寄生线虫对柞蚕幼虫保护酶活性和免疫相关基因表达的影响 被引量:1

摘要 研究线虫寄生对柞蚕幼虫保护酶活性和免疫相关基因表达量的影响,可以从免疫学角度揭示柞蚕寄生线虫的致病机制。测定柞蚕5龄幼虫被线虫寄生后其血淋巴超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)的活性和谷胱甘肽硫转移酶(GST)、酚氧化酶原(PPO)、凝集素(lectin)和溶菌酶(lysozyme)基因的表达变化,结果表明,柞蚕幼虫被线虫寄生后,其血淋巴SOD活性逐渐升高并显著高于对照组,POD活性逐渐升高,于第14天达到最高值45.78 U/mL,随后逐渐下降,被寄生第6天CAT活性略高于对照组,随后寄生组CAT活性显著低于对照组;GST和溶菌酶编码基因表达水平极显著上调,凝集素编码基因的表达水平呈往复波动趋势,PPO编码基因表达被抑制,并且与对照组存在极显著差异。研究结果可为柞蚕的免疫机制研究提供参考。 Studying the effect of nematode parasitization on activity of protective enzymes and expression of immune-related genes in Antheraea pernyi larvae can uncover the mechanism of nematode infection from the perspective of immunology.By determining the activity of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT),and expression levels of glutathione S-transferase(GST),prophenoloxidase(PPO),lectin and lysozyme genes in hemolymph of 5th instar of A.pernyi larvae parasitized by nematode,it was found that activity of SOD in hemolymph gradually increased and was significantly higher than that of control group after parasitization.Activity of POD gradually increased,reached to the highest of 45.78 U/mL on the 14th day,and then decreased gradually.Activity of CAT was little higher than that of control group after parasitization on the 6th day,but later significantly lower than that of control group.Expression levels of GST and lysozyme genes in hemolymph were up-regulated significantly,expression level of lectin gene fluctuated between high and low,and expression level of PPO gene was controlled and extremely significantly different from that of control group.These results could provide valuable information for the study on immune mechanism of tussah.
