负载分子信标的生物矿化纳米载体用于检测microRNA

Molecular Beacon-Loaded Biomineralized Nanocarrier for MicroRNA Detection

摘要 为了提高分子信标(MB)检测肿瘤细胞内微小核糖核酸(miRNA)的灵敏度和准确性,设计了一种酸响应的生物矿化纳米载体BSA/CaP,可高效负载MB制备MB@BSA/CaP纳米探针,用于人源乳腺肿瘤细胞中miRNA-155的检测。BSA/CaP纳米载体为中空球型结构,负载MB后粒径增大,MB负载量最高达到331.24 nmol/L,包封率为82.81%,可在酸性环境中(pH 5.00)释放MB。MB@BSA/CaP纳米探针更容易被肿瘤细胞摄取,其摄取效率是商用脂质体载体的约5倍,可将更多的MB递送至细胞内与miRNA-155结合,显著增强探针对细胞内miRNA-155的荧光成像检测灵敏度。同时,由于BSA/CaP纳米载体可保护MB不被核酸酶降解,减少了假阳性信号的产生,提高了探针检测的准确性。 In order to improve the sensitivity and accuracy of molecular beacon(MB) in the detection of microRNA(miRNA) in tumor cells, the MB@BSA/CaP nanoprobe was obtained by loading MB on the acid-responsive biomineralized BSA/CaP nanocarrier for the detection of miRNA-155 in human breast tumor cells. The size of the nanocarrier with hollow structure increased after loading MB.The loading capacity and encapsulation efficiency of MB@BSA/CaP were calculated to be 331.24 nmol/L and 82.81%. The nanocarrier could release MB under acidic environment(pH 5.00). The cellular uptake efficiency of the nanoprobe was about 5 times as high as that of commercial liposome carriers. Therefore, more MBs could be delivered by the nanocarrier into cells and bind to miRNA-155, thereby significantly enhancing the detection sensitivity of the probe for intracellular miRNA-155 fluorescence imaging. In addition, the BSA/CaP nanocarrier could protect the MB from being degraded by nucleases, reducing the generation of false positive signals and improving the detection accuracy of probe.
