthritis by injection of urate crystals in the joint cavity.All rats were sacrificed after 48 hours of treatment in the Futalin emulsion group and the Modified Jiufen Powder group.Draw peripheral blood of rats,use ELISA method to detect serum TNF-αlevel.The experimental results were analyzed by SPSS23.0.;At the same time,the joint bones were taken,sliced and stained,and the bone pathological changes were compared.Results Compared with the blank group,the TNF-αlevel in the peripheral blood of the model group was significantly higher,which was statistically significant,P<0.01.The level of TNF-αin peripheral blood of the Futalin emulsion group and the Modified Jiufen Powder group was significantly lower than that of the model group,which was statistically significant,P<0.01.Compared with the blank control group,the model group,the Futalin emulsion group and the Modified Jiufen Powder group showed reduced trabeculae,enlarged bone marrow cavity,and increased fat cells compared with the blank control group.Compared with the Futalin emulsion group and the Modified Jiufen Powder group,the model group has reduced joint bone trabeculae,enlarged bone marrow cavity and increased fat cells.Conclusion Modified Jiufen Powder can reduce the level of serum TNF-αin rats with gouty arthritis and play an anti-inflammatory and analgesic effect,thereby reducing joint bone inflammation.
