摘要
目的:建立多元素含量的分析方法,探究雄黄内As对肝脏内源性元素水平的影响。方法:建立电感耦合等离子体质谱(ICP-MS)法测定小鼠肝脏中Na、Mg、K、Ca、Fe、Cu、Zn、Cr、Mn、Co、Ni、Se和As元素含量的分析方法,并将所建方法应用到雄黄染毒肝脏中上述元素的含量测定。使用内标法改善基体效应,标准曲线法计算含量。结果:相关系数(r)〉0.992 3(钾为0.966 6,铁为0.946 1);检出限范围为0.001 5-6.608 2μg/L;仪器精密度RSD〈1.75%;方法重复性RSD〈4.69%;添加不同浓度的混合标准溶液,加标回收率为82.70%-108.13%(钾为53.48%,铁为50.51%),RSD〈5.69%。结论:建立的研究方法检测速度快,检出限低,灵敏度高;雄黄内As能引起肝脏内Mg、Ca、Fe、Cu、Zn、Cr、Mn、Co、Ni和Se水平的变化,对Na和K水平的影响不明显。
Objective : To establish a method of multi - element analysis to investigate the effect of As in realgar on thelevel of endogenous elements in liver. Methods : An analysis method has been established to test 13 endogenous elements (Na, K, Mg, Ca, Fe, Cu, Zn, Se, Mn, Cr, Co, Ni, As) in the liver of mice with inductively coupled plasma mass spectrometry( ICP - MS), and then the contents of these elements in liver of mice exposed to realgar were determined. The contents of elements were calculated according to their calibration curves, and internal standard method was adopted to reduce the matrix effect. Results: The experimental results showed that the correlation coefficient(r) was greater than 0. 992 3 ( K was 0. 966 6, Fe was 0. 942 1 ). The limits detection was 0.001 5-6. 608 2 μg/L. The precision of appara- tus with RSD was lower than 1.75%. The recovery rate was 82.70% - 108.13% ( K was 53.48% , Fe was 50.51% ) with RSD lower than 5.69%. Conclusion : This method was applicable to determinate the contents of endogenous elements which had a high detection speed, low limits detection and high sensitivity. Realgar can cause changes in the levels of Mg, Ca, Fe, Cu, Zn, Cr, Mn, Co, Ni and Se in the liver, but with no obvious influence on the levels of Na and K.
