木犀草苷抑制皮肤鳞状细胞癌细胞的增殖、迁移和侵袭及其机制 被引量:4

摘要 目的:探讨木犀草苷对皮肤鳞状细胞癌细胞增殖、迁移和侵袭的影响及其机制。方法:人皮肤鳞状细胞癌细胞A431购自美国标准生物品收藏中心。分别以80 μmol/L(木犀草苷-低组)、160 μmol/L(木犀草苷-中组)和240 μmol/L(木犀草苷-高组)木犀草苷处理A431人皮肤鳞状细胞癌细胞株,对照组未行木犀草苷处理,实时定量反转录聚合酶链反应(RT-qPCR)检测细胞中肿瘤抑癌基因7-L(ST7L)基因信使核糖核酸(mRNA)的表达,蛋白质印迹法(Western blot)检测ST7L、p21、周期蛋白D1(Cyclin D1)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)等基因蛋白表达水平,细胞计数试剂盒(CCK-8)法测定A431细胞增殖抑制率,Transwell实验检测细胞迁移和侵袭能力。多组间比较采用单因素方差分析(组间比较采用SNK- q检验)。 结果:木犀草苷-低组、中组、高组A431细胞的增殖抑制率上升[(10.22±1.02)%、(23.51±2.36)%、(49.28±4.33)%, F=644.968, P<0.05],迁移[(87.36±8.33)%、(71.02±7.05)%、(55.32±5.43)%, F=81.657, P<0.05]和侵袭细胞数[(73.65±7.22)个、(60.25±6.03)个、(41.25±4.33)个, F=105.695, P<0.05]均降低,细胞中Cyclin D1(0.50±0.05、0.38±0.04、0.24±0.03, F=116.198, P<0.05)、MMP-2(0.57±0.05、0.44±0.04、0.31±0.03, F=107.242, P<0.05)和MMP-9(0.43±0.04、0.31±0.03、0.19±0.02, F=194.667, P<0.05)基因蛋白含量降低,p21(0.46±0.04、0.61±0.06、0.76±0.00, F=117.900, P<0.05)和ST7L(1.61±0.15、2.11±0.21、2.84±0.27, F=118.059, P<0.05)基因蛋白的含量升高。过表达ST7L基因可提高A431细胞的增殖抑制率[(40.56±4.32)%]和p21蛋白(0.73±0.06)含量,降低迁移[(61.58±6.17)%]和侵袭[(52.65±5.23)个]细胞数,下调细胞中Cyclin D1(0.29±0.03)、MMP-2(0.36±0.03)和MMP-9(0.24±0.03)基因蛋白表达,差异均有统计学意义( P<0.05)。抑制ST7L基因表达可逆转木犀草苷对A431细胞增殖、迁移和侵袭的作用。 结论:木犀草苷可抑制皮肤鳞状细胞癌细胞增殖、� Objective To investigate the effects of luteoloside on proliferation,migration and invasion of skin squamous carcinoma cells and the mechanism.Methods A431 cells(From American type culture collection)were treated with luteoloside at 80μmol/L(low-luteoloside group),160μmol/L(median-luteoloside group)and 240μmol/L(high-luteoloside group).The levels of ST7L mRNA in A431 cells was detected by real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR).The expression levels of ST7L,p21,Cyclin D1,matrix metalloproteinase(MMP)-2 and MMP-9 proteins were detected by Western blotting.The proliferation inhibition rate of A431 cells was determined by cell counting kit-8(CCK-8)assay,and the migration and invasion abilities of A431 cells were measured by Transwell assay.Results In the low-luteoloside group,median-luteoloside group and high-luteoloside group,the proliferation inhibition rate of A431 cells were increased[(10.22±1.02)%,(23.51±2.36)%,(49.28±4.33)%,F=644.968,P<0.05],the number of migrating cells[(87.36±8.33)%,(71.02±7.05)%,(55.32±5.43)%,F=81.657,P<0.05]and invading cells(73.65±7.22,60.25±6.03,41.25±4.33,F=105.695,P<0.05)was decreased,the contents of Cyclin D1(0.50±0.05,0.38±0.04,0.24±0.03,F=116.198,P<0.05),MMP-2(0.57±0.05,0.44±0.04,0.31±0.03,F=107.242,P<0.05)and MMP-9(0.43±0.04,0.31±0.03,0.19±0.02,F=194.667,P<0.05)proteins were down-regulated,and the contents of p21(0.46±0.04,0.61±0.06,0.76±0.0,F=117.900,P<0.05)and ST7L(1.61±0.15,2.11±0.21,2.84±0.27,F=118.059,P<0.05)were increased.In A431 cells,over-expression of ST7L increased the inhibition rate of proliferation(40.56±4.32)of A431 cells,reduced the number of migrating cells(61.58±6.17)and invading(52.65±5.23),and down-regulated the levels of Cyclin D1(0.29±0.03),MMP-2(0.36±0.03)and MMP-9(0.24±0.03)proteins.Inhibition of ST7L reversed the effects of luteoloside on proliferation,migration and invasion of A431 cells.Conclusion Luteoloside inhibits proliferation,migration and invasion of A431 cells by promoting the
