皮肤再生医疗技术作用IGF-1/PI3K/Akt信号通路调控难愈合创面血管生成的机制研究 被引量:29

Study on the mechanism of MEBT/MEBO activating the IGF-1/PI3K/Akt signaling pathway to promote angiogenesis in the recovery process of chronic non-healing cutaneous wound

摘要 目的:探讨皮肤再生医疗技术(MEBT/MEBO)作用IGF-1/PI3K/Akt信号通路调控创面血管生成促进体表慢性难愈合创面修复的机制。方法:145只雄性SD大鼠随机分为空白组(21只),对照组(31只),以及慢性难愈合创面组(93只);慢性难愈合创面组造模成功后再随机分为模型组、贝复新组和MEBT/MEBO组,每组各31只。计算创面的愈合时间及愈合率;取干预治疗后第3、7、14天的创面组织,ELISA法检测胰岛素样生长因子1(IGF-1)含量,Western Blot技术测定磷脂酰肌醇3-激酶(PI3K)、磷酸化蛋白激酶B(p-Akt)、磷酸化内皮型一氧化氮合酶(p-eNOS)及血管内皮生长因子受体2(VEGFR2)的蛋白表达量;碱性水浸泡法扫描电子显微镜技术(SEM)观察创面组织全层皮肤愈合过程。结果:与模型组比较,MEBT/MEBO组显著缩短大鼠创面愈合时间和提高创面愈合率(P<0.01)。造模及干预治疗14d后,SEM结果显示MEBT/MEBO组大鼠全层皮肤创面愈合过程较模型组理想。与模型组比较,第7天时MEBT/MEBO组大鼠创面组织IGF-1水平及PI3K、p-Akt、p-eNOS、VEGFR2蛋白表达含量均显著增加(P<0.05,P<0.01)。结论:MEBT/MEBO能较显著促进慢性难愈合性创面的修复,较好改善其全层皮肤创面愈合过程,其作用机制可能是通过激活IGF-1/PI3K/Akt信号通路,促进创面新生血管生成,从而加快创面的愈合速度。 Objective:To investigate whether MEBT/MEBO markedly ameliorated chronic non-healing cutaneous wound by activating the IGF-1/PI3 K/Akt/signaling to promote angiogenesis in the wound.Methods:A total of 145 male SD rats were randomly divided into normal group(21 rats),control group(31 rats),and chronic wound group(93 rats).After the establishment of chronic non-healing cutaneous would model,93 rats were evenly assigned to the model group(31 rats),rb-FGF group(31 rats),and MEBT/MEBO group(31 rats).After initiation of treatment with saline,rb-FGF,or MEBT/MEBO,healing time and rates of each group were recorded and calculated.On the 3 rd,7 th,and 14 th day after initiation of treatment,tissues were collected from each group and supernatant was separated from tissue specimens for measurement of IGF-1 by ELISA.Proteins were extracted from tissue samples to detect expression levels of PI3 K,p-Akt,p-eNOS,and VEGFR by Western Blot.Tissues were also examined by the scan electronic microscopy(SEM).Results:As compared with the model group,rats of the MEBT/MEBO group demonstrated significantly shortened healing time and greater healing rates(P<0.01).Significant improvement in full thickness wound was observed in the SEM images of the MEBT/MEBO group than those of the model group.The MEBT/MEBO group exhibited significantly elevated IGF-1 levels,and markedly greater expression of PI3 K,p-Akt,p-eNOS and VEGFR2 than the model group did(P<0.05,P<0.01)Conclusion:MEBT/MEBO is effective in treating chronic non-healing cutaneous wound.The therapeutic benefits of MEBT/MEBO could result from enhanced angiogenesis via activation of the IGF-1/PI3 K/Akt signaling pathway.
