紫外线照射构建SKH-1小鼠皮肤鳞癌模型过程中皮肤屏障损伤的相关性研究 被引量:6

摘要 目的 紫外线照射SKH-1 小鼠构建皮肤鳞状细胞癌(SCC)动物模型,探讨皮肤鳞癌发生发展过程中皮肤屏障的损伤情况。方法 随机将43 只SKH-l 无毛小鼠分为实验组(33 只)和对照组(10 只)。采用SUV-1000 日光模拟器模拟日光照射实验组小鼠,对照组不做任何处理。采用德国CK 公司多功能皮肤测试仪分别于各时间段检测角质层含水量、经表皮水分丢失(TEWL)、皮肤pH、皮肤黑素和红斑含量。再于第12、18、28 周分别处死实验组小鼠和对照组小鼠行皮肤组织病理检查。结果 12 周后实验组部分小鼠背部皮肤增厚,皮棘隆起;18 周后实验组小鼠背部皮肤出现粟粒性大小丘疹;28 周后成瘤;对照组未见肿瘤形成。照光12 周,实验组的角质层含水量、经皮水分丢失(TEWL)、皮肤黑素含量与对照组的结果出现明显差异;皮肤pH 值和皮肤红斑量在照光2 周后出现明显差异。结论 成功制备小鼠日光性角化病(AK)模型和皮肤鳞状细胞癌(SCC)模型;明确了鳞癌发生发展过程中皮肤屏障损伤的相关性。 Objective Using ultraviolet irradiation of SKH-1 mice to construct an animal model of squamous cell carcinoma of the skin. To investigate the damage of skin barrier during the development of skin squamous cell carcinoma. Methods Forty-nine SKH-l hairless mice were randomly divided into an experimental group(33 rats) and a control group(10 rats).The mice in the experimental group were simulated by the SUV-1000 solar simulator, and the control group was not treated.The CK company’s multi-functional skin tester was used to detect the water content of the stratum corneum, the loss of transepidermal water(TEWL), the PH value of the skin, the melanin and erythema of the skin at various time intervals.The experimental group mice and the control group mice were sacrificed at 12, 18, and 28 weeks for skin histopathological examination. Results After 12 weeks, part of the mouse in the experimental group was thickened in the back skin. After 18 weeks, the mice in the experimental group showed a needle-sized papule on the skin.The experimental group developed tumors after 28 weeks;no tumor formation was observed in the control group. After 12 weeks of irradiation, the water content of the stratum corneum, transepidermal water loss(TEWL), skin melanin content and the results of the control group were significantly different. The pH value of the skin and the amount of erythema on the skin showed significant differences after 2 weeks of illumination. Conclusion The mouse AK model and SCC model were successfully prepared in this experiment.The correlation of skin barrier damage during the development of squamous cell carcinoma was clarified.
