摘要
[目的]了解12份水稻恢复系之间的遗传亲缘关系,以期为水稻恢复系选育和杂交组合选配提供分子依据。[方法]利用40对SSR引物对8份‘明恢’系列恢复系和4份种植广泛的恢复系进行遗传多样性研究。构建该12份恢复系的指纹图谱,并以标记数据为基础计算遗传距离和生成聚类图。[结果](1)40对SSR引物共检测出75个等位位点,平均每对引物产生1.875个;以其中多态性良好的11对标记对12份恢复系进行指纹图谱构建,确定了每份材料的唯一分子身份信息。(2)12份恢复系之间的遗传相似系数变异范围在0.62~0.93之间。在遗传相似系数0.74处可以将12份材料分为6个类群,在0.79处可以将第Ⅰ类群的6份材料分为3个亚类。[结论]12份供试恢复系可以分成6个亚群,明确了相互之间的亲缘关系,从分子层面为恢复系选育和亲本组配提供了较好的遗传信息。
[Purpose]Identification of the genetic relationship between 12 rice restorer lines will provide molecular basis for the breeding of restorer line and selection of hybrid parents.[Method]The genetic diversity of 12 rice restorer lines,including 8 lines of‘Minghui’series and 4 widely planted materials,was studied by means of 40 pairs of simple sequence repeat(SSR)primers.Based on the fingerprint of the 12 lines,the genetic distance was calculated and cluster dendrogram was generated according to the marker data.[Result](1)A total of 75 alleles were detected by the 40 pairs of SSR primers,with an average of 1.875 per primer pair.The fingerprint of the 12 restorer lines were constructed by 11 pairs of SSR markers with good polymorphism,based on which the unique molecular identity of each line was determined.(2)The genetic similarity coefficients varied from 0.62 to 0.93 among the 12 restorer lines.The 12 lines can be divided into 6 groups at the genetic similarity coefficient of 0.74,and at the coefficient of 0.79,group Ⅰ can be further divided into 3 subgroups.[Conclusion]The 12 restorer lines can be divided into 6 subgroups,together with the clarification of the genetic relationship between them,which provided good genetic information for restorer breeding and material matching at the molecular level.
