花生AOC基因家族的鉴定和表达分析

Identification and Expression Analysis of AOC Gene Family in Peanut

摘要 茉莉酸(jasmonic acid,JA)已被认定为新型植物激素,广泛参与植物的生长发育和胁迫响应等过程,丙二烯氧化物环化酶(AOC)是茉莉酸合成途径中的关键酶。为探明花生基因组中AOC基因信息及表达特征,本研究利用生物信息学方法鉴定了花生AOC基因家族,并分析其理化性质、系统进化树、染色体分布、结构特征以及在22个不同组织中和胁迫下的表达模式。结果表明,花生栽培种含有9个AOC基因,野生种各含有4个AOC基因。这些基因不均匀分布在不同染色体的两端,进化树分析显示17个基因聚为4组,且同组基因具有相似的基因结构、外显子数量、基序数量、类型和顺序,但与拟南芥和水稻经历了不同的进化过程。表达分析表明,AOC基因在22个组织中均有表达,在根、雌蕊、果壳中显著高于其他组织,AdAOC3和AiAOC1可被盐处理和干旱处理高度诱导表达,AhAOC在根中表达量高于茎和叶片且被干旱处理高度诱导表达。本研究为进一步探讨花生AOC基因的功能奠定了基础。 Jasmonic acid(JA)has been identified as a new plant hormone and is widely involved in plant growth and development and stress response processes.Allene oxide cyelase(AOC)is a key enzyme in the jasmonate synthesis pathway.To investigate the information and expression characteristics of AOC gene in peanuts,AOC gene family was identified by bioinformatics,and its physicochemical properties,phylogenetic trees,chromosomal distribution,genes structure and gene expression patterns in 22 tissues and under stresses were analyzed.The results showed that peanut cultivars contain 9 AOC genes,two wild species each contain 4 AOC genes.These genes are unevenly distributed at the ends of different chromosomes.The results of gene structures and phylogenetic analysis revealed that 17 AOC genes were classified into 4 groups,and the gene structure,exon number,motif number,type and order were genes similar in genes within the same group,but the evolutionary processes of AOC gene in peanut maybe different from that in Arabidopsis and rice.Expression pattern analysis showed that AOC genes were all expressed in 22 tissues,and the expression in roots,pistils and pericarp were significantly higher than that in other tissues.AdAOC3 and AiAOC1 can be highly induced by salt stress and drought stress.The expression of AhAOC in roots is higher than that in stems and leaves and is highly induced by drought stress.The results of this study have laid a theoretical foundation for further exploring the function of AOC genes in peanuts.
