摘要
目的运用网络药理学方法探究大黄延缓慢性肾脏病的作用机制。方法利用中药系统药理学分析平台(TCMSP)筛选大黄的候选有效成分,导入Pharm Mapper数据库进行候选成分的靶点预测。通过Genecards、OMIM数据库筛选慢性肾脏病相关疾病靶点,通过GO和KEGG进行相关分析并完成数据可视化。结果得到大黄潜在活性成分16个,药物-疾病交集潜在作用靶点49个,靶点主要涉及调节内肽酶活性、酪氨酸激酶活性、丝氨酸肽酶活性及丝氨酸水解酶活性、激素受体结合等功能,且通过调节PI3K-Akt、Ras、Rap1、Fox O等信号通路达到延缓慢性肾脏病进展的作用。结论大黄可能通过PI3K-Akt、Fox O等通路,调节机体内多种蛋白酶活性以延缓慢性肾脏病进展。
Objective To explore the mechanism of rhubarb in slowing the progression of chronic kidney disease by using network pharmacology.Methods TCMSP was used to screen the candidate active ingredients of rhubarb,and the Pharm Mapper database was imported to predict the targets of candidate ingredients.Targets for chronic kidney disease-related diseases were screened through the Genecards and OMIM databases.Analysis and visualization of the data were performed by GO and KEGG.Results 16 rhubarb potential ingredients and 49 potential targets for drug-disease interaction were obtained.The target functions mainly involved the regulation of endopeptidase,tyrosine kinase,serine peptidase,serine hydrolase activities,and hormone receptors.It can be combined with the body and can delay the progression of chronic kidney disease by regulating the signal pathways such as PI3 K-Akt,Ras,Rap1,and Fox O.Conclusion Rhubarb may regulate the activities of various proteases in the body through the pathways such as PI3 K-Akt and Fox O to delay the progression of chronic kidney disease.
