摘要
目的 运用网络药理学方法探索中药威灵仙治疗癌性疼痛“药物成分-作用靶点-信号通路”的作用网络及其潜在调控机制。方法 采用中药系统药理学数据库网络平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)并结合研究报道筛选威灵仙的药物活性成分及作用靶点,通过人类基因信息网站(GeneCards)与在线数据库(Online Mendelian Inheritance in Man,OMIM)筛选出癌性疼痛的疾病靶点,对药物成分与疾病靶点进行匹配并构建“药物-成分-关键靶点-疾病”网络。构建靶蛋白互作网络,利用R语言绘制柱状图找出关键基因,进行基因Symbol与基因ID转换,并对“成分-疾病”的交集基因进行GO及KEGG通路富集分析。结果 本次研究筛选到威灵仙28种潜在有效成分,33个潜在作用靶点。通过生物信息学分析,主要通过胆碱能突触通路、神经活性受体配体相互作用通路、“细胞凋亡-多物种”通路等信号通路发挥治疗癌性疼痛的作用。结论 威灵仙中皂苷类、甾醇类、黄酮及多元酚类可能是治疗癌性疼痛的物质基础,具有“多成分-多靶点”的调控作用,主要作用于中枢镇痛相关通路以及细胞凋亡等相关通路等。
Objective To explore the active compound-action target-signal pathway pharmacological mechanism of Radix Clematidis for the treatment of cancer pain based on the network pharmacology,so as to reveal its underlying regulatory mechanism.Methods The active ingredients and action targets of Radix Clematidis were searched by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)combined with research reports literature review.GeneCards database and Online Mendelian Inheritance in Man(OMIM)database were employed to predict the targets of cancer pain.The drug composition was matched with the disease target by using R language algorithm,and then the network of“herbs-active compounds-key targets disease”was constructed by using Cytoscape software(Version 3.7.2).The target-protein interaction network was constructed by protein-protein interaction network database(String),and the key genes were identified by bar chart drawn with R language.The transformation between gene Symbol and gene ID was carried out by installing Bioconductor installation package in R language,and the component-disease intersection genes were analyzed by GO and KEGG pathway enrichment analysis.Results The results of network analysis showed that 28 active compounds and 33 action targets of Radix Clematidis were obtained.The bioinformatics analysis results showed that the therapeutic mechanism of Radix Clematidis in treating cancer pain is mainly through the cholinergic synaptic pathway,neuroactive ligand-receptor interaction pathway,apoptosis-multispecies pathway.Conclusions Saponins,sterols,flavonoids and polyphenols may be the active compounds of Radix Clematidis for the treatment of cancer pain.The action of Radix Clematidis on cancer pain is multi-component multi-target,and is through the central analgesia related pathways and apoptosis pathway.
