摘要 目的:探讨不同浓度葡聚糖硫酸钠(dextran sulfate sodium,DSS)及不同体重段ICR小鼠对诱导急性UC模型的影响。方法:采用ICR小鼠自由饮用DSS溶液(2%、3%、4%、5%)的方法建立急性UC模型,正常组饮用蒸馏水作为对照。通过疾病活动指数(DAI)和组织学来评价结肠的形态和组织病理学改变。结果:DAI评分随着浓度(2%、3%、4%和5%)的增加而增加,并与小鼠体重相关;死亡率随浓度的增加而升高,随体重增加而降低。结论:DSS模型的临床表现和病理表现与人类UC及其相似,是理想的UC模型。采用ICR小鼠饮用3%DSS(分子量为36000~50000)溶液7 d的方法可建立理想的UC急性模型。 Objective:To investigate the effects of different concentrations of dextran sulfate sodium on mice with different ranges of body weight. Method:The ICR mice were fed with 2%,3%,4% and 5% DSS solution to induce acute colitis,the control group were only fed with distilled water. In all groups,the macroscopic of colon were evaluated by assessing disease active index(DAI). Result:The increase in DAI was noted with increasing concentrations,which is relevant to weight.,in addition,the increase of mortality rate was in positively relevant to the increased DSS concentration,and in negatively relevant to the increased weight of rats. Conclusion:The clinical and histological features of DSS induced acute colitis ressemble with UC in human. ICR mice feed with 3% DSS(molecular weight is 36000~50000)solution for 7 days may establish an ideal acute UC model.
